1. Micro/nano topological modification of TiO2 nanotubes activates Thy-1 signaling to control osteogenic differentiation of stem cells

Abstract

Micro/nano topological modification is critical for improving the in vivo behaviors of bone implants, regulating multiple cellular functions. Titania (TiO2) nanotubes show the capacity of promoting osteoblast-related cell differentiation and induce effective osseointegration, serving as a model material for studying the effects of micro/nano-topological modifications on cells. However, the intracellular signaling pathways by which TiO2 nanotubes regulate the osteogenic differentiation of stem cells are not fully defined. Thy-1 (CD90), a cell surface glycoprotein anchored by glycosylphosphatidylinositol, has been considered a key molecule in osteoblast differentiation in recent years. Nevertheless, whether the micro/nano topology of the implant surface leads to changes in Thy-1 is unknown, as well as whether these changes promote osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs). Here, TiO2 nanotubes of various diameters were prepared by adjusting the anodizing voltage. qPCR and immunoblot were carried out to assess the mechanism by which TiO2 nanotubes regulate Thy-1. The results revealed Ti plates harboring TiO2 nanotubes ∼100-nm diameter (TNT-100) markedly upregulated Thy-1. Subsequently, upregulated Thy-1 promoted the activation of Fyn/RhoA/MLC Ⅱ/F-actin axis, which enhanced the nuclear translocation of YAP. After Thy-1 knockdown by siRNA, the Fyn/RhoA/MLC Ⅱ/F-actin axis was significantly inhibited and TiO2 nanotubes showed decreased effects on osteogenic differentiation. Therefore, Thy-1 upregulation might be a major mechanism by which micro/nano-topological modification of TiO2 nanotubes promotes osteogenic differentiation in BMSCs. This study provides novel insights into the molecular mechanism of TiO2 nanotubes, which may help design improved bone implants for clinical application.

Prealbumin, a nutritional and inflammatory indicator, is a well-established predictor of short- and long-term outcomes in numerous clinical conditions. The current study investigated the association of pre-procedural prealbumin levels with CI-AKI and long-term outcomes in geriatric patients after elective percutaneous coronary intervention (PCI).Patients and methods: A total of 558 patients aged≥75 years, who underwent elective PCI between January 2012 and December 2015, were selected for the current study. Pre-procedural prealbumin levels were measured before PCI. Multivariable logistic regression and Cox proportional hazard regression analyses were performed to identify the independent risk factors for CI-AKI and long-term mortality.Results: Out of 558 patients, 54 developed CI-AKI. The optimal cutoff value of prealbumin for detecting CI-AKI was 185.5 mg/L with 62.7% sensitivity and 70.4% specificity based on the receiver operating characteristic analysis (C-statistic=0.710; 95% confidence interval [CI] 0.673–0.751). Multivariable analysis demonstrated that prealbumin≤185.5 mg/L was significantly associated with CI-AKI (odds ratio [OR] 0.397; 95% CI 0.195–0.808; P=0.011). Abstract

A novel hydrogel, consisting of titanium dioxide nanotubes and graphene oxide (H-TN-GO), was fabricated to remove ciprofloxacin (CIP) from aqueous solution. The adsorbent was characterized by scanning electron microscope (SEM), transmission electron microscope (TEM), X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR) and Raman spectroscopy. Batch adsorption experiments systematically investigated the effect of contact time, temperature and pH on the capacity of H-TN-GO for CIP. Adsorption property of H-TN-GO and its regeneration performance was evaluated. H-TN-GO exhibited high adsorption capacity of 530 mg/g and Langmuir model was more suitable for fitting adsorption behavior. The removal ratio of CIP was more than 99% when initial concentrations were less than 100 mg/L. Besides, the initial pH of CIP solution had a distinct influence on the removal ratio, and adsorption capacity of adsorbance could be recovered by regeneration with mild alkaline condition. Large pore size and abundant oxygen-containing groups on the surface further improved its adsorption performance. The enhancing effect of H-TN-GO mainly attributed to the immobilization of TiO2 onto graphene oxide and π-π interactions stacking between H-TN-GO and graphene oxide. Abstract

Short peptide surfactants (pepfactants) bearing the amphiphilic molecular architecture of hydrophobic and hydrophilic moieties are attractive for a wide range of biological and medical applications. Understanding of structural basis and molecular mechanism underlying the self-assembling behaviour of pepfactants is fundamentally important for rationally designing surfactant-like peptides to function as diverse biomaterials. To date, however, the relationship between the physicochemical properties and self-assembly of pepfactants still remains largely unexplored. In this study, we attempt to elucidate the role of structural flexibility and dynamics in peptide self-assembly. Two fast and reliable quantitative structure–property relationship predictors are carefully developed with the sophisticated genetic algorithm/partial least squares method; the predictors are then applied to estimate molecular flexibility and self-assembling ability for 10,000 randomly generated surfactant-like peptides. As a result, a significant negative correlation between the flexibility and self-assembly is observed, which can be fitted fairly well using an exponential curve. Furthermore, atomistic molecular dynamics simulations also reveal a noticeable difference of flexibility profile between strong and weak self-assembling peptides.
