Ethylene is a gaseous plant growth regulator that controls a multitude of developmental and stress responses. Recently, the levels of Arabidopsis EIN3 protein, a key transcription factor mediating ethylene-regulated gene expression, have been demonstrated to increase in response to the presence of ethylene gas. Furthermore, in the absence of ethylene, EIN3 is quickly degraded through a ubiquitin/proteasome pathway mediated by two F-box proteins, EBF1 and EBF2. Here we report the identification of ETHYLENE-INSENSITIVE5 as the 5' [right arrow] 3' exoribonuclease XRN4. Specifically, we demonstrate that EIN5 is a component of the ethylene signal transduction cascade acting downstream of CTR1 that is required for ethylene-mediated gene expression changes. Furthermore, we find that the ethylene insensitivity of ein5 mutant plants is a consequence of the over-accumulation of EBF1 and EBF2 mRNAs resulting in the under-accumulation of EIN3 even in the presence of ethylene gas. Together, our results suggest that the role of EIN5 in ethylene perception is to antagonize the negative feedback regulation on EIN3 by promoting EBF1 and EBF2 mRNA decay, which consequently allows the accumulation of EIN3 protein to trigger the ethylene response. Genes with unstable transcripts often encode proteins that play important regulatory, roles. ATL2 is a member of a multigene family coding highly related RING-H2 zinc-finger proteins that may function as E3 ubiquitin ligases. ATL2 mRNA accumulation occurs rapidly and transiently after incubation with elicitors of pathogen response. We screened 50,000 [M.sub.2] families from a line that carries a fusion of pATL2 to the GUS reporter gene and isolated five mutants, which we named eca (expresion constitutiva de ATL2), that showed constitutive expression of the reporter gene. One mutant exhibits a drastic stunted phenotype while the other four grow similarly to wild type. Two early chitin-induced genes and known pathogenesis-related genes such as NPR1, PAL, and CHS are activated in all the mutants whereas members of the ATL family and PR-1 and PDF2.1, which are markers of the salicylic acid (SA) jasmonate (JA) defense-response pathways, display differential expression between the mutants. These observations indicate that the ECA gene products may function in the early steps of an elicitor-response pathway, although some of them may function at other stages on the SA or JA defense-response pathways. Likewise, the fact that ATL2 and other members of the ATL family are activated in eca mutants links the induction of this putative class of ubiquitin ligases to plant defense signaling pathways.

In this study we have established the structure of chromosome ends in the basidiomycete fungus Ustilago maydis. We isolated and characterized several clones containing telomeric regions and found that as in other organisms, they consist of middle repeated DNA sequences. Two principal types of sequence were found: UTASa was highly conserved in nucleotide sequence and located almost exclusively at the chromosome ends, and UTASb was less conserved in nucleotide sequence than UTASa and found not just at the ends but highly interspersed throughout the genome. Sequence analysis revealed that UTASa encodes an open reading frame containing helicase motifs with the strongest homology to RecQ helicases; these are DNA helicases whose function involves the maintenance of genome stability in Saccharomyces cerevisiae and in humans, and the suppression of illegitimate recombination in Escherichia coli. Both UTASa and UTASb contain a common region of about 300 bp located immediately adjacent to the telomere repeats that are also found interspersed in the genome. The analysis of the chromosome ends of U. maydis provides information on the general structure of chromosome ends in eukaryotes, and the putative RecQ helicase at UTASa may reveal a novel mechanism for the maintenance of chromosome stability.

DNA from Pseudomonas syringae pv. phaseolicola completely digested with different restriction enzymes, PacI and PmeI, produced 15 and 16 fragments, respectively. Its chromosome was found to be about 5.65 Mb in size. The different fragments were linked into a circular chromosome map for both enzymes. Partial digestions of DNA from various Tn5 mutants 'tagging' specific bands were produced and the complete and partial products of the digestion separated and identified. By computing the size of the partial products, the different bands were linked into a physical map.

Genetic variation in basiodiomycetous fungus Ustilago maydis was investigated by fingerprint analysis with probes containing telomeric DNA. Group differences were observed between the endmost restriction fragments (EFs) and the endmost-associated restriction fragments (EAFs) of the chromosomes within strains. EFs showed similar lengths in two strains but were of different lengths in the other strains. Analysis of the EAFs showed the presence of distinct bands in the terminal and interstitial sites of the chromosomes in all strains.

A study of Ustilago maydis telomeric repeats yields three clones with tandem repeats of a TTAGGG sequence that is evolutionarily conserved. The sequence proximal to the telomeric repeat is composed of 376 bp and shows hybridization pattern resembling the repeat. The similar order and location of the repeat in several chromosomes indicates a common functional role.
