1. Surface contamination rates at different spatial scales in the Columbus module (ISS) during the MATISS campaigns

Abstract

Future long-duration human spaceflights require developments to limit biocontamination of surface habitats. The three MATISS (Microbial Aerosol Tethering on Innovative Surfaces in the International Space Station) campaigns exposed surface treatments over several months in the ISS. To this end, eight sample holders designed were mounted with lamella-bearing FDTS ((1H, 1H, 2H, 2H)-perfluorodecyltrichlorosilane), SiOCH, and parylene hydrophobic coatings, at two different locations, for several months, during three distinct periods from 2016 to 2020. Tile scanning optical microscopy (×3 and ×30 magnifications) detected several thousand particles, indicating a relatively clean environment (a few particles per mm2). In previous studies, exposure rates were analyzed for all the coarse and fine particles detected on the largest total area of the integrated FDTS area exposed in the ISS (several cm2). Here, the contamination rates observed for a smaller constant area unit (the 0.66-cm2 window area of the holder) were statistically analyzed. Therefore, a statistical difference in rate distributions between RGSH (Return Grid Sensor House) and EDR (European Drawer Rack) and between FDTS and either SiOCH or parylene was shown for the coarse particles but not for the fine particles. The contamination rates were found to be low, confirming the efficiency of the long-term air purification system. The rates tend to vary with the astronaut occupancy rates. Surfaces of spacecraft for long-duration exploration left unmanned during dormancy periods can be considered safe from biocontamination.

Abstract

Abstract Future long-duration human spaceflight will require developments to limit biocontamination of surface habitats. The MATISS (Microbial Aerosol Tethering on Innovative Surfaces in the international Space Station) experiments allowed for exposing surface treatments in the ISS (International Space Station) using a sample-holder developed to this end. Three campaigns of FDTS (perFluoroDecylTrichloroSilane) surface exposures were performed over monthly durations during distinct periods. Tile scanning optical microscopy (×3 and ×30 magnifications) showed a relatively clean environment with a few particles on the surface (0.8 to 7 particles per mm2). The varied densities and shapes in the coarse area fraction (50–1500 µm2) indicated different sources of contamination in the long term, while the bacteriomorph shapes of the fine area fraction (0.5–15 µm2) were consistent with microbial contamination. The surface contamination rates correlate to astronauts’ occupancy rates on board. Asymmetric particles density profiles formed throughout time along the air-flow. The higher density values were located near the flow entry for the coarse particles, while the opposite was the case for the fine particles, probably indicating the hydrophobic interaction of particles with the FDTS surface.

Abstract

Despite decades of research on the reduction of surface fouling from biomolecules or micro-organisms, the ultimate antibiofouling surface remains undiscovered. The recent covid-19 pandemic strengthened the crucial need for such treatments. Among the numerous approaches that are able to provide surfaces with antibiofouling properties, chemical, biological, and topographical strategies have been implemented for instance in the marine, medical, or food industries. However, many of these methods have a biocidal effect and, with antibioresistance and biocide resistance a growing threat on humanity, strategies based on reducing adsorption of biomolecules and micro-organism are necessary for long-term solutions. Bioinspired strategies, combining both surface chemistry and topography, are currently at the heart of the best innovative and sustainable solutions. The synergistic effect of micro/nanostructuration, together with engineered chemical or biological functionalization is believed to contribute to the development of antibiofouling surfaces. This review aims to present approaches combining hydrophobic or hydrophilic chemistries with a specific topography to avoid biofouling in various industrial environments and healthcare facilities.

Abstract

In recent years, many biocatalytic processes have been developed for the production of chemicals and pharmaceuticals. In this context, enzyme immobilization methods have attracted attention for their advantages, such as continuous production and increased stability. Here, enzyme immobilization methods and a collection of nitrilases from biodiversity for the conversion of 3-cyanopyridine to nicotinic acid were screened. Substrate conversion over 10 conversion cycles was monitored to optimize the process. The best immobilization conditions were found with cross-linking using glutaraldehyde to modify the PMMA beads. This method showed good activity over 10 cycles in a batch reactor at 30 and 40°C. Finally, production with a new thermostable nitrilase was examined in a continuous packed bed reactor, showing very high stability of the biocatalytic process at a flow rate of 0.12 ml min-1 and a temperature of 50°C. The complete conversion of 3-cyanopyridine was obtained over 30 days of operation. Future steps will concern reactor scale-up to increase the production rate with reasonable pressure drops.

Abstract

International audience; In this work, we present an innovative strategy for the grafting of an antibacterial agent onto nanocellulose materials in supercritical carbon dioxide (scCO2). Dense cellulose nanofibril (CNF) nanopapers were prepared and subsequently functionalized in supercritical carbon dioxide with an aminosilane, N-(6-aminohexyl)aminopropyltrimethoxysilane (AHA-P-TMS). Surface characterization (X-ray photoelectron spectroscopy, contact angle, ζ-potential analysis) evidenced the presence of the aminosilane. The results show that the silane conformation depends on the curing process: a nonpolycondensed conformation of grafted silane with the amino groups facing outwards was favored by curing in an oven, while the curing step performed in scCO2 yielded CNF structures with the alkyl chain facing outwards. The grafted nanopapers exhibited antibacterial activity, and no antibacterial agent was released into the media. Furthermore, these materials proved to benefit from low cytotoxicity. This study offers a proof of concept for the covalent grafting of active species on nanocellulose structures and the control of aminosilane orientation using a green and controlled approach. These newly designed materials could be used for their antibacterial activity in the biomedical field. Thus, perspectives for topical administration and design of wound dressing could be envisaged.

International audience; This study presents the impregnation in supercritical carbon dioxide (scCO 2) of nanocellulose-based structures with thymol as a natural antimicrobial molecule to prepare bioactive biosourced materials. First, cellulose nanofibrils (CNFs) were used to produce four types of materials (nanopapers, cryogels from water or tert-butyl alcohol suspensions and aerogels) of increasing specific surface area up to 160 m 2 .g-1 thanks to the use of different processes, namely vacuum filtration, freeze-drying and supercritical drying. Secondly, these CNF-based structures were impregnated with thymol in scCO 2 medium using relatively low temperature and pressure of 40°C and 100 bars during 1 hour. Abstract

Extracellular Vesicles (EVs) are lipid vesicles secreted by cells that allow intercellular communication. They are decorated with surface proteins, which are membrane proteins that can be targeted by biochemical techniques to isolate EVs from background particles. EVs have recently attracted attention for their potential applications as biomarkers for numerous diseases. This review focuses on the contribution of biomolecules used as ligands in affinity-based biosensors for the detection and isolation of EVs. Capturing biological objects like EVs with antibodies is well described in the literature through different biosensing techniques. However, since handling proteins can be challenging due to stability issues, sensors using non-denaturable biomolecules are emerging. DNA aptamers, short DNA fragments that mimic antibody action, are currently being developed and considered as the future of antibody-like ligands. These molecules offer undeniable advantages: unparalleled ease of production, very high stability in air, similar affinity constants to antibodies, and compatibility with many organic solvents. The use of peptides specific to EVs is also an exciting biochemical solution to target EV membrane proteins and complement other probes. These different ligands have been used in several types of biosensors: electrochemical, optical, microfluidic using both generic probes (targeting widely expressed membrane proteins such as the tetraspanins) and specific probes (targeting disease biomarkers such as proteins overexpressed in cancer).

Abstract

In this paper, strictly speaking aerogels of cellulose nanofibrils (CNFs) and TEMPO-oxidized CNFs (TO-CNFs) were obtained from an optimized supercritical drying processes and cryogels were prepared after freeze-drying. The cryogels and aerogels were characterized and the influence of the preparation process on the resulting properties was studied. Significant differences were observed in the micro- and nanoscale organization of the porous structures. In addition, the specific surface areas measured varied from 25 to 160 m². g-1 for CNF materials, depending on the preparation process. Very high specific surface areas up to 482 m². g-1 among the highest reported for pure cellulose nanofibrils porous materials were achieved for TO-CNF aerogels. Finally, in order to evaluate their aptitudes for wound dressings applications, the capillary water uptake capacities were assessed on skin mimicking layers. From this study, it was revealed that TO-CNF aerogels can absorb almost 120 times their own weight of water.

Abstract

Encapsulation is a critical step in LED packaging to ensure good light extraction and protection. We present a novel mold-free process using a specific functionalization of the substrate to master the shape of the encapsulant dome. By controlling its aspect ratio, it is possible to increase light extraction. Experiments are correlated with Monte Carlo simulations to evidence the impact of surface tension on the dome shape as well as the effect of the refractive index on the light output. A moderate optical gain of 10% is obtained as the refractive index of the encapsulant material increases from 1.42 to 1.64, which is explained with the simulations as a strong effect of the die parameters (roughness and back mirror) on the light extraction.

11. Ice-templated freeze-dried cryogels from tunicate cellulose nanocrystals with high specific surface area and anisotropic morphological and mechanical properties

Abstract

High aspect ratio cellulose nanocrystals (CNCs) extracted from tunicate were used to create so-called cryogels from an ice-templating directional freeze-drying process. The structure of the resulting solid foam was investigated at the micro- and nanoscales by scanning electron microscopy and nitrogen adsorption measurements were used to extract the specific surface area. The mechanical properties were probed by compression tests. To highlight the specificities of tunicate CNC-based cryogels, results were compared with the one obtained from two other types of nanocellulose, namely cellulose nanofibrils (CNFs) from wood and CNCs from cotton, which exhibit different dimensions, aspect ratio, flexibility and crystallinity. While CNF- and cotton CNC-based cryogels exhibited a classical morphology characterized by a sheet-like structure, a particular honeycomb organization with individual particles was obtained in the case of tunicate CNC cryogels. The latter cryogels presented a very high specific surface area of about 122 m2 g−1, which is unexpected for foams prepared from a water-based process and much higher than what was obtained for CNF and cotton CNC cryogels (25 and 4 m2 g−1, respectively). High mechanical resistance and stiffness were also obtained with such tunicate CNC cryogels. These results are explained by the high crystallinity, aspect ratio and rigidity of the tunicate CNCs combined with the particular honeycomb architecture of the cryogel.

Abstract

International audience; The first Very Large Scale Integration process with variable shape beam lithography for optomechanical devices is presented. State of the art performance was obtained with silicon microdisk resonators showing 1 million optical quality factors and 10 -17 m.Hz (-1/2) displacement resolution. Single-particle mass spectrometry could be performed with these optomechanical resonators in vacuum. The devices retained high performance when directly immersed in liquid media, allowing for biosensing experiments. These results open the door to large, dense arrays of optomechanical sensors.

Abstract

The natural biodegradabilty of porous silicon (pSi) in physiological media limits its wider usage for implantable systems. We report the stabilization of porous silicon (pSi) membranes by chemical surface oxidation using RCA1 and RCA2 protocols, which was followed by a PEGylation process using a silane-PEG. These surface modifications stabilized the pSi to allow a long period of immersion in PBS, while leaving the pSi surface sufficiently hydrophilic for good filtration and diffusion of several biomolecules of different sizes without any blockage of the pSi structure. The pore sizes of the pSi membranes were between 5 and 20 nm, with the membrane thickness around 70 μm. The diffusion coefficient for fluorescein through the membrane was 2 × 10−10 cm2 s−1, and for glucose was 2.2 × 10−9 cm2 s−1. The pSi membrane maintained that level of glucose diffusion for one month of immersion in PBS. After 2 months immersion in PBS the pSi membrane continued to operate, but with a reduced glucose diffusion coefficient. The chemical stabilization of pSi membranes provided almost 1 week stable and functional biomolecule transport in blood plasma and opens the possibility for its short-term implantation as a diffusion membrane in biocompatible systems.

Abstract

Hydrogen production through direct sunlight-driven water splitting in photo-electrochemical cells (PECs) is a promising solution for energy sourcing. PECs need to fulfill three criteria: sustainability, cost-effectiveness and stability. Here we report an efficient and stable photocathode platform for H2 evolution based on Earth-abundant elements. A p-type silicon surface was protected by atomic layer deposition (ALD) with a 15 nm TiO2 layer, on top of which a 300 nm mesoporous TiO2 layer was spin-coated. The cobalt diimine–dioxime molecular catalyst was covalently grafted onto TiO2 through phosphonate anchors and an additional 0.2 nm ALD-TiO2 layer was applied for stabilization. This assembly catalyzes water reduction into H2 in phosphate buffer (pH 7) with an onset potential of +0.47 V vs. RHE. The resulting current density is −1.3 ± 0.1 mA cm−2 at 0 V vs. RHE under AM 1.5 solar irradiation, corresponding to a turnover number of 260 per hour of operation and a turnover frequency of 0.071 s−1. Abstract

International audience; Metallic conductive nanowires (NWs) with DNA bundle core are achieved, thanks to an original process relying on double-stranded DNA alignment and physical vapor deposition (PVD) metallization steps involving a silicon substrate. First, bundles of DNA are suspended with a repeatable process between 2 aem high parallel electrodes with separating gaps ranging from 800 nm to 2 mu m. The process consists in the drop deposition of a DNA lambda-phage solution on the electrodes followed by a naturally evaporation step. The deposition process is controlled by the DNA concentration within the buffer solution, the drop volume, and the electrode hydrophobicity. The suspended bundles are finally metallized with various thicknesses of titanium and gold by a PVD e-beam evaporation process. The achieved NWs have a width ranging from a few nanometers up to 100 nm. The electrical behavior of the achieved 60 and 80 nm width metallic NWs is shown to be Ohmic and their intrinsic resistance is estimated according to different geometrical models of the NW section area. For the 80 nm width NWs, a resistance of about few ohms is established, opening exploration fields for applications in microelectronics.

Abstract

Open-surface microfluidic systems, i.e., microflows with a free surface, are finding increasing use in the fields of biotechnology, biology and medicine, especially in the domain of point-of-care and home-care systems. In such systems, the fluids are moved by capillarity and do not require the use of costly and/or bulky pumps, or other actuation systems. Recently, a subcategory of such flows, called “suspended microflows,” has appeared where the liquids flows between vertical walls without supporting walls at the bottom, i.e., surface tension is used to fill and maintain a fluid in microscale structures devoid of a ceiling and floor. The condition for the onset of spontaneous suspended flows has been developed in a preceding article. However, the dynamics of suspended microfluidics has not yet been investigated, and we present here a first analysis of their dynamics. Verifications of the theoretical model have been performed using colored water. Applications to the capillary motion of whole blood and to the motion of polymeric liquids have been done. In particular, we show that capillary filling of a suspended turning channel may be an approach to the realization of in situ giant porous micromembranes.

Abstract

The development of “green” sensors is a challenging task in the field of biomolecule sensing, for example in the detection of cardiac troponin-I (cTnI). In the present work a complete approach in green chemistry was developed to create chemically active patterns for the immobilisation of biological probes. This key technology is discussed on the basis of the twelve green chemistry principles, and is a combination of surface patterning by spotting and surface chemistries modified by molecular vapour deposition. The (1 H ,1 H ,2 H ,2 H )-perfluorodecyltrichlorosilane (FDTS) was used as a novel anti-adsorption layer while the 3,4-epoxybutyltrimethoxysilane (EBTMOS) was used to immobilise probes. Oligonucleotides and the anti-cTnI antibody were studied. The spatially controlled immobilisation of probes was characterised by fluorescence. The demonstrated surface modification has broad applications in areas such as diagnostics and bio-chemical sensing. Moreover, the environmental impacts of surface patterning and surface chemistry were discussed from a “greenness” point of view.

Abstract

Redox active ferrocene derivatives were grafted upon SiO2 surface via a carbon-based linker. The indirect grafting procedure consisted in a silanization followed by a click-chemistry reaction. This immobilization method of ethynyl-ferrocene yields a mixed organic/inorganic charge transfer modulation medium made of a C3 linker ended by a 1,2,3-triazole, bound to a thin SiO2 layer. Such a linker has never been tested before. Its use combines the advantages of a thin dielectric layer (SiO2+ > C3 linker) to the versatility of a two-step process that could easily be transferred to other sensitive or hard to synthesize compounds. Such organic/inorganic linker has been tested to tune the redox properties and charge transfer kinetics of the overall hybrid system and the results could be of importance for hybrid memory devices applications. Cyclic voltammetry studies conducted on this hybrid system were indeed used to confirm ferrocene immobilization, determine the molecular surface coverage and study the redox charge/discharge kinetics. The obtained results show the good surface coverage and charge retention properties, achieved by grafting upon SiO2 via a > C3 linker. For comparison, the ferrocene compound was also immobilized on Si (C2 linker).

Abstract

DNA is under continuous assault by environmental and endogenous reactive oxygen and alkylating species, inducing the formation of mutagenic, toxic and genome destabilizing nucleobase lesions. Due to the implications of such genetic alterations in cell death, aging, inflammation, neurodegenerative diseases and cancer, many efforts have been devoted to developing assays that aim at analyzing DNA repair activities from purified enzymes or cell extracts. The present work deals with the conception and application of a new, miniaturized and parallelized on surface-DNA biosensor to measure base excision repair (BER) activities. Such a bio-analytical tool was built by using the "click chemistry" approach to immobilize, on a glass slide, fluorescent stem-loop DNA probes, which contain a specific nucleobase lesion. The performance of this new high-throughput DNA repair analysis technology was determined by detecting uracil N-glycosylase and AP-endonuclease activities from purified enzymes or in cell extracts. The applications of this device were extended to analyze, in cell extracts, the ability of two inhibitors (Uracil glycosylase inhibitor (Ugi) and methoxyamine (MX)) to block the excision of uracil and the cleavage of AP sites, respectively. Altogether, our results show that this new fluorescent DNA microarray platform provides an easy, rapid and robust method for detecting DNA N-glycosylase and AP-endonuclease activities and evaluating the effects of BER inhibitors in a multiplexed fashion.

Abstract

In this thesis, the interactions between different proteins and small ligands were characterized by surface plasmon resonance spectroscopy (SPR) and fluorescence resonance energy transfer (FRET) based assays. For the C-reactive protein (CRP), a new type of artificial binder was identified which allows designing diagnostic assays superior to commonly used standard assays. Furthermore, an interaction study with the endogenous ligand phosphocholine revealed the importance of the avidity of pentameric CRP for the distinction of different types of lipid membranes. The interaction study with calcium showed how SPR based assays can be used to study ion-protein interactions despite the low atomic weight of ions. The transmembrane protease BACE1, an important drug target for Alzheimer’s disease, was immobilized to an SPR biosensor surface and embedded into a lipid membrane. An interaction study with a set of known BACE1 inhibitors showed that the transmembrane region has only minor effects on the interactions. Furthermore the pH-dependencies of the interactions were investigated and revealed new important conclusions for inhibitor design. Computer aided modelling showed that the protonation state of the aspartic dyad is dependent on the interacting inhibitor which offers new perspectives for in silico screenings.The SPR assay developed for BACE1 was adapted to a more complex membrane protein, the pentameric β3 GABAA receptor. The assay allowed the pharmacological characterisation for histaminergic and GABAergic ligands and gave further evidence for cross-talk between the two signal transduction pathways. This study shows that the immobilisation method used for BACE1 and the s3 GABAA receptor has the potential to become a standard method for handling membrane proteins. The identification of new drug leads from natural sources is a common strategy for drug discovery. A combination of SPR and FRET based activity assays were explored to increase the efficiency of this process. For HIV-1 protease, secreted aspartic protease (SAP) 1, 2 and 3 extracts from a marine vertebrate were identified containing potent inhibitors which interacted with the active site of the enzymes.The studies in this thesis show that the investigation of protein interactions is crucial for understanding protein functions and can help to develop novel drugs for the treatment of different diseases.

Abstract

A hand-held formaldehyde (HCHO) gas detector has been developed. This fluorescence-based system uses a standard microscope glass slide both as a transducer and a sensor. The selective detection of gaseous formaldehyde is performed using the well-known Hantzsch reaction with Fluoral-P, which is chosen as a reagent. The sensor consists of a nanoporous thin film doped with the reagent, which allows capturing the pollutants molecules. The nanoporous films of polymers are prepared using the Sol-Gel method and are coated on the microscope slide by spin-coating, enabling reproducible coatings of various shapes and thicknesses. Experiments were carried out in the laboratory to calibrate the instrument, with the objective to perform measurements in indoor and outdoor conditions. The real-time monitoring allows detecting pollution peaks of formaldehyde.

Abstract

International audience; The functionalization of silicon oxide based substrates using silanes is generally performed through liquid phase methodologies. These processes involve a huge quantity of potentially toxic solvents and present some important disadvantages for the functionalization of microdevices or porous materials, for example the low diffusion. To overcome this drawback, solvent-free methodologies like molecular vapor deposition (MVD) or supercritical fluid deposition (SFD) have been developed. In this paper, the deposition process of 3,4-epoxybutyltrimethoxysilane (EBTMOS) on silicon oxide using supercritical carbon dioxide (scCO2) as a solvent is studied for the first time. The oxirane ring of epoxy silanes readily reacts with amine group and is of particular interest for the grafting of amino-modified oligonucleotides or antibodies for diagnostic application. Then the ability of this specific EBTMOS layer to react with amine functions has been evaluated using the immobilization of amino-modified oligonucleotide probes. The presence of the probes is revealed by fluorescence using hybridization with a fluorescent target oligonucleotide. The performances of SFD of EBTMOS have been optimized and then compared with the dip coating and molecular vapor deposition methods, evidencing a better grafting efficiency and homogeneity, a lower reaction time in addition to the eco-friendly properties of the supercritical carbon dioxide. The epoxysilane layers have been characterized by surface enhanced ellipsometric contrast optical technique, atomic force microscopy, multiple internal reflection infrared spectroscopy and X-ray photoelectron spectroscopy. The shelf life of the 3,4-epoxybutyltrimethoxysilane coating layer has also been studied. Finally, two different strategies of NH2-oligonucleotide grafting on EBTMOS coating layer have been compared, i.e. reductive amination and nucleophilic substitution, SN2. This EBTMOS based coating layer can be used for a wide range of applications such as the preparation of new supported and recoverable catalysts and new integrated silicon microdevices for healthcare purposes.

Abstract

Current detection and identification of micro-organisms is based on either rather unspecific rapid microscopy or on more accurate but complex and time-consuming procedures. In a medical context, the determination of the bacteria Gram type is of significant interest. The diagnostic of microbial infection often requires the identification of the microbiological agent responsible for the infection, or at least the identification of its family (Gram type), in a matter of minutes. In this work, we propose to use terahertz frequency range antennas for the enhanced selective detection of bacteria types. Several microorganisms are investigated by terahertz time-domain spectroscopy: a fast, contactless and damage-free investigation method to gain information on the presence and the nature of the microorganisms. We demonstrate that plasmonic antennas enhance the detection sensitivity for bacterial layers and allow the selective recognition of the Gram type of the bacteria. Abstract

International audience; A hand-held formaldehyde (HCHO) gas detector has been developed. This fluorescence-based system uses a standard microscope glass slide both as a transducer and a sensor. The detection of gaseous formaldehyde is performed using the well-known Sawicki reaction with Fluoral-P, a reagent present in the nanoporous thin film which allows a selective reaction with formaldehyde. The nanoporous film of hybrid organic-inorganic polymer, prepared via the sol-gel method, is coated on the microscope slide by spin-coating, enabling reproducible coatings of various shapes and thicknesses. Experiments were carried out in the laboratory to calibrate the instrument, with the objective to perform measurements indoor and outdoor. The real-time monitoring allows to detect formaldehyde pollution peaks.

International audience; A new process for preparing oligonucleotide arrays is described that uses surface grafting chemistry which is fundamentally different from the electrostatic adsorption and organic covalent binding methods normally employed. Solid supports are modified with a mixed organic/inorganic zirconium phosphonate monolayer film providing a stable, well-defined interface. Oligonucleotide probes terminated with phosphate are spotted directly to the zirconated surface forming a covalent linkage. Specific binding of terminal phosphate groups with minimal binding of the internal phosphate diesters has been demonstrated. On the other hand, the reaction of a bisphosphonate bone resorption inhibitor (Zoledronate) with calcium deficient apatites (CDAs) was studied as a potential route to local drug delivery systems active against bone resorption disorders. A simple mathematical model of the Zoledronate/CDA interaction was designed that correctly described the adsorption of Zoledronate onto CDAs. The resulting Zoledronate-loaded materials were found to release the drug in different phosphate-containing media, with a satisfactory agreement between experimental data and the values predicted from the model.

Abstract

A new approach to oligonucleotide arrays is demonstrated that utilizes zirconium phosphonate-derivatized glass slides. The active slides are prepared by binding Zr(4+) to surfaces terminated with organophosphonate groups previously deposited using either Langmuir-Blodgett or self-assembled monolayer methods. Oligonucleotide probes modified with a terminal phosphate bind strongly to the active zirconium phosphonate monolayer, and arrays for detecting fluorescent targets have been prepared using commercial spotting and scanning instruments. Preferred binding to the surface of the terminal phosphate of the modified probes instead of the internal phosphate diester groups is demonstrated and shown to yield increased fluorescence intensity after hybridization with labeled targets. A significant decrease in background signal is achieved by treating the slides with bovine serum albumin after spotting and before hybridization. A further increase in fluorescence after hybridization is observed when using a poly-guanine spacer between the probe oligomer and the terminal phosphate. Combining these modifications, an intensity ratio of nearly 1000 is achieved when comparing 5'-phosphate-modified 33-mer probes with unmodified probes upon hybridization with fluorescent targets.

Abstract

A novel series of chiral P,N-ligands was obtained by desymmetrization of the achiral meso-N, N′-dimethyl-1,2-diphenylethane-1,2-diamine backbone and resolution of the resulting adducts. This transformation was achieved by the selective introduction of a diphenylphosphine moiety on one of the two nitrogen centers. The resulting palladium complexes were used for the catalytic enantioselective allylic alkylation of 1,3-diphenyl-1-acetoxy-2-propene with dimethyl malonate. In some cases, high conversions (up to 98% after 1 h) along with good chiral inductions (up to 95% e.e.) were obtained, as a result of a control of the inversion of the stereogenic center formed at nitrogen upon coordination to palladium.
