Abstract

Summary: CD8 T cells are crucial adaptive immune cells with cytotoxicity to fight against pathogens or abnormal self-cells via major histocompatibility complex class I-dependent priming pathways. The composition of the memory CD8 T-cell pool is influenced by various factors. Physiological aging, chronic viral infection, and autoimmune diseases promote the accumulation of CD8 T cells with highly differentiated memory phenotypes. Accumulating studies have shown that some of these memory CD8 T cells also exhibit innate-like cytotoxicity and upregulate the expression of receptors associated with natural killer (NK) cells. Further analysis shows that these NK-like CD8 T cells have transcriptional profiles of both NK and CD8 T cells, suggesting the transformation of CD8 T cells into NK cells. However, the specific induction mechanism underlying NK-like transformation and the implications of this process for CD8 T cells are still unclear. This review aimed to deduce the possible differentiation model of NK-like CD8 T cells, summarize the functions of major NK-cell receptors expressed on these cells, and provide a new perspective for exploring the role of these CD8 T cells in health and disease. Abstract

BACKGROUND. Despite an increasing appreciation of the roles that myeloid cells play in tumor progression and therapy, challenges remain in interpreting the tumor-associated myeloid response balance and its translational value. We aimed to construct a simple and reliable myeloid signature for hepatocellular carcinoma (HCC). METHODS. Using in situ immunohistochemistry, we assessed the distribution of major myeloid subtypes in both peri- and intratumoral regions of HCC. A 2-feature-based, myeloid-specific prognostic signature, named the myeloid response score (MRS), was constructed using an L1-penalized Cox regression model based on data from a training subset (n = 244), a test subset (n = 244), and an independent internal (n = 341) and 2 external (n = 94; n = 254) cohorts. RESULTS. The MRS and the MRS-based nomograms displayed remarkable discriminatory power, accuracy, and clinical usefulness for predicting recurrence and patient survival, superior to current staging algorithms. Moreover, an increase in MRS was associated with a shift in the myeloid response balance from antitumor to protumor activities, accompanied by enhanced [CD8.sup.+] T cell exhaustion patterns. Additionally, we provide evidence that the MRS was associated with the efficacy of sorafenib treatment for recurrent HCC. CONCLUSION. We identified and validated a simple myeloid signature for HCC that showed remarkable prognostic potential and may serve as a basis for the stratification of HCC immune subtypes. FUNDING. Abstract

Tumor biomarkers are frequently used in clinical practice for disease screening, disease monitoring, and prognostic diagnosis. An increasing number of studies are now focusing on the impact of tumor biomarkers on the immune system. For example, the tumor-derived AFP has immunosuppressive effects on various cell types, including natural killer (NK) cells, dendritic cells (DCs) and T cells. CEACAM1, CEACAM5 and CEACAM6 play important roles in gastrointestinal tumors. CEACAM5 can facilitate antigen presentation to T cells via CD1d and can transmit inhibitory signals to NK cells through CEACAM1. CEACAM6 also plays a crucial role in the metastatic process of tumors. As the only member of the CEACAM family expressed on activated T cells, CEACAM1 has opposing effects depending on its subtypes (CEACAM1-L/CEACAM1-S) . CA125, also known as MUC16, can inhibit tumor immunity by binding to inhibitory receptors on immune cells such as NK cells and macrophages. On the other hand, MUC16 can promote DCs maturation. Abstract

The expandability of high demands for multimedia applications brings out more and more video standards for improving the coding and compression efficiency. As the most commonly used transform, Discrete cosine transform (DCT) achieves excellent energy compaction property and good compression efficiency. Hardware sharing is the mostly used efficient strategy to reduce the cost for video codec. Based on traditional matrix factorization, this paper makes three observations to direct the design of proposed hardware sharing architecture. The proposed architecture can be generally used to compute 8×8 DCT of AVS, H.264, VC-1 and HEVC in a low cost way, and can be used to decode Full-HD and WQXGA formate video sequences in real time. The design has been synthesized in 0.13μm technology. The synthesis results show that the proposed architecture achieves 76.9% reduction in gate count, 85.6% decrease in power consumption and 35% improvement in operational speed in comparison with other existing designs.

Abstract

Reconfigurable digital filter is being widely used in applications such as communication and signal processing. Its performance, power consumption, and logic resource utilization are the major factors to be taken into consideration when designing the filters. This paper proposes a concise canonic signed digit coefficient grouping method aiming at reducing the number of common subexpressions (CSs). Further, we statistically analyze every CS occurance for numerous sorts of the finite-impulse response (FIR) filters and obtain characterization of the distribution behavior for all the possible CS patterns in a 16-bit coefficient. Thus, a novel processing element structure is proposed to form a medium-grain array for computationally efficient realization of reconfigurable FIR filter. The experiment results suggest such design implementations typically achieve 21% reduction in silicon area, 20% decrease in power consumption, and 14% improvement in operation speed in comparison to other conventional FIR architectures.

Abstract

FPGA-based SoCs have been gaining great favors among traditional applications and expanding the application domains to some new areas. Reusing and sharing components is an attractive and pratical methodology for system designers to reduce the design complexity of the SoC architectures. Open source hardware has become an effective method of improving the design productivity. As the core functional component, processors affects the performance of SoC systems. This paper investigates existing open source processors, and gives an overview. From the points of usability and stability, the main features of open source processors are summarized. Following these features, some open source processors with high usability and stability are selected. These open source processors and existing vendor-provided soft processors are implemented on Stratix V and Virtex-7 FPGAs using corresponding EDA tools, Quart us n and ISE. The implementation results are compared and discussed.

Abstract

The paper presents a suitable functional in the form of a convolution of the magnetic vector potential in the space-time domain for transient nonlinear eddy problem. And then, appropriate element equations have been derived following the usual finite element variational procedure. Furthermore, the element equations are improved. Finally, the improved space-time finite element method (STFEM) has been used to compute the transient nonlinear electromagnetic field and performances of a permanent magnet synchronous motor (PMSM). Numerical illustrations have indicated that the improved space-time finite element method is a valid method to simulate performances of PMSM.
