Abstract: We present a new measurement of the Λ4H hypernuclear lifetime using in-flight K− + 4He → Λ4H + π0 reaction at the J-PARC hadron facility. We demonstrate, for the first time, the effective selection of the hypernuclear bound state using only the γ-ray energy decayed from π0. This opens the possibility for a systematic study of isospin partner hypernuclei through comparison with data from (K−, π−) reaction. As the first application of this method, our result for the Λ4H lifetime, τ(HΛ4)=206±8(stat.)±12(syst.)ps, is one of the most precise measurements to date. We are also preparing to measure the lifetime of the hypertriton (Λ3H) using the same setup in the near future.

Abstract: We measured a set of π±Σ∓, π0Σ0, and π−Σ0 invariant mass spectra below and above the K¯N mass threshold in K−-induced reactions on deuteron. The measured πΣ mass spectral shape is well reproduced via the two-step mechanism, i.e., a neutron knocked out at a forward angle from a deuteron by an incident K−, with the K¯ recoiled backward reacting with the residual nucleon to produce π and Σ. We deduced the S-wave K¯N→πΣ and K¯N→K¯N scattering amplitudes in the isospin 0 channel in the framework of a K¯N and πΣ coupled channel. We find that a resonance pole corresponding to Λ(1405) is located at 1417.7−7.4+6.0 (fitting errors)−1.0+1.1 (systematic errors) + [−26.1−7.9+6.0 (fitting errors)−2.0+1.7 (systematic errors)] i MeV/c2, closer to the K¯N mass threshold than the value determined by the Particle Data Group.

Abstract: Abstract The NA60 experiment has studied low-mass muon pair production in proton–nucleus collisions with a system of Be, Cu, In, W, Pb and U targets, using a 400 GeV proton beam at the CERN SPS. The transverse momentum spectra of the $$\rho /\omega $$ ρ/ω and $$\phi $$ ϕ mesons are measured in the full $$p_{\mathrm {T}}$$ pT range accessible, from $$p_{\mathrm {T}}= 0$$ pT=0 up to $$2 \, {\hbox {GeV/c}}$$ 2GeV/c . The nuclear dependence of the production cross sections of the $$\eta $$ η , $$\omega $$ ω and $$\phi $$ ϕ mesons has been found to be consistent with the power law $$\sigma _{\mathrm {pA}} \propto {\mathrm {A}}^\alpha $$ σpA∝Aα , with the $$\alpha $$ α parameter increasing as a function of $$p_{\mathrm {T}}$$ pT for all the particles, and an approximate hierarchy $$\alpha _\eta \approx \alpha _\phi > \alpha _\omega $$ αη≈αϕ>αω . The cross section ratios $$\sigma _\eta /\sigma _\omega $$ ση/σω , $$\sigma _\rho /\sigma _\omega $$ σρ/σω and $$\sigma _\phi /\sigma _\omega $$ σϕ/σω have been studied as a function of the size A of the production target, and an increase of the $$\eta $$ η and $$\phi $$ ϕ yields relative to the $$\omega $$ ω is observed from p–Be to p–U collisions.

Abstract: We observed a distinct peak in the Λp invariant mass spectrum of He3(K−,Λp)n, well below mK+2mp, i.e., the mass threshold of the K− to be bound to two protons. By selecting a relatively large momentum-transfer region q=350∼650 MeV/c, one can kinematically separate the peak from the quasi-free process, K‾N→K‾N followed by the non-resonant absorption by the two spectator-nucleons K‾NN→ΛN. We found that the simplest fit to the observed peak gives us a Breit–Wigner pole position at BKpp=47±3(stat.)−6+3(sys.) MeV having a width ΓKpp=115±7(stat.)−20+10(sys.) MeV, and the S-wave Gaussian reaction form-factor parameter QKpp=381±14(stat.)−0+57(sys.) MeV/c, as a new form of the nuclear bound system with strangeness – “K−pp”. Abstract: The SIDDHARTA-2 experiment installed at the DA$\mathrm{\Phi}$NE collider of INFN-LNF performed, for the first time, measurements of high-n transitions in intermediate mass kaonic atoms during the data taking campaigns of 2021 and 2022. Kaonic carbon, oxygen, nitrogen and aluminium transitions, which occur in the setup materials, were measured by using the kaons stopped in the gaseous helium target cell with aluminium frames and Kapton walls, and are reported in this paper. These new kaonic atoms measurements add valuable input to the kaonic atoms transitions data base, which is used as a reference for theories and models of the low-energy strong interaction between antikaon and nuclei. Moreover, these results pave the way for future dedicated kaonic atoms measurements through the whole periodic table and to a new era for the antikaon-nuclei studies at low energy., Comment: 8 pages, 7 figures

Abstract: In the last decade, heavy-ion based experiments (HypHI, STAR and ALICEi ) announced surprisingly short lifetime for 3 ΛH mesonic weak decay (MWD), which is difficult to interpret given the fact that 3 ΛH is a very loosely bound system. It will be very interesting to study this issue with a different experimental approach. We propose a direct measurement for 3 ΛH hypernucleus MWD lifetime with ∼10% precision as J-PARC E73 experiment; 4 ΛH hypernucleus lifetime will also be measured as a feasibility test for our experimental approach. In this proceeding, we will introduce the experimental method and the current status of E73 experiment.

Abstract: We have measured the 3d→2p transition x rays of kaonic ^{3}He and ^{4}He atoms using superconducting transition-edge-sensor microcalorimeters with an energy resolution better than 6 eV (FWHM). We determined the energies to be 6224.5±0.4(stat)±0.2(syst) eV and 6463.7±0.3(stat)±0.1(syst) eV, and widths to be 2.5±1.0(stat)±0.4(syst) eV and 1.0±0.6(stat)±0.3(stat) eV, for kaonic ^{3}He and ^{4}He, respectively. These values are nearly 10 times more precise than in previous measurements. Our results exclude the large strong-interaction shifts and widths that are suggested by a coupled-channel approach and agree with calculations based on optical-potential models.

Abstract: The nuclear E2 resonance effect occurs when an atomic de-excitation energy is closely matched by a nuclear excitation energy. It produces an attenuation of some of the atomic X-ray lines in the resonant isotope target. Investigating the nuclear E2 resonance effect in kaonic atoms, important information about kaon-nucleus strong interaction can be provided. The only K − − 42 98 Mo nuclear resonance effect was measured by G. L. Goldfrey, G- K. Lum and C. E. Wiegand at Lawrence Berkeley Laboratory, in 1975. The nuclear E2 resonance effect was observed in 25 hours of data taking, not enough to provide a conclusive result. In four kaonic Molybdenum isotopes ( 42 94 Mo , 42 96 Mo , 42 98 Mo and 42 100 Mo ), the nuclear E2 resonance effect is expected at the same transition, with similar energy values. The KAMEO (Kaonic Atoms Measuring nuclear resonance Effects Observables) experiment plans to study the E2 nuclear resonance effect in kaonic Molybdenum isotopes at the DAΦNE e+e− collider, during the SIDDHARTA-2 experiment. The experimental strategy consists of exposing four solid strip targets, each enriched with one Molybdenum isotope, to negatively charged kaons, using a germanium detector for X-ray transition measurements. A further exposure of a non-resonant 42 92 Mo isotope solid strip target will be used as reference for standard non-resonant transitions.

Abstract: The SIDDHARTA-2 collaboration is aiming to perform the challenging measurement of kaonic deuterium X-ray transitions to the ground state. This will allow to extract the isospin-dependent antikaon-nucleon scattering lengths, providing input to the theory of Quantum Chromodynamics (QCD) in the non-perturbative regime with strangeness. This work describes the SIDDHARTA-2 experimental apparatus and presents the results obtained during the commissioning phase realized with kaonic helium measurements. In particular, the first observation of the kaonic helium transitions to the 3s level (M-lines), reported in this work, represents a new source of information to study the kaonic helium cascade process and demonstrates the potential of the SIDDHARTA-2 apparatus, in the view of the ambitious kaonic deuterium measurement.

Abstract: ΔFosB is a stable transcription factor which accumulates in the nucleus accumbens (NAc), a key part of the brain's reward circuitry, in response to chronic exposure to cocaine or other drugs of abuse. While ΔFosB is known to heterodimerize with a Jun family member to form an active transcription factor complex, there has not to date been an open-ended exploration of other possible binding partners for ΔFosB in the brain. Here, by use of yeast two-hybrid assays, we identify PSMC5-also known as SUG1, an ATPase-containing subunit of the 19S proteasomal complex-as a novel interacting protein with ΔFosB. We verify such interactions between endogenous ΔFosB and PSMC5 in the NAc and demonstrate that both proteins also form complexes with other chromatin regulatory proteins associated with gene activation. We go on to show that chronic cocaine increases nuclear, but not cytoplasmic, levels of PSMC5 in the NAc and that overexpression of PSMC5 in this brain region promotes the locomotor responses to cocaine. Together, these findings describe a novel mechanism that contributes to the actions of ΔFosB and, for the first time, implicates PSMC5 in cocaine-induced molecular and behavioral plasticity.

Abstract: Objective: In the largest avascular low-nutrient intervertebral disc, resident cells would utilize autophagy, a stress-response survival mechanism by self-digestion and recycling wastes. Our goal was to elucidate the involvement of autophagy in disc homeostasis through RNA interference of autophagy-related gene 5 (Atg5). Design: In vitro, small interfering RNAs (siRNAs) targeting autophagy-essential Atg5 were transfected into rat disc cells. Cell viability with levels of autophagy including Atg5 expression, apoptosis, and senescence was assessed under serum starvation and/or pro-inflammatory interleukin-1 beta (IL-1β) stimulation. In vivo, time-course autophagic flux was monitored following Alexa Fluor® 555-labeled Atg5-siRNA injection into rat tail discs. Furthermore, 24-h temporary static compression-induced disruption of Atg5 siRNA-injected discs was observed by radiography, histomorphology, and immunofluorescence. Results: In disc cells, three different Atg5 siRNAs consistently suppressed autophagy with Atg5 protein knockdown (mean 44.4% [95% confidence interval: −51.7, −37.1], 51.5% [−80.5, −22.5], 62.3% [−96.6, −28.2]). Then, Atg5 knockdown reduced cell viability through apoptosis and senescence not in serum-supplemented medium (93.6% [−0.8, 21.4]) but in serum-deprived medium (66.4% [−29.8, −8.6]) further with IL-1β (44.5% [−36.9, −23.5]). In disc tissues, immunofluorescence detected intradiscal signals for the labeled siRNA even at 56-d post-injection. Immunoblotting found 56-d autophagy suppression with prolonged Atg5 knockdown (33.2% [−52.8, −5.3]). With compression, Atg5 siRNA-injected discs presented radiographic height loss ([−43.9, −0.8]), histological damage ([−5.5, −0.2]), and immunofluorescent apoptosis ([2.2, 22.2]) and senescence ([4.1, 19.9]) induction compared to control siRNA-injected discs at 56 d. Conclusions: This loss-of-function study suggests Atg5-dependent autophagy-mediated anti-apoptosis and anti-senescence. Autophagy could be a molecular therapeutic target for degenerative disc disease.

Abstract: We have constructed a new laser-Compton-scattering facility, called the LEPS2 beamline, at the 8-GeV electron storage ring, SPring-8. This facility provides a linearly polarized photon beam in a tagged energy range of 1.3--2.4 GeV. Thanks to a small divergence of the low-emittance storage-ring electrons, the tagged photon beam has a size (sigma) suppressed to about 4 mm even after it travels about 130 m to the experimental building that is independent of the storage ring building and contains large detector systems. This beamline is designed to achieve a photon beam intensity higher than that of the first laser-Compton-scattering beamline at SPring-8 by adopting the simultaneous injection of up to four high-power laser beams and increasing a transmittance for the long photon-beam path up to about 77%. The new beamline is under operation for hadron photoproduction experiments.

Abstract: We measured missing mass spectrum of the ^{12}C(γ,p) reaction for the first time in coincidence with potential decay products from η^{'} bound nuclei. We tagged an (η+p) pair associated with the η^{'}N→ηN process in a nucleus. After applying kinematical selections to reduce backgrounds, no signal events were observed in the bound-state region. Abstract: The prevalence of carbapenem-resistant Enterobacteriaceae (CRE) has been reported to be lower in Japan than in many other countries. However, extensive surveillance for CRE carriage has not been performed in Japan.To investigate the prevalence of CRE carriage in Japan among convalescent patients considered to be at high risk of being CRE carriers using an improved selective culture medium.A cross-sectional survey was conducted in 22 acute care hospitals (ACHs) and 21 long-term care hospitals (LTCHs) in northern Osaka from December 2015 to January 2016. Patients who used incontinence aids, an enteral feeding tube or a urinary catheter were enrolled. Faecal specimens were examined using the newly developed M-ECC for imipenemase (IMP)-producing CRE, which is the most prevalent form of CRE in Japan. The positive isolates were analysed by polymerase chain reaction and sequencing. Risk factors associated with carriage were analysed by logistic regression.Among 1507 patients, 184 (12.2%) carried CRE. The percentage of positive patients was significantly higher in LTCHs (14.9%) than in ACHs (3.6%) (P0.001). Risk factors for CRE carriage were longer hospital stay [odds ratio (OR) 2.59; 95% confidence interval (CI) 1.87-3.60], enteral feeding (OR 3.03, 95% CI 2.08-4.42) and antibiotic exposure (OR 2.00, 95% CI 1.40-2.87). Among the 233 CRE isolates identified, 223 were IMP producers; the remaining isolates did not produce carbapenemase.This is the first Japanese report to demonstrate the significant spread of CRE in both ACHs and LTCHs using an improved selective medium. A coordinated regional approach may help to prevent further spread.

Abstract: The kaonic deuterium measurement at J-PARC and DAΦNE will provide a piece of information still missing to the antikaon-nucleon interaction close to threshold, providing valuable information to answer one of the most fundamental problems in hadron physics today - to the yet unsolved puzzle of how the hadron mass is generated. For this a new X-ray detector system has been developed to measure the shift and width of the 2p → 1s transition of kaonic deuterium with a precision of 60 eV and 140 eV, respectively.

Abstract: We have developed multi-gap resistive plate chambers (MRPCs) with 2 . 5 × 200 cm 2 readout strips for the time-of-flight (TOF) detector system of the LEPS2 experiment at SPring-8. These chambers consist of two float glass stacks with 5 gas gaps each, in a mirrored configuration. A time resolution of σ ≃ 80 ps was achieved for any position within a strip (at above 99% detection efficiency), improved to 60 ps after correcting for a relatively small time-charge slewing effect. This latter fact seems to stem from the suppression of modal dispersion, achieved through convenient design choices.

Abstract: Expression of the FosB gene has been studied extensively in many fields using a variety of tools. However, previous techniques have had a variety of caveats, from potential off-target effects (e.g., overexpression of FosB, ΔFosB, or a dominant negative mutant of JunD, termed ΔJunD) or confounding developmental effects (e.g., the constitutive FosB knockout mouse). Therefore, we sought to create a floxed FosB mouse line that will allow true silencing of the FosB gene with both spatial and temporal control. Here, we detail the cloning strategy, production, and validation of the floxed FosB mouse. We demonstrate methodology for breeding and genotyping, and show that viral-mediated expression of Cre recombinase in a targeted, discrete brain region ablates expression of the FosB gene in floxed but not wild type mice. Thus, the floxed FosB mouse presented here represents an important new tool for the continued investigation of this critical gene.

Abstract: Tyrosine kinase inhibitor (TKI)-sensitive and TKI-resistant mutations of epidermal growth factor receptor (EGFR) are associated with lung adenocarcinoma. EGFR mutants were previously shown to exhibit ligand-independent activation. We have previously demonstrated that pulmonary surfactant protein D (SP-D, SFTPD) suppressed wild-type EGFR signaling by blocking ligand binding to EGFR. We herein demonstrate that SFTPD downregulates ligand-independent signaling in cells harboring EGFR mutations such as TKI-sensitive exon 19 deletion (Ex19del) and L858R mutation as well as TKI-resistant T790M mutation, subsequently suppressing cellular growth and motility. Lectin blotting and ligand blotting in lung cancer cell lines suggested that EGFR mutants express oligomannose-type N-glycans and interact with SFTPD directly. Cross-linking assay indicated that SFTPD inhibits ligand-independent dimerization of EGFR mutants. We also demonstrated that SFTPD reduced dimerization-independent phosphorylation of Ex19del and T790M EGFR mutants using point mutations that disrupted the asymmetric dimer interface. It was confirmed that SFTPD augmented the viability-suppressing effects of EGFR-TKIs. Furthermore, retrospective analysis of 121 patients with lung adenocarcinoma to examine associations between serum SFTPD levels and clinical outcome indicated that in TKI-treated patients with lung cancer harboring EGFR mutations, including Ex19del or L858R, high serum SFTPD levels correlated with a lower number of distant metastases and prolonged overall survival and progression-free survival. These findings suggest that SFTPD downregulates both TKI-sensitive and -resistant EGFR mutant signaling, and SFTPD level is correlated with clinical outcome. These findings illustrate the use of serum SFTPD level as a potential marker to estimate the efficacy of EGFR-TKIs.

Abstract: Regulating Opioid Responses Different drugs of abuse are thought to highjack similar reward systems in the brain using common mechanisms. However, Koo et al. (p. 124 ) now observe that some of the neural mechanisms that regulate opiate reward can be both different and even opposite to those that regulate reward by stimulant drugs. While knockdown of brain-derived neurotrophic factor (BDNF) in the ventral tegmental area in mice antagonized the response to cocaine, the same manipulation strengthened the potential of opiates to increase dopamine neuron excitability. Optogenetic stimulation of dopaminergic terminals in the nucleus accumbens could counteract the effects of BDNF on morphine reward blockade.

Abstract: Recently, the shift of the kaonic helium-4 2p state was precisely determined by the E570 and SIDDHARTA experiments. Prior to the experiment by E570, the average of three earlier experimental results showed - 43±8 eV, while most of the theoretical calculations give ∼ 0 eV. This five-sigma discrepancy between theory and experiment was known as the "kaonic helium puzzle". A recent theoretical model showed a possible resonance-like shift of maximum 10 eV for a certain value of a deep antikaon-nucleon interaction potential, which is different in helium 3 and helium 4. The E570 experiment determined the shift of the kaonic helium-4 2p state as +2±2 (stat)±2 (sys) eV in 2007. The SIDDHARTA experiment determined the shift as 0±6 (stat)±2 (sys) eV in 2009. The results of these experiments resolved the long-standing puzzle. A new experiment of the kaonic helium-3 X-ray measurement is being prepared by the J-PARC E17 collaborators, and the kaonic helium-3 X-ray data taken very recently by the SIDDHARTA experiment are on the way to be analyzed. The results of the E570, E17 and SIDDHARTA experiments examine the strong interaction for light nuclei with different isospin, and test furthermore recent theoretical predictions. © 2010 American Institute of Physics.

Abstract: The Toyobo-National Cardiovascular Center pneumatic ventricular assist device (Toyobo-NCVC VAD) is widely used in Japan; however, the current pneumatic drivers have some drawbacks, including their large size, heavy weight, and high power consumption. These issues cause difficulty with mobility and contribute to an unsatisfactory quality of life for patients. Because it is urgently necessary to improve patients' safety and quality of life, we have developed a compact, low-noise, portable VAD driver by utilizing an electrohydraulic actuator consisting of a brushless DC motor and a regenerative pump. This unit can be actuated for as long as 2 h with two rechargeable lightweight batteries as well as with external AC power. It is compact in size (33 x 25 x 43 cm) and light in weight (13 kg), and the unit is carried on a mobile wheeled cart. In vitro testing with a Toyobo-NCVC VAD demonstrated a sufficient pumping capacity of up to 8 l/min. We conclude that this newly-developed compact portable driver can provide a better quality of life and improved safety for patients using protracted pneumatic VAD support.

Abstract: The transcription factor DeltaFosB (Delta FosB) accumulates in a region-specific manner in the brain during chronic exposure to stress, drugs of abuse or other chronic stimuli. Once induced, DeltaFosB persists in the brain for at least several weeks following cessation of the chronic stimulus. The biochemical basis of the persistent expression of DeltaFosB has remained unknown. Here, we show that the FosB C-terminus, absent in DeltaFosB as a result of alternative splicing, contains two degron domains. Pulse-chase experiments of C-terminal truncation mutants of full-length FosB indicate that removal of its most C-terminal degron increases its half-life approximately fourfold, and prevents its proteasome-mediated degradation and ubiquitylation, properties similar to DeltaFosB. In addition, removal of a second degron domain, which generates DeltaFosB, further stabilizes FosB approximately twofold, but in a proteasome-independent manner. These data indicate that alternative splicing specifically removes two destabilizing elements from FosB in order to generate a longer-lived transcription factor, DeltaFosB, in response to chronic perturbations to the brain.

Abstract: We investigated the effect of Li doping into Er-doped GaAs and Er, O-codoped GaAs on the enhancement of photoluminescence (PL) intensity. The Li doping into Er-doped GaAs increases the integrated intensity by 50 times higher than that from the Er-doped layer without Li doping. Main peak position of the Er, Li-codoped layer was slightly smaller than that of the Er-doped layer. This difference of peak position means that Li doping induced Er-related active centers different from those in Er-doped layer are formed. In the previous study, we have revealed that oxygen doping into Er-doped GaAs produced sharp and strong PL peak intensity due to Er-2O center. The Li doping into Er, O-codoped GaAs increases the integrated intensity twice higher than that from the Er, O-codoped layer without Li doping. However, the peak intensity decreased and the PL spectrum shape becomes broader. These results indicated that Li doping partly modified the Er-2O center and introduced many kinds of luminescent Er centers.

Abstract: Direct growth of high-quality, thick CdTe (211) epilayers, with thickness up to 100 µm, on Si (211) substrates in a vertical metalorganic vapor phase epitaxy system is reported. In order to obtain homo-orientation growth on Si substrates, pretreatment of the substrates was carried out in a separate chamber by annealing them together with pieces of GaAs at 800–900°C in a hydrogen environment. Grown epilayers had very good substrate adhesion. The full-width at half-maximum (FWHM) value of the x-ray double-crystal rocking curve from the CdTe (422) reflection decreased rapidly with increasing layer thickness and remained between 140–200 arcsec for layers >18 µm. Photoluminescence measurement at 4.2 K showed high-intensity, bound excitonic emission and very small defect-related deep emissions, indicating the high crystalline quality of the grown layers. Furthermore, a CdTe/n+-Si heterojunction diode was fabricated that exhibited clear rectifying behavior.

Abstract: 大腸ポリープの既往を有する2症例に対して,経過観察のために施行した内視鏡検査中,いずれも回盲部に糸状で白色調を呈し,一端は粘膜に迷入する寄生虫と思われる虫体を検出した.1例目(雌)は偶然にも虫体が視野内にみられ,アニサキスなどとの鑑別がつかないまま鉗子にて摘出したのち,専門家の判断を仰ぎ鞭虫と判明した.その4カ月後に定期の大腸内視鏡検査下に2例目(雄)の虫体を確認し直ちに本症と診断しえた.自験2例には雌雄それぞれが含まれ,一度経験することによりその後の鑑別も可能となった.鞭虫症は,嗜好や食習慣の変化にともない,大腸内視鏡下に遭遇する機会も想定され,回虫,蟯虫,アニサキスなどと共に日常の診療に当たって念頭に置くべき疾患と考える.

Abstract: The NA60 experiment at the CERN SPS studies the production of open charm and prompt dimuons in collisions induced by proton and heavy ion beams on nuclear targets. The experimental setup includes a silicon vertex telescope, placed inside a dipole magnetic field, to match charged particle tracks with the muon tracks measured by a muon spectrometer. In p-A collisions the vertex telescope is mostly made of silicon microstrip planes, whereas in A-A interactions the higher multiplicity imposes the use of high granularity silicon pixel detectors. The high interaction rate required for the study of rare processes imposes the use of radiation tolerant pixel detectors. NA60 took its first data in the year 2002, with 400 GeV protons and with Pb ions of 20 and 30 GeV per nucleon. The proton data have been used for the study of dimuon production, while the heavy ion data were devoted to the commissioning of the pixel detectors and to the study of charged particle pseudorapidity densities. After an overview of the detector concept, this paper presents the first results obtained with the 2002 data.

Abstract: The configuration of the vaned diffuser of the integrated heart lung assist device (IHLAD) has been revised to reduce mechanical blood trauma caused by the device. The flow visualization study of the flow near the diffuser vanes revealed the existence of a rotating stall which deteriorates the hydrodynamic performance of the device and augments the chance for blood cells to pass through the regions with intense shearing forces. Design changes of the diffuser included decrease in vane number from 7 to 5 and decrease in passage width from 3 mm to 2 mm. This design change was effective to suppress initiation of a rotating stall. Improvement of hydrodynamic performance and antihemolytic properties was confirmed with the newly designed configuration based on the flow visualization study.

Abstract: The integrated heart lung assist device (IHLAD) has been developed to overcome the problems of currently available extracorporeal membrane oxygenation devices. The integrated structure of a centrifugal blood pump and cylindrical bundle of polyolefin hollow-fibers has allowed a remarkably compact size for the device. This study deals with the design change of the IHLAD that added to the vaned diffuser between the impeller of the centrifugal pump and the hollow-fiber bundle with a view to enhancing the gas-transfer performance. Ex vivo gas-transfer performance tests were carried out, as well as hydrodynamic characteristics and hemolysis test using fresh goat blood. The oxygen transfer rate was generally improved, and the carbon dioxide removal rate was slightly improved. Intolerable amount of hemolysis (index of hemolysis= 0.177) was caused by the IHLAD, which must be resolved by improving the design in the future.

Abstract: Aortic wall thinness was one of the most characteristic changes observed in experimental animals under prolonged continuous flow left heart bypass. The goal of this study was to determine the roles of smooth muscle cells in the vascular remodeling process in cases demonstrating aortic wall thinness under prolonged continuous flow left heart bypass. The aortic samples from three goats in which continuous flow left heart bypass was performed were subjected to histological and immunohistochemical analyses. After 4 weeks of observation, the pulse pressure in the goats under the continuous flow left heart bypass was clearly lower than that in the normal healthy goats. The aortic walls of these goats became thinner, an effect caused by the dilation of their internal diameter. These aortic smooth muscle cells maintained contractile formation due to the fact that they contained abundant alpha-smooth muscle actin (SMA) and smooth muscle myosin heavy chain (SMMS). These cells also synthesized redundant matrix metalloproteinase-2 and -9, and the ratio of the SMMS-positive to the SMA-positive area was significantly lower (0.76) than that observed in the control goat (1.00; P < 0.05). The smooth muscle cells demonstrated synthetic-dedifferentiated formation, which is one of the phenotypes of smooth muscle cell function. In conclusion, aortic wall thinness under prolonged continuous flow left heart bypass is caused by over-synthesis of matrix metalloproteinase in smooth muscle cells, and this refers the vascular remodeling process of the extracellular matrix.

Abstract: Electrohydraulic total artificial heart (EHTAH) and electrohydraulic ventricular assist device (EHVAD) systems have been developed in our institute. The EHTAH system comprises a pumping unit consisting of blood pumps and an actuator, as well as an electronic unit consisting of an internal controller, internal and external batteries, and transcutaneous energy transfer (TET) and optical telemetry (TOT) subunits. The actuator, placed outside the pericardial space, reciprocates and delivers hydraulic silicone oil to the alternate blood pumps through a pair of flexible oil conduits. The pumping unit with an external controller was implanted in 10 calves as small as 55 kg. Two animals survived for more than 12 weeks in a good general condition. The assumed cardiac output ranged between 6 and 10 L/min, the power consumption was 12-18 W, and the energy efficiency was estimated to be 9-11%. Initial implantation of subtotal system including electronic units was further conducted in another calf weighing 73 kg. It survived for 3 days with a completely tether free system. The EHVAD system is developed by using the left blood pump and the actuator of the EHTAH, which were packaged in a compact metal casing with a compliance chamber. In vitro testing demonstrated maximum output more than 9 L/min and more than 13% maximum efficiency. The initial animal testing lasted for 25 days. These results indicate that our EHTAH and EHVAD have the potential to be totally implantable systems.

Abstract: In ovariectomized Wistar rats, 1- or 2-mm wide knife cuts were placed in a coronal plane from the surface of the cortex to the floor of the cranial cavity to interrupt posterior efferents of the ventromedial nucleus of the hypothalamus (VMN). Sham-operated females had the same knife lowered to a depth of 7 mm. Ovariectomized nonoperated females were also used. After recovery, all rats received a single injection of 2 microg oestradiol benzoate, and were tested 48 h later for male and female sexual behaviour and partner preference. When placed with highly receptive stimulus females, the rats with the 2-mm cut showed a significantly higher incidence of mounting with ejaculatory thrusts than any other groups. When placed with stud males, 1-mm cut, as well as sham-operated females, had increased lordosis quotients. Similarly, both 1-mm and 2-mm cuts and sham operation enhanced the incidence of ear wiggling. Despite the display of a transsexual behaviour (i.e. vigorous mounting), all females with the cut showed heterosexual partner preference. Thus, the cut in the present study removed the inhibitory neural effect on mounting, which presumably descends from the VMN. In the absence of this inhibition, minute amounts of oestrogen sufficed to induce vigorous mounting. Sham operation in the present study appeared to interfere with certain inhibitory neural circuitry for lordosis.
