Organs transplanted across donor-specific HLA antibodies (DSA) are associated with a variety of clinical outcomes, including a high risk of acute kidney graft rejection. Unfortunately, the currently available assays to determine DSA characteristics are insufficient to clearly discriminate between potentially harmless and harmful DSA. To further explore the hazard potential of DSA, their concentration and binding strength to their natural target, using soluble HLA, may be informative. There are currently a number of biophysical technologies available that allow the assessment of antibody binding strength. However, these methods require prior knowledge of antibody concentrations. Our objective within this study was to develop a novel approach that combines the determination of DSA-affinity as well as DSA-concentration for patient sample evaluation within one assay. We initially tested the reproducibility of previously reported affinities of human HLA-specific monoclonal antibodies and assessed the technology-specific precision of the obtained results on multiple platforms, including surface plasmon resonance (SPR), bio-layer interferometry (BLI), Luminex (single antigen beads; SAB), and flow-induced dispersion analysis (FIDA). While the first three (solid-phase) technologies revealed comparable high binding-strengths, suggesting measurement of avidity, the latter (in-solution) approach revealed slightly lower binding-strengths, presumably indicating measurement of affinity. We believe that our newly developed in-solution FIDA-assay is particularly suitable to provide useful clinical information by not just measuring DSA-affinities in patient serum samples but simultaneously delivering a particular DSA-concentration. Here, we investigated DSA from 20 pre-transplant patients, all of whom showed negative CDC-crossmatch results with donor cells and SAB signals ranging between 571 and 14899 mean fluorescence intensity (MFI). DSA-concentrations were found in the range between 11.2 and 1223 nM (median 81.1 nM), and their measured affinities fall between 0.055 and 24.7 nM (median 5.34 nM; 449-fold difference). In 13 of 20 sera (65%), DSA accounted for more than 0.1% of total serum antibodies, and 4/20 sera (20%) revealed a proportion of DSA even higher than 1%. To conclude, this study strengthens the presumption that pre-transplant patient DSA consists of various concentrations and different net affinities. Validation of these results in a larger patient cohort with clinical outcomes will be essential in a further step to assess the clinical relevance of DSA-concentration and DSA-affinity., HLA, ISSN:2059-2310, ISSN:2302

L'étude a pour objectif d'identifier des sources de matériaux renouvelables et de démontrer les possibilités de fabriquer des matériaux d'utilité et de valeur à partir de biomatériaux. En revanche, l'exploitation des matières premières renouvelables pour la production d'énergie ne fait pas l'objet de ce projet. Cela implique notamment d'identifier des procédés techniques prometteurs pour la fabrication et la transformation, ainsi que de prendre en considération les questions de sécurité d'approvisionnement et de durabilité. Il convient en outre d'évaluer les perspectives d'évolution pour la Suisse: quels sont nos points forts et nos points faibles? Où existent-ils des possibilités de subventions pour la recherche et le développement? Partant de là, il s'agit alors de tirer les conséquences qui en découlent pour la politique suisse en Recherche & Développement. Il est intéressant notamment de savoir dans quelle mesure les biomatériaux sont utilisés aujourd'hui dans les entreprises, si les matières à base biologique jouent un rôle dans les objectifs et les stratégies de l'entreprise, et de quelle manière les objectifs sont poursuivis en termes d'innovation. Parallèlement, le groupe de travail a recherché les hautes écoles suisses exerçant des activités de recherche et de développement susceptibles de contribuer à la réalisation de ces objectifs. Cette brochure propose une synthèse des résultats obtenus et formule des recommandations quant à la politique de recherche suisse.

Diese Studie soll Quellen für erneuerbare Materialien eruieren und Möglichkeiten aufzeigen, wie aus Biomaterialien Gebrauchs- und Wertstoffe hergestellt werden können. Die Nutzung nachwachsender Rohstoffe für die Energiegewinnung war nicht Gegenstand des Projekts. Es sollen erfolgversprechende technische Verfahren für die Herstellung und Umwandlung identifiziert und dabei Fragen der Wirtschaftlichkeit, der Versorgungssicherheit und der Langfristigkeit berücksichtigt werden. Weiter sollen Entwicklungsperspektiven für die Schweiz evaluiert werden: Wo liegen unsere Stärken und Schwächen? Wo bestehen Fördermöglichkeiten für die Forschung und Entwicklung? Darauf basierend sollen Folgerungen für die schweizerische F&E-Politik aufgezeigt werden. Dabei interessierte es, wie weit in den Unternehmen heute Biomaterialien verwendet werden, ob in den Unternehmerzielen und -strategien biobasierte Materialien eine Rolle spielen und auf welche Weise entsprechende Innovationshorizonte verfolgt werden. Gleichzeitig wurde untersucht, wo an Schweizer Hochschulen Forschungs- und Entwicklungsarbeiten geleistet werden, die einen Beitrag zu dieser Zielsetzung leisten können. Die Broschüre fasst die Resultate zusammen und schliesst mit Empfehlungen für die schweizerische Forschungspolitik.

Aujourd’hui, la recherche biotechnologique n’est plus l’apanage des laboratoires spécialisés. Une communauté croissante de biologistes, de bricoleurs et de passionnés de technique s’adonne à des expériences dans des cuisines, des ateliers et des laboratoires individuels. Certains considèrent cette démocratisation de la biotechnologie comme un danger, d’autres comme une chance d’améliorer la compréhension des relations scientifiques complexes au sein de la société.

Biotechnologische Forschung findet heute nicht mehr nur in spezialisierten Labors statt. Eine wachsende Gemeinschaft von Biologen, Bastlern und Technikbegeisterten experimentiert in Küchen, Werkstätten und Eigenbau-Labors. Einige sehen in der Demokratisierung der Biotechnologie eine Gefahr; andere die Chance für ein besseres Verständnis von komplexen wissenschaftlichen Zusammenhängen in der Gesellschaft.

patent submitter:BioVersys AG The invention relates to compounds and pharmaceutical compositions useful in combination with tetracyclines in the treatment of bacterial infections caused by Gram- positive and Gram-negative pathogens, with particular efficacy in tetracycline resistant strains. These compounds specifically bind to TetR and therefore prevent the transcriptional activation of tet resistance genes. The compounds have a potentiating effect on the activity of members of the tetracycline family, in particular of tetracycline, minocycline, doxycycline and tigecycline, in the treatment of tetracycline susceptible, intermediate and tetracycline resistant pathogens.

Nogo-A is a membrane protein of the central nervous system (CNS) restricting neurite growth and synaptic plasticity via two extracellular domains: Nogo-66 and Nogo-A-Δ20. Receptors transducing Nogo-A-Δ20 signaling remained elusive so far. Here we identify the G protein-coupled receptor (GPCR) sphingosine 1-phosphate receptor 2 (S1PR2) as a Nogo-A-Δ20-specific receptor. Nogo-A-Δ20 binds S1PR2 on sites distinct from the pocket of the sphingolipid sphingosine 1-phosphate (S1P) and signals via the G protein G13, the Rho GEF LARG, and RhoA. Deleting or blocking S1PR2 counteracts Nogo-A-Δ20- and myelin-mediated inhibition of neurite outgrowth and cell spreading. Blockade of S1PR2 strongly enhances long-term potentiation (LTP) in the hippocampus of wild-type but not Nogo-A−/− mice, indicating a repressor function of the Nogo-A/S1PR2 axis in synaptic plasticity. A similar increase in LTP was also observed in the motor cortex after S1PR2 blockade. We propose a novel signaling model in which a GPCR functions as a receptor for two structurally unrelated ligands, a membrane protein and a sphingolipid. SCS-FH Awards at the ILMAC 2013 Creoptix GmbH has developed a novel and innovative technology for label-free detection of molecules based on grating-coupled interferometry (GCI). GCI is a proprietary technology characterized by a very high sensitivity at low technical complexity. One main application will be the measurement of binding affinities in research and development projects such as drug discovery. Goal of an interdisciplinary CTI project with the partners from ZHAW, FHNW and CSEM together with Creoptix is the development of a first GCI instrument including disposables and the optimization of its functionality, followed by field tests to pave the way to market introduction.

Die Synthetische Biologie ist eine relativ junge Disziplin, die versucht, gezielt Zellen mit bestimmten Eigenschaften zu konstruieren. Noch steht sie am Anfang, doch die ersten Erfolge geben einen Eindruck des Potenzials, das in ihr steckt. Aber auch der Herausforderungen, die es zu meistern gilt.

Die Ausbildung zum Chemiker ist an den Hochschulen unterschiedlich ausgeprägt und eingebettet. Gemeinsam ist den Programmen die Vermittlung einer soliden theoretischen und praktischen Ausbildung, die auf eine gute Arbeitsmarktfähigkeit der Studierenden abzielt. Die Leichtigkeit, mit der die Absolvierenden Stellen finden, weist darauf hin, dass die erworbenen Kompetenzen von der Industrie nachgefragt werden.
