BackgroundSarcomatoid urothelial carcinoma (SUC) is a rare renal malignancy. Its biological malignancy is high, the prognosis is poor, diagnostic and treatment options are few, and there is no standard treatment plan.Case presentationIn this case, a 64-year-old woman was hospitalized with fever and lower back pain one week previously. The preliminary diagnosis was a right kidney stone with a urinary tract infection. After the anti-infection treatment, a percutaneous right nephrostomy was performed. The intraoperative biopsy (renal pelvis) finding was infiltrating urothelial carcinoma with a sarcomatoid variation. Subsequently, radical surgery was performed for cancer of the right renal pelvis. Implant metastasis of the abdominal wall and adjacent abdominal cavity occurred half a month after the surgery. The lesion was resected again, and two cycles of doxorubicin plus carboplatin chemotherapy were administered. However, the disease progressed more rapidly after the chemotherapy. With the written consent of the patient, the treatment was altered to targeted immune therapy with toripalimab plus anlotinib. A clinical cure was achieved after nine cycles of treatment with no obvious lesions on imaging. The maintenance therapy was administered consecutively for over a year, and the patient is at present still in good condition with a disease-free survival exceeding two years.ConclusionThis case proves that the combination of toripalimab and anlotinib is effective in the treatment of recurrent renal SUC. To the best of our knowledge, this is the first reported case of a patient with advanced recurrent urothelial carcinoma of the renal pelvis sarcomatoid cured with this therapy.

The death of cells can occur through various pathways, including apoptosis, necroptosis, mitophagy, pyroptosis, endoplasmic reticulum stress, oxidative stress, ferroptosis, cuproptosis, and disulfide-driven necrosis. Increasing evidence suggests that mitophagy and ferroptosis play crucial regulatory roles in the development of stroke. In recent years, the incidence of stroke has been gradually increasing, posing a significant threat to human health. Hemorrhagic stroke accounts for only 15% of all strokes, while ischemic stroke is the predominant type, representing 85% of all stroke cases. Ischemic stroke refers to a clinical syndrome characterized by local ischemic-hypoxic necrosis of brain tissue due to various cerebrovascular disorders, leading to rapid onset of corresponding neurological deficits. Currently, specific therapeutic approaches targeting the pathophysiological mechanisms of ischemic brain tissue injury mainly include intravenous thrombolysis and endovascular intervention. Despite some clinical efficacy, these approaches inevitably lead to ischemia-reperfusion injury. Therefore, exploration of treatment options for ischemic stroke remains a challenging task. In light of this background, advancements in targeted therapy for cerebrovascular diseases through mitophagy and ferroptosis offer a new direction for the treatment of such diseases. In this review, we summarize the progress of mitophagy and ferroptosis in regulating ischemia-reperfusion injury in stroke and emphasize their potential molecular mechanisms in the pathogenesis. Importantly, we systematically elucidate the role of medicinal plants and their active metabolites in targeting mitophagy and ferroptosis in ischemia-reperfusion injury in stroke, providing new insights and perspectives for the clinical development of therapeutic drugs for these diseases.

Four kinds of manganese oxides were successfully prepared by hydrothermal and redox precipitation methods, and the obtained oxides were used for CIP removal from water by activating PMS. The microstructure and surface properties of four oxides were systematically characterized. The results showed that ε-MnO2 prepared by the redox precipitation method had large surface area, low crystallinity, high surface Mn(III)/Mn(Ⅳ) ratio and the highest activation efficiency for PMS, that is, when the concentration of PMS was 0.6 g/L, 0.2 g/L ε-MnO2 could degrade 93% of CIP within 30 min. Multiple active oxygen species, such as sulfate radical, hydroxyl radical and singlet oxygen, were found in CIP degradation, among which sulfate radical was the most important one. The degradation reaction mainly occurred on the surface of the catalyst, and the surface hydroxyl group played an important role in the degradation. The catalyst could be regenerated in situ through the redox reaction between Mn4+ and Mn3+. The ε-MnO2 had the advantages of simple preparation, good stability and excellent performance, which provided the potential for developing new green antibiotic removal technology. HIGHLIGHTS Epsilon-MnO2 nanoflowers were simply prepared by the redox precipitation method.; Epsilon-MnO2 nanoflowers were used to activate PMS to degrade CIP in water.; The high activity of ε-MnO2 was due to its large surface area, low crystallinity and high surface Mn(III)/Mn(Ⅳ) ratio.; The –OH groups on the surface could promote the activation of PMS.;

Abstract The strong excitonic effects widely exist in polymer-semiconductors and the large exciton binding energy (E b) seriously limits their photocatalysis. Herein, density functional theory (DFT) calculations are conducted to assess band alignment and charge transfer feature of potential donor-acceptor (D-A) covalent organic frameworks (COFs), using 1,3,5-tris(4-aminophenyl)triazine (TAPT) or 1,3,5-tris(4-aminophenyl)benzene (TAPB) as acceptors and tereph-thaldehydes functionalized diverse groups as donors. Given the discernable D-A interaction strengths in the D-A pairs, their E b can be systematically regulated with minimum E b in TAPT-OMe. Guided by these results, the corresponding D-A COFs are synthesized, where TAPT-OMe-COF possesses the best activity in photocatalytic H2 production and the activity trend of other COFs is associated with that of calculated E b for the D-A pairs. In addition, further alkyne cycloaddition for the imine linkage in the COFs greatly improves the stability and the resulting TAPT-OMe-alkyne-COF with a substantially smaller E b exhibits ~20 times higher activity than the parent COF.

Steering charge kinetics is a key to optimizing quantum efficiency. Advancing the design of photocatalysts (ranging from single semiconductor to multicomponent semiconductor junctions) that promise improved photocatalytic performance for converting solar to chemical energy, entails mastery of increasingly more complicated processes. Indeed, charge kinetics become more complex as both charge generation and charge consumption may occur simultaneously on different components, generally with charges being transferred from one component to another. Capturing detailed charge dynamics information in each heterojunction would provide numerous significant benefits for applications and has been needed for a long time. Here, the steering of charge kinetics by modulating charge energy states in the design of semiconductor–metal‐interface‐based heterogeneous photocatalysts is focused. These phenomena can be delineated by separating heterojunctions into classes exhibiting either Schottky/ohmic or plasmonic effects. General principles for the design and construction of heterojunction photocatalysts, including recent advances in the interfacing of semiconductors with graphene, carbon quantum dots, and graphitic carbon nitride are presented. Their limitations and possible future outlook are brought forward to further instruct the field in designing highly efficient photocatalysts.

The Yellow River flows through Lanzhou city and is the only drinking water source for 3.6 million people. However, people are not clear about the water environmental quality and safety in Lanzhou. To address this problem. Water samples were collected from different sites within this section during the high water period, normal water period and dry water period, and the environmental quality and health risk of the surface water were evaluated using the Nemerow index and health risk assessment method. The results are as follows: first, none of the pollutants exceeded the standard, except for total nitrogen; second, the highest comprehensive evaluation score was 1.04, so the water quality level was good; third, the health risk assessment showed that health risk value of water quality in the Lanzhou section of the Yellow River is on the high side, which is mainly caused by Chromium(Cr); fourth, the carcinogenic risk is five orders of magnitude higher than the non-carcinogenic risk, and the total carcinogenic risk is higher than the maximum acceptable risk level (10−5 a−1), while the total non-carcinogenic risk is lower than the acceptable health risk level (10−6 a−1). Therefore, to ensure the safety of its drinking water, Cr pollutants in the Lanzhou section of the Yellow River should be properly treated and controlled. HIGHLIGHTS We combined water quality analysis with health risk assessment for the Yellow River and Lanzhou area, and this kind of research is less studied in the Yellow River and Lanzhou area.; The research concluded that the concentration of conventional pollutants in the Yellow River is low. There are no excessive amounts of heavy metal pollutants in the surface water, but the carcinogenic risk of the Cr element is the highest.;

Purpose. Dendritic cells (DC) are specialized antigen-presenting cells, and cytokine-induced killer (CIK) cells have a specific killing activity to a variety of tumors. However, the underlining mechanism and function of DC-CIK cells in acute myeloid leukemia (AML) remain largely elusive. Methods. Gene expression profiles of leukemia patients were obtained from TCGA, DC cell components were evaluated using the quanTIseq method, and cancer stem cell scores were estimated using machine learning methods. The transcriptomes were obtained in DC-CIK cells from normal and AML patients by high-throughput sequencing. Large differentially expressed mRNAs were verified by RT-qPCR assay, and MMP9 and CCL1 were selected for subsequent studies in vivo and in vitro experiments. Results. Significant positive correlations were found with DC versus cancer stem cells (p=0.008) and the expression of MMP9 versus cancer stem cells (p=0.018). MMP9 and CCL1 were found to be highly expressed in DC-CIK cells from AML patients. DC-CIK cells with MMP9 and CCL1 knockout alone had little effect on leukemia cells, while knockdown of MMP9 and CCL1 in DC-CIK cells increased cytotoxicity, suppressed proliferation, and induced apoptosis of leukemia cells. In addition, we proved that MMP9- and CCL1-silenced DC-CIK cells significantly elevated the CD3+CD4+ and CD3+CD8+ cells and lowered the CD4+PD-1+ and CD8+PD-1+ T cells. Meanwhile, blockage of MMP9 and CCL1 in DC-CIK cells dramatically increased IL-2 and IFN-γ, increased CD107aþ (LAMP-1) and granzyme B (GZMB), and downregulated PD-1, CTLA4, TIM3, and LAG3 T cells from AML patients and AML model mice. Furthermore, activated T cells in DC-CIK cells knocking down MMP9 and CCL1 also prevented proliferation and accelerated apoptosis of AML cells. Conclusion. Our findings demonstrated that blockage of MMP9 and CCL1 in DC-CIK cells could markedly enhance the therapeutic efficiency in AML via activating T cells.

Advanced oxidation process (AOP) has attracted widespread attention because it can effectively remove antibiotics in water, but its practical engineering application is limited by the problems of the low efficiency and difficult recovery of the catalyst. In the study, nano-spinel CoFe2O4 was prepared by hydrothermal method and served as the peroxymonosulfate (PMS) catalyst to degrade antibiotic amoxicillin (AMX). The reaction parameters such as CoFe2O4 dosage, AMX concentration, and initial pH value were also optimized. The reaction mechanism was proposed through free radical capture experiment and possible degradation pathway analysis. In addition, the magnetic recovery performance and stability of the catalyst were evaluated. Results showed that 85.5% of AMX could be removed within 90 min at optimal conditions. Sulfate radicals and hydroxyl radicals were the active species for AMX degradation. Moreover, the catalyst showed excellent magnetism and stability in the cycle experiment, which has great potential in the AOP treatment of antibiotic polluted wastewater. ObjectivesReinfection of human papillomavirus (HPV) among men who have sex with men (MSM) after clearing the infection of HPV has not been well characterized. To understand the reinfection of HPV among human immunodeficiency virus (HIV) negative MSM without HPV vaccine, we analyzed the reinfection of nine-valent HPV vaccine (9v-HPV) types.MethodsData were acquired from a prospective cohort study among HIV-negative MSM in Urumqi of Xinjiang from 1 April 2016 to 14 July 2020. Participants were recruited through a local non-government organization using a website advertisement. Self-administered questionnaires and self-collected anal swabs were collected at baseline and every 6 months. The incidence rates of reinfection was calculated based on the number of events divided by person-months of observation of event risk. 95% confidence intervals (CIs) were calculated based on the Poisson distribution.ResultsA total of 515 HIV-negative unvaccinated MSM were included with a mean age of 30.93 years (SD 7.85), and 27.68% were reinfected with any 9v-HPV type after clearance. The reinfection incidence rate of any 9v-HPV was 14.47 per 1000 person-months (95% CI: 11.52–17.87). HPV52 was the most common type of reinfection, with a reinfection rate of 17.96 per 1,000 person months (95% CI: 11.58–26.33). Univariate analysis showed that MSM over the age of 30 had a slightly higher risk of reinfection with any 9v-HPV (Hazard ratio (HR): 1.57; 95% CI: 1.01–2.45), but no significant association was observed in multivariate analysis.ConclusionsOur study showed MSM without HPV vaccine will become reinfected following the natural clearance of specific HPV types. It is also suggested that HPV vaccination is recommended not only prior to sexual debut but also after viral clearance for MSM to reduce HPV prevalence.

ObjectiveMen who have sex with men (MSM) are at increased risk for Human papillomavirus (HPV) infection compared to women and heterosexual men. We aimed to assess the incidence, clearance and duration of anal human papillomavirus (HPV) infection in HIV-negative MSM and the influencing factors in a 5-year prospective cohort study.MethodsFrom April 2016 to April 2021, HIV-negative MSM were recruited and followed every 6 months in Urumqi, Xinjiang, China. Questionnaires and anal swabs were collected at baseline and every 6 months. We detected 37 anal HPV genotypes using the HPV Geno Array Diagnostic Kit Test. Incidence and clearance rates of anal HPV infection and the influencing factors were estimated using a two-state Markov model.ResultsA total of 585 MSM were included with a median age of 37 years [interquartile range (IQR): 31–43 years] and were followed for a median 2.8 years (IQR: 1.8–3.6 years). Incidence rates for any HPV and high-risk HPV (Hr-HPV) were 53.4 [95% confidence interval (CI): 49.1–58.0] and 39.0 (95% CI: 35.7–42.5)/1,000 person-months. Median duration of infection was 9.67 (95% CI: 8.67–10.86) and 8.51 (95% CI: 7.57–9.50) months, respectively. Clearance rates for any HPV and Hr-HPV were 50.9 (95% CI: 46.7–55.3) and 62.1 (95% CI: 56.8–66.7)/1,000 person-months, respectively. HPV16 and HPV6 had the highest incidence, lowest clearance rate and longest duration of infection among Hr-HPV and low-risk HPV (Lr-HPV) types, respectively. Receptive anal sex is a risk factor for any HPV [hazard ratio (HR) = 1.66, 95% CI: 1.16–2.38] and Hr-HPV infection (HR = 1.99, 95% CI:1.39–2.85). Recent anal sex without condom use was significantly associated with any HPV (HR = 1.80, 95% CI: 1.10–2.94) and Hr-HPV infection (HR = 2.60, 95% CI: 1.42–4.77). Age ≥35 years was significantly associated with Lr-HPV HPV infection only (HR = 1.40, 95% CI: 1.02–1.93). Both inserted and receptive anal sex (HR = 0.60, 95% CI: 0.40–0.89) and anal sex ≥2 times per week (HR = 0.61, 95% CI: 0.43–0.87) were associated with reduced Hr-HPV clearance. Six of the nine-valent vaccine types (HPV6, 11, 16, 18, 52 and 58) occurred most frequently, which indicates the need for high vaccination coverage in MSM.ConclusionsIn this cohort study, high incidence and low clearance of any HPV, Hr-HPV and individual HPV infections emphasize the importance of MSM vaccination. Modifiable behavioral factors such as condoms and drug use should be incorporated into HPV prevention strategies.

Background Post‐resuscitation syndrome, involves a severe inflammatory response following successful cardiopulmonary resuscitation. The potential mechanism of Vitamin C (VitC) after cardiopulmonary resuscitation on myocardial and cerebral function, duration of survival is undefined. Methods and Results A first set of experiments were done in 18 male Sprague‐Dawley rats for the investigation of short‐term follow‐up, randomized into 3 groups: (1) sham; (2) controls; (3) VitC. Ventricular fibrillation was electrically induced and untreated for 6 minutes. Cardiopulmonary resuscitation including chest compression and mechanical ventilation were then initiated and continued for 8 minutes followed by defibrillation. At 5 minutes after return of spontaneous circulation, either VitC (200 mg/kg) or placebo was administered by intravenous infusion with a syringe pump for half an hour. There were significant improvements in myocardial function and buccal microcirculation in rats treated with VitC after return of spontaneous circulation 4 hours compared with controls. As high-performance computing (HPC) systems have scaled up, resilience has become a great challenge. To guarantee resilience, various kinds of hardware and software techniques have been proposed. However, among popular software fault-tolerant techniques, both the checkpoint-restart approach and the replication technique face challenges of scalability in the era of peta- and exa-scale systems due to their numerous processes. In this situation, algorithm-based approaches, or algorithm-based fault tolerance (ABFT) mechanisms, have become attractive because they are efficient and lightweight. Although the ABFT technique is algorithm-dependent, it is possible to implement it at a low level (e.g., in libraries for basic numerical algorithms) and make it application-independent. However, previous ABFT approaches have mainly aimed at achieving fault tolerance in integrated circuits (ICs) or at the architecture level and are therefore not suitable for HPC systems; e.g., they use checksums of rows and columns of matrices rather than checksums of blocks to detect errors. Furthermore, they cannot deal with errors caused by node failure, which are common in current HPC systems. To solve these problems, this paper proposes FT-PBLAS, a PBLAS-based library for fault-tolerant parallel linear algebra computations that can be regarded as a fault-tolerant version of the parallel basic linear algebra subprograms (PBLAS), because it provides a series of fault-tolerant versions of interfaces in PBLAS. To support the underlying error detection and recovery mechanisms in the library, we propose a block-checksum approach for non-fatal errors and a scheme for addressing node failure, respectively. We evaluate two fault-tolerant mechanisms and FT-PBLAS on HPC systems, and the experimental results demonstrate the performance of our library.

Head and neck cancer is the sixth most frequent cancer all over the world, with the majority of subtypes of head and neck squamous cell carcinoma (HNSCC). Cellular senescence-associated genes have been confirmed to play a critical role in cancer and have the potential to be prognostic biomarkers for cancer. Clinical information of HNSCC samples and expression data were acquired from public databases. Expression profiles of genes related to cellular senescence were used to identify molecular subtypes by consensus clustering. To screen differentially expressed genes (DEGs) between different subtypes, differential analysis was performed. We used the univariate Cox regression to identify prognostic DEGs and performed least absolute shrinkage and selection operator (LASSO) to optimize and construct a prognostic model. CIBERSORT, ESTIMATE, and TIDE tools were applied to estimate immune characteristics. Four molecular subtypes were established based on cellular senescence-associated genes. Differential prognosis was observed among different subtypes with C4 having the longest overall survival and C1 having the worst prognosis. C4 subtype also showed the highest immune infiltration. We screened a total of eight cellular senescence prognosis-related genes and established a cellular senescence-related signature score (CSRS.Score) that could stratify samples into high-CSRS.Score and low-CSRS.Score groups. The high-CSRS.Score group had worse prognosis, lower immune infiltration, and lower response to immunotherapy. We further improved the prognostic model and survival prediction by combining CSRS.Score with clinicopathological features using a decision tree model, which had high predictive accuracy and survival prediction. This study demonstrated an important role of cellular senescence in HNSCC. The identified eight cellular senescence-associated genes have the potential to provide ideas for adjuvant treatment and personalized treatment of HNSCC patients.

The physiology and physiopathology process of mitochondrial function following cardiac arrest remains poorly understood. We aimed to assess mitochondrial respiratory function on the heart and brain homogenates from cardiac arrest rats. The expression level of SIRT1/PGC-1α pathway was measured by immunoblotting. 30 rats were assigned to the CA group and the sham group. Rats of CA were subjected to 6 min of untreated ventricular fibrillation (VF) followed by 8 min of cardiopulmonary resuscitation (CPR). Mitochondrial respiratory function was compromised following CA and I/R injury, as indicated by CIL (451.46 ± 71.48 vs. 909.91 ± 5.51 pmol/min*mg for the heart and 464.14 ± 8.22 vs. 570.53 ± 56.33 pmol/min*mg for the brain), CI (564.04 ± 64.34 vs. 2729.52 ± 347.39 pmol/min*mg for the heart and 726.07 ± 85.78 vs. 1762.82 ± 262.04 pmol/min*mg for the brain), RCR (1.88 ± 0.46 vs. 3.57 ± 0.38 for the heart and 2.05 ± 0.19 vs. 3.49 ± 0.19, for the brain) and OXPHOS coupling efficiency (0.45 ± 0.11 vs. 0.72 ± 0.03 for the heart and 0.52 ± 0.05 vs. 0.71 ± 0.01 for the brain). However, routine respiration was lower in the heart and comparable in the brain after CA. CIV did not change in the heart but was enhanced in the brain. Furthermore, both SIRT1 and PGC-1α were downregulated concurrently in the heart and brain. The mitochondrial respiratory function was compromised following CA and I/R injury, and the major affected respiratory state is complex I-linked respiration. Furthermore, the heart and the brain respond differently to the global I/R injury after CA in mitochondrial respiratory function. Inhibition of the SIRT1/PGC-1α pathway may be a major contributor to the impaired mitochondrial respiratory function.

This paper presents the design and process of two highly sensitive sensors working in the terahertz band. The sensors comprise the quartz substrate, medium, reflection plate, and metal resonant layer with a symmetrical single-slot patch array. The devices help study the electrically induced permittivity of two liquid crystals in different frequency bands and at different voltages, and the experimental data verify that both liquid crystals have a large birefringence. Based on experimental results, the sensitivity of the fabricated sensor is 47.03 GHz/RIU in the frequency range 90–140 GHz. Similarly, the other fabricated sensor has a sensitivity of 112.47 GHz/RIU in the frequency range 325–500 GHz. The results show that both sensors have superior sensing properties and potential applications in biological and chemical liquid sensing.

Abstract The fluorescence emission from green fluorescent protein (GFP) is known to be heavily influenced by hydrogen bonding between the core fluorophore and the surrounding side chains or water molecules. Yet how to utilize this feature for modulating the fluorescence of GFP chromophore or GFP-like fluorophore still remains elusive. Here we present theoretical calculations to predict how hydrogen bonding could influence the excited states of the GFP-like fluorophores. These studies provide both a new perspective for understanding the photophysical properties of GFP as well as a solid basis for the rational design of GFP-based fluorophores.

We present new results on the fault tolerability of $k$ -ary $n$ -cube (denoted $Q_{n}^{k}$ ) networks. $Q_{n}^{k}$ is a topological model for interconnection networks that has been extensively studied since proposed, and this paper is concerned with the structure/substructure connectivity of $Q_{n}^{k}$ networks, for paths and cycles, two basic yet important network structures. Let $G$ be a connected graph and $T$ a connected subgraph of $G$ . The $T$ -structure connectivity $\kappa (G; T)$ of $G$ is the cardinality of a minimum set of subgraphs in $G$ , such that each subgraph is isomorphic to $T$ , and the set’s removal disconnects $G$ . The $T$ -substructure connectivity $\kappa ^{s}(G; T)$ of $G$ is the cardinality of a minimum set of subgraphs in $G$ , such that each subgraph is isomorphic to a connected subgraph of $T$ , and the set’s removal disconnects $G$ . In this paper, we study $\kappa (Q_{n}^{k}; T)$ and $\kappa ^{s}(Q_{n}^{k}; T)$ for $T=P_{i}$ , a path on $i$ nodes (resp. $T=C_{i}$ , a cycle on $i$ nodes). Lv et al. determined $\kappa (Q_{n}^{k}; T)$ and $\kappa ^{s}(Q_{n}^{k}; T)$ for $T\in \{P_{1},P_{2},P_{3}\}$ . Our results generalize the preceding results by determining $\kappa (Q_{n}^{k}; P_{i})$ and $\kappa ^{s}(Q_{n}^{k}; P_{i})$ . In addition, we have also established $\kappa (Q_{n}^{k}; C_{i})$ and $\kappa ^{s}(Q_{n}^{k}; C_{i})$ .

Background To investigate the therapeutic potential of combined therapy with polyethylene glycol‐20k (PEG‐20k) and MCC950 on post‐resuscitation myocardial function in a rat model of cardiac arrest. Methods and Results Thirty rats were randomized into 5 groups: Sham, Control, PEG‐20k, MCC950, PEG‐20k+ MCC950. Except for sham, animals were subjected to 6 minutes of ventricular fibrillation followed by 8 minutes cardiopulmonary resuscitation. Two milliliters PEG‐20k was administered by intravenous injection coincident with the start of cardiopulmonary resuscitation; MCC950 (10 mg/kg), a highly selective NLRP3 inflammasome inhibitor, was delivered immediately after restoration of spontaneous circulation. Myocardial function, sublingual microcirculation, mitochondrial function, plasma cardiac troponin I, and interleukin‐1β, expression of proteins in SIRT1 (sirtuin 1)/PGC‐1α (peroxisome proliferator‐activated receptor gamma coactivator 1‐alpha) and NLRP3 (the NOD‐like receptor family protein 3) inflammasome pathways were evaluated. Following cardiopulmonary resuscitation, myocardial function was compromised with a significantly decreased cardiac output, ejection fraction, and increased myocardial performance index, cardiac troponin I. Sublingual microcirculation was disturbed with impaired perfused vessel density and microvascular flow index. Cardiac arrest reduced mitochondrial routine respiration, Complex I‐linked respiration, respiratory control rates and oxidative phosphorylation coupling efficiency. PEG‐20k or MCC950 alone restored mitochondrial respiratory function, restituted sublingual microcirculation, and preserved myocardial function, whereas a combination of PEG‐20k and MCC950 further improved these aspects. PEG‐20k restored the expression of SIRT1 and PGC‐1α, and blunted activation of NLRP3 inflammasomes. MCC950 suppressed expression of cleaved‐caspase‐1/pro‐caspase‐1, ASC (apoptosis‐associated speck‐like protein), GSDMD [gasdermin d], and interleukin‐1β. Conclusions Combined therapy with PEG‐20k and MCC950 is superior to either therapy alone for preserving post‐resuscitated myocardial function, restituting sublingual microcirculation at restoration of spontaneous circulation at 6 hours. The responsible mechanisms involve upregulated expression of SIRT1/PGC1‐α in tandem with inhibition of NLRP3 inflammasomes.

Accumulating evidence demonstrated that administration of ω-3 polyunsaturated fatty acid (ω-3 PUFA) or ascorbic acid (AA) following cardiac arrest (CA) improves survival. Therefore, we investigate the effects of ω-3 PUFA combined with AA on myocardial function after CA and cardiopulmonary resuscitation (CPR) in a rat model.Thirty male rats were randomized into 5 groups: (1) sham; (2) control; (3) ω-3 PUFA; (4) AA; (5) ω-3 PUFA + AA. Ventricular fibrillation (VF) was induced and untreated for 6 min followed by defibrillation after 8 min of CPR. Infusion of drug or vehicle occurred at the start of CPR. Myocardial function and sublingual microcirculation were measured at baseline and after return of spontaneous circulation (ROSC). Heart tissues and blood were collected 6 h after ROSC. Myocardial function and sublingual microcirculation improvements were seen with ω-3 PUFA or AA compared to control after ROSC (p < 0.05). ω-3 PUFA + AA shows a better myocardial function than ω-3 PUFA or AA (p < 0.05). ω-3 PUFA or AA decreases pro-inflammatory cytokines, cTnI, myocardium malondialdehyde (MDA) and 4-hydroxynonenal (4-HNE) modified proteins compared to control (p < 0.05). ω-3 PUFA and AA combined have lower MDA and 4-HNE modified proteins than alone (p < 0.05). ω-3 PUFA or AA treatment reduces the severity of post-resuscitation myocardial dysfunction, improves sublingual microcirculation, decreases lipid peroxidation and systemic inflammation in the early phase of recovery following CA and resuscitation. A combination of ω-3 PUFA and AA treatment confers an additive effect in suppressing lipid peroxidation and improving myocardial function.

In this article, we present a study of the equivalent dielectric constant of two nematic liquid crystals (LCs) with different thicknesses, in the region from 90 to 140 GHz. The equivalent dielectric constant of the LCs was measured using a frequency selective surface (FSS). The LC-based tunable FSS with 22 × 25 unit cells was printed on a quartz substrate with an area of 4 × 4 cm2 and a thickness of 480 µm; the LC layer with thicknesses ranging from 30 to 100 µm acted as a substrate. The FSS featured a maximum frequency-shifting range of 8.15 GHz and 30 µm-thick LC layers with mutually orthogonal rubbing directions were deposited on it. The results show that the initial equivalent dielectric constant of the LC layer increased with the LC layer thickness, while the saturation-equivalent dielectric constant remained almost constant. This work provides LC parameters that can be useful for the design of LC-based devices in the millimeter and terahertz ranges.

Abstract Isaria farinosa is a pathogen of alpine Thitarodes larvae that are hosts for the Chinese medicinal fungus, Ophiocordyceps sinensis. A matrix analysis indicated that the optimal culture conditions for the mycelial growth of I. farinosa are a 50-mL liquid broth in a 250-mL flask at more than 100-rpm rotation and 15–25 °C. Illumination does not affect the mycelial growth. The optimal nutrition requirements are D-(+)-galactose and D-(−)-fructose as carbon resources and D-cysteine as well as yeast powder, peptone, and beef extract as nitrogen resources at a carbon-to-nitrogen ratio of 1:1 to 1:7. The mineral component and vitamins also significantly increase the mycelial growth of I. farinosa. Based on the optimal culture conditions and nutrition requirements for the mycelial growth of I. farinosa, the effects of altitude on mycelial growth and its metabolome were evaluated using quadrupole-time-of-flight/mass spectrometry, principal component analysis and partial least squares discriminant analysis. The altitude did not affect the mycelial production but significantly regulated its metabolome. The study presents a new approach to better select a method for producing more useful metabolites and highlights the necessity of establishing standards for culturing methods related to altitude to preserve fungal quality; additionally, the results indicate that the use of a fermenter may meet the demands of large-scale mycelial production.

In this paper, we propose a new technique to distinguish the reason for program failure between hardware malfunctions and program bugs, which mitigates the impact of shorter mean time between failures to the debugging process on the future exa-scale supercomputers and improves the productivity of debugging large-scale parallel programs. Our technique detects program failures by observing the abnormal message passing behaviors with distributed monitors and leverages event-driven mechanism to trigger global status checking among different node groups concurrently. Besides, both coarse-grained execution snapshots and fine-grained failure events can be provided for further failure diagnosis and bug analysis. We implement this technique as a user-space library named failure cause resolver (FCR). Experimental results on the Tianhe-2 supercomputer demonstrate that the latency of FCR for failure detection is acceptable with negligible overhead. In addition, FCR does not require administrative privilege and can be easily integrated into existing large-scale parallel programs.

Photocatalytic water splitting is an attractive route to hydrogen production, but safely isolating hydrogen and oxygen products remains a challenge. Here, the authors propose a carbon nitride/grapheme (GR)-based sandwich structure, providing safe and effective photocatalytic hydrogen generation and capsule storage.

Abstract Al-doped ZnO (AZO) thin films were deposited directly on p-GaN substrates by using a two-step deposition consisting of polymer assisted deposition (PAD) and atomic layer deposition (ALD) methods. Ohmic contacts of the AZO on p-GaN have been formed. The lowest sheet resistance of the two-step prepared AZO films reached to 145 Ω/sq, and the specific contact resistance reduced to 1.47 × 10−2 Ω·cm2. Transmittance of the AZO films remained above 80% in the visible region. The combination of PAD and ALD technique can be used to prepare p-type ohmic contacts for optoelectronics.

Abstract GaN-based metal-oxide-semiconductor capacitors with ZrO2 as the dielectric layer have been prepared by atomic layer deposition. The accumulation and depletion regions can be clearly distinguished when the voltage was swept from −4 to 4 V. Post-annealing results suggested that the capacitance in accumulation region went up gradually as the annealing temperature increased from 300 to 500 °C. A minimum leakage current density of 3 × 10−9 A/cm2 at 1 V was obtained when O3 was used for the growth of ZrO2. Leakage analysis revealed that Schottky emission and Fowler-Nordheim tunneling were the main leakage mechanisms.

With five histone deacetylase (HDAC) inhibitors approved for cancer treatment, proteolysis-targeting chimeras (PROTACs) for degradation of HDAC are emerging as an alternative strategy for HDAC-targeted therapeutic intervention. Herein, three bestatin-based hydroxamic acids (P1, P2 and P3) were designed, synthesized and biologically evaluated to see if they could work as HDAC degrader by recruiting cellular inhibitor of apoptosis protein 1 (cIAP1) E3 ubiquitin ligase. Among the three compounds, the bestatin-SAHA hybrid P1 exhibited comparable even more potent inhibitory activity against HDAC1, HDAC6 and HDAC8 relative to the approved HDAC inhibitor SAHA. It is worth noting that although P1 could not lead to intracellular HDAC degradation after 6 h of treatment, it could dramatically decrease the intracellular levels of HDAC1, HDAC6 and HDAC8 after 24 h of treatment. Intriguingly, the similar phenomenon was also observed in the HDAC inhibitor SAHA. Cotreatment with proteasome inhibitor bortezomib could not reverse the HDAC decreasing effects of P1 and SAHA, confirming that their HDAC decreasing effects were not due to protein degradation. Moreover, all three bestatin-based hydroxamic acids P1, P2 and P3 exhibited more potent aminopeptidase N (APN, CD13) inhibitory activities than the approved APN inhibitor bestatin, which translated to their superior anti-angiogenic activities. Taken together, a novel bestatin-SAHA hybrid was developed, which worked as a potent APN and HDAC dual inhibitor instead of a PROTAC.

Rice false smut has become an increasingly serious fungal disease in rice (Oryza sativa L.) production worldwide. Ustilaginoidins are bis-naphtho-γ-pyrone mycotoxins previously isolated from the rice false smut balls (FSBs) infected by the pathogen Villosiclava virens in rice spikelets on panicles. To investigate the main ustilaginoidins and their distribution in rice FSBs, five main bis-naphtho-γ-pyrones, namely ustilaginoidins A (1), G (2), B (3), I (4) and C (5), were isolated and identified by NMR and high-resolution mass spectrometry as well as by comparison with the data in the literature. The rice FSBs at early, middle and late maturity stages were divided into their different parts and the contents of five main ustilaginoidins for each part were determined by HPLC analysis. The results revealed that the highest levels of ustilaginoidins were in late stage rice FSBs, followed by those at middle stage. Most ustilaginoidins, 96.4% of the total quantity, were distributed in the middle layer at early stage. However, ustilaginoidins were mainly distributed in the outer and middle layers at middle and late stages. Small amounts of ustilaginoidins A (1) and G (2) were found in the inner part of rice FSBs at each maturity stage. The contents of ustilaginoidins A (1) and G (2) without hydroxymethyl groups at C-2 and C-2’ of the γ-pyrone rings in rice FSBs were relatively high at early stage, while the contents of ustilaginoidins B (3), I (4), and C (5) with hydroxymethyl groups at C-2 or C-2’ were relatively high at late stage.

The Botryosphaeriaceae represents an important, cosmopolitan family of pathogens infecting woody plants. Grapevines (Vitis vinifera) suspected of being affected by Botryosphaeria dieback were collected from different Provinces of China and several Botryosphaeriaceae species were identified. This research was conducted to further study the species of Botryosphaeriaceae associated with grapevines in China and to estimate the prevalence and severity of the disease. Symptoms were characterized by partial or total death of grapevine cordons, with brown U-shaped necrotic sectors and brownish-black spots in cross-sections of affected trunks and arms. A fungus isolated from diseased vines in Henan and Anhui Provinces was identified as Neofusicoccum mangiferae on the basis of its morphological and cultural characteristics as well as an analysis of combined sequence data from the internal transcribed spacer (ITS) cluster of the ribosomal DNA together with partial sequences of the β-tubulin (BT) and translation elongation factor (EF1-α) genes. Pathogenicity was assessed by inoculating the fungus onto detached green grapevine shoots under controlled laboratory conditions and mature wood of standing vines cv. Summer Black in a greenhouse. This is the first record of N. mangiferae associated with grapevine dieback in the world.

Ultrathin, transparent and flexible capacitors using graphene as the bottom electrodes were directly fabricated on polyethylene naphthalate (PEN) substrates. ZrO2 dielectric films were deposited on the treated surface of graphene by atomic layer deposition (ALD). The deposition process did not introduce any detectible defects in the graphene, as indicated by Raman measurements, guaranteeing the electrical performances of the graphene electrodes. The Aluminum-doped zinc oxide (AZO) films were prepared as the top electrodes using the ALD technique. The capacitors presented a high capacitance density (10.3 fF/μm2 at 10 kHz) and a relatively low leakage current (5.3 × 10−6 A/cm2 at 1 V). Bending tests revealed that the capacitors were able to work normally at an outward bending radius of 10 mm without any deterioration of electrical properties. The capacitors exhibited an average optical transmittance of close to 70% at visible wavelengths. Thus, it opens the door to practical applications in transparent integrated circuits.

Sorbicillinoids are important hexaketide metabolites derived from fungi. They have a variety of biological activities including cytotoxic, antioxidant, antiviral and antimicrobial activity. The unique structural features of the sorbicillinoids make them attractive candidates for developing new pharmaceutical and agrochemical agents. About 90 sorbicillinoids have been reported in the past few decades. This mini-review aims to briefly summarize their occurrence, structures, and biological activities.
