Multiple organs are affected by the complex autoimmune illness known as systemic sclerosis (SSc), which has a high fatality rate. Genes linked to autophagy have been linked to the aetiology of SSc. It is yet unknown, though, whether autophagy-related genes play a role in the aetiology of SSc. After using bioinformatics techniques to examine two databases (the GSE76885 and GSE95065 datasets) and autophagy-related genes, we were able to identify 12 autophagy-related differentially expressed genes that are linked to the pathophysiology of SSc. Additional examination of the receiver operating characteristic curve revealed that SFRP4 (AUC = 0.944, P < 0.001) and CD93 (AUC = 0.904, P < 0.001) might be utilized as trustworthy biomarkers for the diagnosis of SSc. The SSc group’s considerably greater CD93 and SFRP4 expression levels compared to the control group were further confirmed by qRT-PCR results. The autophagy-related genes SFRP4 and CD93 were found to be viable diagnostic indicators in this investigation. Our research sheds light on the processes by which genes linked to autophagy affect the pathophysiology of SSc.

The dynamic characteristics of buildings are the basis for studying the vibration of buildings under wind loads, while the innovative design of wind-resistant structures is an important guarantee for the safety and comfort of buildings under wind loads. In this paper, we calculate the building’s dynamic characteristics by establishing the vibration equations of the structure. We then convert the wind load into an equivalent static load and establish the necessary conditions to construct an optimized wind-resistant structural design model. The finite element modeling method is used to generate the building model of this paper and conduct research on its dynamic characteristics and wind-resistant design. In this paper, the model in the vicinity of 0.6Hz and the wind load along the parallel direction of the building, x direction appears to reach the peak amplitude response of 4.22cm. The pressure coefficients at each measurement point of the building are found to increase gradually with the increase of Re and reach the critical condition at Re = 1.02×106 under 0° wind angle. Through the implementation of simulated wind tunnel tests on the building, this paper provides a preliminary assessment of the existing wind design methods and highlights the problems in these methods. Using the comparative analysis method, it can be obtained that the optimized wind-resistant structure model of the building designed in this paper reduces the base shear force in the crosswind direction by 3.73% and the base shear force in the downwind direction by 5.53%. At the same time, the inter-story displacement angle of the building is significantly reduced after optimizing the wind-resistant design.

The compression deformation test on extruded-solid solution 7A43 alloy (Al-6.0Zn-2.1Mg-0.15Cu-0.15Zr, mass fraction/%) was conducted by Gleeble-3500 machine with strain rate of 0.001 s-1 to 1 s-1 at room temperature, and the microstructural features were characterized by scanning electron microscopy (SEM), backscattered electron microscopy (EBSD) and X-ray differaction analysis (XRD). It is revealed that with the increase of strain rate, the average grain size decreases due to severe microstructural distortion, whilst the lattice strain and dislocation density in the alloy gradually increase. Dynamic recovery substructure components in the microstructure are enhanced, and the coarse deformed grains are replaced by fine equiaxed grains under high deformation rate conditions. A Fields-Backofen (F-B) constitutive model was constructed based on the experimental stress-strain data, the correlation coefficient (R) and average absolute relative error (AARE) between the experimental and predicted values are 0.991069 and 3.667%, respectively, which provide a good description of the model for deformation rheological behavior of 7A43 aluminum alloy at room temperature.

As the depth increases, the ratio of horizontal to vertical stress on the coal rock strata will gradually decrease, which affects the crack propagation behavior around hydro-fracture injection hole. In order to explore the characteristics of hydro-fracture crack turning behavior at different depths, based on the mechanical model and adsorption and desorption model of coal seam, COMSOL was used to analyze the stress distribution and crack initiation pressure around the injection hole at different depths. The results show that at shallow depths, the ratio of horizontal stress to vertical stress is greater than 1, the cracks extend mainly horizontally, and the fracture is more likely to turn when the stress ratio increases. At a deeper depth, the ratio of horizontal stress to vertical stress is less than 1, the cracks extend mainly vertically, and the fracture is more likely to turn when the stress ratio decreases.

Found in as many as 80% of women, uterine leiomyomas are a frequent cause of abnormal uterine bleeding, pelvic pain, and infertility. Despite their significant clinical impact, the mechanisms responsible for driving leiomyoma growth remain poorly understood. After obtaining IRB permission, expression of ecto‐5′‐nucleotidase (NT5E, CD73) was assessed in matched specimens of myometrium and leiomyoma by real‐time qPCR, Western blot, and immunohistochemistry (IHC). Adenosine concentrations were measured by enzyme‐linked assay. Primary cultures were used to assess the impact of adenosine and/or adenosine receptor agonists on proliferation, apoptosis, and patterns of intracellular signaling in vitro. When compared to matched specimens of healthy myometrium, uterine leiomyomas were characterized by reduced CD73 expression. Largely limited to thin‐walled vascular structures and the pseudocapsule of leiomyomas despite diffuse myometrial distribution. Restricted intra‐tumoral CD73 expression was accompanied by decreased levels of intra‐tumoral adenosine. In vitro, incubation of primary leiomyoma cultures with adenosine or its hydrolysis‐resistant analog 2‐chloro‐adenosine (2‐CL‐AD) inhibited proliferation, induced apoptosis, and reduced proportion of myocytes in S‐ and G2‐M phases of the cell cycle. Decreased proliferation was accompanied by reduced expression of phospho‐Akt, phospho‐Cdk2‐Tyr15, and phospho‐Histone H3. Enforced expression of the A2B adenosine receptor (ADORA2B) and ADORA2B‐selective agonists similarly suppressed proliferation and inhibited Akt phosphorylation. Collectively, these observations broadly implicate CD73 and reduced extracellular concentrations of adenosine as key regulators of leiomyoma growth and potentially identify novel strategies for clinically managing these common tumors. Over the past forty years of reform and opening up, the urban center system in the Pearl River Delta Region has been rapidly restructured. Its two central cities, Guangzhou and Shenzhen, have been fiercely competing in the field of economic development. Dealing with the functional positioning of the two cities has constituted a long-term concern for regional planning and policy making. Under the new circumstances of the Guangdong-Hong Kong-Macao Greater Bay Area, there is an urgent need to clarify the functional positioning of the two central cities of Guangzhou and Shenzhen. Further, there is a need to coordinate the relationship between competition and cooperation, the premise of which lies in the dynamic and accurate identification of urban functional characteristics. Previous studies directly used economic and industrial statistical data to study and analyze urban functions. However, due to the relative low frequency and high cost of data collection and analysis, it is difficult to carry out real-time analysis using these data. Based on the theoretical understanding of the projection of city functions to the city network, this article uses inter-city air passenger flow as its research medium. Further, it uses the air passenger flow data extracted from Baidu location-based services (LBS) big data. Taking the Chinese city network as its overall scope, it adopts the following two approaches. First, it compares the functional characteristics of the cities where the air passenger flows toward Guangzhou and Shenzhen are sourced. Second, it compares the functional characteristics of the employment places in the two megacities of Beijing and Shanghai, where the air passenger flows toward Guangzhou and Shenzhen are sourced. Therefore, the functional attributes and the functional areas of the source cities are used as the basis for identifying the comparative functional features of Guangzhou and Shenzhen to judge their overall functional characteristics in the Chinese urban network. The research results show the following. 1) First, compared with Shenzhen, Guangzhou's network connection with cities across China and Beijing and Shanghai focuses on administrative management and business functions. It plays the role of an administrative and business center within the spatial scale of the Pearl River Delta urban agglomeration. 2) Second, compared with Guangzhou, Shenzhen focuses on technological innovation, finance, and high-tech manufacturing functions, and plays the role of a technological innovation and financial center and high-tech manufacturing base under the scale of the Pearl River Delta. 3) Third, as the two major service industry centers in the Pearl River Delta region, Guangzhou focuses on public and general producer services, and Shenzhen focuses on advanced producer services such as finance and technological research and development. Shenzhen has a higher status within the Chinese urban network system based on the producer service industry. The method provided by this article, which involves locational big data to flexibly extract urban functional characteristics from the city network, is novel. Further, it can serve as a supplementary approach to the studies on urban functions which relied on traditional economic and industrial statistics. Meanwhile, the research results of this article provide evidence for deepening the theoretical understanding of the urban function-space-network mapping relationship.
