The thesis deals with the topic of inclusive education from the perspective of primary school teachers. The aim of this work is to evaluate the attitude of teachers to this issue, to reveal their needs and to propose measures that could satisfy these needs. The thesis contains two main parts: theoretical and practical one. The theoretical part introduces to readers the position of persons with disabilities in the past and in the present and presents a way of anchoring the fundamental rights of these persons in international treaties and legislative actions. Furthermore, the basic concepts related to inclusive education are defined in this section and one chapter is devoted to the teacher himself and the attitudes he develops within the classroom. The research part introduces the researches on a similar topic, describes the methodology used in the survey and presents data obtained through interviews with primary school teachers. The results of the research point out the shortcomings in the way of education of primary school teachers, in particular reveal the absence of interconnection of the teaching and special education fields. The Diploma thesis The Semiotic Analysis of campaign spots of parliamentary election in 2013 uses the semiotic analysis to analyse campaign spots broadcasted by the Czech Television before the elections in 2013. With regards of the purpose of this paperwork we chose the spots of victorious political parties, the ones who have exceeded the minimal level of 5% of total votes to gain the mandate, thus ČSSD, ANO, KSČM, Top 09, ODS, Úsvit and KDU- ČSL. We emphasise signs and means of expression used in chosen campaign spots. The aim of this paperwork is to answer the question what message campaign spots address to voters. Moreover, the diploma thesis tries to answer the question, if all victorious campaign spots contain the same signs and means of expression which could help them to influence the result of elections. The diploma thesis deals with the issue of an autonomy in the environment of Célestin Freinet's pedagogy. As the principal aim of the thesis the author has chosen to demonstrate the potential of this pedagogy in the sphere of the pupils' autonomy development. The theoretical part is divided into two thematic areas. The first area deals with the concept of autonomy. At first, it seeks to define the concept at a general level and then in the context of education. This part also includes a description of the self-determination theory and its bases for a self-regulation. The second area focuses on C. Freinet's pedagogy. There are characterized both his personality and the basic pedagogical principles. In the conclusion there are presented the opportunities for the autonomy development in the environment of Freinet's schools. The practical part contains three chapters, each focused on a different research method. The first chapter consists of a case study of the École des Bruyères, which applies the C. Freinet's pedagogical concept, and of the observation that the author has made in the 3rd and 4th grade of this school. Abctract In my diploma thesis I deal with actions that happen around bulk containers, that are intended to collect larger pieces of domestic waste and that are periodically placed in public space in Prague. I consider the bulk container as an actor, which leads us to particular practices of guarding, disposession and collecting stuff (from the container). I use method of multi-sited ethnography to investigate how the value of disposed objects change according to actions around container and according to change of the context in which it is newly placed. I identify several kinds of value, that are inscribed to wasted objects by their new owners. Waste can become valuable with regard to its economic potencial (as a thing or material that can be sold), with regard to possibility of its further practic use (as a household equipment or material), with regard to its estethic value (as an indicator of taste or as an artefact) or it can gain some specific kind of value in socially engaged context. The activation of bone marrow-derived mast cells (BMMCs) induces a number of cell processes such as degranulation, proliferation and cytoskeleton rearrangements. Although microtubules are important in these processes, molecular mechanisms that control changes in microtubule organisation during cell activation are unknown. Activation of BMMCs can be achieved in several ways. Under physiological conditions, the aggregation of IgE receptors (FcRI) on the surface of BMMCs leads to the initiation of specific signaling pathways. Cells can be also activated nonspecifically by a tyrosine phosphatase inhibitor pervanadate, or by thapsigargin that inhibits Ca2+ ATPase pumps located on the endoplasmic reticulum. In this diploma thesis it was found out that rapid morphological changes can be monitored when BMMC are immobilised on the fibronectin before their activation. It was proved that specific and nonspecific activation events lead to microtubule reorganization, as well as to generation of a large number of microtubule-dependent protrusions. The theme of this bachelor's work is analysis of the poetry in czech sign language (ČZJ). It can be useful for linguists who are concerned with czech sign language but also for the general public. This work is supporting the fact that czech sign language is a real language as the spoken language is, because of the evidence of art signing, especially the existence of the poetry in czech sign language. The aim of this bachelor's work is also to overcome the prejudice against czech sign language that are still existing till nowadays. Examples of sign language poetry can be seen on DVD which is attached to this work. The author of this bachelor's work discusses the question of the production the poetry in czech sign language and tries to compare it with the production of spoken language poetry (in its oral and also in the written form); explains the history of czech sign language poetry and the foreign sign language poetry. In this bachelor's work is also a short chapter that concerns of the comparation between the music, poetry and strorytelling. Mezenchymal migration strategy is a mode of individual cell invasion, along with an ameboid migration strategy. It is dependent on cell adhesion structures formation and traction forces generation. This work is focused on integrin-mediated cell to extracellular matrix connections called focal adhesions. Focal adhesions are very complex and comprise of many proteins. The clusters of integrin dimers make up the focal adhesion core that binds extracellular matrix. Their intracellular domains indirectly interact with actin stress fibres throught plaque proteins talin and vinculin. Focal adhesion assembly is force dependent and its commponents turnover is also regulated by focal adhesion kinase and prolin-rich tyrosin kinase 2. These kinases are probably recruited to focal adhesions by paxillin and then linked to signaling complexes by adaptor proteins as p130Cas. The 3D structure is what defines the options of interaction among participating proteins. Therefore, this work summarizes 3D structures of six proteins of interest, deals with their interactions and impact on focal adhesions. PDB codes of all available structures of these six proteins are enclosed.
