It was the aim of this study to examine whether the usage of the vaccine COXEVAC ® (Ceva Santé Animale) could reduce the consumption of antibiotics in Q-fever-positive dairy farms. Additionally, the effects of other herd-level factors on the consumption of antibiotics were investigated. A total of 36 farms with vaccination and 13 farms without vaccination participated in this longitudinal cohort study. In all herds, Coxiella burnetii had been directly or indirectly diagnosed. To compare the treatment frequency of antibiotics between the vaccinated group and the non-vaccinated group, the consumption of antibiotics for each farm was collected using the veterinary documents about the application and delivery of antibiotics. To gather detailed information about herd data, nutrition, milking management, housing, and animal health, the farmers were interviewed with the help of a questionnaire. The results thereof suggest that there might be an association between the vaccination against Q fever and a reduced consumption of antibiotics. Neither herd size nor milk yield level influenced the consumption of antibiotics in the study herds. Type of flooring and udder-cluster disinfection while milking were associated with a lower and higher therapy frequency, respectively. Further studies are necessary to elucidate the cause-effect relationship between vaccination and the consumption of antibiotics.

The aim of this study was to investigate the influence of calving-related injuries of the vestibulum vaginae and the vagina on fertility measures in heifers. German Holstein heifers ( n = 236) were checked for vestibulum vaginae and vaginal injuries. These were scored according to localization, depth and length. The healing process was assessed until day 42 post partum. Calving ease and the occurrence of metritis and endometritis were evaluated. In 160 heifers, the following fertility measures were calculated to assess the reproductive performance of heifers: mean interval from calving to first insemination, mean days open, mean interval from first insemination to conception, mean calving interval, mean pregnancy index, percentage of animals pregnant at 200 days p.p., and first service conception rate. On the one hand, dystocia was a risk factor for injuries of the soft birth canal, and, on the other hand, those injuries were a risk factor for metritis and endometritis. In this study, calving-related injuries of the vestibulum vaginae and the vagina had no statistically significant effect on the reproductive performance of heifers. One reason for this outcome was probably the overall good healing tendencies of those injuries in heifers.

Introduction: The aim of this study was to record the course of peripartal udder oedema with ultrasonography in dairy cows and to investigate the therapeutic effects of diuretic furosemide. For this purpose, a device with a force sensor for the ultrasound probe was developed, which ensured the generation of data under similar pressure conditions and thereby repeatable and comparable results. In a pretrial with ten cows, ultrasonographic measurements were performed daily at four locations per udder quarter beginning 14 days ante partum until 14 d post partum. The main study included 50 dairy cows. The experimental group (n=25) received 500 mg furosemide intramuscularly on the day of calving, as well as on the first and second day post partum. The control group (n=25) was treated with 10 mL 0,9 % sodium chloride solution (NaCl) at the same timepoints. The experimental period was extended from 21 days ante partum until 21 days post partum and included 15 ultrasonographic measurements at three-day intervals. The measurements were performed at the teat base, which turned out to be the most suitable location in the pretrial. Quarter milk samples were collected on the day of calving, 7 and 14 days post partum. The average thickness of the udder oedema between the group treated with furosemide and the control group did not differ. In conclusion, a method for the ultrasonographic measurement of udder oedema under comparable applied forces was established. The teat base turned out to be a suitable location to monitor the characteristic temporal course of udder oedema. In the present study, treatment with furosemide did not have a measurable, positive effect on the severity of the udder oedema post partum in the study animals. Finally, further studies with a higher sample size are necessary to confirm this relationship.

In order to increase fertility in modern dairy farming, the interval between first insemination and conception should be as short as possible. Therefore, one approach is to diagnose non pregnant animals early to re-inseminate them as soon as possible. Commercial milk pregnancy assays are available to detect throphoblast derived bovine pregnancy-associated glycoproteins (bPAG) in milk. The aim of the present study was first to evaluate pre-analytical factors interfering with the correct detection of bPAG in milk. To achieve this aim, the stability of the bPAG was tested after repeated freezing and thawing cycles, as well as after a storage of seven days at room temperature and at 37°C. Secondly, the diagnostic performance of a commercially available PAG-ELISA was evaluated for pregnancy diagnosis between 28 to 60 days after artificial insemination by comparing the results with transrectal ultrasonography in a field study (n = 291 cows). After one freezing and thawing procedure the optical density (OD) increased and afterwards stayed stable. The storage of milk samples at room temperature had no effect on the OD, but after five days of storage at 37 °C the OD dropped sharply. It is therefore recommended to add an appropriate microbicidal preservative for shipment and storage of milk samples for bPAG analysis . The results of the field study showed that PAG-ELISA results and ultrasonography results agreed in 95.7% of the cases. Nine samples were tested as "false negative"and five as "false positive", however a re-check revealed that all "false positive"and "false negative" tested cows suffered from embryonic/fetal mortality. In conclusion, the pregnancy diagnostic by bPAG detection in milk samples is an accurate method to diagnose early pregnancy in lactating cows.

It was the aim of this study to record common diseases in dairy cows in Schleswig-Holstein, Germany, and to describe associations between lactation number (LN: 1, 2, > 2), first test-day milk yield (TD1: < 30 kg, ≥ 30 kg) and herd milk yield (HM: ≤ 7500 kg, > 7500 kg), the latter parameter serving as a proxy for herd management (extensive vs. intensive). Data of 98 dairy herds (6439 lactations) were processed on cow level using mixed logistic regression models with LN< TD1, HM and calving season as fixed effects and herd as random effect. Lactational incidences were as follows: hypocalcaemia (5.0%), dystocia (13.2%), retained placental membranes (7.2%), clinical metritis/endometritis (4.9%), clinical mastitis (15.3%), subclinical mastitis (61.9%), ketosis (1.6%), displaced abomasum (0.4%), lameness (15.4%). Number of lactation (2, > 2 vs. 1) was a risk factor for hypocalcaemia (OR 3.715, 23.047), retained placental membranes (OR 1.764, 2.479), clinical mastitis (> 2 vs. 1 OR 2.118), subclinical mastitis (OR 1.668,4.397), ketosis (> 2 vs. 1 OR 3.936) and lameness (OR 1.275, 2.070). Older cows had a lower risk for dystocia (OR 0.373, 0.357). TD1 (≥ 30 kg) was not a risk factor of disease except for subclinical mastitis in first parity animals (OR 1.319). Herd milk yield (> 7500 kg) was a risk factor for clinical metritis/endometritis (OR 1.971), displaced abomasum (OR 7.764), lameness (OR 1.618) and hypocalcaemia (cows with high TD1 [OR 2.273]). In conclusion, not individual milk yield, but herd milk yield as an indicator of differences in intensity of herd management as well as number of lactation seemed to influence the frequency of common diseases in dairy cows.

Ante- and postpartum antioxidative/oxidative profiles in blood plasma of cows with and without retained placental membranes (RFM) were investigated. Twenty-two healthy pregnant cows were included in the study. Seven animals out of 22 suffered from RFM. Blood samples were obtained at 4, 3, 2, 1 weeks and 5 days antepartum (a.p.), at parturition and 1, 3, and 5 weeks postpartum. The following antioxidative parameters were measured using spectrophotometric methods: total antioxidant activity (TAC), β-carotene, vitamin A, vitamin C, and ceruloplasmin. The oxidative profile was based on the content of intermediates and end products of lipid and protein peroxidative processes which were measured by spectrophotometric and spectrofluorimetric methods. The examined parameters revealed a dynamic profile within the experimental period. The highest antioxidant and oxidant activity was noted at 2 and 1 week a.p. with a drop towards parturition suggesting the presence of oxidative stress during this time period and an apparent appropriate metabolic response of the macroorganism. Except for TAC and vitamin A, the contents of oxidative and antioxidative blood constituents did not differ between cows with and without RFM. A TAC and vitamin A by time interaction indicated higher antepartal concentrations of TAC and vitamin A in cows without RFM than in cows with RFM suggesting a possible role of antioxidative/oxidative imbalances in the aetiology of RFM., (Copyright © 2010 Elsevier Inc.
