Background and purpose: Long non-coding RNA small nucleolar RNA host gene 5 (lncRNA SNHG5) plays a cancer-promoting role in many cancers, however its effect on colorectal cancer (CRC) and its regulatory mechanism are not clear. This study aimed to explore the mechanism of lncRNA SNHG5/miR-26a-5p/metadherin (MTDH) signal axis promoting metastasis of CRC. Methods: The data of The Cancer Genome Atlas (TCGA) database was analyzed, the abnormal expression of lncRNA in CRC was explored and analyzed the survival. Samples of CRC, paracancerous tissues and complete clinical data of patients who underwent surgical resection from October 2012 to October 2016 were collected. The expression levels of SNHG5 and miR-26a-5p in lncRNA were detected by real-time fluorescence quantitative polymerase chain reaction (RTFQ-PCR), and the expression level of MTDH was detected by immunohistochemistry. The relationship between the relative expression level of lncRNA SNHG5 in CRC and clinicopathological features and survival time was analyzed. The effects of lncRNA SNHG5 on the proliferation, migration and invasion of CRC cells were detected by cell counting kit-8 (CCK-8), clone formation, scratching assays, transwell test and in vivo xenotransplantation. The relationship between CRC cell metastasis, the expression level of epithelial-mesenchymal transition related molecules and lncRNA SNHG5 expression level by Western blot and immunohistochemical detection were explored. The physical interaction between SNHG5 and miR-26a-5p, MTDH and miR-26a-5p was studied by RNA pull-down test, double luciferase reporter gene detection and RNA co-immunoprecipitation. The functional relationship among the three was verified by CCK-8, EdU and transwell experiments. The effect of SNHG5, miR-26a-5p and MTDH expression on migration and invasion related molecules was analyzed by Western blot. Results: The results of TCGA database analysis showed that lncRNA SNHG5 was significantly upregulated in CRC. The drag torque produced by the viscous shearing action of lubricating oil is an important component of the power loss of wet clutches. Studying the prediction model and its influencing factors will provide important theoretical support for reducing the drag torque. Taking a single friction pair wet clutch as the research object, this research obtained the oil film shrinkage under different working conditions based on the VOF model. On this basis, a drag torque calculation model considering the surface tension and shrinkage of oil film is established, in which the drag torque is composed of the torque transmitted by oil film in the oil film continuous zone and the torque transmitted by oil-liquid and gas-liquid in the oil film rupture area, respectively. Comparing the theoretical drag torque with the simulation and experimental results, the variation law of drag torque with the speed difference under different oil film thicknesses and different supply flow is obtained, and the validity of the theoretical model is also verified. Results show that increasing the oil film thickness and decreasing the supply flow will lead to a decrease in drag torque, which provides a useful reference for reducing the drag torque. Objectives Polymorphism in a coding region of deoxyribonuclease I-like III (DNASE1L3), causing amino acid substitution of Arg-206 to Cys (R206C), is a robustly replicated heritable risk factor for SSc and other autoimmune diseases. DNASE1L3 is secreted into the circulation, where it can digest genomic DNA (gDNA) in apoptosis-derived membrane vesicles (AdMVs). We sought to determine the impact of DNASE1L3 R206C on digestion of circulating gDNA in SSc patients and healthy controls (HCs). Methods The ability of DNASE1L3 to digest AdMV-associated gDNA was tested in vitro. The effect of R206C substitution on extracellular secretion of DNASE1L3 was determined using a transfected cell line and primary monocyte-derived dendritic cells from SSc patients. Plasma samples from SSc patients and HCs with DNASE1L3 R206C or R206 wild type were compared for their ability to digest AdMV-associated gDNA. The digestion status of endogenous gDNA in plasma samples from 123 SSc patients and 74 HCs was determined by measuring the proportion of relatively long to short gDNA fragments. Results The unique ability of DNASE1L3 to digest AdMV-associated gDNA was confirmed. Extracellular secretion of DNASE1L3 R206C was impaired. Plasma from individuals with DNASE1L3 R206C had reduced ability to digest AdMV-associated gDNA. The ratio of long: short gDNA fragments was increased in plasma from SSc patients with DNASE1L3 R206C, and this ratio correlated inversely with DNase activity. Conclusion Our results confirm that circulating gDNA is a physiological DNASE1L3 substrate and show that its digestion is reduced in SSc patients with the DNASE1L3 R206C variant. In order to improve the properties of laser overlap welded T-joint, laser-additional current hybrid welding process is put forward. In this paper, the welding characteristics of laser-additional current hybrid welded aluminum and titanium alloy T-joint were conducted and compared. The weld width at faying surface increase, which results in tensile shear load increasing compared with those of laser welding for both aluminum and titanium alloy, but the effect of current on aluminum alloy is more obvious. The porosity defect within the laser-additional current hybrid welded joint sharply reduces compared with that within laser welding. The tensile shear load of aluminum alloy and titanium alloy hybrid welded joints respectively increase 21% and 15%. The effects of additional current on welding characteristics of aluminum alloy and titanium alloy are compared and analyzed.
