Abstract Background Phaseolus vulgaris is a warm-season crop sensitive to low temperatures, which can adversely affect its growth, yield, and market value. Exogenous growth regulators, such as diethyl aminoethyl hexanoate (DA-6), have shown potential in alleviating stress caused by adverse environmental conditions. However, the effects that DA-6 has on P. vulgaris plants subjected to low-temperature stress are not well understood. This study aimed to investigate the impact DA-6 has on the growth, photosynthesis, antioxidant system, and gene expression in cold-tolerant (YJ009763) and cold-sensitive (Baibulao) P. vulgaris seedlings under low-temperature stress. Results To simulate low-temperature stress, P. vulgaris seedlings were exposed to 5 °C, and 25 mg/L DA-6 solution applied to their leaves. This study revealed that DA-6 significantly enhanced the growth and photosynthetic performance of P. vulgaris seedlings under low-temperature stress. Specifically, DA-6 increased chlorophyll content and photosynthetic rates, reducing stomatal limitation and enhancing carbon assimilation. It also improved the photosynthetic efficiency by boosting electron transport in the reaction center. The antioxidant enzyme activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) were markedly increased following DA-6 treatment. After 24 h of low-temperature stress, the cold-tolerant seedlings showed a 68.95% increase in POD activity, whereas the cold-sensitive seedlings displayed a 160.63% increase in SOD activity and an 85.56% increase in CAT activity. In addition, DA-6 significantly reduced the production rate of superoxide anion radical generation, with a 25.24% reduction in cold-tolerant seedlings and a 49.38% reduction in cold-sensitive seedlings. Under low-temperature stress, exogenous DA-6 could upregulate the relative expression of antioxidant enzyme-related genes, such as PvSOD and PvAPX. DA-6 also promoted the expression of key antioxidant genes, including PvMDHAR and PvDHAR2, which accelerated the ascorbate-glutathione cycle and mitigated oxidative stress. Conclusion Exogenous application of DA-6 effectively alleviates low-temperature stress in P. vulgaris by enhancing photosynthetic capacity and regulating the antioxidant defense system. Cold-tolerant varieties exhibited a stronger response to DA-6, demonstrating a greater ability to withstand cold stress. These findings suggest that DA-6 treatment could serve as a promising approach for improving the resilience of P. vulgaris to low temperatures.

Abstract Background Consumption of leafy vegetables is a primary route of cadmium (Cd) exposure in the human body. Salicylic acid (SA) is a major stress signaling molecule that alleviates Cd toxicity in various plants. Our study aimed to investigate the effects of different SA concentrations on spinach growth, cadmium accumulation, and stress resistance physiology under cadmium stress (50 µmol/L). Results Cd stress significantly markedly decreased spinach growth and biomass, reduced its photosynthetic efficiency, increased activities of antioxidative enzymes, and upregulated the relative expression of several genes involved in cadmium absorption and transport compared to the control. The exogenous application of SA mitigated the harmful effects of Cd in spinach. 0.8 and 1.6 mmol/L SA significantly increased spinach root length, plant height, and biomass and decreased the Cd content in shoots by 30.03 and 17.35% compared to the Cd-treated group. Moreover, SA alleviated the yellowing of leaves caused by Cd stress. Exogenous SA ameliorated Cd toxicity in spinach by reducing reactive oxygen species, malondialdehyde, proline, and soluble protein levels. Exogenous SA application reduced Cd absorption in spinach leaves by downregulating the expression of genes involved in Cd transport, such as SoHMA4-like, SoNramp3.1-like, SoNramp6-like, and SoNramp7.2-like. Principal component analysis and correlation analysis showed that exogenous SA application under Cd stress was correlated with plant Cd content, photosynthetic pigment content, and relative expression of Cd absorption and transportation-related genes. Conclusions To summarize, these findings indicate that SA mitigates Cd toxicity in spinach by reversing the adverse effects of Cd stress on plant growth and reducing Cd accumulation in the shoots.

Abstract Sodium sulfide (Na2S) as an initial cathode material in room-temperature sodium-sulfur batteries is conducive to get rid of the dependence on Na-metal anode. However, the micron-sized Na2S that accords with the practical requirements is obstructed due to poor kinetics and severe shuttle effect. Herein, a subtle strategy is proposed via regulating Na2S redeposition behaviours. By the synergistic effect from both conductive structure and cuprous sulfide (Cu2S) catalysis, the micron-sized Na2S particles are broken down and redeposited to nano-size during the initial cycle which can be fully utilized in subsequent cycles. Consequently, the Na2S/CPVP@Cu2S||Na cell delivers excellent cyclability (670 mAh gS −1 after 500 cycles) with a remarkable average Coulombic efficiency over 99.7% and rate capability (480 mAh gS −1 at 4 A gS −1). Besides, the Na-free anodes are used to prove the application prospects. This work provides an innovative idea for utilizing micron-sized Na2S and offers insights into its conversion pathway.

The ‘Okitsu No. 58’ citrus variety is highly prone to fruit cracking, which jeopardizes yield and results in economic losses. In this study, we investigated the impacts of spraying 5 distinct concentrations (0.1, 0.2, 0.3, 0.4, and 0.5 g/L) of chelated calcium (Ca) or silicon (Si) fertilizers at the young fruit stage (60-90 days after flowering, DAF) on fruit cracking and quality in the citrus variety ‘Okitsu No. 58’. The results showed either Ca or Si fertilizer treatments reduced fruit cracking. We found that all Ca and partial Si treatments (0.4 and 0.5 g/L) significantly promoted the accumulation of Ca content in the peel. Notably, Ca or Si treatments significantly reduced polygalacturonase (PG) activity and inhibited the production of water-soluble pectin (WSP) in the peel. Additionally, Ca or Si treatments elevated the superoxide dismutase (SOD) activity and decreased the malondialdehyde (MDA) content of the peels. Changes in these parameters likely contributed to strengthening the durability of peel cell wall constituents, thus enhancing the fruit’s resistance to fruit cracking. Overall, except for the C3 (0.3 g/L of Ca), Ca or Si fertilizers contributed to fruit conventional quality, mainly in terms of higher soluble sugars (SS) and SS/TA (titratable acid). Therefore, our findings will provide a reference for the prevention and control of citrus fruit cracking and the development of new fertilizers.

Volatile compounds are important determinants affecting fruit flavor. Previous study has identified a bud mutant of ‘Ehime 38’ (Citrus reticulata) with different volatile profile. However, the volatile changes between WT and MT during fruit development and underlying mechanism remain elusive. In this study, a total of 35 volatile compounds were identified in the pulps of WT and MT at five developmental stages. Both varieties accumulated similar and the highest levels of volatiles at stage S1, and showed a downward trend as the fruit develops. However, the total volatile contents in the pulps of MT were 1.4–2.5 folds higher than those in WT at stages S2-S5, which was mainly due to the increase in the content of d-limonene. Transcriptomic and RT-qPCR analysis revealed that most genes in MEP pathway were positively correlated with the volatile contents, of which DXS1 might mainly contribute to the elevated volatiles accumulation in MT by increasing the flux into the MEP pathway. Moreover, temporal expression analysis indicated that these MEP pathway genes functioned at different developmental stages. This study provided comprehensive volatile metabolomics and transcriptomics characterizations of a citrus mutant during fruit development, which is valuable for fruit flavor improvement in citrus.

‘Fengtang‘ plums soften quickly and lose flavor after harvest. This study comprehensively evaluated the effect of exogenous melatonin on the fruit quality of ‘Fengtang’ plums. According to our findings, exogenous melatonin prevented plum fruit from losing water, delayed the decline in firmness, and preserved a high TSS/TA level. Additionally, exogenous melatonin also enhanced the activity of antioxidant enzymes and increased the non-enzymatic antioxidants, thereby further increasing the antioxidant capacity of plum fruit. Notably, exogenous melatonin delayed the degradation of covalent soluble pectin (CSP), cellulose, and hemicellulose, as well as the rise in water-soluble pectin (WSP) concentration and the activity of cell wall degrading enzymes. Further investigation using atomic force microscopy (AFM) revealed that the chain-like structure of ionic-soluble pectin (ISP) and the self-assembly network structures of CSP were depolymerized, and melatonin treatment retarded the depolymerization of pectin structures. Our results showed that exogenous melatonin preserved the postharvest quality of plum fruits by controlling fruit softness and antioxidant capacity during storage.

The unprecedented infrared spectroscopic capabilities of JWST have provided high-quality interstellar medium metallicity measurements and enabled characterization of the gas-phase mass–metallicity relation (MZR) for galaxies at z ≳ 5 for the first time. We analyze the gas-phase MZR and its evolution in a high-redshift suite of FIRE-2 cosmological zoom-in simulations at z = 5–12 and for stellar masses M _* ∼ 10 ^6 –10 ^10 M _⊙ . These simulations implement a multichannel stellar feedback model and produce broadly realistic galaxy properties, including when evolved to z = 0. The simulations predict very weak redshift evolution of the MZR over the redshift range studied, with the normalization of the MZR increasing by less than 0.01 dex as redshift decreases from z = 12 to z = 5. The median MZR in the simulations is well approximated as a constant power-law relation across this redshift range given by $\mathrm{log}(Z/{Z}_{\odot })=0.37\mathrm{log}({M}_{* }/{{\rm{M}}}_{\odot })-4.3$ . We find good agreement between our best-fit model and recent observations made by JWST at high redshift. The weak evolution of the MZR at z > 5 contrasts with the evolution at z ≲ 3, where increasing normalization of the MZR with decreasing redshift is observed and predicted by most models. The FIRE-2 simulations predict increasing scatter in the gas-phase MZR with decreasing stellar mass, in qualitative agreement with some observations.

Sugars and organic acids are the main factors determining the flavor of citrus fruit. The WRKY transcription factor family plays a vital role in plant growth and development. However, there are still few studies about the regulation of citrus WRKY transcription factors (CsWRKYs) on sugars and organic acids in citrus fruit. In this work, a genome-wide analysis of CsWRKYs was carried out in the citrus genome, and a total of 81 CsWRKYs were identified, which contained conserved WRKY motifs. Cis-regulatory element analysis revealed that most of the CsWRKY promoters contained several kinds of hormone-responsive and abiotic-responsive cis-elements. Furthermore, gene expression analysis and fruit quality determination showed that multiple CsWRKYs were closely linked to fruit sugars and organic acids with the development of citrus fruit. Notably, transcriptome co-expression network analysis further indicated that three CsWRKYs, namely, CsWRKY3, CsWRKY47, and CsWRKY46, co-expressed with multiple genes involved in various pathways, such as Pyruvate metabolism and Citrate cycle. These CsWRKYs may participate in the metabolism of fruit sugars and organic acids by regulating carbohydrate metabolism genes in citrus fruit. These findings provide comprehensive knowledge of the CsWRKY family on the regulation of fruit quality.

The GLK gene family plays a crucial role in the regulation of chloroplast development and participates in chlorophyll synthesis. However, the precise mechanism by which GLK contributes to citrus’s chlorophyll synthesis remains elusive. The GLK gene family causes variations in the photosynthetic capacity and chlorophyll synthesis of different citrus varieties. In this study, we identified tissue-specific members and the key CcGLKs involved in chlorophyll synthesis. A total of thirty CcGLK transcription factors (TFs) were discovered in the citrus genome, distributed across all nine chromosomes. The low occurrence of gene tandem duplication events and intronic variability suggests that intronic variation may be the primary mode of evolution for CcGLK TFs. Tissue-specific expression patterns were observed for various GLK family members; for instance, CcGLK12 and CcGLK15 were specifically expressed in the skin, while CcGLK30 was specific to the ovary, and CcGLK10, CcGLK6, CcGLK21, CcGLK2, CcGLK18, CcGLK9, CcGLK28, and CcGLK8 were specifically expressed in the leaves. CcGLK4, CcGLK5, CcGLK11, CcGLK23, CcGLKl7, CcGLK26, and CcGLK20 may participate in the regulation of the ALA, prochlorophylate, protoporphyrin IX, Mg-protoporphyrin IX, Chl b, T-Chl, MG-ProtoIX ME, and POR contents in citrus.

IntroductionSalicylic acid (SA) is a multi-functional endogenous phytohormone implicated in the growth, development, and metabolism of many plant species.MethodsThis study evaluated the effects of different concentrations of SA (0, 25, 100, 200, and 500 mg/L) on the growth and cadmium (Cd) content of lettuce (Lactuca sativa L.) under Cd stress. The different concentrations of SA treatments were administered through foliar application. ResultsOur results showed that 100-200 mg/L SA significantly increased the plant height and biomass of lettuce under Cd stress. When SA concentration was 200 mg/L, the plant height and root length of lettuce increased by 19.42% and 22.77%, respectively, compared with Cd treatment alone. Moreover, 200 mg/L and 500mg/L SA concentrations could reduce peroxidase (POD) and superoxide dismutase (SOD) activities caused by Cd stress. When the concentration of exogenous SA was 500 mg/L, the POD and SOD activities of lettuce leaves decreased by 15.51% and 19.91%, respectively, compared with Cd treatment. A certain concentration of SA reduced the uptake of Cd by the lettuce root system and the transport of Cd from the lettuce root system to shoots by down-regulating the expression of Nramp5, HMA4, and SAMT, thus reducing the Cd content of lettuce shoots. When the concentration of SA was 100 mg/L, 200 mg/L, and 500 mg/L, the Cd contents of lettuce shoots were 11.28%, 22.70%, and 18.16%, respectively, lower than that of Cd treatment alone. Furthermore, principal component and correlation analyses showed that the Cd content of lettuce shoots was correlated with plant height, root length, biomass, antioxidant enzymes, and the expression level of genes related to Cd uptake.DiscussionIn general, these results provide a reference for the mechanism by which SA reduces the Cd accumulation in vegetables and a theoretical basis for developing heavy metal blockers with SA components.

Citrus cultivation involves the widespread practice of grafting, which has a significant impact on citrus development and fruit quality and yield. However, understanding the effect of flavonoid compounds after different rootstock grafting have been limited. Flavonoid compounds, found at the highest levels in citrus peels, contribute to improving fruit quality and nutritional value. In this study, scion-rootstock interaction was investigated at various developmental stages when sweet orange 'Newhall' was hetero-grafted with two commonly used rootstocks (Poncirus trifoliate population, C. junos Siebold ex Tanaka). Physiological index detection showed a higher concentration of total flavonoid content in peels of sweet orange 'Newhall' grafted on Poncirus trifoliate population (ct) than C. junos Siebold ex Tanaka (cj). Further metabolomic analysis identified 703 flavonoid compounds, including flavones, flavonols, and flavanones. Out of the 25 flavonoids affected by different rootstock grafting and developmental stages, most were flavones. Transcriptomic analysis identified 8,562 differentially expressed genes (DEGs). Co-expression and Pearson's correlation analysis discovered six hub structure genes and 19 transcription factors (TFs) that affected flavonoid biosynthesis. In addition to increasing the transcript levels of genes that synthesize flavones, flavonols, and flavanones, the scion-rootstock interaction also affected the expression of many TFs. Taken together, our findings suggested that hetero-grafting could promote the accumulation of flavonoid compounds in citrus peels during the development stages. These results offered fresh perspectives on grafting's application usefulness and the enhancement of the accumulation of nutritive flavonoid components by grafting in citrus.

Sweet orange ‘Newhall’ (C. sinensis) is a popular fruit in high demand all over the world. Its peel and pulp are rich in a variety of nutrients and are widely used in catering, medicine, food and other industries. Grafting is commonly practiced in citrus production. Different rootstock types directly affect the fruit quality and nutritional flavor of citrus. However, the studies on citrus metabolites by grafting with different rootstocks are very limited, especially for amino acids (AAs). The preliminary test showed that there were significant differences in total amino acid content of two rootstocks (Poncirus trifoliata (CT) and C. junos Siebold ex Tanaka (CJ)) after grafting, and total amino acid content in the peel was higher than flesh. However, the molecular mechanism affecting amino acid differential accumulation remains unclear. Therefore, this study selected peel as the experimental material to reveal the amino acid components and differential accumulation mechanism of sweet orange ‘Newhall’ grafted with different rootstocks through combined transcriptome and metabolome analysis. Metabolome analysis identified 110 amino acids (AAs) and their derivatives in sweet orange ‘Newhall’ peels, with L-valine being the most abundant. L-asparagine was observed to be affected by both developmental periods and rootstock grafting. Weighted gene co-expression network analysis (WGCNA) combined with Redundancy Analysis (RDA) revealed eight hub structural genes and 41 transcription factors (TFs) that significantly influenced amino acid biosynthesis in sweet orange ‘Newhall’ peels. Our findings further highlight the significance of rootstock selection in enhancing the nutritional value of citrus fruits and might contribute to the development of functional citrus foods and nutritional amino acid supplements.

Fruit cracking affects both the yield and economic efficiency of citrus; however, the underlying mechanism remains unclear. Therefore, this study focused on resistant and susceptible cultivars to identify the mechanisms underlying fruit cracking. The results showed that in ‘Mingrijian’, pectin morphological transformation and hemicellulose and lignin degradation in the pericarp were important contributing factors. During the critical fruit-cracking period (115–150 days after flowering), the water-soluble pectin, protopectin, and lignin contents in the pericarp of ‘Daya’ presented inverse changes relative to those in ‘Mingrijian’, thus enhancing the mechanical properties and resistance of pericarp. From 115 to 150 days after flowering, the soluble sugar content in the pulp of ‘Mingrijian’ increased rapidly by 97.35%, aiding in pulp water absorption and expansion. Moreover, the soluble protein content in the pericarp of ‘Mingrijian’ exhibited a declining trend and was lower than that of ‘Daya’, thus affecting the overall metabolism. The superoxide dismutase (SOD) activity in the pericarp of ‘Mingrijian’ gradually decreased from 115 to 180 days after flowering, while the peroxidase (POD) activity remained at a low level, resulting in weaker antioxidant capacity and lower environmental resistance. This study provides valuable insights into the mechanisms of citrus fruit cracking, laying the foundation for preventive and control strategies.

Bursty star formation—a key prediction for high-redshift galaxies from cosmological simulations explicitly resolving stellar feedback in the interstellar medium—has recently been observed to prevail among galaxies at redshift z ≳ 6. Line intensity mapping (LIM) of the 158 μ m [C ii ] line as a star formation rate (SFR) indicator offers unique opportunities to tomographically constrain cosmic star formation at high redshift, in a way complementary to observations of individually detected galaxies. To understand the effects of bursty star formation on [C ii ] LIM, which have remained unexplored in previous studies, we present an analytic modeling framework for high- z galaxy formation and [C ii ] LIM signals that accounts for bursty star formation histories induced by delayed supernova feedback. We use it to explore and characterize how bursty star formation can impact and thus complicate the interpretation of the [C ii ] luminosity function and power spectrum. Our simple analytic model indicates that bursty star formation mainly affects low-mass galaxies by boosting their average SFR and [C ii ] luminosity, and in the [C ii ] power spectrum it can create a substantial excess in the large-scale clustering term. This distortion results in a power spectrum shape that cannot be explained by invoking a mass-independent logarithmic scatter. We conclude that burstiness must be accounted for when modeling and analyzing [C ii ] data sets from the early Universe, and that in the extreme, the signature of burstiness may be detectable with first-generation experiments such as TIME, CONCERTO, and CCAT-DSS.

Sweet orange ‘Newhall’ peels (SOPs) are abundant in flavonoids, making them increasingly popular in the realms of nutrition, food, and medicine. However, there is still much unknown about flavonoid components in SOPs and the molecular mechanism of flavonoid biosynthesis when subjected to magnesium stress. The previous experiment conducted by the research group found that the total flavonoid content of Magnesium deficiency (MD) was higher than Magnesium sufficiency (MS) in SOPs. In order to study the metabolic pathway of flavonoids under magnesium stress, an integrative analysis of the metabolome and transcriptome was performed in SOPs at different developmental stages, comparing MS and MD. A comprehensive analysis revealed the identification of 1,533 secondary metabolites in SOPs. Among them, 740 flavonoids were classified into eight categories, with flavones identified as the dominant flavonoid component. The influence of magnesium stress on flavonoid composition was evaluated using a combination of heat map and volcanic map, which indicated significant variations between MS and MD varieties at different growth stages. The transcriptome detected 17,897 differential genes that were significantly enriched in flavonoid pathways. Further analysis was performed using Weighted gene correlation network analysis (WGCNA) in conjunction with flavonoid metabolism profiling and transcriptome analysis to identify six hub structural genes and ten hub transcription factor genes that play a crucial role in regulating flavonoid biosynthesis from yellow and blue modules. The correlation heatmap and Canonical Correspondence Analysis (CCA) results showed that CitCHS had a significant impact on the synthesis of flavones and other flavonoids in SOPs, as it was the backbone gene in the flavonoid biosynthesis pathway. The qPCR results further validated the accuracy of transcriptome data and the reliability of candidate genes. Overall, these results shed light on the composition of flavonoid compounds in SOPs and highlight the changes in flavonoid metabolism that occur under magnesium stress. This research provides valuable insights for improving the cultivation of high-flavonoid plants and enhancing our understanding of the molecular mechanisms underlying flavonoid biosynthesis.

Recent discoveries of a significant population of bright galaxies at cosmic dawn $\left(z\gtrsim 10\right)$ have enabled critical tests of cosmological galaxy formation models. In particular, the bright end of the galaxys’ UV luminosity functions (UVLFs) appear higher than predicted by many models. Using approximately 25,000 galaxy snapshots at 8 ≤ z ≤ 12 in a suite of FIRE-2 cosmological “zoom-in” simulations from the Feedback in Realistic Environments (FIRE) project, we show that the observed abundance of UV-bright galaxies at cosmic dawn is reproduced in these simulations with a multichannel implementation of standard stellar feedback processes, without any fine-tuning. Notably, we find no need to invoke previously suggested modifications, such as a nonstandard cosmology, a top-heavy stellar initial mass function, or a strongly enhanced star formation efficiency. We contrast the UVLFs predicted by bursty star formation in these original simulations to those derived from star formation histories (SFHs) smoothed over prescribed timescales (e.g., 100 Myr). The comparison demonstrates that the strongly time-variable SFHs predicted by the FIRE simulations play a key role in correctly reproducing the observed, bright-end UVLFs at cosmic dawn: the bursty SFHs induce order-or-magnitude changes in the abundance of UV-bright ( M _UV ≲ −20) galaxies at z ≳ 10. The predicted bright-end UVLFs are consistent with both the spectroscopically confirmed population and the photometrically selected candidates. We also find good agreement between the predicted and observationally inferred integrated UV luminosity densities, which evolve more weakly with redshift in FIRE than suggested by some other models.

IntroductionHeat stress can limit vegetable growth, and this can lead to constraints on agricultural production. Grafting technologies, however, can be used to alleviate various plant stresses.MethodsIn this study, the differences in the heat stress impacts and recovery abilities of pumpkin and luffa rootstocks for bitter gourd were analyzed in terms of their antioxidant activity and carbon and nitrogen metabolism.ResultsCompared with the un-grafted and self-grafted bitter gourd, which suffered from heat stress at 40°C for 24 h, heterologously grafted bitter gourd showed higher heat stability of the cell membrane (relative conductivity and malondialdehyde content were reduced), reduced oxidative stress (antioxidant enzyme activity was increased and the reactive oxygen species content reduced), and increased enzyme activity (sucrose phosphate synthase, sucrose synthase, neutral invertase, and acid invertase) and sugar content (soluble sugar, sucrose, fructose, and glucose) in carbon metabolism. The enzyme activity (nitrate reductase, nitrite reductase, and glutamine synthetase) and product content (nitrate and nitrite) of nitrogen metabolism were also found to be increased, and this inhibited the accumulation of ammonium ions. After the seedlings were placed at 25°C for 24 h, the heterogeneous rootstocks could rapidly restore the growth of the bitter gourd seedlings by promoting the antioxidant and carbon and nitrogen metabolism systems. When luffa was used as rootstock, its performance on the indexes was better than that of pumpkin. The correlation between the various indicators was demonstrated using a principal component and correlation analysis.DiscussionThe luffa rootstock was found to be more conducive to reducing cell damage and energy loss in bitter gourd seedlings caused by heat induction through the maintenance of intracellular redox homeostasis and the promotion of carbon and nitrogen metabolism.

Asparagus bean (Vigna unguiculata ssp. sesquipedialis) is an important cowpea subspecies. We assembled the genomes of Ningjiang 3 (NJ, 550.31 Mb) and Dubai bean (DB, 564.12 Mb) for comparative genomics analysis. The whole-genome duplication events of DB and NJ occurred at 64.55 and 64.81 Mya, respectively, while the divergence between soybean and Vigna occurred in the Paleogene period. NJ genes underwent positive selection and amplification in response to temperature and abiotic stress. In species-specific gene families, NJ is mainly enriched in response to abiotic stress, while DB is primarily enriched in respiration and photosynthesis. We established the pan-genomes of four accessions (NJ, DB, IT97K-499-35 and Xiabao II) and identified 20,336 (70.5%) core genes present in all the accessions, 6,507 (55.56%) variable genes in two individuals, and 2,004 (6.95%) unique genes. The final pan genome is 616.35 Mb, and the core genome is 399.78 Mb. The variable genes are manifested mainly in stress response functions, ABC transporters, seed storage, and dormancy control. In the pan-genome sequence variation analysis, genes affected by presence/absence variants were enriched in biological processes associated with defense responses, immune system processes, signal transduction, and agronomic traits. The results of the present study provide genetic data that could facilitate efficient asparagus bean genetic improvement, especially in producing cold-adapted asparagus bean.

Harumi tangor fruit with Ponkan as an interstock contains significantly higher levels of total soluble solids compared to Harumi tangor fruit cv.with no interstock. Transcriptome analysis of two graft combinations (Harumi/Hongjv (HP) and cv. cv.Harumi/Ponkan/Hongjv (HPP)) was conducted to identify the genes related to use of the Ponkan interstock. Soluble sugars and organic acids were also measured in the two graft combinations. The results showed that the contents of sucrose, glucose, and fructose were higher in the fruits of HPP than in those of HP; additionally, the titratable acid levels were lower in grafts with interstocks than in grafts without interstocks. Transcriptome analysis of HPP and HP citrus revealed that the interstock regulated auxin and ethylene signals, sugar and energy metabolism, and cell wall metabolism. Trend and Venn analyses suggested that genes related to carbohydrate-, energy-, and hormone-metabolic activities were more abundant in HPP plants than in HP plants during different periods. Moreover, weighted gene co-expression network analysis demonstrated that carbohydrates, hormones, cell wall, and transcription factors may be critical for interstock-mediated citrus fruit development and ripening. The contents of ethylene, auxin, cytokinin, transcription factors, starch, sucrose, glucose, fructose, and total sugar in HPP plants differed considerably than those in HP fruits. Interstocks may help to regulate the early ripening and quality of citrus fruit through the above-mentioned pathways. These findings provide information on the effects of interstock on plant growth and development.

Temperature pretreatment is one of the most important factors which significantly affects the postharvest quality of citrus fruit. In this study, late-ripening Valencia orange (citrus sinensis) fruits were used to investigate the effect of short-term treatment at low (6°C), room (20°C), and high (40°C) temperatures on fruit quality. Our results revealed that both low and room-temperature treatments maintained the content of sugars and organic acids, whereas high-temperature treatments elevated the accumulation of sugars but decreased the content of citric acid. In fruit peel (flavedo and albedo), the accumulation of sugars and organic acids responding to temperatures was diverse and mostly different from that in the pulp. Meanwhile, GABA and several amino acids were upregulated under short-term high-temperature treatment but downregulated in response to low-temperature treatment in both peel and pulp. Furthermore, PCA and correlation analysis revealed that the short-term temperature treatments changed the metabolic flow, and GABA was positively correlated with sugars and organic acids. Our study analyzed the metabolic changes of fruit peel and pulp in response to short-term temperature treatments and revealed that GABA may act as a signaling molecular involved in temperature-controlled quality changes.

We present LIMFAST, a seminumerical code for simulating high-redshift galaxy formation and cosmic reionization as revealed by multitracer line intensity mapping (LIM) signals. LIMFAST builds upon and extends the 21cmFAST code widely used for 21 cm cosmology by implementing state-of-the-art models of galaxy formation and evolution. The metagalactic radiation background, including the production of various star formation lines, together with the 21 cm line signal tracing the neutral intergalactic medium (IGM), is self-consistently described by photoionization modeling and stellar population synthesis coupled to the galaxy formation model. We introduce basic structure and functionalities of the code, and demonstrate its validity and capabilities by showing broad agreements between the predicted and observed evolution of cosmic star formation, IGM neutral fraction, and metal enrichment. We also present the LIM signals of 21 cm, Ly α , H α , H β , [O ii ], and [O iii ] lines simulated by LIMFAST, and compare them with results from the literature. We elaborate on how several major aspects of our modeling framework, including models of star formation, chemical enrichment, and photoionization, may impact different LIM observables and thus become testable once applied to observational data. LIMFAST aims at being an efficient and resourceful tool for intensity mapping studies in general, exploring a wide range of scenarios of galaxy evolution and reionization and frequencies over which useful cosmological signals can be measured.

The epoch of reionization (EoR) offers a unique window into the dawn of galaxy formation, through which high-redshift galaxies can be studied by observations of both themselves and their impact on the intergalactic medium. Line intensity mapping (LIM) promises to explore cosmic reionization and its driving sources by measuring intensity fluctuations of emission lines tracing the cosmic gas in varying phases. Using LIMFAST, a novel seminumerical tool designed to self-consistently simulate LIM signals of multiple EoR probes, we investigate how building blocks of galaxy formation and evolution theory, such as feedback-regulated star formation and chemical enrichment, might be studied with multitracer LIM during the EoR. On galaxy scales, we show that the star formation law and the feedback associated with star formation can be indicated by both the shape and redshift evolution of LIM power spectra. For a baseline model of metal production that traces star formation, we find that lines highly sensitive to metallicity are generally better probes of galaxy formation models. On larger scales, we demonstrate that inferring ionized bubble sizes from cross-correlations between tracers of ionized and neutral gas requires a detailed understanding of the astrophysics that shape the line luminosity–halo mass relation. Despite various modeling and observational challenges, wide-area, multitracer LIM surveys will provide important high-redshift tests for the fundamentals of galaxy formation theory, especially the interplay between star formation and feedback by accessing statistically the entire low-mass population of galaxies as ideal laboratories, complementary to upcoming surveys of individual sources by new-generation telescopes.

The interstock, a stock between the rootstock and scion, has a significant regulatory effect on the stock and scion, and its function is highly dependent on graft compatibility. To assess the graft compatibility of the interstock and scion, ‘Yuanxiaochun’ was top grafted onto ‘Ponkan’, ‘Shiranuhi’, ‘Harumi’, ‘Tarocco’, and ‘Kumquat’. The results showed significant differences in the survival ratio and preservation ratio among different combinations. Grafting compatibility in the early stages of grafting was associated with the genetic relationship. The biomass accumulation revealed that the interstock could influence both the rootstock and the scion. The physiological and biochemical traits analysis suggested that SPAD reading and soluble sugar could be employed as preselected indices to evaluate graft compatibility in the late stage of grafting. These results indicated that the evaluation of graft compatibility was a dynamic process. The findings provided a new approach for studying the stock and scion interaction mechanisms mediated by interstock, and directly provided a theoretical and practical basis for the high-grafting of ‘Yuanxiaochun’ citrus.

Cold storage preserves lemon fruit quality; however, it can result in significant chilling injury (CI). The effects of pre- and post-harvest methyl jasmonate (MeJA) treatments at four concentrations (0, 0.1, 0.3, and 0.5 mM) on CI and sensory quality of lemons during 80 d of storage at 7–10 °C were investigated. Both pre- and post-harvest MeJA treatments reduced CI, weight loss (WL) and maintained higher firmness, total soluble solids (TSS), and total acidity (TA) than in the controls. Antioxidant enzyme activities decreased in the control fruit but increased in both pre- and post-harvest MeJA-treated fruit. In addition, phospholipase D (PLD) and lipoxygenase (LOX) activities and malondialdehyde (MDA) content were higher in the control than in the MeJA-treated fruit. Pre-harvest MeJA treatment generally preserved fruit better than post-harvest MeJA treatment, with the best results observed when MeJA was applied at 0.3 mM, which enhanced the antioxidant system of the lemon fruits, thus reducing the post-harvest incidence of chilling injury. These results have important implications for improved fruit quality post-harvest.

Blood orange (Citrus sinensis L. Osbeck) is a rare commercial citrus fruit containing abundant anthocyanins and has numerous health benefits. Blood orange rootstock determines the fruit yield and quality. This study evaluated the effect of the three most commonly used rootstocks on the fruit features, color index, physicochemical parameters, anthocyanin accumulation, the anthocyanin biosynthetic gene expression, and the associated enzymes during the fruit development and ripening of ‘Tarocco’ blood orange. The highest anthocyanin content at harvest was found in blood orange trees grafted onto ‘Trifoliate orange’ (Poncirus trifoliata L. Raf., Pt) rootstock. Molecular analyses revealed that the rootstock affects the anthocyanin accumulation in the blood orange. Additionally, there was a strong correlation between the anthocyanin content and the expression and the activity of related genes and enzymes, respectively. Based on gene expression and enzymatic activity analyses, Pt rootstock promotes a very high anthocyanin accumulation in ‘Tarocco’ blood orange fruit. Accordingly, Pt is the promising rootstock for producing good quality and highly nutritious ‘Tarocco’ blood orange fruit for commercial purposes.

Nitrogen metabolism in citrus has received increased attention due to its effects on plant growth and productivity. However, little is known about the effects of nitrogen fertilization on nitrogen metabolism in young trees of citrus cultivar 'Huangguogan' (Citrus reticulata × Citrus sinensis). Here, genes encoding nitrate reductase (NR), nitrite reductase (NiR), glutamine synthetase (GS), glutamate dehydrogenase (GDH), and asparagine synthetase (AS), represented as HgNR, HgNiR, HgGS, HgGDH, and HgAS, respectively, were cloned from Huangguogan. Deduced protein sequences were analyzed and proteins were confirmed to be localized in their respective cellular organelles. Moreover, pot-cultured 'Huangguogan' seedlings were fertilized with 0 (N1), 1.36 (N2), 1.81 (N3), 2.26 (N4), or 2.72 (N5) kg N/year, for 12 months. Enzyme activity and enzyme-gene expression were studied in roots, leaves, and fruits at different stages. Finally, the effects of N application rate on root activity, leaf N, soluble protein, yield, and fruit quality at the ripening stage were measured. The results showed that: 1) HgNR, HgNiR, HgGDH, and HgAS gene products were found mainly in the cytoplasm and plasma membrane, while HgGS gene product was found mainly in cytoplasm and mitochondria. 2) Gene expression and enzyme activity differed among plant organs. As the root is in permanent direct contact with the soil we suggest that root gene expression and enzyme activity can be used as reference to determine N application rate. 3) Yield, fruit quality, enzyme activity, and enzyme-related gene expression were considerably lower at low than at high-N supply. However, they were all inhibited by excess nitrogen (i.e., 2.72 kg/year). Therefore, we recommend 1.81 kg N/year as the optimal N application rate for young 'Huangguogan' trees.

Considering the known effects of xyloglucan endotransglycosylase (XET) on plant growth and development, we aimed to determine whether XETs help to regulate the growth and elongation of Huangguogan shoots and roots. We confirmed a possible role for XET during seedling etiolation. Our results revealed that the roots of etiolated seedlings (H-E) were longer than those of green seedlings (H-G). However, shoot length exhibited the opposite pattern. We also observed positive and negative effects on the xyloglucan-degrading activity of XET in the root sub-apical region and shoots of etiolated Huangguogan seedling, respectively. There was a significant down-regulation in CitXET expression in the etiolated shoots at 15 days after seed germination. On the contrary, it was significantly increased in the root sub-apical region of etiolated and multicolored seedlings at 15 days after seed germination. The XET coding sequence (i.e., CitXET) was cloned from Huangguogan seedlings using gene-specific primers. The encoded amino acid sequence was predicted by using bioinformatics-based methods. The 990-bp CitXET gene was highly homologous to other XET genes. The CitXET protein was predicted to contain 319 amino acids, with a molecular mass of 37.45 kDa and an isoelectric point of 9.05. The predicted molecular formula was C1724H2548N448O466S14, and the resulting protein included only one transmembrane structure. The CitXET secondary structure consisted of four main structures (i.e., 21% α-helix, 30.72% extended strand, 9.09% β-turn, and 39.18% random coil). Analyses involving the NCBI Conserved Domains Database (NCBI-CDD), InterPro, and ScanProsite revealed that CitXET was a member of the glycosyl hydrolase family 16 (GH16), and included the DEIDFEFLG motif. Our results indicate that the differed degrees of etiolation influenced the CitXET expression pattern and XET activity in Huangguogan seedlings. The differential changes in XET activity and CitXET expression levels in Huangguogan seedlings may influence the regulation of root and shoot development, and may be important for seedling etiolation.
