Abstract

Posttranslational amyloid-β (Aβ) modification is considered to play an important role in Alzheimer's disease (AD) etiology. An N-terminally modified Aβ species, pyroglutamate-amyloid-β (pE3–Aβ), has been described as a major constituent of Aβ deposits specific to human AD but absent in normal aging. Formed via cyclization of truncated Aβ species by glutaminyl cyclase (QC;QPCT) and/or its isoenzyme (isoQC;QPCTL), pE3–Aβ aggregates rapidly and is known to seed additional Aβ aggregation. To directly investigate pE3–Aβ toxicityin vivo, we generated and characterized transgenic TBA2.1 and TBA2.2 mice, which express truncated mutant human Aβ. Along with a rapidly developing behavioral phenotype, these mice showed progressively accumulating Aβ and pE3–Aβ deposits in brain regions of neuronal loss, impaired long-term potentiation, microglial activation, and astrocytosis. Illustrating a threshold for pE3–Aβ neurotoxicity, this phenotype was not found in heterozygous animals but in homozygous TBA2.1 or double-heterozygous TBA2.1/2.2 animals only. A significant amount of pE3–Aβ formation was shown to be QC-dependent, because crossbreeding of TBA2.1 with QC knock-out, but not isoQC knock-out, mice significantly reduced pE3–Aβ levels. Hence, lowering the rate of QC-dependent posttranslational pE3–Aβ formation can, in turn, lower the amount of neurotoxic Aβ species in AD.

Abstract

The bioactivity of neuropeptide Y (NPY) is either N-terminally modulated with respect to receptor selectivity by dipeptidyl peptidase 4 (DP4)-like enzymes or proteolytic degraded by neprilysin or meprins, thereby abrogating signal transduction. However, neither the subcellular nor the compartmental differentiation of these regulatory mechanisms is fully understood. Using mass spectrometry, selective inhibitors and histochemistry, studies across various cell types, body fluids, and tissues revealed that most frequently DP4-like enzymes, aminopeptidases P, secreted meprin-A (Mep-A), and cathepsin D (CTSD) rapidly hydrolyze NPY, depending on the cell type and tissue under study. Novel degradation of NPY by cathepsins B, D, L, G, S, and tissue kallikrein could also be identified. The expression of DP4, CTSD, and Mep-A at the median eminence indicates that the bioactivity of NPY is regulated by peptidases at the interphase between the periphery and the CNS. Detailed ex vivo studies on human sera and CSF samples recognized CTSD as the major NPY-cleaving enzyme in the CSF, whereas an additional C-terminal truncation by angiotensin-converting enzyme could be detected in serum. The latter finding hints to potential drug interaction between antidiabetic DP4 inhibitors and anti-hypertensive angiotensin-converting enzyme inhibitors, while it ablates suspected hypertensive side effects of only antidiabetic DP4-inhibitors application.

Abstract

The anatomical dissection course takes place in the first year of preclinical training at the Medical School of Hannover. In two consecutive years, one out of eight groups (each about 40 students) received an intensified introduction supported by electronic slides and videos at the beginning of each day of dissection. In both years, the new introduction was evaluated. In the second year, modifications resulting from the first evaluation were incorporated. In both years, the students rated the new approach very positively. These results are discussed in respect to educating future medical doctors and to being well prepared for the preclinical exams. Der Präparierkurs wird an der Medizinischen Hochschule Hannover im ersten und zweiten vorklinischen Semester durchgeführt. In zwei aufeinander folgenden Jahren wurde in jeweils einer von insgesamt acht Gruppen mit etwa 40 Studierenden immer unmittelbar vor Kursbeginn eine intensivierte Einleitung mit elektronischem Bildmaterial und Filmen durchgeführt. In beiden Jahren wurden die Einführungen evaluiert. Aus den Ergebnissen der ersten Befragung ableitbare Veränderungen wurden im zweiten Jahr bereits eingearbeitet. Die neue Vorgehensweise wurde in beiden Jahren sehr positiv bewertet. Anhand der Resultate werden Möglichkeiten einer optimierten Durchführung des Präparierkurses im Hinblick auf die Ausbildung zum Arzt und als Vorbereitung auf die ärztliche Vorprüfung diskutiert.
