Abstract

Objectives Rippling muscle disease (RMD) is characterized by muscle stiffness, muscle hypertrophy, and rippling muscle induced by stretching or percussion. Hereditary RMD is due to sequence variants in the CAV3 and PTRF/CAVIN1 genes encoding Caveolin-3 or Cavin-1, respectively; a few series of patients with acquired autoimmune forms of RMD (iRMD) associated with AChR antibody–positive myasthenia gravis and/or thymoma have also been described. Recently, MURC/caveolae-associated protein 4 (Cavin-4) autoantibody was identified in 8 of 10 patients without thymoma, highlighting its potential both as a biomarker and as a triggering agent of this pathology. Here, we report the case of a patient with iRMD-AchR antibody negative associated with thymoma. Methods We suspected a paraneoplastic origin and investigated the presence of specific autoantibodies targeting muscle antigens through a combination of Western blotting and affinity purification coupled with mass spectrometry–based proteomic approaches. Results We identified circulating MURC/Cavin-4 autoantibodies and found strong similarities between histologic features of the patient's muscle and those commonly reported in caveolinopathies. Strikingly, MURC/Cavin-4 autoantibody titer strongly decreased after tumor resection and immunotherapy correlating with complete disappearance of the rippling phenotype and full patient remission. Discussion MURC/Cavin-4 autoantibodies may play a pathogenic role in paraneoplastic iRMD associated with thymoma.

Lactobacillus plantarum ctsR was characterized, ctsR was found to be cotranscribed with clpC and induced in response to various abiotic stresses, ctsR deletion conferred a heat-sensitive phenotype with peculiar cell morphological features. The transcriptional pattern of putative CtsR regulon genes was examined in the [DELTA]ctsR mutant. Abstract

Cyclopropane fatty acid (CFA) synthesis was investigated in Oenococcus oeni. The data obtained demonstrated that acid-grown cells or cells harvested in the stationary growth phase showed changes in fatty acid composition similar to those of ethanol-grown cells. An increase of the CFA content and a decrease of the oleic acid content were observed. The biosynthesis of CFAs from unsaturated fatty acid phospholipids is catalysed by CFA synthases. Quantitative real-time-PCR experiments were performed on the cfa gene of O. oeni, which encodes a putative CFA synthase. The level of cfa transcripts increased when cells were harvested in stationary phase and when cells were grown in the presence of ethanol or at low pH, suggesting transcriptional regulation of the cfa gene under different stress conditions. In contrast to Escherichia coli, only one functional promoter was identified upstream of the cfa gene of O. oeni. The function of the cfa gene was confirmed by complementation of a cfa-deficient E. coli strain. Nevertheless, the complementation remained partial because the conversion percentage of unsaturated fatty acids into CFA of the complemented strain was much lower than that of the wild-type strain. Moreover, a prevalence of cycC19 : 0 was observed in the membrane of the complemented strain. This could be due to a specific affinity of the CFA synthase from O. oeni. In spite of this partial complementation, the complemented strain of E. coli totally recovered its viability after ethanol shock (10%, v/v) whereas its viability was only partly recovered after an acid shock at pH 3.0.

19. CtsR is the master regulator of stress response gene expression in Oenococcus oeni

Abstract

Although many stress response genes have been characterized in Oenococcus oeni, little is known about the regulation of stress response in this malolactic bacterium. The expression of eubacterial stress genes is controlled both positively and negatively at the transcriptional level. Overall, negative regulation of heat shock genes appears to be more widespread among gram-positive bacteria. We recently identified an ortholog of the ctsR gene in O. oeni. In Bacillus subtilis, CtsR negatively regulates expression of the clp genes, which belong to the class III family of heat shock genes. The ctsR gene of O. oeni is cotranscribed with the downstream clpC gene. Sequence analysis of the O. oeni IOB 8413 (ATCC BAA-1163) genome revealed the presence of potential CtsR operator sites upstream from most of the major molecular chaperone genes, including the clp genes and the groES and dnaK operons. Using B. subtilis as a heterologous host, CtsR-dependent regulation of O. oeni molecular chaperone genes was demonstrated with transcriptional fusions. No alternative sigma factors appear to be encoded by the O. oeni IOB 8413 (ATCC BAA-1163) genome. Moreover, apart from CtsR, no known genes encoding regulators of stress response, such as HrcA, could be identified in this genome. Unlike the multiple regulatory mechanisms of stress response described in many closely related gram-positive bacteria, this is the first example where dnaK and groESL are controlled by CtsR but not by HrcA.

Abstract

A locus containing the clpP and clpL genes in the lactic acid bacterium Oenococcus oeni was studied. Real-time reverse transcription-PCR analysis revealed different induction factors involved in expression of these genes during stress. According to the conditions, clpP and clpL genes could be transcripted as two distinct transcripts or cotranscripted. The clpP promoter depended on the CtsR regulator, but surprisingly the clpL promoter did not. The amount of the clpL transcript depended on mRNA stability. This clp ATPase gene is at least controlled at the posttranscriptional level.

The gene encoded a 45,650 Da protein, exhibited ATPase activity, expressed maximally in the exponential growth phase, and showed increased synthesis in response to a shift in temperature from 30 to 42 degreesC.

A study was conducted to analyze the nucleotide sequence of a chromosomal DNA fragment encoding the thioredoxin of Oenococcus oeni. The size of the transcript and the transcriptional start site were determined. The expression of the gene in response to oxidative and heat stresses were then analyzed. Experimental results indicated that the trxA gene encodes a functional thioredoxin.

The hsp18 gene encodes a small heat shock 18-kilodalton polypeptide Lo18, that is peripherally associated with the membrane of Leuconostoc oenos. The transcription of this heat shock gene increases with an increase in temperature. The gene forms a unique transcription unit in L. oenos that is similar to the unit in Clostridium acetobutylicum and Streptomyces albus. The gene has a putative promoter whose structure is similar to the consensus promoter sequence of vegetative genes of both gram-negative and gram-positive bacteria.

The characterization of the mle locus from Leuconostoc oenos shows a 2.6-kb transcript, indicating that it may have an operon structure containing the malolactic enzyme gene mleA and malate permease gene mleP. The mle operon has one transcription start site situated 17 nucleotides upstream of the ATG translation start site for the mleA gene. An 894-bp open reading frame (RF), that transcribes divergently from the mle operon, is located upstream of the mleA gene. This ORF encodes a polypeptide belonging to the LysR-type regulatory protein family, and has a molecular mass of 34 kDa.

Abstract

The genes that code for the malolactic enzyme and the malate permease in the bacterium Leuconostoc oenos are arranged in a cluster. A 324-bp DNA fragment amplified by PCR from Leuconostoc oenos was used to screen the genome bank. Of the two open reading frames, the first protein has highly conserved protein regions. The expression of the gene in Escherichia coli and Saccharomyces cerevisiae produced malolactic activity in the organisms. The second open reading frame codes a protein that has properties of a carrier protein and probably has 10 membrane-spanning segments.

The system, which is functional in the gastrointestinal pathogen Listeria monocytogenes EGD-e, is determined and autoinducer 2 (AI-2) bioassay is performed with culture supernatants. The results show that Lux1 accumulates more S-adenosyl homocysteine (SAH) and S-ribosyl homocysteine (SRH) in culture supernatant than the parental strain acccumulates and that SRH, but not SAH or AI-2, is able to modify the number of attached cells.

Experimental in-vessel biowaste composts are used to study the survival of seeded Listeria monocytogenes, Salmonella enterica subsp. enterica serotype Enteritidis, and Escherichia coli. Results suggest that like composted sewage sludge or manure, domestic waste composts might support pathogen survival and survival is not related to the physicochemical characteristics of the composts.

Abstract

Autophagy is an intracellular catabolic pathway essential for the recycling of proteins and larger substrates such as aggregates, apoptotic corpses, or long-lived and superfluous organelles whose accumulation could be toxic for cells. Because of its unique feature to engulf part of cytoplasm in double-membrane cup-shaped structures, which further fuses with lysosomes, autophagy is also involved in the elimination of host cell invaders and takes an active part of the innate and adaptive immune response. Its pivotal role in maintenance of the inflammatory balance makes dysfunctions of the autophagy process having important pathological consequences. Indeed, defects in autophagy are associated with a wide range of human diseases including metabolic disorders (diabetes and obesity), inflammatory bowel disease (IBD), and cancer. In this review, we will focus on interrelations that exist between inflammation and autophagy. We will discuss in particular how mediators of inflammation can regulate autophagy activity and, conversely, how autophagy shapes the inflammatory response. Impact of genetic polymorphisms in autophagy-related gene on inflammatory bowel disease will be also discussed.
