Abstract

Chikungunya virus (CHIKV) is a mosquito-borne alphavirus that induces in humans a disease characterized by fever, rash, and pain in muscles and joints. The recent emergence or reemergence of CHIKV in the Indian Ocean Islands and India has stressed the need to better understand the pathogenesis of this disease. Previous CHIKV disease models have used young or immunodeficient mice, but these do not recapitulate human disease patterns and are unsuitable for testing immune-based therapies. Herein, we describe what we believe to be a new model for CHIKV infection in adult, immunocompetent cynomolgus macaques. CHIKV infection in these animals recapitulated the viral, clinical, and pathological features observed in human disease. In the macaques, long-term CHIKV infection was observed in joints, muscles, lymphoid organs, and liver, which could explain the long-lasting CHIKV disease symptoms observed in humans. In addition, the study identified macrophages as the main cellular reservoirs during the late stages of CHIKV infection in vivo. Abstract

Human adipose tissue-derived stem cells (hATSC) have been contemplated as reparative cells for cartilage engineering. Chondrogenic differentiation of hATSC can be induced by an enriched culture medium and a three-dimensional environment. Given that bone is vascularized and cartilage is not, oxygen tension has been suggested as a regulatory factor for osteochondrogenic differentiation. Our work aimed at determining whether hypoxia affects the osteochondrogenic potential of hATSC, hATSC were cultured in chondrogenic or osteogenic medium for 28 days, in pellets or monolayers, and under 5% or 20% oxygen tension. Cell differentiation was monitored by real-time PCR (COL2A1, aggrecan, Runx2, and osteocalcin). The chondrogenic differentiation was further evaluated by Alcian blue and immunohistological staining for glycosaminoglycans (GAGs) and type II collagen, respectively. Osteogenic differentiation was also assessed by the staining of mineralized matrix (Alizarin Red) and measurement of alkaline phosphatase (ALP) activity. The expression of chondrogenic markers was upregulated when hATSC were exposed to hypoxia in chondrogenic medium. Conversely, osteocalcin expression, mineralization, and ALP activity were severely reduced under hypoxic conditions even in the presence of osteogenic medium. Our data strongly suggest that hypoxia favors the chondrogenic differentiation of hATSC as evidenced by the expression of the chondrogenic markers, whereas it could alter their osteogenic potential. Our results highlight the differential regulatory role of hypoxia on the chondrogenic and osteogenic differentiation processes of hATSC. Abstract

Abstract Background Several cases of myopathies have been observed in the horse Norman Cob breed. Muscle histology examinations revealed that some families suffer from a polysaccharide storage myopathy (PSSM). It is assumed that a gene expression signature related to PSSM should be observed at the transcriptional level because the glycogen storage disease could also be linked to other dysfunctions in gene regulation. Thus, the functional genomic approach could be conducted in order to provide new knowledge about the metabolic disorders related to PSSM. We propose exploring the PSSM muscle fiber metabolic disorders by measuring gene expression in relationship with the histological phenotype. Results Genotypying analysis of GYS1 mutation revealed 2 homozygous (AA) and 5 heterozygous (GA) PSSM horses. In the PSSM muscles, histological data revealed PAS positive amylase resistant abnormal polysaccharides, inflammation, necrosis, and lipomatosis and active regeneration of fibers. Ultrastructural evaluation revealed a decrease of mitochondrial number and structural disorders. Extensive accumulation of an abnormal polysaccharide displaced and partially replaced mitochondria and myofibrils. The severity of the disease was higher in the two homozygous PSSM horses. Gene expression analysis revealed 129 genes significantly modulated (p < 0.05). The following genes were up-regulated over 2 fold: IL18, CTSS, LUM, CD44, FN1, GST01. The most down-regulated genes were the following: mitochondrial tRNA, SLC2A2, PRKCα, VEGFα. Data mining analysis showed that protein synthesis, apoptosis, cellular movement, growth and proliferation were the main cellular functions significantly associated with the modulated genes (p < 0.05). Several up-regulated genes, especially IL18, revealed a severe muscular inflammation in PSSM muscles. The up-regulation of glycogen synthase kinase-3 (GSK3β) under its active form could be responsible for glycogen synthase (GYS1) inhibition and hypoxia-inducible factor (HIF1α) destabilization. Conclusion The main disorders observed in PSSM muscles could be related to mitochondrial dysfunctions, glycogenesis inhibition and the chronic hypoxia of the PSSM muscles.

Abstract

These properties due to the multiphotonic excitation represent very promising approach to explore the state of various degenerative diseases associated with abnormal fibrotic and fat tissue. In this report, we use SHG/THG to explore the fibrotic and fat tissue in muscular dystrophy context using rat and dog model. By comparison with classical used immunolabeling dedicated to collagen, we confirmed that SHG microscopy has strong potentiality to follow the fibrosis content in the disease. We also demonstrated that THG can be used to follow the distribution of lipid bodies in cells and dystrophic tissue. Thanks to these properties, the combination of fluorescence and harmonic signals represent a promising way to monitor the course of the neuro muscular diseases as well as the efficacy of therapeutic strategies.

Abstract

International audience; Currently, the clinically most advanced strategy to treat Duchenne muscular dystrophy (DMD) is the exon skipping strategy. Whereas antisense oligonucleotide-based clinical trials are underway for DMD, it is essential to determine a dystrophin restoration threshold needed to ensure improvement of muscle physiology at the molecular level. A preclinical trial was recently conducted in golden retriever muscular dystrophy (GRMD) dogs treated in a forelimb by locoregional delivery of rAAV8-U7snRNA to promote exon skipping on the canine dystrophin messenger. Here, we exploited the rAAV8-U7snRNA transduced GRMD muscle samples, well-characterized for their percentage of dystrophin-positive fibers, in the aim to define a threshold of dystrophin rescue necessary for normalization of the status of the neuronal nitric oxide synthase mu (nNOSµ), the inducible nitric oxide synthase (iNOS), and the ryanodine receptor-calcium release channel type 1 (RyR1), crucial actors for an efficient contractile function. Results showed that the restoration of dystrophin in 40% of muscle fibers is needed to decrease the abnormal cytosolic nNOSµ expression and to reduce the overexpression of iNOS, these two parameters leading to a reduction of the NO level into the muscle fiber. Furthermore, the same percentage of dystrophin-positive fibers of 40 % was associated with the normalization of the RyR1 nitrosylation status and to a stabilization of the RyR1/calstabin1 complex that is required to facilitate coupled gating. We concluded that a minimal threshold of 40% of dystrophin-positive fibers is necessary for the reinstatement of central proteins needed for a proper muscle contractile function, and thus identified a rate of dystrophin expression significantly improving, at the molecular level, the dystrophic muscle physiology.

Abstract

Numerous studies have demonstrated the efficacy of the Adeno-Associated Virus (AAV)-based gene delivery platform in vivo. The control of transgene expression in many protocols is highly desirable for therapeutic applications and/or safety reasons. To date, the tetracycline and the rapamycin dependent regulatory systems have been the most widely evaluated. While the long-term regulation of the transgene has been obtained in rodent models, the translation of these studies to larger animals, especially to nonhuman primates (NHP), has often resulted in an immune response against the recombinant regulator protein involved in transgene expression regulation. These immune responses were dependent on the target tissue and vector delivery route. Here, using AAV vectors, we evaluated a doxycyclin-inducible system in rodents and macaques in which the TetR protein is fused to the human Kru ̈ ppel associated box (KRAB) protein. We demonstrated long term gene regulation efficiency in rodents after subretinal and intramuscular administration of AAV5 and AAV1 vectors, respectively. However, as previously described for other chimeric transactivators, the TetR-KRAB-based system failed to achieve long term regulation in the macaque after intramuscular vector delivery because of the development of an immune response. Thus, immunity against the chimeric transactivator TetR-KRAB emerged as the primary limitation for the clinical translation of the system when targeting the skeletal muscle, as previously described for other regulatory proteins. New developments in the field of chimeric drug-sensitive transactivators with the potential to not trigger the host immune system are still needed.

Abstract

Morphological features and chronology of myocytes changes after denervation were studied over 35 days period in 2 heavy (HW) and light-­weight (LW) strains of 5-­week-­old male turkeys. Denervation caused progressive atrophy in posterior latissimus dorsi (PLD). By day 28, the weight of the PLD muscle had reached about 62% of the non-­denervated contralateral muscle weight in both strains. On the contrary, ALD muscle mass increase progressively after denervation. Thus the maximum hypertrophy of the ALD, expressed as a percentage of contralateral muscle, was respectively about 67% and 37%,day 21 in the HW strain and day 28 in LW strain after denervation. ALD hypertrophy ceased apparently after day 21 (HW strain) and day 28 (LW strain). Morphometric analysis revealed that fast twitch (type II) fibers were atrophied after denervation, whereas slow tonic (type III) fibers were hypertrophied from day 7.Coagulative necrosis with fragmentation and lysis associated with moderate infiltration of inflammatory cells, were similar in both strains. Irregularities in mitochondrial distribution occurred mainly in type III fibers of ALD muscle at day 7. Seven and 14 days after denervation, immunolabelling of proliferating cell nuclear antigen (PCNA) revealed satellite cell activation in denervated muscles. The number of activated satellite cells was greater in the LW than HW mainly in ALD muscles

Abstract

Le formaldéhyde, fixateur des tissus biologiques, a été classé « cancérogène de groupe I » par le Centre International de Recherche sur le Cancer (CIRC). La réglementation française impose donc sa substitution, sous condition que cela soit techniquement possible. Nous avons évalué la possibilité de substituer le formol dans les laboratoires d'anatomie pathologique, en prenant compte des contraintes liées à une pratique vétérinaire, domaine d'exercice où les protections sont particulièrement peu répandues. Trois substituts aux compositions représentatives des différentes solutions sur le marché (acide acétique, glyoxal, mélange glyoxal/acide acétique) ont été testés, en comparaison au formol, sur un panel de dix tissus biologiques d’animaux. L'influence de la taille de l’échantillon et de la durée de fixation a aussi été envisagée. La qualité du prélèvement a été évaluée en aveugle, au moment de la recoupe des échantillons, de la coupe au microtome et de l'observation microscopique après coloration topographique de routine et de trois colorations spéciales. Tous les substituts utilisés possèdent des qualités de fixation suffisantes pour une observation histologique et permettent de poser un diagnostic de routine, après coloration Hémalun-Eosine-Safran (HES). Le substitut à base de glyoxal permet d'obtenir des préparations histologiques comparables à celles observées avec le formol, assurant même une meilleure qualité de fixation sur certains tissus réputés difficiles. Les essais d’immunomarquages ont révélé un signal interprétable pour trois anticorps, sur un panel de cinq utilisés en routine. Aucun substitut de fixation ne permet d'utiliser tous les anticorps testés sans une adaptation technique préalable. Chaque laboratoire devra donc évaluer la possibilité de substituer le formol en fonction des techniques qu'il utilise couramment.
