1. High-flux high-energy ion beam production from stable collisionless shock acceleration by intense petawatt-picosecond laser pulses

Abstract: A scheme to achieve stable collisionless shock acceleration (CSA) of ions from a near-critical plasma by intense petawatt-picosecond laser pulses is proposed, where the plasma is confined in a high-Z solid tube. The application of the tube, on the one hand, restrains the plasma from transverse thermal expansion, helping to sustain sufficient density steepening required for shock formation and maintenance; on the other hand, due to the induced sheath field along its wall, pinches hot electrons for recirculation near laser axis, aiding to reach efficient plasma heating that is crucial to have a strong shock velocity for ion reflection. Consequently, stable ion CSA can be maintained for picosecond time scales, resulting in production of high-flux high-energy ion beams. Two-dimensional PIC simulations show that proton beams with narrow energy spread between 50 and 80 MeV and high flux with particle number about 10 ^12 are produced by a laser pulse at intensity 8.8 × 10 ^19 W cm ^−2 and duration 1 ps. By extending the pulse duration to 3 ps, over 100 MeV high-flux proton beams are obtained.

Abstract: ABSTRACT The bursa of Fabricius (BF) is the central humoral immune organ unique to birds. The present study investigated the possible difference on a molecular level between two duck breeds. The digital gene expression profiling (DGE) technology was used to enrich the differentially expressed genes (DEGs) in BF between the Jianchang and Nonghua-P strains of ducks. DGE data identified 195 DEGs in the bursa. Gene Ontology (GO) analysis suggested that DEGs were mainly enriched in the metabolic pathways and ribosome components. Pathways analysis identified the spliceosome, RNA transport, RNA degradation process, Jak-STAT signaling pathway, TNF signaling pathway and B cell receptor signaling pathway. The results indicated that the main difference in the BF between the two duck strains was in the capabilities of protein formation and B cell development. These data have revealed the main divergence in the BF on a molecular level between genetically different duck breeds and may help to perform molecular breeding programs in poultry in the future.

Abstract: ABSTRACT The carbohydrate response element-binding protein (ChREBP) is an important nuclear factor that regulates glycolysis and de novo lipogenesis. However, the role of ChREBP in fatty liver development in geese remains unclear. In order to understand the function of ChREBP in lipid metabolism of geese, we first cloned the complete cDNA of the ChREBP of the Sichuan White goose (Anser cygnoides) using RT-PCR, 5’ RACE and 3’ RACE, and analyzed goose ChREBP expression in nine different tissues using real-time PCR technology. The results showed that the goose ChREBP CDS consists of 945bp nucleotides that encode 314 amino acids, and the sequence has high similarities with the swan goose (Anser cygnoides domesticus) and duck (Anas platyrhynchos) sequences, both at the nucleotide and amino acid levels. The predicted ChREBP protein had a molecular mass of 35.64 kDa with pI value of 5.36. The phylogenetic analysis indicated its evolutionary relationships with corresponding orthologous sequences in swan geese and ducks. The qPCR assays revealed that ChREBP is highly expressed in liver in the Sichuan White goose. Together, these results indicate that goose ChREBP may play an important role in the development of hepatic steatosis.

7. Molecular Authentication of Meats from Three Terrestrial Birds Based on Pcr-Rflp Analysis of the Mitochondrial 12S rRNA Gene

Abstract: ABSTRACT In this study, a method utilizing PCR-restriction fragment length polymorphism (PCR-RFLP) of a mitochondrial gene was developed for the identification of chicken (Gallus gallus), quail (Coturnix coturnix), and common pigeon (Columba livia) meat. PCR products of ~440 bp were obtained from the 12S rRNA gene of these three birds using a pair of universal primers. The three terrestrial birds can be distinguished using one restriction endonuclease, Alu I, which was selected based on species-specific variations in the mt 12S rRNA gene sequence using 9 newly-obtained and 44 published chicken, quail and pigeon sequences. This method was also successfully used to identify commercial quail and pigeon meat products, which were found to be adulterated with chicken meat. Additionally, our method had relatively high sensitivity for detecting a meat mixture. Ten percent of chicken meat in the mixed quail and pigeon sample was detectable. This assay can be useful for the accurate identification of meats from terrestrial birds, avoiding mislabeling or fraudulent species substitution in meat products.

Abstract: Prostate cancer is one of the most heritable cancers. Hundreds of germline polymorphisms have been linked to prostate cancer diagnosis and prognosis. Polygenic risk scores can predict genetic risk of a prostate cancer diagnosis. Although these scores inform the probability of developing a tumor, it remains unknown how germline risk influences the tumor molecular evolution. We cultivated a cohort of 1250 localized European-descent patients with germline and somatic DNA profiling. Men of European descent with higher genetic risk were diagnosed earlier and had less genomic instability and fewer driver genes mutated. Higher genetic risk was associated with better outcome. These data imply a polygenic “two-hit” model where germline risk reduces the number of somatic alterations required for tumorigenesis. These findings support further clinical studies of polygenic risk scores as inexpensive and minimally invasive adjuncts to standard risk stratification. Further studies are required to interrogate generalizability to more ancestrally and clinically diverse populations.

Abstract: Interface shear characteristics have an important impact on the stability of geosynthetically reinforced soil structures. The shear characteristics of three-dimensional (3D) geogrid–sand interfaces were investigated using large-scale direct shear tests and the discrete element method (DEM). Geogrids were manufactured by 3D printing. The effect of mesh pattern and transverse-rib thickness on the stress–displacement relationship, strength parameters, coordination number and porosity distribution were evaluated. The results showed that the mesh pattern and transverse-rib thickness have an impact on the interface shear characteristics. The peak and residual interface shear strength of the modified geogrid mesh pattern exceeded that of biaxial geogrids. The average coordination numbers of the modified geogrid mesh pattern were greater than those of biaxial geogrids. The variability of particle compactness, as characterised by the porosity distribution, shows how the modified mesh pattern increases the interface shear strength. The interface shear strength of the geogrid–sand interface was improved by thickening the transverse ribs of the modified geogrid mesh pattern.

20. Infection Characteristics and Physical Prevention Strategy of Panax notoginseng Round Spot Disease Caused by Mycocentrospora acerina

Abstract: Panax notoginseng round spot disease (PRSD), caused by Mycocentrospora acerina, is the main leaf disease occurring in cultured P. notoginseng. Aiming to find a safe and efficient control method for PRSD, we studied the disease characteristics of PRSD and the optimal growth conditions of M. acerina and evaluated the efficacy of rain-shelter cultivation in PRSD control. Moreover, we described M. acerina based on morphological characterization and molecular analyses (ITS, ACT, LSU, and TEF-1α). The optimum temperature for M. acerina conidial germination was found to be 14 to 22°C. Furthermore, leaf surface wetness for at least 4 h is required for conidial germination, and conidia can successfully infect P. notoginseng when the leaf wetness lasts for more than 8 h. Additionally, rainwater splashing determines the conidial transfection distance, which is less than 2 m. Finally, our study revealed that rain-shelter cultivation is an effective and simple physical prevention strategy to control PRSD, with an average efficacy of up to 100%.

Abstract: International audience; A search for the nonresonant production of Higgs boson pairs (HH ) via gluon-gluon and vector boson fusion processes in final states with two bottom quarks and two tau leptons is presented. The search uses data from proton-proton collisions at a center-of-mass energy of s=13TeV recorded with the CMS detector at the LHC, corresponding to an integrated luminosity of 138fb−1. Events in which at least one tau lepton decays hadronically are considered and multiple machine learning techniques are used to identify and extract the signal. The data are found to be consistent, within uncertainties, with the standard model (SM) predictions. Upper limits on the HH production cross section are set to constrain the parameter space for anomalous Higgs boson couplings. The observed (expected) upper limit at 95% confidence level corresponds to 3.3 (5.2) times the SM prediction for the inclusive HH cross section and to 124 (154) times the SM prediction for the vector boson fusion HH cross section. At 95% confidence level, the Higgs field self-coupling is constrained to be within −1.7 and 8.7 times the SM expectation, and the coupling of two Higgs bosons to two vector bosons is constrained to be within −0.4 and 2.6 times the SM expectation.

22. A hub-and-spoke model: the role of satellite cardiac electrophysiology laboratory units in mitigating the waiting list pressures for catheter ablation for atrial fibrillation

Background Since setting up the cardiac electrophysiology (EP) service at our tertiary centre (UH) in 2010, there has been a significant increase in referrals for EP procedures especially catheter ablation for atrial fibrillation (AF). UH serves a catchment area of above 1 million people and the increasing demand coupled with limited resources in a publicly funded health system has led to a huge supply-demand mismatch in our AF ablation waiting list. This mismatch was compounded by the COVID-19 pandemic. Aim To share our experience of how we attempted to mitigate the waiting list pressures on our AF ablation procedures before and during the pandemic by utilising satellite cardiac EP laboratory units. Methods Firstly, we made cryoballoon pulmonary vein isolation (PVI) the initial ablation strategy for all of our paroxysmal and persistent AF patients undergoing catheter ablation (due to shorter procedure times and standardised workflow). All cryoballoon PVI cases were performed under local anaesthesia. In 2016, we transitioned from overnight-stay to same-day AF ablation and demonstrated its safety and cost-effectiveness. Finally, we increased our EP laboratory capacity by developing a hub-and-spoke model in collaboration with 2 secondary care centres regionally (WH & StX). From May 2019, cryoballoon PVI cases were performed at 3 sites: UH (hub) and 2 satellite units (WH & StX). During the pandemic, 3 national lockdowns were declared (23/3/20, 5/11/20, 6/1/21) and national healthcare services were restructured. Urgent inpatient cardiac EP cases were still performed at UH but elective EP procedures across all three sites (UH, WH & StX) were cancelled during the first national lockdown. One of the steps taken to mitigate the impact of the pandemic on our increasing AF ablation waiting list was to collaborate with the private sector (MH) which functioned as a ‘clean’ site for elective EP procedures. Cryoballoon PVI cases resumed within 2 months of the first national lockdown. From September 2020, all 3 sites [UH and its two satellite units (WH & MH)] were performing cryoballoon PVI. Data from every EP case performed in all centres was entered into an electronic database. Data was extracted from the 1st January 2019 (pre-pandemic) to the 31st December 2021 (post-pandemic). UH was labeled as ‘tertiary unit’ while WH, StX and MH were pooled together and labeled as ‘satellite units’. Results The implementation of three national lockdowns led to a transient decrease in the number of electrophysiology procedures (Figure 1). However, the ‘clean’ patient pathway that we rapidly established at our satellite unit (MH) facilitated the sustained delivery of our cardiac electrophysiology service throughout the pandemic from 2020 to 2021. There were no deaths or differences in complication rates between the 2 units (Table 1). Conclusion Our hub-and-spoke model is a safe and practical system in tackling the AF ablation waiting list even throughout the pandemic.

26. Macroeconomic, Political and Institutional Factors of Economic Growth

Abstract: Broad theoretical and empirical literature discusses how economic development is affected by different economic, political, institutional, social, and other factors. The classic macroeconomic growth theory argues that poorer states must have higher rates of economic growth compared to more prosperous states due to the so-called ‘catch-up effect’. The concept of ‘institutional sclerosis’, suggested by Olson, claims that economic performance is worse in countries that have powerful interest groups. Some scholars suggest that economic growth is conditioned by ‘institutional coherence’ when one institution in a country promotes the efficiency of another. In this research, using regression analysis we estimate the effect of different factors on economic growth in 187 countries from 2001 to 2018. Our findings do not fully support Olson’s argument that economic decline in some countries was conditioned by the influence of interest groups. Our econometric results do not confirm the importance of the effectiveness of state governance and the quality of public policy. At the same time, the most robust result of our analysis is a statistically significant negative relationship between the level of a country’s initial economic development and its following medium- and long-term economic growth. This research contributes to the existing literature by testing Olson’s institutional sclerosis effect for the last two decade,; estimating the combined effect of interacted variables that explains predictions of both ‘interest groups’ and ‘varieties of capitalism’ theories; considering various macroeconomic and social factors along with political ones in one multivariate model.

27. Factors Associated with Early Career Research Productivity after Ophthalmology Residency

Abstract: Background Few studies have evaluated associations between ophthalmology trainee characteristics and performance with postgraduate research productivity. Purpose This article evaluates factors associated with post-residency research productivity among U.S. ophthalmology graduates. Methods Publicly available information of residents graduating between 2009 and 2014 from 30 randomly selected U.S. ophthalmology programs was collected from June to September 2020. Differences in publications between the 5 years post-residency and pre-residency/residency period were used as metrics of productivity. Residents with incomplete records were excluded. Results A total of 758 of 768 residents, 306 females (40.4%) and 452 males (59.6%), met inclusion criteria. The mean (standard deviation [SD]) number of pre-residency publications was 1.7 (4.0), residency was 1.3 (2.2), and post-residency was 4.0 (7.3). Mean (SD) H-index was 4.2 (4.9). Top-ranked residency (p = 0.001), Alpha Omega Alpha (AOA) medical honor status (p = 0.002), U.S. medical school graduates (p 4). There was a positive correlation between both pre-residency/residency and post-residency publications (rho = 0.441; p 2 residency publications (OR = 2.89; p 4 postgraduation publications. Conclusion Higher post-residency productivity was associated with multiple factors, with choice of an academic career, Heed fellowship, and residency productivity playing key roles.

Abstract: Methotrexate (MTX) is the first-line medicine to treat psoriasis. So far, there has been less research on protein biomarkers to predict its efficacy by the proteomic technique.To evaluate differentially expressed proteins in peripheral mononuclear cells (PBMCs) between good responders (GRs) and non-responders (NRs) after MTX treatment, compared with normal controls (NCs).We quantified protein expression of PBMCs with four GRs and four NRs to MTX and four NCs by isobaric tags for relative and absolute quantification (iTRAQ), analysing and identifying proteins related to efficacy of MTX in 18 psoriatic patients.A total of 3177 proteins had quantitative information, and 403 differentially expressed proteins (fold change ≥1.2, P 0.05) were identified. Compared to NCs, upregulated proteins (ANXA6, RPS27A, EZR, XRCC6), participating in the activation of NF-κB, the JAK-STAT pathway and neutrophil degranulation were detected in GRs. The proteins (GPV, FN1, STOM), involving platelet activation, signalling and aggregation as well as neutrophil degranulation were significantly downregulated in GRs. These proteins returned to normal levels after MTX treatment. Furthermore, Western blotting identified the expression of ANXA6 and STAT1 in PBMCs, which were significantly downregulated in GRs, but not in NRs.We identified seven differentially expressed and regulated proteins (ANXA6, GPV, FN1, XRCC6, STOM, RPS27A and EZR) as biomarkers to predict MTX efficacy in NF-κB signalling, JAK-STAT pathways, neutrophil degranulation, platelet activation, signalling and aggregation.

31. DEEP LEARNING-BASED METHOD TO EXTEND THE TIME SERIES OF GLOBAL ANNUAL VIIRS-LIKE NIGHTTIME LIGHT DATA

Abstract: The nighttime light (NTL) remote sensed imagery has been applied in monitoring human activities from many perspectives. As the two most widely used NTL satellites, the Defense Meteorological Satellite Program (DMSP) Operational Linescan System and the Suomi National Polar-orbiting Partnership (NPP)-Visible Infrared Imaging Radiometer Suite (VIIRS) have different spatial and radiometric resolutions. Thus, some long-time series analysis cannot be conducted without effective and accurate cross-calibration of these two datasets. In this study, we proposed a deep-learning based model to simulate VIIRS-liked DMSP NTL data by integrating the enhanced vegetation index (EVI) data product from MODIS. By evaluating the spatial pattern of the results, the modified Self-Supervised Sparse-to-Dense networks delivered satisfying results of spatial resolution downscaling. The quantitative analysing of the simulated VIIRS-liked DMSP NTL with original VIIRS NTL showed a good consistency at the pixel level of four selected sub datasets with R2 ranging from 0.64 to 0.76, and RMSE ranging from 3.96-9.55. Our method presents that the deep learning model can learn from relatively raw data instead of fine processed data based on expert knowledge to cross-sensor calibration and simulation NTL data.

32. A SHORT-CUT CONNECTIONS-BASED NEURAL NETWORK FOR BUILDING EXTRACTION FROM HIGH RESOLUTION ORTHOIMAGERY

Abstract: Extracting building footprints utilizing deep learning-based (DL-based) methods for high-resolution remote sensing images is one of the current research interest areas. However, the extraction results suffer from blurred edges, rounded corners and detail loss in general. Hence, this article presents a detail-oriented deep learning network named eU-Net (enhanced U-Net). The method adopted in this study, imagery send into the pre-module, which consists of the Canny edge detector, Principal Component Analysis (PCA) and the inter-band ratio operations, before feeding them into the network. Then, process skips connections used in the network to reduce the loss of details during edge and corner detection. The encoding and decoding modules, in this network, are redesigned to expand the perceptual field with shortcut connections and stacked layers. Finally, a Dropout module is added in the bottom layer of the network to avoid the over-fitting problem. The experimental results indicate that the methods used in this study outperform other commonly used and state-of-the-art methods of FCN-8s, U-net, DeepLabv3 and Fast SCNN.

33. IMPACT OF DEEP LEARNING-BASED SUPER-RESOLUTION ON BUILDING FOOTPRINT EXTRACTION

Abstract: Automated building footprints extraction from High Spatial Resolution (HSR) remote sensing images plays important roles in urban planning and management, and hazard and disease control. However, HSR images are not always available in practice. In these cases, super-resolution, especially deep learning (DL)-based methods, can provide higher spatial resolution images given lower resolution images. In a variety of remote sensing applications, DL based super-resolution methods are widely used. However, there are few studies focusing on the impact of DL-based super-resolution on building footprint extraction. As such, we present an exploration of this topic. Specifically, we first super-resolve the Massachusetts Building Dataset using bicubic interpolation, a pre-trained Super-Resolution CNN (SRCNN), a pre-trained Residual Channel Attention Network (RCAN), a pre-trained Residual Feature Aggregation Network (RFANet). Then, using the dataset under its original resolution, as well as the four different super-resolutions of the dataset, we employ the High-Resolution Network (HRNet) v2 to extract building footprints. Our experiments show that super-resolving either training or test datasets using the latest high-performance DL-based super-resolution method can improve the accuracy of building footprints extraction. Although SRCNN based building footprint extraction gives the highest Overall Accuracy, Intersection of Union and F1 score, we suggest using the latest super-resolution method to process images before building footprint extraction due to the fixed scale ratio of pre-trained SRCNN and low speed of convergence in training.

34. Recent advances in synthesis of water-stable metal halide perovskites and photocatalytic applications

Abstract: Solar-driven photocatalytic reactions have attracted wide interest as a viable method to generate green energy and alleviate environmental challenges posed by fossil fuels. Although, various classes of photocatalysts have been explored during the past decades, the pursuit towards even more efficient ones is still ongoing. Metal halide perovskites (MHPs) have been recently proposed as novel photocatalysts owing to their wide light absorption range and excellent optoelectronic properties. However, the instability of MHPs in water is the main obstacle that impedes their applications in practice and prompts stabilization strategies to be developed. This review focuses on the recent approaches for stabilizing MHPs in water, including surface engineering, common-ion effect, and intrinsic water stability. The photocatalytic applications of water-stable MHPs are summarized and an outlook with perspectives over the current challenges are provided.

Abstract: Tooth loss, which usually leads to malnutrition, is common in the elderly. However, limited information is available regarding its association with sarcopenia. This study aimed to investigate the relationship between loss of occlusal pairs of tooth and sarcopenia.A cross-sectional retrospective study was performed.The elderly who participated in the National Basic Public Health Project in the Maigaoqiao Community Medical Center in Nanjing, Jiangsu Province, China.A total of 2850 individuals aged ≥60 years were enrolled.Sarcopenia was defined according to the criteria proposed by the Asian Working Group for Sarcopenia. A trained dentist assessed oral health status and counted the number of present teeth. Logistic regression analyses were performed to evaluate the association between the loss of occlusal pairs and sarcopenia.The prevalence of sarcopenia was 7.1% (201/2850). Univariate logistic regression analysis showed that loss of occlusal pairs was associated with sarcopenia [anterior occlusal pairs (AOPs): odd ratio (OR) = 1.292, 95% confidence interval (CI) = 1.158-1.442; posterior occlusal pairs (POPs): OR = 1.147, 95% CI = 1.018-1.221]. Multivariate logistic regression analysis indicated that loss of POPs was still an independent risk for sarcopenia (OR = 1.108, 95% CI = 1.007-1.220) after adjustment for traditional confounders. Subgroup analysis showed that loss of POPs was more significantly linked to sarcopenia in those with advanced age (≥80years) (OR = 1.307, 95% CI = 1.116-1.532) and in females (OR = 1.165, 95%CI = 1.038-1.308). Compared to individuals with ≥5 occluding pairs of POPs, those with5 occluding pairs of POPs had a higher incidence of sarcopenia.Loss of POPs is associated with an increased risk of sarcopenia in the elderly in a Chinese population. Further research on the mechanism of the observed causal relationship is needed.

36. Liguowuite, WO3, a new member of the A-site vacant perovskite type minerals from the Panzhihua–Xichang region, China

Abstract: A new member of the non-stoichiometric perovskite group mineral species, liguowuite, ideally WO3, has been found in the Neoproterozoic Sinian biotite-quartz monzonite in the southern part of the Panzhihua–Xichang region (Nanyang village: 26∘46′8.21′′ N, 101∘27′13.86′′ E), China. It is associated with hornblende, pargasite, ferro-hornblende, annite, hydrobiotite, phlogopite, orthoclase, microcline, albite, quartz, kaolinite, ilmenite, goethite, hematite, magnetite, pyrite, zircon, zoisite, titanite, epidote, diopside, tourmaline, almandine, fluorapatite, monazite-(Ce), allanite-(Ce), bastnäsite-(Ce), xenotime-(Y), scheelite, moissanite, tellurite, wumuite, and tewite. Liguowuite occurs as greenish yellow prisms, ranging from 0.05 to 0.1 mm in diameter, and it is transparent with a greasy luster and a white streak. Liguowuite exhibits a triclinic pseudomorphism composed of nanoparticles with many cracks and nanovoids. This mineral is brittle and has a Mohs hardness value of approximately 3–4. The calculated density is 7.22 g cm−3. Electron microprobe analyses gave (average weight percent, wt %, of 15 spot analyses of 4 samples) K2O = 0.01, WO3 = 99.23, TeO2 = 0.03, CaO = 0.06, Na2O = 0.04, and total 99.37, yielding the empirical formula W1.00O3, based on Oapfu=3 and ideally WO3. The strongest five diffraction lines (d Å (I)(hkl)) are 3.8552 (88)(002), 3.7685 (88)(020), 3.6590 (100)(–200), 2.6928 (43)(022), and 2.6258 (60)(202). Liguowuite is monoclinic and is in space group P21/n, with a= 7.32582(18) Å, b= 7.54767(18) Å, c= 7.71128(18) Å, β= 90.678(3)∘, V= 426.348(19) Å3, and Z= 8. The crystal structure data were refined using the Rietveld refinement method and X-ray powder diffraction data. The reliability factors (R factors) were Rwp= 0.0604 and Rp= 0.0454, with χ2 = 1.707. Liguowuite consists of corner-sharing distorted and tilted [WO6] octahedra, in which the W atoms are off-center and form six W–O bonds ranging from 1.7 to 2.2 Å with the surrounding oxygen atoms. According to the hierarchical scheme for perovskite supergroup minerals, liguowuite is the first reported example of A-site vacant single oxide, i.e., a new perovskite subgroup.

38. Characterization of ovarian tissue oocytes from transgender men reveals poor calcium release and embryo development, which might be overcome by spindle transfer

Abstract: STUDY QUESTION Can spindle transfer (ST) overcome inferior embryonic development of in vitro matured ovarian tissue oocytes (OTO-IVM) originating from testosterone-treated transgender men? SUMMARY ANSWER ST shows some potential to overcome the embryo developmental arrest observed in OTO-IVM oocytes from transgender men. WHAT IS KNOWN ALREADY OTO-IVM is being applied as a complementary approach to increase the number of oocytes/embryos available for fertility preservation during ovarian tissue cryopreservation in cancer patients. OTO-IVM has also been proposed for transgender men, although the potential of their oocytes remains poorly investigated. Currently, only one study has examined the ability of OTO-IVM oocytes originating from transgender men to support embryo development, and that study has shown that they exhibit poor potential. STUDY DESIGN, SIZE, DURATION Both ovaries from 18 transgender men undergoing oophorectomy were collected for the purposes of this study, from November 2020 to September 2022. The patients did not wish to cryopreserve their tissue for fertility preservation and donated their ovaries for research. All patients were having testosterone treatment at the time of oophorectomy and some of them were also having menses inhibition treatment. PARTICIPANTS/MATERIALS, SETTING, METHODS Sibling ovaries were collected in either cold or warm medium, to identify the most optimal collection temperature. Cumulus oocyte complexes (COCs) from each condition were isolated from the ovarian tissue and matured in vitro for 48 h. The quality of OTO-IVM oocytes was assessed by calcium pattern releasing ability, embryo developmental competence following ICSI, and staining for mitochondrial membrane potential. In vitro matured metaphase I (MI) oocytes, germinal vesicle (GV) oocytes, and in vivo matured oocytes with aggregates of smooth endoplasmic reticulum (SERa) were donated from ovarian stimulated women undergoing infertility treatment and these served as Control oocytes for the study groups. ST was applied to overcome poor oocyte quality. Specifically, enucleated mature Control oocytes served as cytoplasmic recipients of the OTO-IVM spindles from the transgender men. Embryos derived from the different groups were scored and analysed by shallow whole genome sequencing for copy number variations (CNVs). MAIN RESULTS AND THE ROLE OF CHANCE In total, 331 COCs were collected in the cold condition (OTO-Cold) and 282 were collected in the warm condition (OTO-Warm) from transgender men. The maturation rate was close to 54% for OTO-Cold and 57% for OTO-Warm oocytes. Control oocytes showed a calcium releasing ability of 2.30 AU (n = 39), significantly higher than OTO-Cold (1.47 AU, P = 0.046) oocytes (n = 33) and OTO-Warm (1.03 AU, P = 0.036) oocytes (n = 31); both values of calcium release were similar between the two collection temperatures. Mitochondrial membrane potential did not reveal major differences between Control, OTO-Warm, and OTO-Cold oocytes (P = 0.417). Following ICSI, 59/70 (84.2%) of Control oocytes were fertilized, which was significantly higher compared to 19/47 (40.4%) of OTO-Cold (P LARGE SCALE DATA N/A. LIMITATIONS, REASONS FOR CAUTION Due to the limited access to human oocytes and ovarian tissue, our results should be interpreted with some caution, as only a limited number of human oocytes and embryos could be investigated. WIDER IMPLICATIONS OF THE FINDINGS The results of this study, clearly indicate that OTO-IVM oocytes originating from transgender patients are of inferior quality, which questions their use for fertility preservation. The poor quality is likely to be related to cytoplasmic factors, supported by the increased blastocyst numbers following application of ST. Future research on OTO-IVM from transgender men should focus on the cytoplasmic content of oocytes or supplementation of media with factors that promote cytoplasmic maturation. A more detailed study on the effect of the length of testosterone treatment is also currently missing for more concrete guidelines and guidance on the fertility options of transgender men. Furthermore, our study suggests a potentially beneficial role of experimental ST in overcoming poor embryo development related to cytoplasmic quality. Abstract: Androgen receptor (AR) signaling is a key driver of prostate cancer, and androgen-deprivation therapy (ADT) is a standard treatment for patients with advanced and metastatic disease. However, patients receiving ADT eventually develop incurable castration-resistant prostate cancer (CRPC). Here, we report that the chromatin modifier LSD1, an important regulator of AR transcriptional activity, undergoes epigenetic reprogramming in CRPC. LSD1 reprogramming in this setting activated a subset of cell-cycle genes, including CENPE, a centromere binding protein and mitotic kinesin. CENPE was regulated by the co-binding of LSD1 and AR to its promoter, which was associated with loss of RB1 in CRPC. Notably, genetic deletion or pharmacological inhibition of CENPE significantly decreases tumor growth. Our findings show how LSD1-mediated epigenetic reprogramming drives CRPC, and they offer a mechanistic rationale for its therapeutic targeting in this disease. Abstract: Neuroendocrine prostate cancer (NEPC) is a lethal subtype of prostate cancer arising mostly from adenocarcinoma via neuroendocrine transdifferentiation following androgen deprivation therapy. Mechanisms contributing to both NEPC development and its aggressiveness remain elusive. In light of the fact that hyperchromatic nuclei are a distinguishing histopathologic feature of NEPC, we utilized transcriptomic analyses of our patient-derived xenograft (PDX) models, multiple clinical cohorts, and genetically engineered mouse models to identify 36 heterochromatin-related genes that are significantly enriched in NEPC. Longitudinal analysis using our unique, first-in-field PDX model of adenocarcinoma-to-NEPC transdifferentiation revealed that, among those 36 heterochromatin-related genes, heterochromatin protein 1α (HP1α) expression increased early and steadily during NEPC development and remained elevated in the developed NEPC tumor. Its elevated expression was further confirmed in multiple PDX and clinical NEPC samples. HP1α knockdown in the NCI-H660 NEPC cell line inhibited proliferation, ablated colony formation, and induced apoptotic cell death, ultimately leading to tumor growth arrest. Its ectopic expression significantly promoted NE transdifferentiation in adenocarcinoma cells subjected to androgen deprivation treatment. Mechanistically, HP1α reduced expression of androgen receptor and RE1 silencing transcription factor and enriched the repressive trimethylated histone H3 at Lys9 mark on their respective gene promoters. These observations indicate a novel mechanism underlying NEPC development mediated by abnormally expressed heterochromatin genes, with HP1α as an early functional mediator and a potential therapeutic target for NEPC prevention and management.Significance: Heterochromatin proteins play a fundamental role in NEPC, illuminating new therapeutic targets for this aggressive disease. Abstract: Purpose:Chromosomal instability (CIN) is a common phenomenon in colorectal cancer, but its role and underlying cause remain unknown. We have identified that mitotic regulator microtubule-associated protein 9 (MAP9) is a critical regulator of CIN in colorectal cancer. We thus studied the effect of MAP9 loss on colorectal cancer in Map9-knockout mice and in cell lines.Experimental Design:We generated colon epithelial–specific Map9-knockout mice and evaluated colorectal cancer development. Effect of Map9 knockout on colorectal cancer progression was determined in chemical or ApcMin/+-induced colorectal cancer. Molecular mechanism of MAP9 was determined using spectral karyotyping, microtubule assays, and whole-genome sequencing (WGS). Clinical significance of MAP9 was examined in 141 patients with CRC.Results:Spontaneous colonic tumors (9.1%) were developed in colon epithelium–specific Map9-knockout mice at 17 months, but none was observed in wild-type littermates. Map9 deletion accelerated colorectal cancer formation both in ApcMin/+ mice and azoxymethane-treated mice, and reduced survival in ApcMin/+ mice. Mechanistically, MAP9 stabilized microtubules and mediated mitotic spindle assembly. MAP9 also maintained the spindle pole integrity and protected K-fiber from depolymerization at spindle poles. MAP9 loss induced severe mitosis failure, chromosome segregation errors, and aneuploidy, leading to transformation of normal colon epithelial cells. WGS confirmed enhanced CIN in intestinal tumors from Map9 knockout ApcMin/+ mice. In patients with colorectal cancer, MAP9 was frequently silenced and its downregulation was associated with poor survival.Conclusions:MAP9 is a microtubule stabilizer that contributes to spindle stability and inhibits colorectal tumorigenesis, supporting the role of MAP9 as a tumor suppressor for preventing CIN in colorectal cancer.
