Levoglucosan is a typical orientationally disordered crystalline material (the molecules are long-range positionally ordered but orientationally disordered). At present, experimental research on its glass transition is mainly focused on polycrystals, while the study of the corresponding single crystals is rarely reported. Centimetre-size (maximum dimensions of 2.1 × 0.9 × 0.4 cm) levoglucosan single crystals of high quality were grown by the solution method, laying a necessary foundation for experimental measurements of the glass transition mechanism in the material.

Its goal was to see how a transdermal fentanyl patch combined with accelerated recovery after surgery (ERAS) affected the treatment efficacy and analgesic effect of liver cancer, as well as to help patients with liver cancer choose the right analgesic treatment and nursing mode. 150 patients with liver cancer were divided into group A (transdermal fentanyl patch), group B (ERAS), and group C (transdermal fentanyl patch combined with ERAS). Patients in the three groups were compared in terms of pain, survival, psychological status, adverse responses, postoperative recovery, and patient satisfaction. The results showed that under different treatment and nursing methods, the number of patients with mild cancer pain in the three groups was increased, especially the number of patients with mild cancer pain in group C ( P < 0.05 ). Besides, the quality of life score of patients in each group was decreased. Patients who received the combination analgesia had a significantly higher quality of life than those who received simply a transdermal fentanyl patch or ERAS ( P < 0.05 ). The scores of both the Hamilton anxiety scale (HAMA) and Hamilton depression rating scale (HAMD) of patients with the combined analgesia were decreased signally ( P < 0.05 ). There were few patients with combined analgesia who had adverse reactions ( P < 0.05 ). After surgery, the time of the first anal exhaust, first defecation, and first ambulation in group C were shorter than those in the other two groups ( P < 0.05 ). To summarize, combining the two techniques aided in the recovery of gastrointestinal function as well as the physical recovery of patients following surgery. Furthermore, combining the two approaches produced a clear analgesic impact, which could improve patients’ quality of life while also having a favorable clinical adoption effect.

Taken together, this study examined the effects of arbuscular mycorrhizal fungi (AMF) and rhizobial combinations on the growth of Al-tolerant and Al-sensitive soybeans as well as their associated microbial communities in acidic soils and concluded that AMF enhances soybean growth and Al stress tolerance by recruiting PGPR and altering the root-associated microbiome assembly in a host-dependent manner. In the future, these findings will help us better understand the impacts of AMF on rhizosphere microbiome assembly and will contribute to the development of soybean breeding techniques for the comprehensive use of PGPR in sustainable agriculture.

Chlamydomonas reinhardtii is a nonpathogenic, nontoxigenic green algae used as a sustainable source of protein in foods. In order to mimic meat-like qualities, a strain rich in protoporphyrin IX (PPIX), an endogenous heme/chlorophyll precursor, was developed using an evolution and selection strategy, and investigations were carried out to evaluate the safety of the novel strain, C. reinhardtii (red), strain TAI114 (TAI114). Digestibility and proteomic evaluations were conducted to determine whether any potentially allergenic or toxic proteins occurred as the result of the mutation process. The genotoxic potential of pure PPIX was evaluated using a bacterial reverse mutation test, an in vitro mammalian chromosomal aberration test, and an in vivo mammalian micronucleus test. Finally, the novel TAI114 biomass was evaluated for general toxicity and identification of target organs in a 90-day repeated-dose oral toxicity study in rats. All proteins were rapidly degraded in pepsin at pH 2.0 suggesting low allergenic potential. The proteomic evaluation indicated that TAI114 biomass contains typical C. reinhardtii proteins. PPIX was unequivocally negative for genotoxic potential and no target organs or adverse effects were observed in rats up to the maximum feasible dose of 4000 mg/kg bw/day TAI114 biomass, which was determined to be the no-observed-adverse-effect-level (NOAEL). These results support the further development and risk characterization of TAI114 biomass as a novel ingredient for use in the meat analogue category of food.

Drought stress severely threatens the yield of cereal crops. Therefore, understanding the molecular mechanism of drought stress response of plants is crucial for developing drought-tolerant cultivars. NAC transcription factors (TFs) play important roles in abiotic stress of plants, but the functions of NAC TFs in sorghum are largely unknown. Here, we characterized a sorghum NAC gene, SbNAC9, and found that SbNAC9 can be highly induced by polyethylene glycol (PEG)-simulated dehydration treatments. We therefore investigated the function of SbNAC9 in drought stress response. Sorghum seedlings overexpressing SbNAC9 showed enhanced drought-stress tolerance with higher chlorophyll content and photochemical efficiency of PSII, stronger root systems, and higher reactive oxygen species (ROS) scavenging capability than wild-type. In contrast, sorghum seedlings with silenced SbNAC9 by virus-induced gene silencing (VIGS) showed weakened drought stress tolerance. Furthermore, SbNAC9 can directly activate a putative peroxidase gene SbC5YQ75 and a putative ABA biosynthesis gene SbNCED3. Silencing SbC5YQ75 and SbNCED3 led to compromised drought tolerance and reduced ABA content of sorghum seedlings, respectively. Therefore, our findings revealed the important role of SbNAC9 in response to drought stress in sorghum and may shed light on genetic improvement of other crop species under drought-stress conditions.

A digital twin basin serves as a virtual representation of a physical basin, enabling synchronous simulation, virtual–real interaction, and iterative optimization. The construction of a digital twin basin requires a basin database characterized by large-scale coverage, high-precision, high-resolution, and low-latency attributes. The advancements in remote sensing technology present a new technical means for acquiring essential variables of the basin. The purpose of this paper was to provide a comprehensive overview and discussion of the retrieval principle, data status, evaluation and inter-comparison, advantages and challenges, applications, and prospects of remote sensing technology in capturing seven essential variables, i.e., precipitation, surface temperature, evapotranspiration, water level, river discharge, soil moisture, and vegetation. It is indicated that remote sensing can be applied in some digital twin basin functions, such as drought monitoring, precipitation forecasting, and water resources management. However, more effort should be paid to improve the data accuracy, spatiotemporal resolution, and latency through data merging, data assimilation, bias correction, machine learning algorithms, and multi-sensor joint retrieval. This paper will assist in advancing the application of remote sensing technology in constructing a digital twin basin.

The pathogenesis of central nervous system involvement (CNSI) in patients with acute lymphoblastic leukaemia (ALL) remains unclear and a robust biomarker of early diagnosis is missing. An untargeted cerebrospinal fluid (CSF) metabolomics analysis was performed to identify independent risk biomarkers that could diagnose CNSI at the early stage. Thirty-three significantly altered metabolites between ALL patients with and without CNSI were identified, and a CNSI evaluation score (CES) was constructed to predict the risk of CNSI based on three independent risk factors (8-hydroxyguanosine, l-phenylalanine and hypoxanthine). This predictive model could diagnose CNSI with positive prediction values of 95.9% and 85.6% in the training and validation sets respectively. Moreover, CES score increased with the elevated level of central nervous system (CNSI) involvement. In addition, we validated this model by tracking the changes in CES at different stages of CNSI, including before CNSI and during CNSI, and in remission after CNSI. The CES showed good ability to predict the progress of CNSI. Finally, we constructed a nomogram to predict the risk of CNSI in clinical practice, which performed well compared with observed probability. This unique CSF metabolomics study may help us understand the pathogenesis of CNSI, diagnose CNSI at the early stage, and sequentially achieve personalized precision treatment.

Both aldosterone and arginine vasopressin (AVP) are produced in the heart and may participate in cardiac fibrosis. However, their relationship remains unknown. This study aims to demonstrate the regulation and role of AVP in aldosterone synthesis in the heart. Rats were subjected to a sham operation or myocardial infarction (MI) by ligating the coronary artery. Cardiac function and fibrosis were assessed using echocardiography and immunohistochemical staining, respectively. In addition, the effects of AVP stimulation on cardiac microvascular endothelial cells (CMECs) were studied using ELISA, real-time PCR, and Western blotting. Compared with the rats having undergone a sham operation, the MI rats had an increased LVMI, type I collagen composition, and concentrations of aldosterone and AVP in the heart but decreased cardiac function. As the MI rats aged, the LVMI, type I collagen, aldosterone, and AVP increased, while the LVMI decreased. Furthermore, AVP time-dependently induced aldosterone secretion and CYP11B2 mRNA expression in CMECs. The p-CREB levels were significantly increased by AVP. Nevertheless, these effects were completely blocked by SR49059 or partially inhibited by KN93. This study demonstrated that AVP could induce the secretion of local cardiac aldosterone, which may involve CaMK and CREB phosphorylation and CYP11B2 upregulation through V1 receptor activation.

PDK1 is one of the key enzymes in the glucose metabolism pathway, which is abnormally high expressed in breast cancer tissues and can promote tumor proliferation and metastasis. PDK1 and the PDHC/PDK axis are important targets for regulating glucose metabolism and anti-tumor activity. In this study, we evaluated the anti-tumor activities of a series of semi-synthesized shikonin (SK) derivatives against human breast cancer cells.The anti-proliferation activity of SK derivatives against human breast cancer cell lines was tested by CCK-8 and EdU assay. Flow cytometry was utilized to evaluate cell apoptosis, reactive oxygen species and cell cycle distribution. Cell migration ability was determined by wound healing and trans-well assay. PDK1 targeting effect was confirmed by western bolting, molecular docking, bio-layer interferometry and PDK1 enzyme activity assay. Nude-mouse transplanted tumor model was used to evaluate their anti-tumor effect in vivo.Findings revealed that SK derivatives had good anti-proliferation ability against MDA-MB-231 cell. They induced cell apoptosis by regulating the mitochondrial apoptosis and death receptor pathway. They also inhibited cell migration by suppressing EMT progression. Molecular docking, PDK1 affinity and enzyme activity demonstrated their PDK1 targeting. In vivo antitumor experiment showed that E2 could significantly inhibit tumor growth with lower side-effect on mice than SK.In conclusion, the novel SK derivatives E2 and E5 inhibited tumor glycolysis by targeting PDK1 and ultimately induced apoptosis. Our data demonstrated that E2 would be a good lead compound for the treatment of human TNBC as a novel PDK1 inhibitor.

Skillful sub-seasonal precipitation forecasts can provide valuable information for both flood and drought disaster mitigations. This study evaluates both deterministic and probabilistic sub-seasonal precipitation forecasts of ECMWF, ECCC, and UKMO models derived from the Sub-seasonal to Seasonal (S2S) Database at various spatiotemporal scales over China during the boreal summer monsoon. The Multi-Source Weighted-Ensemble Precipitation, version 2 (MSWEP V2), is used as the reference dataset to evaluate the forecast skills of the models. The results suggest that skillful deterministic sub-seasonal precipitation forecasts are found when the lead time is within 2 weeks. The deterministic forecast skills reduce quickly when the lead time is beyond 2 weeks. Positive ranked probability skill scores (RPSS) are only found when the lead time is within 2 weeks for probabilistic forecasts as well. Multimodel ensembling helps to improve forecast skills by removing large negative skill scores in northwestern China. The forecast skills are also improved at larger spatial scales or longer temporal scales. However, the improvement is only observed for certain regions where the predictable low frequency signals remain at longer lead times. The composite analysis suggests that both the El Niño–Southern Oscillation (ENSO) and Madden–Julian Oscillation (MJO) have an impact on weekly precipitation variability over China. The forecast skills are found to be enhanced during active ENSO and MJO phases. In particular, the forecast skills are found to be enhanced during active MJO phases.

Both convection and land surface parameterization influence seasonal precipitation forecasts. In this study, the sensitivity of dynamical downscaling seasonal precipitation forecasts to convection and land surface parameterization was investigated by nesting the Weather Research and Forecasting (WRF) model into the NCEP’s Climate Forecast System version 2 (CFSv2) retrospective forecasts with four convective schemes: Kain–Fritsch (KF), Betts–Miller–Janjic (BMJ), Grell–Freitas (GF), and new simplified Arakawa–Schubert (NSAS) schemes, and two land surface schemes: Noah and simplified Simple Biosphere (SSiB) schemes over the Han River basin. The CFSv2 model biases are reduced when the KF convective scheme is used in the wet summer season. However, negative biases still exist especially when the combination of BMJ and SSiB schemes is used. Compared with CFSv2 reforecasts and other combinations of schemes, the forecast skills of spatial patterns of precipitation anomalies are highest when the combination of KF and Noah schemes is used in summer. In contrast, the combination of BMJ and SSiB schemes shows lowest forecast skills in summer. To understand the causes of the differences in precipitation forecasts using different parameterization schemes, the simulated moisture flux convergence, thermodynamic parameters at different pressure levels, convective available potential energy (CAPE), convective inhibition (CIN), and heat fluxes are compared with the data in the ERA-5 reanalysis dataset. The WRF model-simulated moisture flux convergence is closer to that of the ERA-5 reanalysis compared with that of the CFSv2 reforecasts in summer. The vertical thermodynamic profiles also suggest that the combination of the KF and Noah schemes has caused a more unstable atmosphere, which is crucial for precipitation. In contrast, the combination of BMJ and SSiB schemes shows a less unstable atmospheric environment in summer, which explains the lower forecast skills compared with other schemes. The spatial patterns of CAPE are also improved when using the WRF model, which further enhances the precipitation forecast skills over the Han River basin.

Background Experimental and clinical trials have demonstrated the efficiency of bone marrow-derived mesenchymal stromal/stem cells (bMSCs) in the treatment of myocardial infarction. However, after intravenous injection, the ineffective migration of engrafted bMSCs to the hearts remains an obstacle, which has an undesirable impact on the efficiency of cell-based therapy. Therefore, we attempted to identify a marker that could distinguish a subpopulation of bMSCs with a promising migratory capacity. Methods Here, CD51-negative and CD51-positive cells were isolated by flow cytometry from Ter119−CD45−CD31−bMSCs and cultured in specifically modified medium. The proliferation ability of the cells was evaluated by 5-ethynyl-2′-deoxyuridine (EdU) staining or continuously monitored during culture, and the differentiation potential was assessed by culturing the cells in the appropriate conditioned media. Wound healing assays, transwell assays and quantitative polymerase chain reaction (qPCR) were used to measure the migratory ability. The mice were subjected to a sham operation or myocardial infarction (MI) by permanently occluding the coronary artery, and green fluorescent protein (GFP)-labelled cells were transplanted into the mice via intravenous infusion immediately after MI. Heart function was measured by echocardiography; infarct myocardium tissues were detected by triphenyl tetrazolium chloride (TTC) staining. Additionally, immunofluorescence staining was used to verify the characteristics of CD51+bMSCs and inflammatory responses in vivo. Statistical comparisons were performed using a two-tailed Student’s t test. Results In this study, the isolated CD51−bMSCs and CD51+bMSCs, especially the CD51+ cells, presented a favourable proliferative capacity and could differentiate into adipocytes, osteocytes and chondrocytes in vitro. After the cells were transplanted into the MI mice by intravenous injection, the therapeutic efficiency of CD51+bMSCs in improving left ventricular ejection fraction (LVEF) and left ventricular fractional shortening (LVFS) was better than that of CD51−bMSCs. Compared with CD51−bMSCs, CD51+bMSCs preferentially migrated to and were retained in the infarcted hearts at 48 h and 8 days after intravenous injection. Accordingly, the migratory capacity of CD51+bMSCs exceeded that of CD51−bMSCs in vitro, and the former cells expressed higher levels of chemokine receptors or ligands. Interestingly, the retained CD51+bMSCs retained in the myocardium possessed proliferative potential but only differentiated into endothelial cells, smooth muscle cells, fibroblasts or cardiomyocytes. Transplantation of CD51+bMSCs partially attenuated the inflammatory response in the hearts after MI, while the potential for inflammatory suppression was low in CD51−bMSC-treated mice. Conclusions These findings indicated that the CD51-distinguished subpopulation of bMSCs facilitated proliferation and migration both in vitro and in vivo, which provided a novel cell-based strategy to treat acute MI in mice by intravenous injection.

Genetically engineered (GE) maize has been thoroughly studied regarding its agro-environmental impact; however, its concerns for the soil environment remain. This work was aimed to decode rhizosphere microbe interactions and potential ecological hazards associated with GE maize. Rhizobacterial communities of field grown transgenic insect-resistant 2A5 maize carrying mcry1Ab and mcry2Ab genes were compared with control Z58 using PacBio sequencing platform. Also full-length 16S rDNA gene sequencing was used to verify the partial (V3-V4) sequencing results obtained in 2017. Measures of α-diversity displayed transgenic 2A5 to be significantly lower in species richness at the flowering stage; however, diversity remained undisturbed. β-diversity was least affected by genetic modifications where similar community profiles were shared by transgenic 2A5 and control Z58. In addition, root exudation patterns were found to drive variations in bacterial assemblages based on developmental stages. For example, genus Massilia successfully colonized the rhizosphere at jointing stage, while Mucilaginobacter showed higher relative abundance in flowering stages of both 2A5 and Z58. These members are known to possess attributes related to plant growth. The impact of dual-transgene insertion on nifH gene abundance was also analyzed where no apparent significant difference in nifH gene copy number was observed. Our results confirmed that full-length 16S rDNA sequencing was sufficient to provide higher taxonomic resolution. Also, results of our 2-year field trials confirmed that there is no significant impact of mcry gene integration on belowground biomasses. Therefore, GE insect-resistant 2A5 maize carrying mcry1Ab and mcry2Ab genes can continue to benefit human populations by increasing crop productivity. In future, further research needs to be catalyzed to analyze the impact of Bt-insertion on microbial community structure across the years for ecosystem sustainability.

Allogeneic hematopoietic stem cell transplantation (allo-HSCT) is recommended for adults acute lymphoblastic leukemia (ALL) with minimal residual disease (MRD) negative during their first complete remission (CR1). However, the role of pre-transplant consolidation chemotherapy remains unclear. We evaluated 78 CR1/MRD-negative patients, the consolidation and non-consolidation groups had similar 5-year OS (74.8% [95% CI: 62.2-87.3%] vs. 74.2% [95% CI: 53.2-95.1%]

In the past thirty years, the biosafety of the aboveground part of crops, including horizontal gene transferal through pollen dispersal and hybridization, has been the focus of research; however, microbial communities in the underground part are attracting increasing attention. In the present study, the soybean root-associated bacterial communities of the G2-EPSPS plus GAT transgenic soybean line Z106, its recipient variety ZH10, and Z106 treated with glyphosate (Z106J) were compared at the seedling, flowering, and seed filling stages by high-throughput sequencing of the V4 hypervariable regions of 16S rRNA gene amplicons using Illumina MiSeq. The results obtained showed no significant differences in the alpha/beta diversities of root-associated bacterial communities at the three stages among ZH10, Z106, and Z106J under field growth conditions; however, the relative abundance of four main nitrogen-fixing bacterial genera significantly differed among ZH10, Z106, and Z106J. Ternary plot results indicated that in the root compartment, the proportional contributions of rhizobial nitrogen-fixing Ensifer fredii and Bradyrhizobium elkanii, which exhibit an extremely broad nodulation host range, markedly differed among the three treatments at the three stages. Thus, the present results indicate that transgenic G2-EPSPS and GAT soybean may induce different changes in functional bacterial species in soil, such as E. fredii and B. elkanii, from ZH10, which were compensated for/enriched at the flowering and seed filling stages, respectively, to some extent through as of yet unknown mechanisms by transgenic soybean treated with glyphosate.

OsGhd7 gene was discovered by screening our rice activation tagging population. CRISPR–Cas9 created knockouts of OsGhd7 conferred early flowering and early maturity in rice varieties across multiple geographical locations in China. Our research shows that OsGhd7 is a good target for breeding early maturity rice varieties, and an excellent example of the advantages of applying the CRISPR–Cas9 technology for trait improvement. Flowering time (heading date) is an important trait for crop cultivation and yield. In this study, we discovered a late flowering gene OsGhd7 by screening our rice activation tagging population, and demonstrated that overexpression of OsGhd7 delayed flowering time in rice, and the delay in flowering time depended on the transgene expression level. OsGhd7 is a functional allele of the Ghd7 gene family; knockouts of OsGhd7 generated by CRISPR–Cas9 significantly accelerated flowering time and the earliness of the flowering time depended on field location. The homozygous OsGhd7 knockout lines showed approximately 8, 10, and 20 days earlier flowering than controls at three different locations in China (Changsha City, Sanya City, and Beijing City, respectively) that varied from 18.25° N to 39.90° N. Furthermore, knockouts of OsGhd7 also showed an early flowering phenotype in different rice varieties, indicating OsGhd7 can be used as a common target gene for using the CRISPR technology to modulate rice flowering time. The importance of OsGhd7 and CRISPR technology for breeding early maturity rice varieties are discussed.

Sub-seasonal precipitation forecasts during the boreal summer monsoon season are very valuable for flood and drought mitigation over China. Here, we evaluate the sub-seasonal precipitation forecast skills of 11 dynamic models from the Sub-seasonal to Seasonal (S2S) Prediction Project at various spatial and temporal scales. For ensemble mean forecasts, most models show significant correlations with observations at both grid and basin scales with lead time up to 2 weeks. When the lead time is beyond week-2, significant correlations are only observed over southeast and western China at the grid scale. Spatial aggregation helps improve week-3-4 average forecast skills at basin scales; significant correlations can be found for all hydroclimatic regions over China. For ensemble forecasts, most S2S models produce skilful forecasts at basin scale as measured by discrimination scores. Both the El Niño-Southern Oscillation (ENSO) and the Madden-Julian Oscillation (MJO) have an impact on precipitation forecast skills at week-3-4. In particular, forecast skill improvement is most pronounced when the forecasts are initialized during active MJO center located in Maritime Continent (Phase 4~5). The results here will help inform the usefulness of sub-seasonal forecasts for hydrological modelling for drought and flood mitigation.

Vertically downward motion of air current is physically drought-inducing, which has the potential of being a simple and universal drought indicator. The core objective of the present study is to employ vertical motion to simulate and predict droughts after investigating dynamically drought-inducing mechanism. Season-scale drought processes and spatial distributions during 2009–2016 are our concerns, and all the drought study regions of China were chosen as the research areas. Three-month SPI (SPI3) updated daily was used to identify drought processes, and original vertical motion and associated horizontal divergence were also transformed to season-scale standardized anomalies (SA) with a daily running window. In situ observation, ERA-Interim reanalysis, and CFSv2 forecast products were comprehensively employed for drought simulation and prediction. To date, the main results and conclusions are as follow: (1) Atmospheric dynamical anomalies during drought processes and key phases were uncovered. Dynamically drought-inducing features are generally characterized as the typically anomalous upper-convergence–lower-divergence patterns and the intensified downward vertical motion as expected. Signal intensities and vertical configurations are time-varying and seemingly coincide with evolution of regional processes. Particularly, vertical velocity exhibited universally strengthened downward anomalies over almost all the droughts. (2) On the basis of dynamically vertical features uncovered above, the SVVAI (Standardized Vertical Velocity Anomaly Index) is newly proposed. The SVVAI is calculated using SA-based values of vertical motion at multiple pressure levels in the troposphere. The SVVAI_ave and SVVAI_max, corresponding to the vertically average- and maximum-based computation schemes, can be adopted. (3) Drought processes and spatial distributions were simulated with the SVVAI_ave and SVVAI_max. They commonly show highly positive correlations with realistic ones over most regions, and the SVVAI_ave outperformed the SVVAI_max. (4) To further understand difference of simulation capacity, temporal correlation coefficients (TCC) of the SVVAI_ave against observed SPI3 at the grid scale were used for analysis. Positive TCCs above +0.3 occupies most areas to the east of 110° E, while large-area low TCCs (−0.1 ~ +0.3) appear to the west of 110° E over China. It is notably seen that East China and Northeast China are the two regions with highly positive TCCs (+0.6 ~ +0.8). (5) Drought prediction using the SVVAI_ave was preliminarily explored. Regarding the prospective 60-day process prediction, the predicted SVVAI_ave was equally matched with or a little better than the forecasted SPI3 in most cases. Predicted spatial distribution is preliminarily assessed via the example of the 2011 summer–autumn drought over Southwest China, and prediction performance at the occurrence, peak and termination times are inconsistent. (6) Overall, the novel SVVAI herein may be complementary to current approaches of operational drought monitoring and prediction. Further study could be focused on the two following aspects: One is index applicability, that is to say, to explore when and where the predicted SVVAI outperforms the forecasted SPI3. The other is to further explore antecedent drought-inducing signals of atmospheric/oceanic anomalies with the bridge of vertical motion, which may provide a fundamental approach for drought prediction with long lead times.

Purpose According to the Chinese Stock Exchange rules, the listed companies’ management earnings forecasts (MEFs) are divided into mandatory and voluntary earnings forecasts. Different information disclosure mechanisms may bring different economic consequences. Compared with the former, when, how frequently and what kind of voluntary earnings forecasts are disclosed almost entirely depends on the discretion of managers and the major shareholders[1]. The purpose of this paper is to examine whether listed companies’ voluntary earnings forecasts have self-benefited motives before the major shareholders’ selling of original non-tradable shares and how the capital market reacts in China. Design/methodology/approach This paper uses multiple regression analyses to examine the influence of the major shareholders’ non-tradable shares selling motives on MEFs’ type and frequency of A-share listed companies and makes robust tests using the difference in difference model (DID). Findings In the paper, it is found that before the major shareholders’ selling of original non-tradable shares, managers of listed companies are prone to release positive voluntary MEFs; during the shares reduction year of the major shareholders, the disclosure frequency of MEFs is much higher; these forecasts before the major stockholders’ selling have significant higher excess market returns. The evidence suggests that voluntary positive MEFs are for the major shareholders’ self-interested motive rather than for the open, fair and just disclosure purpose that damages the allocation efficiency of the capital market. Originality/value This paper enriches the understanding of voluntary MEFs’ incentives literature and provides scientific evidence to improve the supervision of information disclosure and insider trading in Chinese security market.

Reliable drought prediction is fundamental for water resource managers to develop and implement drought mitigation measures. Considering that drought development is closely related to the spatial–temporal evolution of large-scale circulation patterns, we developed a conceptual prediction model of seasonal drought processes based on atmospheric and oceanic standardized anomalies (SAs). Empirical orthogonal function (EOF) analysis is first applied to drought-related SAs at 200 and 500 hPa geopotential height (HGT) and sea surface temperature (SST). Subsequently, SA-based predictors are built based on the spatial pattern of the first EOF modes. This drought prediction model is essentially the synchronous statistical relationship between 90-day-accumulated atmospheric–oceanic SA-based predictors and SPI3 (3-month standardized precipitation index), calibrated using a simple stepwise regression method. Predictor computation is based on forecast atmospheric–oceanic products retrieved from the NCEP Climate Forecast System Version 2 (CFSv2), indicating the lead time of the model depends on that of CFSv2. The model can make seamless drought predictions for operational use after a year-to-year calibration. Model application to four recent severe regional drought processes in China indicates its good performance in predicting seasonal drought development, despite its weakness in predicting drought severity. Overall, the model can be a worthy reference for seasonal water resource management in China.

Post-processing outputs from Coupled Global Circulation Models (CGCM) is required to generate skillful and reliable sub-seasonal precipitation forecasts. However, it is not currently known at what spatiotemporal scale can sub-seasonal precipitation forecasts be improved after post-processing. In this study, a Bayesian joint probability (BJP) method is used to post-process European Centre for Medium-Range Weather Forecasts (ECMWF) sub-seasonal precipitation forecasts at various spatiotemporal scales across China during the boreal summer monsoon. The relationships between forecasts and observations are built at daily, weekly, and fortnight temporal scales for each grid cell and each hydroclimatic region after normalization. The results suggest that the skills and reliability of ECMWF raw daily precipitation forecasts are improved after post-processing. The forecast skills are further improved at larger spatial scales or longer temporal scales, especially in the Upper and Middle Yangtze River, Southeast Rivers, and Pearl River. Although the forecast skills decrease rapidly as lead time increases, the uncertainty spread of the BJP calibrated sub-seasonal precipitation forecasts is highly reliable at all lead times and spatiotemporal scales. This work will help to realize the potential of CGCM forecasts at sub-seasonal time scales. Future works will investigate the benefits of post-processing precipitation forecasts for sub-seasonal streamflow predictions.

Seasonal pluvial-drought transition processes are unique natural phenomena. To explore possible mechanisms, we considered Southwest China (SWC) as the study region and comprehensively investigated the temporal evolution or spatial patterns of large-scale and regional atmospheric variables with the simple method of Standardized Anomalies (SA). Some key procedures and results include: (1) Because regional atmospheric variables are more directly responsible for the transition processes, we investigate it in detail. The temporal evolution of net vertical integral water vapor flux (net VIWVF) across SWC, together with vertical SA-based patterns of regional horizontal divergence (D) and vertical motion (ω), coincide well with pluvial-drought transition processes. (2) With respect to large-scale circulation patterns, a well-organized Eurasian (EU) Pattern is one important feature during the pluvial-drought transitions over SWC. (3) Based on these large-scale and regional atmospheric anomalous features, relevant SA-based indices were built, to explore the possibility of simulating drought development using previous pluvial anomalies. As a whole, simulated drought development only with SA-indices of large-scale circulation patterns does not perform well. Further, it can be improved a lot when SA-based indices of regional D and net VIWVF are introduced. (4) In addition, the potential drought prediction using pluvial anomalies, together with the deep understanding of physical mechanisms responsible for pluvial-drought transitions, need to be further explored.

A high-fat diet is a major risk factor for coronary heart diseases. Matrix metalloprotease (MMP) expression is changed in many cardiovascular diseases. Selenium, which is an important trace element in animals, has a close relationship with cardiovascular diseases. The TGFβ1/Smad signalling pathway is ubiquitous in diverse tissues and cells, and it is also associated with the occurrence and development of cardiovascular diseases. Therefore, in this study, we aimed to determine selenium's effect on lipid metabolism, atherosclerotic plaque formation, and MMP2 expression, as well as the underlying functional mechanism.In vivo tests: 24 male New Zealand white rabbits were randomly divided into 4 groups: regular diet, high-fat diet, high-fat diet+selenium and regular diet+selenium groups. The high-fat diet induced the lipid disturbances of rabbits at week 12. Selenium supplementation lowered total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C) and triglyceride (TG) levels (p0.01). Selenium supplementation also suppressed MMP2 over-expression in thoracic aortas. In vitro tests: Human umbilical vein endothelial cells (HUVECs) were treated with different concentrations of selenium or ox-LDL. Ox-LDL promoted MMP2 expression by increasing TGFβ1, pSmad2, pSmad3 and Smad3 expression (p0.01). Selenium attenuated MMP2 over-expression by regulating the TGFβ1/Smad signalling pathway.Selenium suppressed high-fat diet-induced MMP2 over-expression in vivo by improving lipid metabolism. In vitro, selenium attenuated MMP2 over-expression through the TGFβ1/Smad signalling pathway.

The Xijiang River is known as the Golden Watercourse because of its role in the development of the Pearl River Delta Regional Economic System in China, which was made possible by its abundant water resources. At present, the hydrological regime of the Xijiang River has now become complicated, the water shortages and successive droughts pose a threat to regional economic development. However, the complexity of hydroclimatological processes with emphasizes on drought has not been comprehended. In order to effectively predict and develop the adaptation strategies to cope with the water scarcity damage caused by hydrological droughts, it is essential to thoroughly analyze the relationship between hydrological droughts and pre/post-dependent hydroclimatological factors. To accomplish this, the extreme-point symmetric mode decomposition method (ESMD) was utilized to reveal the periodic variation in hydrological droughts that is characterized by the Standardized Drought Index (SDI). In addition, the cross-wavelet transform method was applied to investigate the correlation between large-scale climate indices and drought. The results showed that hydrological drought had the most significant response to spring ENSO (El Nino-Southern Oscillation), and the response lags in sub-basins were mostly 8–9 months except that in Yujiang River were mainly 5 or 8 months. Signal reservoir operation in the Yujiang River reduced drought severity by 52–95.8% from January to April over the 2003–2014 time period. Similarly, the cascade reservoir alleviated winter and spring droughts in the Hongshuihe River Basin. However, autumn drought was aggravated with severity increased by 41.9% in September and by 160.9% in October, so that the land surface models without considering human intervention must be used with caution in the hydrological simulation. The response lags of the VCI (Vegetation Condition Index) to hydrological drought were different in the sub-basins. The response lag for the Hongshuihe, Yujiang, and Liujiang River Basins were mostly 0–4 months, 0–1 months, and 2–3 months, respectively, but there was no obvious regular change pattern in the Guijiang River Basin.

Management of water resources may benefit from seasonal precipitation forecasts, but for obtaining high enough resolution, dynamical downscaling is necessary. This study investigated the downscaling capability of the Weather Research and Forecasting (WRF) Model ARW, version 3.5, on seasonal precipitation forecasts for the Hanjiang basin in China during 2001–09, which was the water source of the middle route of the South-to-North Water Diversion Project (SNWDP). The WRF Model is forced by the National Centers for Environmental Prediction Operational Climate Forecast System, version 2 (CFSv2), and it performs at a high horizontal resolution of 10 km with four selected convection schemes. The National Oceanic and Atmospheric Administration’s Climate Prediction Center global daily precipitation data were employed to evaluate the WRF Model on multiple scales. On average, when large biases were removed, the WRF Model slightly outperformed the CFSv2 in all seasons, especially summer. In particular, the Kain–Fritsch convective scheme performed best in summer, whereas little difference was found in winter. The WRF Model showed similar results in monthly precipitation, but no time-dependent characteristics were observed for all months. The spatial anomaly correlation coefficient showed greater uncertainty than the bias and the temporal correlation coefficient. In addition, the performance of the WRF Model showed considerable regional variations. The upper basin always showed better agreement with observations than did the middle and lower parts of the basin. A comparison of the forecast and observed daily precipitation revealed that the WRF Model can provide more accurate extreme precipitation information than the CFSv2.

Our previous study demonstrated that angiotensin II (Ang II) upregulates the expression of Kv1.5, a promising target for atrial fibrillation (AF) therapy, by activating ROS-dependent P-Smad2/3 and P-ERK 1/2. A recent study showed that hydrogen sulfide (H2S) may modulate the effects of angiotensin II (Ang II) by inhibiting the NADPH oxidase 4 (Nox4)-ROS signaling in the heart. The present study aimed to determine whether H2S is involved in the regulation of atrial Kv1.5 via ROS-related mechanisms in AF. Cultured neonatal rat atrial myocytes and a beagle model of AF were used for this study. In the neonatal rat atrial myocytes, quantitative PCR and enzyme immunoassays revealed that the mRNA expression levels of angiotensinogen, angiotensin-converting enzyme, and Ang II type I receptor (AT1R) and the Ang II supernatant concentration were significantly increased by hydrogen peroxide (H2O2) incubation, and these H2O2-induced alterations were reversed by diphenyleneiodonium, apocynin and H2S supplementation. Flow cytometry and Western blotting revealed that blockade of H2S biosynthesis using dl-propargylglycine increased ROS production and the expression of Ang II and Kv1.5. Sodium hydrosulfide (an exogenous H2S donor) and Nox4 siRNA inhibited Ang II-induced ROS production and Ang II-induced expression of Kv1.5, P-Smad2/3, P-ERK 1/2. Sodium hydrosulfide suppressed the Ang II-induced upregulation of Nox4. In our beagle AF model, 24 h of rapid atrial pacing (RAP) increased the atrial Ang II concentration, ROS production and the protein expression of Nox4, Kv1.5, P-Smad2/3 and P-ERK 1/2. These RAP-induced changes were inhibited by H2S supplementation and losartan (an AT1R blocker) pretreatment. In conclusion, our study indicates that H2S downregulates Ang II-induced atrial Kv1.5 expression by attenuating Nox4-related ROS-triggered P-Smad2/3 and P-ERK 1/2 activation during AF. H2S supplementation would be beneficial for AF treatment via the suppression of atrial Kv1.5 expression.
