Aquaculture is one of the fastest growing food production sectors in the world and further expansion is expected throughout the 21st century. However, climate change is threatening the development of the sector and action is needed to prepare the industry for the coming challenges. Using downscaled temperature projections based on the Intergovernmental Panel on Climate Change (IPCC) climate projection (Shared Socioeconomic Pathway, SSP2-4.5), we analysed potential future temperatures at a selected Atlantic cod (Gadus morhua L.) farm site in Northern Norway. Results showed that the farming area may experience increased temperatures the next 10-15 years, including more days with temperatures above 17°C. Based on the predicted future conditions, we designed a study with Atlantic cod (Gadus morhua L.) to evaluate effects from high temperature alone and in combination with Fransicella noatunensis infection. Fish were kept at 12°C and 17°C for eight weeks and samples of skin and spleen collected at different timepoints were analysed with transcriptomics, histology, scanning electron microscopy and immunohistochemistry. Results showed that high temperature had a stronger effect on the barrier functions of skin than the infection. Increased temperature induced gene expression changes in skin and spleen, heat shock protein 47 and cold inducible RNA binding protein were identified as potential gene markers for thermal stress. The effect of bacterial challenge was small at 12°C. At high temperature, the development of severe pathology in spleen coincided with a significant decrease of immunoglobulins transcripts, which contrasted with the activation of multiple immune genes. In addition, we used an in vitro model of skin biopsies and scale explants exposed to hydrogen peroxide (H2O2) to assess the effects of thermal and oxidative stress. High temperature and H2O2 reduced proliferation and migration of keratocytes, and increased expression of stress markers, and compounding effects were observed with combined stressors. Results suggest that the projected increased seawater temperature will pose a significant threat to Norwegian cod farming, affecting various biological processes and making fish more vulnerable to stressors and pathogens. In the present study we investigated the ability of high-throughput FTIR spectroscopy in combination with multivariate data analysis to reveal if any combinatory effects of fatty acids in mixture are present in liver HepG2 cell cultures after three days of exposure. For this investigation we used an experimental mixture design containing three different octadecenoic acids (oleic acid: C18 : 1 cis- 9, elaidic acid: C18 : 1 trans- 9 and vaccenic acid: C18 : 1 trans- 11) of a total concentration of 100 μM. The results obtained revealed both additive and non-additive effects of individual fatty acids when combined in mixtures. Furthermore, we demonstrate by use of scanning electron microscopy that cells are preserved as intact structures ensuring that FTIR measurements are obtained on whole cell keeping cell compounds in their natural surroundings during measurements. When high fat (40 g oil /100 g) food dressings emulsified by whey protein were heat-filled (80°C) into plastic-bottles, large visible cracks developed in the dressings after cooling. The occurrence of cracks was dependent on the length of the heat treatment, pH and protein concentration. Heating (2 h at 80°C), low pH (pH 3) and high protein concentrations (1.5 g/100 g) increased the number of cracks. Dressings with cracks were more viscous and had a broader particle size distribution than dressings devoid of cracks. During heating at 80°C the complex modulus (G*) measured at low deformation increased sharply, signifying the formation of a three-dimensional network as a result of aggregation of whey protein bridging the fat-droplets. Confocal laser scanning microscopy revealed differences in microstructure dependent on cooling rate and pH. At slow cooling and pH 3 the network structure was inhomogeneous with large voids, while at pH 4, closer to the isoelectrical point, the structure was more compact aggregated and homogeneous. The gels at pH 3 were strain sensitive and seemed more prone to localized fracture. A strong (r=0.86) relationship between a visible quality defect called cracks and the gel properties of the emulsion was found. Avoiding cracks called for a strict control of protein concentration, pH and holding time at higher temperatures. [Copyright &y& Elsevier]

This chapter outlines a method for sample preparation for bottom-up proteomics by LC-MSMS analysis of in vitro proliferated bovine primary skeletal muscle cells. The protocol describes the isolation of bovine primary skeletal muscle cells, extraction of proteins, proteolytic digestion of proteins, and desalting of the final peptide samples. The final peptide samples can be analyzed using various LC-MSMS systems after reconstitution in a suitable elution buffer.
