Abstract The electrocatalytic valorization of polyethylene terephthalate-derived ethylene glycol to valuable glycolic acid offers considerable economic and environmental benefits. However, conventional methods face scalability issues due to rapid activity decay of noble metal electrocatalysts. We demonstrate that a dynamic potential cycling approach, which alternates the electrode potential between oxidizing and reducing values, significantly mitigates surface deactivation of noble metals during electrochemical oxidation of ethylene glycol. This method enhances catalyst activity by 20 times compared to a constant-potential approach, maintaining this performance for up to 60 h with minimal deactivation. In situ Raman and X-ray absorption spectroscopy show that this effectiveness results from efficient removal of surface oxide during the reaction. The strategy is applicable to polyethylene terephthalate hydrolysates and various noble metals, such as palladium, gold, and platinum, with palladium showing a high conversion rate in recent studies. Our approach offers an efficient and durable method for electrochemical upcycling of biomass-derived compounds.

Abstract While chronic limb-threatening ischemia is a serious peripheral artery disease, the lack of an appropriate stent significantly limits the potential of interventional treatment. In spite of much progress in coronary stents, little is towards peripheral stents, which are expected to be both long and biodegradable and thus require a breakthrough in core techniques. Herein, we develop a long and biodegradable stent with a length of up to 118 mm based on a metal-polymer composite material. To achieve a well-prepared homogeneous coating on a long stent during ultrasonic spraying, a magnetic levitation is employed. In vivo degradation of the stent is investigated in rabbit abdominal aorta/iliac arteries, and its preclinical safety is evaluated in canine infrapopliteal arteries. First-in-man implantation of the stent is carried out in the below-the-knee artery. The 13 months’ follow-ups demonstrate the feasibility of the long and biodegradable stent in clinical applications.

Iron is considered as an attractive alternative material for bioresorbable scaffolds (BRS). The sirolimus eluting iron bioresorbable scaffold (IBS), developed by Biotyx Medical (Shenzhen, China), is the only iron-based BRS with an ultrathin-wall design. The study aims to investigate the long-term efficacy, safety, biocompatibility, and lumen changes during the biodegradation process of the IBS in a porcine model. A total of 90 IBSs and 70 cobalt-chromium everolimus eluting stents (EES) were randomly implanted into nonatherosclerotic coronary artery of healthy mini swine. The multimodality assessments including coronary angiography, optical coherence tomography, micro-computed tomography, magnetic resonance imaging, real-time polymerase chain reaction (PCR), and histopathological evaluations, were performed at different time points. There was no statistical difference in area stenosis between IBS group and EES group at 6 months, 1year, 2 years and 5 years. Although the scaffolded vessels narrowed at 9 months, expansive remodeling with increased mean lumen area was found at 3 and 5 years. The IBS struts remained intact at 6 months, and the corrosion was detectable at 9 months. At 5 years, the iron struts were completely degraded and absorbed in situ, without in-scaffold restenosis or thrombosis, lumen collapse, aneurysm formation, and chronic inflammation. No local or systemic toxicity and abnormal histopathologic manifestation were found in all experiments. Results from real-time PCR indicated that no sign of iron overload was reported in scaffolded segments. Therefore, the IBS shows comparable efficacy, safety, and biocompatibility with EES, and late lumen enlargement is considered as a unique feature in the IBS-implanted vessels.

Abstract Objective and background The early detection of diabetic ketoacidosis (DKA) in patients with type 2 diabetes (T2D) plays a crucial role in enhancing outcomes. We developed a nomogram prediction model for screening DKA in T2D patients. At the same time, the input variables were adjusted to reduce misdiagnosis. Methods We obtained data on T2D patients from Mimic-IV V0.4 and Mimic-III V1.4 databases. A nomogram model was developed using the training data set, internally validated, subjected to sensitivity analysis, and further externally validated with data from T2D patients in Aviation General Hospital. Results Based on the established model, we analyzed 1885 type 2 diabetes patients, among whom 614 with DKA. We further additionally identified risk factors for DKA based on literature reports and multivariate analysis. We identified age, glucose, chloride, calcium, and urea nitrogen as predictors in our model. The logistic regression model demonstrated an area under the curve (AUC) of 0.86 (95%CI: 0.85–0.90]. To validate the model, we collected data from 91 T2D patients, including 15 with DKA, at our hospital. The external validation of the model yielded an AUC of 0.68 (95%CI: 0.67–0.70). The calibration plot confirmed that our model was adequate for predicting patients with DKA. The decision-curve analysis revealed that our model offered net benefits for clinical use. Conclusions Our model offers a convenient and accurate tool for predicting whether DKA is present. Excluding input variables that may potentially hinder patient compliance increases the practical application significance of our model.

Abstract In situ exploration of the dynamic structure evolution of catalysts plays a key role in revealing reaction mechanisms and designing efficient catalysts. In this work, PtCu/MgO catalysts, synthesized via the co-impregnation method, outperforms monometallic Pt/MgO and Cu/MgO. Utilizing quasi/in-situ characterization techniques, it is discovered that there is an obvious structural evolution over PtCu/MgO from Pt x Cu y O z oxide cluster to PtCu alloy with surface CuO x species under different redox and CO oxidation reaction conditions. The synergistic effect between PtCu alloy and CuO x species enables good CO oxidation activity through the regulation of CO adsorption and O2 dissociation. At low temperatures, CO oxidation is predominantly catalyzed by surface CuO x species via the Mars-van Krevelen mechanism, in which CuO x can provide abundant active oxygen species. As the reaction temperature increases, both surface CuO x species and PtCu alloy collaborate to activate gaseous oxygen, facilitating CO oxidation mainly through the Langmuir-Hinshelwood mechanism.

PD-1/PD-L1 is an important signaling pathway in the adaptive immune system. The CD47/SIRPα signaling pathway is a crucial “do not eat me” signal for innate immunity. This study evaluated the anti-tumor mechanism of IMM2520 in vitro and in vivo. IMM2520 was generated using the “mab-trap” platform. IMM2520 showed high affinity to PD-L1 and relatively lower affinity to CD47, displaying preferential binding to PD-L1 on tumor cells. IMM2520 had the potent ability to inhibit the PD-1/PD-L1 and CD47/SIRPα signaling pathways and killed tumor cells through ADCC and ADCP. Importantly, IMM2520 did not bind to human red blood cells or induce erythrocyte agglutination. IMM2520 demonstrated a tendency to bind to CD47+/PD-L1+ tumor cells, reducing its binding to CD47 single-positive cells. In mouse transplantation models, compared with the first-generation CD47/PD-L1 BsAb (IMM2505), IMM2520 exhibited stronger and dose-dependent antitumor activity. These findings imply that IMM2520 may offer a novel therapeutic alternative for cancer patients.

Land Use/ Cover Change (LUCC) plays a crucial role in influencing hydrological processes, nutrient cycling, and sediment transport in watersheds, ultimately impacting water quality on both spatial and temporal scales. Accurately predicting changes in watershed water quality is beneficial for the sustainable management of water resources. Current models often lack the ability to effectively predict water quality changes in a dynamic spatio-temporal context, particularly in complex watershed environments. The overall purpose of the study is to establish a comprehensive and dynamic modeling framework that links LUCC with water quality, allowing for accurate predictions of future water quality under varying land use scenarios. The model, which uses water quality as the dependent variable and LUCC as the independent variable, was developed to quantitatively predict changes in watershed water quality. To achieve this, annual multi-period remote sensing images from Landsat-5, Landsat-8 or Sentinel-2 satellites spanning from 1992 to 2022 were analyzed. Random Forest (achieving a Kappa coefficient of 0.9468) were employed to classify land use within the watershed. Based on classification results, a Cellular Automata-Markov chain model (CA-Markov) was constructed to simulate and predict the spatio-temporal patterns of land use, incorporating driving factors such as proximity to water systems, roads, elevation, and slope. Validation of the model using LUCC data from 2020 yielded a high prediction accuracy with a Kappa coefficient of 0.9505. The CA-Markov model was further utilized to project LUCC under three different scenarios—natural development, ecological protection, and arable land protection—between 2023 and 2033. Based on these projections, the coupled water quality and LUCC model was employed to predict water quality changes in the watershed over the same period. Key findings indicate that water quality is likely to improve under ecological protection scenario, while deterioration is expected under natural development scenario and cropland protection scenario due to urban expansion, agricultural practices, and water diversion for irrigation. This study provides a robust framework for watershed management, offering scientific guidance for source management and water purification efforts, thereby contributing significantly to the sustainable development of water resources.

Background and objective Lung adenocarcinoma (LUAD) is a highly morbid and fatal cancer. Despite advancements in modern medical treatment, the 5-year survival rate of patients remains suboptimal. Our previous study revealed that zinc finger SWIM-type containing 1 (ZSWIM1), a novel protein, promotes the proliferation, migration, and invasion of LUAD cells. The aim of this study is to investigate the impact of E3 ubiquitin ligase tripartite motif protein 21 (TRIM21) on ZSWIM1-mediated cell proliferation and migration. Methods The interaction and co-localization between TRIM21 and ZSWIM1 were verified using co-immunoprecipitation (Co-IP) and immunofluorescence (IF). The effects of TRIM21 and ZSWIM1 on the proliferation and migration of LUAD cells were assessed through MTT and Transwell assays, respectively. Western blot (WB) analysis was conducted to evaluate the impact of TRIM21 and ZSWIM1 on the expression of epithelial-mesenchymal transition (EMT) markers in LUAD cells. The influence of TRIM21 on the ubiquitination of ZSWIM1 was examined using Co-IP combined with WB. Results TRIM21 was found to interact and co-localize with ZSWIM1. Overexpression of TRIM21 inhibited the proliferation and migration of LUAD cells. Overexpression of TRIM21 reduced the promoting effect of ZSWIM1 on the proliferation, migration, and invasion of lung adenocarcinoma cells, and reversed the impact of ZSWIM1 on the expression of E-cadherin and Vimentin. Conversely, knockdown of TRIM21 further enhanced the promoting effect of ZSWIM1 on the proliferation and migration of LUAD cells. Mechanistically, we observed that overexpression of TRIM21 significantly enhanced the ubiquitination level of ZSWIM1, leading to a decrease in ZSWIM1 protein expression. Conclusion TRIM21 binds to and promotes the ubiquitination of ZSWIM1, resulting in reduced protein expression of ZSWIM1, which leads to the inhibition of ZSWIM1-mediated promotion of proliferation, migration, and invasion in LUAD cells.

The forest fire environment is complex and volatile, posing a serious threat to the lives of firefighters at any time. Aiming at how people choose escape routes when facing these dangers, this paper proposed an escape route planning method based on the improved A* algorithm. Taking the forest fire that occurred in Xintian County, Yongzhou City, Hunan Province on October 17, 2022, as the research object, this study collected data from multi-temporal remote sensing imagery including GF-4, and Sentinel-2 to obtain 11 factors affecting the escape route planning. We used the FAHP-CRITIC combination weighting method to analyze the weights of the escape impact factors. Forest fire scenes were categorized into five classes based on escape risk coefficient from high to low. The heuristic function and weight coefficients of the traditional A* algorithm were reconstructed to obtain the improved A* algorithm. The computing time of both A* algorithm is similar in the same fire scene. But the percentage of the escape route length, located inside the high-risk zones, in the total escape route planned by the improved algorithm decreased by 53.63% than that planned by the traditional A* algorithm. The escape risk coefficient was reduced by 24.22%, and the escape safety was significantly improved. On this basis, the study combined the improved A* algorithm with the dynamic escape window method to search for the nearest safe area to the escapees and compute the corresponding escape routes. The real-time safety of the algorithm was verified using GF-1 remote sensing images obtained 7 and 35 min after the previous fire burning moment. Results revealed that the escape paths planned by the improved A* algorithm remained in a safe state and were able to be updated in real time according to the trend of the fire. This demonstrates the ability of the improved A* algorithm to adapt to real-time changes in forest fire scenes. It can provide multiple reliable escape options for escapees, thus scientifically and effectively reducing human casualties.

IntroductionThe behavior of forest fire is a complex phenomenon, and accurate simulation of forest fire is conducive to emergency response management after ignition. In order to further understand the characteristics of forest fire spread and the applicability of WRF-Fire in China, which is a coupled fire-atmospheric wildfire model, this study simulated a high-intensity forest fire event that occurred on October 17, 2022 in Xintian County, southern Hunan Province.MethodsBased on the fire-atmosphere coupled WRF-Fire model, we used high-resolution geographic information, meteorological observation and fuel classification data to analyze the forest fire behavior. At the same time, the simulation results are compared with the fire burned area observed by satellite remote sensing forest fire monitoring data.ResultsThe study found that, the simulated wind speed, direction and temperature trends are similar to the observation results, but the simulated wind speed is overestimated, the dominant wind direction is N, and the temperature is slightly underestimated. The simulated wind field is close to the actual wind field, and the simulation results can show the spatial and temporal variation characteristics of the local wind field under complex terrain while obtaining the high-resolution wind field. The simulated fire burned area is generally overestimated, spreading to the north and southwest compared with the observed fires, but it can also capture the overall shape and spread trend of the fire well.DiscussionThe results show that the model can accurately reproduce the real spread of fire, and it is more helpful to forest fire management.

Forest fires can occur suddenly and have significant environmental, economic, and social consequences. The timely and accurate evaluation and prediction of their progression, particularly the spread speed in difficult-to-access areas, are essential for emergency management departments to proactively implement prevention strategies and extinguish fires using scientific methods. This paper provides an analysis of models for predicting forest fire spread in China and globally. Incorporating remote sensing (RS) technology and forest fire science as the theoretical foundation, and utilizing the Wang Zhengfei forest fire spread model (1983), which is noted for its broad adaptability in China as the technical framework, this study constructs a forest fire spread model based on remote sensing interpretation. The model improves the existing model by adding elevation an factor and optimizes the method for acquiring certain parameters. By considering regional landforms (ridge lines, valley lines, and slopes) and vegetation coverage, this paper establishes three-dimensional visual interpretation markers for identifying hotspots; the orientation of the hotspots can be identified to simulate the spread of the fire uphill, downhill, in the direction of the wind, left-level slope, and right-level slope. Then, the data of Sentinel-2 and DEM were used to invert the fuel humidity and slope of pixels in the fire line areas. The statistical inversion data from pixels, which replaced fixed-point values in traditional models, were utilized for predicting forest fire spread speed. In this paper, the model was applied to the case of a forest fire in Mianning County, Sichuan Province, China, and verified using high-time-resolution Himawari-8 data, Gaofen-4 data, and historical data. The results demonstrate that the direction and maximum speed of fire spread for the fire lines in Baifen Mountai, Jiaguer Villageand, Muchanggou, Xujiabaozi, and Zhaizigou are uphill, 16.5 m/min; wind direction, 17.32 m/min; wind direction, 1.59 m/min; and wind direction, 5.67 m/min. The differences are mainly due to the locations of the fire lines, moisture content of combustibles, and maximum slopes being different. Across the entire fire line area, the average rate of increase in the area of open flames within one hour was 3.257 hm2/10 min (square hectares per 10 min), closely matching the average increase rate (3.297 hm2/10 min) monitored by the Himawari-8 satellite in 10 min intervals. In contrast, conventional fixed-point fire spread models predicted an average rate of increase of 3.5637 hm2/10 min, which shows a larger discrepancy compared to the Himawari-8 satellite monitoring results. Moreover, when compared to the fire spot monitoring results from the Gaofen-4 satellite taken 54 min after the initial location of the fire line, the predictions from the RS-enabled fire spread model, which integrates remote sensing interpretations, closely matched the actual observed fire boundaries. Although the predictions from the RS-enabled fire spread model and the traditional model both align with historical data in terms of the overall fire development trends, the RS-enabled model exhibits higher reliability and can provide more accurate information for forest fire emergency departments, enabling effective pre-emptive measures and scientific firefighting strategies.

BackgroundCD38 and CD47 are expressed in many hematologic malignancies, including multiple myeloma (MM), B-cell non-Hodgkin lymphoma (NHL), B-cell acute lymphoblastic leukemia (ALL), and B-cell chronic lymphocytic leukemia (CLL). Here, we evaluated the antitumor activities of CD38/CD47 bispecific antibodies (BsAbs).MethodsFive suitable anti-CD38 antibodies for co-targeting CD47 and CD38 BsAb were developed using a 2 + 2 “mAb-trap” platform. The activity characteristics of the CD38/CD47 BsAbs were evaluated using in vitro and in vivo systems.ResultsUsing hybridoma screening technology, we obtained nine suitable anti-CD38 antibodies. All anti-CD38 antibodies bind to CD38+ tumor cells and kill tumor cells via antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP). Five anti-CD38 antibodies (4A8, 12C10, 26B4, 35G5, and 65A7) were selected for designing CD38/CD47 BsAbs (IMM5605) using a “mAb-trap” platform. BsAbs had higher affinity and binding activity to the CD38 target than those to the CD47 target, decreasing the potential on-target potential and off-tumor effects. The CD38/CD47 BsAbs did not bind to RBCs and did not induce RBC agglutination; thus, BsAbs had much lower blood toxicity. The CD38/CD47 BsAbs had a greater ability to block the CD47/SIRPα signal in CD38+/CD47+ tumor cells than IMM01 (SIRPα Fc fusion protein). Through Fc domain engineering, CD38/CD47 BsAbs were shown to kill tumors more effectively by inducing ADCC and ADCP. IMM5605–26B4 had the strongest inhibitory effect on cellular CD38 enzymatic activity. IMM5605–12C10 had the strongest ability to directly induce the apoptosis of tumor cells. The anti-CD38 antibody 26B4 combined with the SIRPα-Fc fusion proteins showed strong antitumor effects, which were better than any of the mono-therapeutic agents used alone in the NCI-H929 cell xenograft model. The CD38/CD47 BsAbs exhibited strong antitumor effects; specifically, IMM5605–12C10 efficiently eradicated all established tumors in all mice.ConclusionA panel of BsAbs targeting CD38 and CD47 developed based on the “mAb-tarp” platform showed potent tumor-killing ability in vitro and in vivo. As BsAbs had lower affinity for binding to CD47, higher affinity for binding to CD38, no affinity for binding to RBCs, and did not induce RBC agglutination, we concluded that CD38/CD47 BsAbs are safe and have a satisfactory tolerability profile.

Forest disaster fires pose a serious threat to forest resources and people's lives and property, and may also cause ecological disasters and social crises. There is a phenomenon of forest fire omission error, due to combustibles incomplete combustion in the forest fires early stage, low ground temperature and weak infrared radiation energy, and influence of forest canopy shading. Smoke detection based on satellite imagery is imperative for forest fire detection. Therefore, when forest fire occurs, forest fire smoke detection combined with forest fire infrared radiation monitoring can greatly reduce the phenomenon of forest fire omission error. Meteorological satellite imagery is commonly used for near real-time forest fire monitoring, thanks to its high temporal resolution. However, due to the low spatial resolution of meteorological satellite imagery and a large number of mixed pixels, it cannot accurately locate forest fire, and we introduced the concept of subpixel mapping, which is studied in detail in the article (https://doi.org/10.3390/rs14102460) we published. In this paper, the Himawari-9 satellite imagery is used to detect forest fire smoke at subpixel scale based on the Modified Pixel Swapping Algorithm (MPSA) and detect forest fire infrared radiation at subpixel scale based on the Mixed-Pixel Unmixing integrated with Pixel-Swapping Algorithm (MPU-PSA), respectively, obtaining forest fire smoke detection and infrared radiation monitoring results, and average value of forest fire identification accuracy is 41.78 % and 83.96 %, respectively. The Threshold-Weighted Fusion (TWF) method is used to fuse subpixel scale forest fire smoke detection result and forest fire infrared radiation monitoring result, and average value of forest fire identification accuracy is increased from 83.96 % (forest fire infrared radiation monitoring alone) to 93.97 %. Results show that fusion of subpixel scale forest fire smoke detection result and forest fire infrared radiation monitoring result can greatly improve spatial positioning accuracy of forest fire monitoring and reduce phenomenon of forest fire omission error.

Amidst the escalating threat of global warming, which manifests in more frequent forest fires, the prompt and accurate detection of forest fires has ascended to paramount importance. The current surveillance algorithms employed for forest fire monitoring—including, but not limited to, fixed threshold algorithms, multi-channel threshold algorithms, and contextual algorithms—rely primarily upon the degree of deviation between the pixel temperature and the background temperature to discern pyric events. Notwithstanding, these algorithms typically fail to account for the spatial heterogeneity of the background temperature, precipitating the consequential oversight of low-temperature fire point pixels, thus impeding the expedited detection of fires in their initial stages. For the amelioration of this deficiency, the present study introduces a spatial feature-based (STF) method for forest fire detection, leveraging Himawari-8/9 imagery as the main data source, complemented by the Shuttle Radar Topography Mission (SRTM) DEM data inputs. Our proposed modality reconstructs the surface temperature information via selecting the optimally designated machine learning model, subsequently identifying the fire point through utilizing the difference between the reconstructed surface temperatures and empirical observations, in tandem with the spatial contextual algorithm. The results confirm that the random forest model demonstrates superior efficacy in the reconstruction of the surface temperature. Benchmarking the STF method against both the fire point datasets disseminated by the China Forest and Grassland Fire Prevention and Suppression Network (CFGFPN) and the Wild Land Fire (WLF) fire point product validation datasets from Himawari-8/9 yielded a zero rate of omission errors and a comprehensive evaluative index, predominantly surpassing 0.74. These findings show that the STF method proposed herein significantly augments the identification of lower-temperature fire point pixels, thereby amplifying the sensitivity of forest surveillance.

Satellite remote sensing has become an important means of forest fire monitoring because it has the advantages of wide coverage, few ground constraints and high dynamics. When utilizing satellites for forest fire hotspot monitoring, two types of ground hotspots, agricultural and other fire hotspots can be ruled out through ground object features. False forest fire hotspots within forested areas must be excluded for a more accurate distinction between forest fires and non-forest fires. This study utilizes spatio-temporal data along with time-series classification to excavate false forest fire hotspots exhibiting temporal characteristics within forested areas and construct a dataset of such false forest fire hotspots, thereby achieving a more realistic forest fire dataset. Taking Hunan Province as the research object, this study takes the satellite ground hotspots in the forests of Hunan Province as the suspected forest fire hotspot dataset and excludes the satellite ground hotspots in the forests such as fixed heat sources, periodic heat sources and recurring heat sources which are excavated. The validity of these methods and results was then analyzed. False forest fire hotspots, from satellite ground hotspots extracted from 2019 to 2023 Himawari-8/9 satellite images, closely resemble the official release of actual forest fires data and the accuracy rate in the actual forest fire monitoring is 95.12%. This validates that the method employed in this study can improve the accuracy of satellite-based forest fire monitoring.

Objective To explore the clinical features, diagnosis and treatment regimen of adrenal cryptococcosis so as to improve awareness of this disease for clinicians. Methods The clinical manifestations, laboratory indicators, imaging data and postoperative pathological results of a patient with adrenocortical insufficiency caused by Cryptococcus neoformans infection admitted to our hospital in June 2022 were collected and analyzed, and the related literature was reviewed. Results This patient presented with dizziness, fatigue, and multiple pigmentations throughout the whole body.Upper abdominal plain CT scan on June 24, suggested multiple nodular soft tissue shadows in the bilateral adrenal glands, and the scan on July 1 suggested mild inhomogeneous enhancement of bilateral adrenal nodules, which may be adrenal tumors.Retroperitoneal laparoscopic partial adrenalectomy was performed on July 13.Postoperative pathology indicated cryptococcosis.The antifungal treatment with itraconazole was stopped after 2 weeks by himself.In 3 months after the operation, CT revealed multiple adrenal masses on both sides, and itraconazole was taken again for treatment. Conclusion Patients with adrenal cryptococcosis usually have symptoms of adrenocortical insufficiency such as dizziness, fatigue, and multiple pigmentations throughout the whole body.Abdominal enhanced CT usually indicates adrenal nodules with inhomogeneous enhancement.The diagnosis depends on pathological examination, and antifungal therapy can be considered after adrenalectomy.

Abstract Aim To examine the demographic characteristics associated with stress response of fever outpatients and children's families during normalisation of the COVID‐19 epidemic and to examine the relationship between stress response, coping style and resilience. Design Online cross‐sectional study. Methods A total of 541 fever clinic participants from Yiwu, China, were recruited via WeChat from February to November 2021. Online self‐administered questionnaires were used to collect data. Data were analysed using t‐tests, one‐way analyses of variance, Pearson's correlation analyses and multiple linear regression analyses. Results There were apparent physical and emotional responses among the fever outpatients, especially the adult patients. The main coping style was negative coping, and the degree of psychological resilience was low. Income, comorbidities, religious beliefs, tenacity, negative coping and positive coping were independent predictors of stress response.

With rapid social and economic development, land use/land cover change (LUCC) has intensified with serious impacts on water quality in the watershed. In this study, we took Dongjiang Lake watershed as the study area and obtained measured data on water quality parameters from the watershed’s water quality monitoring stations. Based on Landsat-5, Landsat-8, or Sentinel-2 remote sensing data for multiple periods per year between 1992 and 2022, the sensitive satellite bands or band combinations of each water quality parameter were determined. The Random Forest method was used to classify the land use types in the watershed into six categories, and the area proportion of each type was calculated. We established machine learning regression models and polynomial regression models with WQI as the dependent variable and the area proportion of each land use type as the independent variable. Accuracy test results showed that, among them, the quadratic cubic polynomial regression model with grassland, forest land, construction land, and unused land as its independent variables was the best model for coupling watershed water quality with LUCC. This study’s results provide a scientific basis for monitoring spatial and temporal changes in water quality caused by LUCC in the Dongjiang Lake watershed.

The pull-out method is a simple and effective method for detecting the preload of suspension bridge cable clamp bolts. However, research on the pull-out method is currently limited. The force principles governing the bolt during the pulling process are unclear, and the relationship between tension force and the desired preload remains uncertain. This paper aims to explore the force principles of bolts during the pull-out method detection process through a combined approach of theoretical analysis, full-scale test, and finite element simulation. The results indicate that the bolt preload increases during the pulling process. The preload detected by the pull-out method is not the initial preload of the bolt, but rather it exceeds the initial preload. The force relationships among various components are determined as follows: the preload subtracts the change value of the force exerted by the nut at the tension end, which equals the change value of tension force. Additionally, an analysis of the impact of the length of the bolt clamping section and the bolt area on the preload was conducted. Under the same bolt area, a shorter clamping section length corresponds to a greater increase in preload. With the same clamping section length, the increment of preload increases with the bolt area. These findings can serve as references for detecting and specifying the preload of the bolts.

Abstract Tumor necrosis factor receptors (TNFRs) are promising targets for cancer therapy. However, activating their downstream signaling requires cross‐linking of TNFRs. Herein, to devise strong agonists of TNFRs, ligands targeting TNFRs, such as OX40L and tumor necrosis factor‐related apoptosis‐inducing ligand (TRAIL), were fused with a multivalent protein scaffold (MV) to prepare multivalent agonists for cross‐linking TNFRs. The nano‐clustered multivalent‐OX40L (MV‐OX40L) and MV‐TRAIL could promote T cell activation and directly induce tumor cell apoptosis. Moreover, to develop a universal nano‐adaptor for the rapid preparation of multivalent agonists of different TNFRs, the Fc receptor that could immobilize antibodies was fused with MV to prepare MV‐FcR, which could multimerize commercial agonist antibodies targeting TNFRs, such as anti‐OX40 antibody (αOX40). Simply incubating αOX40 with MV‐FcR could prepare MV‐αOX40 to enhance its antitumor efficacy. In addition, MV‐FcR could multimerize with other therapeutic antibodies, such as anti‐PD‐L1 antibody, to enhance their valency. This study provides a promising strategy for engineering multivalent antitumor protein drugs.

ObjectiveTo analyze the effect of different types of disinfectants for perineum and urethral meatus cleaning in preventing catheter-associated asymptomatic bacteriuria and catheter-associated urinary tract infection (CAUTI).MethodsChinese and English databases were searched to collect randomized controlled trials (RCTs) of different disinfectants for perineum and urethral meatus cleaning to prevent CAUTI, and the positive rates of urine culture with different cleaning methods were contrasted by network meta-analysis.ResultsA total of 18 RCTs were included in this study to analyze the effect of 10 disinfectants in preventing CAUTI. The disinfectants were 0.1% chlorhexidine (CHG), 0.5% CHG, 2% CHG, 0.1% benzalkonium bromide (DBDAB), 0.05% iodophor, 0.5% iodophor, acidic oxidizing potential water (EOW), normal saline (NS), Shehuang lotion, and sterile water. Meta-analysis showed that the positive rates of urine culture in the 0.5% CHG cluster, EOW cluster, 0.5% iodophor cluster, and Shehuang lotion cluster were remarkably lower than that in the 0.1% DBDAB cluster (P < 0.05). The positive rate of urine culture in the 0.5% CHG cluster was remarkably lower than that in the 0.05% iodophor cluster (P < 0.05), whereas the positive rates of urine culture in the 0.5% CHG cluster, 0.5% iodophor cluster, Shehuang lotion cluster, and EOW were remarkably lower than that in the normal saline cluster (P < 0.05). The positive rate of urine culture in the 0.5% CHG cluster and EOW cluster were remarkably lower than those in the sterile water cluster (P < 0.05). The results of the area under the cumulative ranking probability plot (SUCRA) analysis showed that the probability ranking of the preventive effect of different disinfectants was as follows: 0.5% CHG > Shehuang lotion > EOW > 0.5% Iodophor > 2% CHG > 0.1% CHG > 0.05% Iodophor > Sterile Water > 0.1% DBDAB > Saline. Conclusion0.5% CHG, Shehuang lotion, EOW, and 0.5% iodophor can be used to clean the perineum and urethral meatus in patients with indwelling catheters.Systematic review registrationChinese Clinical Trial Registry (ChiCTR), identifier ChiCTR2100052260.

BackgroundA significant level of CD70 can be detected in various types of tumor tissues and CD27 is expressed on Treg cells, but CD70 expression is low in normal tissues. The interaction between CD70 and CD27 can stimulate the proliferation and survival of cancer cells and increase the level of soluble CD27, which is associated with poor prognosis in patients with lymphoma and certain solid tumors. Thus, it is a promising therapeutic target for the treatment of many major CD70+ cancer indications, including CD70+ lymphoma, RCC, NSCLC, HNSCC and OC.MethodsIMM40H was obtained through hybridoma screening and antibody humanization techniques. IMM40H was evaluated for its binding, blocking, Fc-dependent effector functions and antitumor activity characteristics in various in vitro and in vivo systems. The safety and tolerability profile of IMM40H were evaluated through single and repeated administration in cynomolgus monkeys.ResultsIn vitro cell-based assays demonstrated that IMM40H had considerably stronger CD70-binding affinity than competitor anti-CD70 antibodies, including cusatuzumab, which enabled it to block the interaction of between CD70 and CD27 more effectively. IMM40H also exhibited potent Fc-dependent effector functions (ADCC/CDC/ADCP), and could make a strong immune attack on tumor cells and enhance therapeutic efficacy. Preclinical findings showed that IMM40H had potent antitumor activity in multiple myeloma U266B1 xenograft model, and could eradicate subcutaneously established tumors at a low dose of 0.3 mg/kg. IMM40H (0.3 mg/kg) showed therapeutic effects faster than cusatuzumab (1 mg/kg). A strong synergistic effect between IMM01 (SIRPα-Fc fusion protein) and IMM40H was recorded in Burkitt’s lymphoma Raji and renal carcinoma cell A498 tumor models. In cynomolgus monkeys, the highest non-severely toxic dose (HNSTD) for repeat-dose toxicity was up to 30 mg/kg, while the maximum tolerated dose (MTD) for single-dose toxicity was up to 100 mg/kg, confirming that IMM40H had a good safety and tolerability profile.ConclusionIMM40H is a high-affinity humanized IgG1 specifically targeting the CD70 monoclonal antibody with enhanced Fc-dependent activities. IMM40H has a dual mechanism of action: inducing cytotoxicity against CD70+ tumor cells via various effector functions (ADCC, ADCP and CDC) and obstructs the proliferation and activation of Tregs by inhibiting CD70/CD27 signaling.

Abstract Aim This study aimed to evaluate the effects of using the teach-back method among women with limited maternal health literacy (LMHL) on maternal health literacy(MHL), postpartum health behaviours and maternal-infant health outcomes. Methods A randomized controlled study was conducted in the obstetrics department of Anhui Provincial Hospital, China. A total of 258 pregnant women with LMHL were recruited at the point of admission to the hospital for birth and randomly assigned to the control group (n = 130), where women received routine education sessions, and the teach-back group (n = 128), where women received routine education sessions plus a teach-back intervention. The two groups were assessed in terms of MHL before and after the intervention, breastfeeding execution, uptake of 42-day postpartum check-ups, complete uptake of one-time recommended vaccines, and physical health outcomes. Statistical tests were employed for data analysis. Results There was no significant difference between the two groups in terms of MHL and other social, demographic, and medical status at baseline. Abstract A novel recombinant SIRPα-Fc fusion protein, IMM01, was constructed and produced using an in-house developed CHO-K1 cell expression system, and the anti-tumor mechanism of IMM01 targeting the CD47-SIRPα pathway was explored. The phagocytosis and in vitro anti-tumor activity of IMM01 were evaluated by antibody-dependent cellular phagocytosis (ADCP), antibody-dependent cell-mediated cytotoxicity (ADCC), and complement-dependent cytotoxicity (CDC) assays. In vivo mouse tumor model studies were used to explore therapeutic efficacy as well as the mechanism of action. An in vitro binding assay revealed that IMM01 has a strong binding affinity to CD47 with an EC50 of 0.4967 nM. IMM01 can induce strong ADCP and moderate ADCC, but not CDC. IMM01-induced strong phagocytosis against tumor cells was attributed to dual activities of blocking the "don’t eat me" signal and activating the "eat me" signal, and IMM01 exhibits strong and robust in vivo anti-tumor activities either as monotherapy on hematological malignancies, or in combination therapy with PD-L1 monoclonal antibody (mAb), PD-1 mAb, and HER-2 mAb on solid tumors. Finally, IMM01 demonstrated a favorable safety profile with no human RBC binding activity or hemagglutination induction. IMM01 inhibits the growth of tumor cells by the following three possible mechanisms: (1) directly activating macrophages to phagocytize tumor cells; (2) activated macrophages degrade phagocytized tumor cells and present tumor antigens to T cells through MHC molecules to activate T cells; (3) activated macrophages can convert "cold tumors" into "hot tumors" and increase the infiltration of immune cells through chemotaxis by secreting some cytokines and chemokines.

This study aimed to investigate the long-term biocompatibility, safety, and degradation of the ultrathin nitrided iron bioresorbable scaffold (BRS) in vivo, encompassing the whole process of bioresorption in porcine coronary arteries. Fifty-two nitrided iron scaffolds (strut thickness of 70 μm) and 28 Vision Co–Cr stents were randomly implanted into coronary arteries of healthy mini-swine. The efficacy and safety of the nitrided iron scaffold were comparable with those of the Vision stentwithin 52 weeks after implantation. In addition, the long-term biocompatibility, safety, and bioresorption of the nitrided iron scaffold were evaluated by coronary angiography, optical coherence tomography, micro-computed tomography, scanning electron microscopy, energy dispersive spectrometry and histopathological evaluations at 4, 12, 26, 52 weeks and even at 7 years after implantation. In particular, a large number of struts were almost completely absorbed in situ at 7 years follow-up, which were first illustrated in this study. The lymphatic drainage pathway might serve as the potential clearance way of iron and its corrosion products.

This study used the ITS approach based on Illumina MiSeq sequencing to assess the endosphere and rhizosphere fungal communities in healthy and diseased faba bean plants. The findings indicate that the most predominant phyla in all samples were Ascomycota (49.89–99.56%) and Basidiomycota (0.33–25.78%). In healthy endosphere samples, Glomeromycota (0.08–1.17%) was the only predominant phylum. In diseased endosphere samples, Olpidiomycota (0.04–1.75%) was the only predominant phylum. At the genus level, Penicillium (0.47–35.21%) was more abundant in rhizosphere soil, while Paraphoma (3.48–91.16%) was predominant in the endosphere roots of faba bean plants. Significant differences were observed in the alpha diversity of rhizosphere samples from different germplasm resources (p < 0.05). The fungal community structures were clearly distinguished between rhizosphere and endosphere samples and between healthy and diseased endosphere samples (p < 0.05). Saccharomyces was significantly enriched in diseased endosphere samples, whereas Apiotrichum was enriched in healthy endosphere samples. Vishniacozyma and Phialophora were enriched in diseased rhizosphere samples, while Pseudogymnoascus was enriched in healthy rhizosphere samples. Diseased samples displayed more strongly correlated genera than healthy samples. Saprotrophs accounted for a larger proportion of the fungal microbes in rhizosphere soil than in endosphere roots. This study provides a better understanding of the composition and diversity of fungal communities in the rhizosphere and endosphere of faba bean plants as well as a theoretical guidance for future research on the prevention or control of faba bean root rot disease.

Abstract Poststent restenosis is caused by insufficient endothelialization and is one of the most serious clinical complications of stenting. We observed a rapid endothelialization rate and increased fibrin deposition on the surfaces of the corroded iron stents. Thus, we hypothesized that corroded iron stents would promote endothelialization by increasing fibrin deposition on rough surfaces. To verify this hypothesis, we conducted an arteriovenous shunt experiment to analyze fibrin deposition in the corroded iron stents. We implanted a corroded iron stent in both the carotid and iliac artery bifurcations to elucidate the effects of fibrin deposition on endothelialization. Co‐culture experiments were conducted under dynamic flow conditions to explore the relationship between fibrin deposition and rapid endothelialization. Our findings indicate that, from the generation of corrosion pits, the surface of the corroded iron stent was rough, and numerous fibrils were deposited in the corroded iron stent. Fibrin deposition in corroded iron stents facilitates endothelial cell adhesion and proliferation, which, in turn, promotes endothelialization after stenting. Our study is the first to elucidate the role of iron stent corrosion in endothelialization, pointing to a new direction for preventing clinical complications caused by insufficient endothelialization.

Forest fires seriously jeopardize forestry resources and endanger people and property. The efficient identification of forest fire smoke, generated from inadequate combustion during the early stage of forest fires, is important for the rapid detection of early forest fires. By combining the Convolutional Neural Network (CNN) and the Lightweight Vision Transformer (Lightweight ViT), this paper proposes a novel forest fire smoke detection model: the SR-Net model that recognizes forest fire smoke from inadequate combustion with satellite remote sensing images. We collect 4,000 satellite remote sensing images, 2,000 each for clouds and forest fire smoke, from Himawari-8 satellite imagery located in forest areas of China and Australia, and the image data are used for training, testing, and validation of the model at a ratio of 3:1:1. Compared with existing models, the proposed SR-Net dominates in recognition accuracy (96.9%), strongly supporting its superiority over benchmark models: MobileNet (92.0%), GoogLeNet (92.0%), ResNet50 (84.0%), and AlexNet (76.0%). Model comparison results confirm the accuracy, computational efficiency, and generality of the SR-Net model in detecting forest fire smoke with high temporal resolution remote sensing images.

Abstract Coexistence of ammonium (NH4+) with manganese (Mn) in acid soils may facilitate the alleviation of Mn toxicity to plants. However, the effect of NH4+ on Mn toxicity and the corresponding mechanisms are unclear. In this study, the effects of NH4+ and nitrate (NO3‐) on Mn toxicity, cell wall properties, and nitric oxide (NO) signaling in sugarcane were compared. NH4+ alleviated Mn‐induced chlorosis in sugarcane seedlings and increased seedling biomass compared with NO3‐. Exogenous application of NH4+ decreased the root cell wall pectin content and methyl esterase (PME) activity, but increased the degree of root pectin esterification (PMD). These changes were accompanied by reductions in the Mn content in roots, leaves, root cell wall, and cell wall pectin. An analysis of adsorption kinetic revealed less Mn‐adsorption capacity in cell walls extracted from NH4+‐fed than from NO3‐‐fed sugarcane. Mn induced NO accumulation in sugarcane roots, but NH4+‐fed seedlings accumulated less NO. Exogenous application of the NO donor sodium nitroprusside increased the Mn content of root cell wall pectin in NH4+‐fed sugarcane, while the NO scavenger 2‐(4‐carboxyphenyl)‐4,4,5,5‐tetramethylimidazoline‐1‐oxyl‐3‐oxid decreased the Mn content in NO3‐‐fed sugarcane. These treatments eliminated the difference in the pectin Mn content between NH4+‐fed and NO3‐‐fed sugarcane, as did a similar treatment with the nitrate reductase inhibitor tungstate, which decreased root cell wall pectin content and NO accumulation. These results suggest that (i) NH4+ alleviates Mn toxicity in sugarcane by reducing root pectin accumulation and root cell wall PME activity, thereby increasing cell wall PMD and decreasing both the Mn‐binding capacity of cell wall and Mn accumulation, and (ii) NO mediates the accumulation of both pectin and Mn in response to different forms of nitrogen. The physiological mechanisms underlying the alleviation of ammonium on Mn phytotoxicity were clarified, which provided important implication for agricultural production and ecosystem functioning in acid soil.

An intelligent reflecting surface (IRS) is proposed to enhance the physical layer security in the Rician fading channel wherein the angular direction of the eavesdropper (ED) is aligned with a legitimate user. A two-phase communication system under active attacks and passive eavesdropping is considered in this scenario. The base station avoids direct transmission to the attacked user in the first phase, whereas other users cooperate in forwarding signals to the attacked user in the second phase, with the help of IRS and energy harvesting technology. Under the occurrence of active attacks, an outage-constrained beamforming design problem is investigated under the statistical cascaded channel error model, which is solved by using the Bernstein-type inequality. An average secrecy rate maximization problem for the passive eavesdropping is formulated, which is then addressed by a low-complexity algorithm. The numerical results of this study reveal that the negative effect of the ED’s channel error is larger than that of the legitimate user.

Abstract The continuous emergence of SARS-CoV-2 variants highlights the need of developing vaccines with broad protection. Here, according to the immune-escape capability and evolutionary convergence, the representative SARS-CoV-2 strains carrying the hotspot mutations were selected. Then, guided by structural and computational analyses, we present a mutation-integrated trimeric form of spike receptor-binding domain (mutI-tri-RBD) as a broadly protective vaccine candidate, which combined heterologous RBDs from different representative strains into a hybrid immunogen and integrated immune-escape hotspots into a single antigen. When compared with a homo-tri-RBD vaccine candidate in the stage of phase II trial, of which all three RBDs are derived from the SARS-CoV-2 prototype strain, mutI-tri-RBD induced significantly higher neutralizing antibody titers against the Delta and Beta variants, and maintained a similar immune response against the prototype strain. Pseudo-virus neutralization assay demonstrated that mutI-tri-RBD also induced broadly strong neutralizing activities against all tested 23 SARS-CoV-2 variants. The in vivo protective capability of mutI-tri-RBD was further validated in hACE2-transgenic mice challenged by the live virus, and the results showed that mutI-tri-RBD provided potent protection not only against the SARS-CoV-2 prototype strain but also against the Delta and Beta variants.

The fruit peel color is an important factor that affects its quality. However, genes involved in regulating pericarp color in bottle gourd (Lagenaria siceraria) have not been explored to date. Genetic analysis of color traits in bottle gourd peel through a genetic population of six generations demonstrated that the green color of peels is inherited as a single gene dominant trait. Combined phenotype-genotype analysis of recombinant plants using BSA-seq mapped the candidate gene to a 22.645 Kb interval at the head end of chromosome 1. We observed that the final interval contained only one gene, LsAPRR2 (HG_GLEAN_10010973). Sequence and spatiotemporal expression analyses of LsAPRR2 unraveled two nonsynonymous mutations (A→G) and (G→C) in the parental CDS sequences. Further, LsAPRR2 expression was higher in all green-skinned bottle gourds (H16) at various stages of fruit development than in white-skinned bottle gourds (H06). Cloning and sequence comparison of the two parental LsAPRR2 promoter regions indicated 11 bases insertion and 8 SNPs mutations in the region -991~-1033, upstream of the start codon in white bottle gourd. Proof of GUS reporting system, Genetic variation in this fragment significantly reduced the expression of LsAPRR2 in the pericarp of white bottle gourd. In addition, we developed a tightly linked (accuracy 93.88%) InDel marker for the promoter variant segment. Overall, the current study provides a theoretical basis for comprehensive elucidation of the regulatory mechanisms underlying the determination of bottle gourd pericarp color. This would further help in the directed molecular design breeding of bottle gourd pericarp.

Background: Ulcerative colitis (UC) is a chronic recurrent inflammatory bowel disease (IBD). The conventional drugs for UC may induce severe side effects. Herbal medicine is considered as a complementary and alternative choice for UC.Purpose: This study aims to estimate the effect of natural polyphenol gallic acid (GA) on the NLRP3 inflammasome with dextran sulfate sodium (DSS)-induced colitis in mice.Study design: The body weights and symptoms of BALB/c mice were recorded. Histological evaluation, ELISA, q-PCR, immunohistochemistry, and western blotting were carried out to observe the morphology, cytokine contents, mRNA expressions, and protein expressions, respectively. Lipopolysaccharide (LPS)-induced RAW264.7 macrophage was used to probe GA’s effect on relative protein expression.Results: GA attenuated weight loss (p < 0.05), relieved symptoms, and ameliorated colonic morphological injury (p < 0.05) in mice with colitis induced by DSS. GA also lowered the contents of TNF-α, IL-1β, IL-18, IL-33, and IFN-γ in the serum and colon of mice, which were elevated by DSS, downregulated protein, and mRNA expressions of the NLRP3 pathway in the colon tissue. Furthermore, GA downregulated the expressions of NLRP3 (p < 0.05), iNOS (p < 0.01), COX2 (p < 0.01), and P-p65 (p < 0.05), and suppressed NO release (p < 0.001) in LPS-induced RAW264.7 cells.Conclusion: GA ameliorated DSS-induced UC in mice via inhibiting the NLRP3 inflammasome. These findings furnish evidence for the anti-inflammatory effect of herbal medicines containing GA on UC.

BackgroundPulmonary arterial hypertension (PAH) is a fatal disease characterized by pulmonary vascular remodeling and increased pulmonary artery pressure, leading to impaired lung oxygenation, right heart failure, and even death. Although great advances have been made in PAH-targeted medications for pediatric patients, the efficacy and safety of these treatments are controversial.MethodsWe retrieved relevant articles from electronic databases including PubMed, EMBASE, Web of Science, and Cochrane Library until 12 April 2022. To compare the effectiveness and safety of endothelin receptor antagonists (ERAs), phosphodiesterase type 5 Inhibitors (PDE-5i), and prostaglandins (ProA) in the treatment of pediatric PAH, we investigated six hemodynamic parameters, four respiratory parameters, intensive care unit (ICU) stay duration, length of hospital stay, and two safety outcomes.ResultsA total of 27 randomized controlled trials (RCTs) were included in the meta-analysis with 1,574 pediatric participants. The duration of mechanical ventilation was shorter for patients using bosentan, sildenafil, and ProsA, compared with that for patients using the placebo. Bosentan helped to shorten more time for mechanical ventilation than ProsA did, while ProsA was more effective than sildenafil in this respect. As for the length of stay in the ICU, patients administered by ProsA or sildenafil needed shorter ICU stay, compared to those using the placebo, while ProsA was more effective for shortening ICU stay time. In light of safety outcomes, there was a statistically significant difference between the sildenafil and the placebo group. Sildenafil surpassed ProsA in reducing the incidence of pulmonary hypertension (PH) crisis.ConclusionsERAs were more effective than ProsA in shortening the duration of mechanical ventilation, while ProsA were better for shortening the duration of mechanical ventilation and ICU stay than PDE-5i. In-stent restenosis after interventional therapy remains a severe clinical complication. Current evidence indicates that neointimal hyperplasia induced by vascular smooth muscle cell (VSMC) proliferation is a major cause of restenosis. Thus, inhibiting VSMC proliferation is critical for preventing in-stent restenosis. The incidence of restenosis was reduced in nitrided iron-based stents (hereafter referred to as iron stents). We hypothesized that the corroded granules produced by the iron stent would prevent in-stent restenosis by inhibiting VSMC proliferation. To verify this hypothesis, we introduced a dynamic circulation device to analyze the components of corroded granules. To investigate the effects of corroded granules on VSMC proliferation, we implanted the corroded iron stent into the artery of the atherosclerotic artery stenosis model. Moreover, we explored the mechanism underlying the inhibition of VSMC proliferation by iron corroded granules. The results indicated that iron stent produced the corroded granules after implantation, and the main component of the corrosion granules was iron oxide. Remarkably, the corroded granules reduced the neointimal hyperplasia in an atherosclerotic artery stenosis model, and iron corroded granules decreased the neointimal hyperplasia by inhibiting VSMC proliferation. In addition, we revealed that corroded granules reduced VSMC proliferation by activating autophagy through the AMPK/mTOR signaling pathway. Importantly, safety of iron corroded granules was evaluated and proved to be satisfactory hemocompatibility in rabbit model. Overall, the role of corroded granules in restenosis prevention was described for the first time. This finding highlighted the implication of corroded granules produced by iron stent in inhibiting VSMC proliferation, pointing to a new direction to prevent in-stent restenosis.

Hypervirulent Klebsiella pneumoniae (hvKp) can cause serious infections and has been increasingly reported clinically. However, we still lack the knowledge to what degree hvKp colonize the community. In this study, we investigated colonization of hvKp in healthy human gut and the relationship between gut hvKp and clinically important invasive strains. We compile global genomes of gut K. pneumoniae for in-depth genetic analysis and found most hvKp genomes originated from Chinese datasets; therefore, we collected gut K. pneumoniae isolates from healthy people around China. The results revealed a moderate carriage rate of hvKp in the healthy population (4%–5.19%). Phylogenetic analysis indicated a close relationship between gut hvKp and fatal clinical strains. These results demonstrate that the human gut may serve as a reservoir of hvKp and that gut hvKp can play a role in infection of other body parts.

Objective: To meet the market demand, high concentration PUFA oil microcapsules were developed. Methods: The microencapsulated oil was prepared by spray drying method with β-cyclodextrin /gelatin as a composite wall material and high content polyunsaturated fatty acid oil sunflower seed oil as a core material, and the preparation process parameters of oil microcapsules were optimized by single factor experiment and response surface experiment. Results: The best technological conditions of oil microcapsule were solid content of 25.2%, and the mass ratio of composite wall material β-cyclodextrin: gelatin was 6.7∶1. The dosage of emulsifier was 1%, and the mass ratio of core wall material was 1∶2. Under this condition, the prepared oil microcapsule particles were round. The particle size was uniform, and the embedding rate was 94.33%. The content of polyunsaturated fatty acid was 21.73%. Conclusion: This preparation method is feasible and can be used to prepare high concentration polyunsaturated fatty acid oil microcapsules.

Affected by global warming and increased extreme weather, Hunan Province saw a phased and concentrated outbreak of forest fires in 2022, causing significant damage and impact. Predicting the occurrence of forest fires can enhance the ability to make early predictions and strengthen early warning and responses. Currently, fire prevention and extinguishing in China’s forests and grasslands face severe challenges due to the overlapping of natural and social factors. Existing forest fire occurrence prediction models mostly take into account vegetation, topographic, meteorological and human activity factors; however, the occurrence of forest fires is closely related to the forest fuel moisture content. In this study, the traditional driving factors of forest fire such as satellite hotspots, vegetation, meteorology, topography and human activities from 2004 to 2021 were introduced along with forest fuel factors (vegetation canopy water content and evapotranspiration from the top of the vegetation canopy), and a database of factors for predicting forest fire occurrence was constructed. And a forest fire occurrence prediction model was built using machine learning methods such as the Random Forest model (RF), the Gradient Boosting Decision Tree model (GBDT) and the Adaptive Augmentation Model (AdaBoost). The accuracy of the models was verified using Area Under Curve (AUC) and four other metrics. The RF model with an AUC value of 0.981 was more accurate than all other models in predicting the probability of forest fire occurrence, followed by the GBDT (AUC = 0.978) and AdaBoost (AUC = 0.891) models. The RF model, which has the best accuracy, was selected to predict the monthly forest fire probability in Changsha in 2022 and combined with the Inverse Distance Weight Interpolation method to plot the monthly forest fire probability in Changsha. We found that the monthly spatial and temporal distribution of forest fire probability in Changsha varied significantly, with March, April, May, September, October, November and December being the months with higher forest fire probability. The highest probability of forest fires occurred in the central and northern regions. In this study, the core drivers affecting the occurrence of forest fires in Changsha City were found to be vegetation canopy evapotranspiration and vegetation canopy water content. The RF model was identified as a more suitable forest fire occurrence probability prediction model for Changsha City. Meanwhile, this study found that vegetation characteristics and combustible factors have more influence on forest fire occurrence in Changsha City than meteorological factors, and surface temperature has less influence on forest fire occurrence in Changsha City.

The Shuttle Radar Topographic Mission (SRTM) digital elevation model (DEM) is a widely utilized product for geological, climatic, oceanic, and ecological applications. However, the accuracy of the SRTM DEM is constrained by topography and vegetation. Using machine learning models to correct SRTM DEM with high-accuracy reference elevation observations has been proven to be useful. However, most of the reference observation-aided approaches rely on either parametric or non-parametric regression (e.g., a single machine learning model), which may lead to overfitting or underfitting and limit improvements in the accuracy of SRTM DEM products. In this study, we presented an algorithm for correcting SRTM DEM using a stacking ensemble machine learning algorithm. The proposed algorithm is capable of learning how to optimally combine the predictions from multiple well-performing machine learning models, resulting in superior performance compared to any individual model within the ensemble. The proposed approach was tested under varying relief and vegetation conditions in Hunan Province, China. The results indicate that the accuracy of the SRTM DEM productions improved by approximately 46% using the presented algorithm with respect to the original SRTM DEM. In comparison to two conventional algorithms, namely linear regression and artificial neural network models, the presented algorithm demonstrated a reduction in root-mean-square errors of SRTM DEM by 28% and 12%, respectively. The approach provides a more robust tool for correcting SRTM DEM or other similar DEM products over a wide area.

Background: For foamy virus, the transactivator of spumaretrovirus (Tas) could bind directly to target DNA sequences termed as Tas responsive elements and trigger the viral internal promoter (IP) and long terminal repeat (LTR) promoters. The cellular endogenous factors also play an important role in viral gene expressions. We hypothesized that except the viral transcription factor Tas, the cellular endogenous factors also affect the viral gene expression. Methods: The full length of the prototype foamy virus (PFV) genome (U21247) was used to predict the potential binding sites of the transcription factors by online software JASPAR (http://jaspar.genereg.net) and Softberry (http://linux1.softberry.com/berry.phtml?topic=index&group=programs&subgroup=promoter). The Dual-Luciferase® Reporter Assay System (Promega, USA) was used to confirm the relative luciferase activities of the test groups. The different representative activating agents or inhibitors of each canonical signal pathway were used to identify the impact of these pathways on PFV 5′LTR and IP promoters. Results: The results showed different cellular endogenous factors might have respective effects on PFV 5′LTR and IP. It is worth mentioning that activator protein-1 and BCL2-associated athanogene 3, 2 kinds of vital proteins associated with NF-κB and PKC pathways, could activate the basal activity of 5′LTR and IP promoters but inhibit the Tas-regulated activity of both promoters. Furthermore, PFV Tas was identified to trigger the transcription of the NF-κB promoter. Conclusion: NF-κB had a negative effect on PFV 5′LTR and IP promoter activities, the PKC pathway might upregulate 5′LTR and IP promoter activities, and the JNK and NF-AT signal pathway could increase the Tas-regulated promoter activity of PFV 5′LTR. This study sheds light on the interaction between PFV and the host cell and may help utilize the viral promoters in retroviral vectors designed for gene transfer experiments.

Large-scale populations in the world have been vaccinated with COVID-19 vaccines, however, breakthrough infections of SARS-CoV-2 are still growing rapidly due to the emergence of immune-evasive variants, especially Omicron. It is urgent to develop effective broad-spectrum vaccines to better control the pandemic of these variants. Here, we present a mosaic-type trimeric form of spike receptor-binding domain (mos-tri-RBD) as a broad-spectrum vaccine candidate, which carries the key mutations from Omicron and other circulating variants. Tests in rats showed that the designed mos-tri-RBD, whether used alone or as a booster shot, elicited potent cross-neutralizing antibodies against not only Omicron but also other immune-evasive variants. Neutralizing antibody ID50 titers induced by mos-tri-RBD were substantially higher than those elicited by homo-tri-RBD (containing homologous RBDs from prototype strain) or the BIBP inactivated COVID-19 vaccine (BBIBP-CorV). Our study indicates that mos-tri-RBD is highly immunogenic, which may serve as a broad-spectrum vaccine candidate in combating SARS-CoV-2 variants including Omicron.

AbstractThe hydrogeochemical characteristics and isotopic analyses of geothermal water are effective in the genetic study of a geothermal system. This study systematically investigated the geochemical and isotopic compositions of the geothermal water in the Qutan and Reshuizhou geothermal fields. The geothermal water in these two fields is of HCO3 (·F)-Ca·Na type and has undergone protracted water runoff and deep thermal cycle. Based on the mineral-water solubility equilibrium, the reservoir in the Qutan geothermal field has a temperature of 60–74°C and a depth range of 770–1003 m. The geothermal water in the Reshuizhou geothermal field has been mixed with cold water, which accounts for 58% in weight. The reservoir in the Reshuizhou geothermal field has a temperature range of 131-150°C and a depth range of 1953–2287 m. The geothermal water in the Qutan and Reshuizhou geothermal fields have 14C ages of 8251–7961 BC and 3514–2254 BC, respectively, which are consistent with the result of the 3H isotopic analyses. When recharging and transiting to geothermal water, the meteoric water underwent changes in geochemical compositions, which were dominantly controlled by water-rock interactions, including the precipitation of chalcedony and muscovite and the dissolution of calcite, fluorite, K-feldspar, and albite. In the Reshuizhou geothermal field, the hydrogeochemical compositions of the geothermal water are affected by cold water mixing.

Detection of in vivo biodegradation is critical for development of next-generation medical devices such as bioresorbable stents or scaffolds (BRSs). In particular, it is urgent to establish a nondestructive approach to examine in vivo degradation of a new-generation coronary stent for interventional treatment based on mammal experiments; otherwise it is not available to semi-quantitatively monitor biodegradation in any clinical trial. Herein, we put forward a semi-quantitative approach to measure degradation of a sirolimus-eluting iron bioresorbable scaffold (IBS) based on optical coherence tomography (OCT) images; this approach was confirmed to be consistent with the present weight-loss measurements, which is, however, a destructive approach. The IBS was fabricated by a metal-polymer composite technique with a polylactide coating on an iron stent. The efficacy as a coronary stent of this new bioresorbable scaffold was compared with that of a permanent metal stent with the name of trade mark Xience, which has been widely used in clinic. The endothelial coverage on IBS was found to be greater than on Xience after implantation in a rabbit model; and our well-designed ultrathin stent exhibited less individual variation. We further examined degradation of the IBSs in both minipig coronary artery and rabbit abdominal aorta models. The present result indicated much faster iron degradation of IBS in the rabbit model than in the porcine model. The semi-quantitative approach to detect biodegradation of IBS and the finding of the species difference might be stimulating for fundamental investigation of biodegradable implants and clinical translation of the next-generation coronary stents.

Forest fires can destroy millions of acres of land at shockingly fast speeds. The forest fire points identification algorithm is the most critical step in the forest fire monitoring process. Most traditional forest fire monitoring methods use fixed thresholds, ignoring background pixels, and have low recognition rates, which could lead to many problems, such as false reporting and low recognition rate. This paper proposes and tests an adaptive forest fire points identification algorithm using Himawari-8 data. By calculating the three-dimensional histogram of brightness temperature, an adaptive threshold that can automatically identify potential forest fire points is obtained. Based on this three-dimensional Otsu method, the contextual test algorithm has also been adopted to specify forest fire points. The experimental results show that the omission rate of the improved algorithm is about 10% lower than that of the previous algorithm in small-scale fire incidents. The improved algorithm can quickly and effectively extract fire point information, and it is also sensitive to small and low-temperature fires, which provides an efficient means for monitoring fire disasters.

The detection of the depth and temperature of deep geothermal reservoirs suffers great uncertainty if it is completed using the simple combination of traditional geophysical and geochemical methods. Given this, this study investigated the combined utilization of multiple methods including the multi-information joint detection using the controlled source audio frequency magnetotellurics (CSAMT) method and geothermometers in the Qutan geothermal field. First, the reservoir temperature and the depth of geothermal water circulation in the geothermal field were estimated using the geochemical method. Afterward, the relationship between the CSAMT resistivity and temperature of strata was explored through borehole temperature measurement. Based on this, the depth and temperature of geothermal reservoirs in the geothermal field were estimated again. According to the comprehensive analysis and comparison of the thermal reservoir temperatures and depths obtained using the two methods, the shallow reservoirs in the Qutan geothermal field have a top burial depth of approximately 423 m, a thickness of about 300 m, a temperature of about 79°C, and a fluid circulation depth of about 1959 m.

Background:Reynoutria multiflora (Thunb.) Moldenke (PM) is a widely-used medicinal plant in China, whose root and stem are included in the Chinese Pharmacopoeia as Polygoni Multiflori Radix (RPM), Polygoni Multiflori Radix Preparata (PMP), and Polygoni Multiflori Caulis (PMC). The hepatotoxicity of RPM and PMP is concerned by the public, while the risk of PMC is ignored.Purpose: Here, we investigate the potential risks for PMC-induced liver injury from clinical, chemical, and animal features.Study design: First, we analyzed the 12-month usage of RPM, PMP, and PMC in Longhua Hospital. Second, we determined the contents of gallic acid, cis-2,3,5,4′-tetrahydroxy-stilbene-2-O-β-D-glucoside (cis-SG), trans-2,3,5,4′-tetrahydroxy-stilbene-2-O-β-D-glucoside (trans-SG), emodin-8-O-β-D-glucoside (EG), physcion-8-O-β-D-glucoside (PG), emodin, and physcion in the water extracts from 15 batches of RPM, PMP, and PMC. Third, we probed the hepatotoxic effect of RPM, PMP, and PMC in mice and explored the mechanism of cis-SG and trans-SG causing the liver injury at the dosages based on our results from the first and second parts.Results: PMC had nearly five times the amount of usage in both outpatient prescriptions and inpatient orders than RPM and PMP. Overall, 68% dosage of PMC was 30 g. The contents of cis-SG, trans-SG, and emodin in PMC water extracts were significantly lower than those in RPM and PMP water extracts. PMC induced milder idiosyncratic liver injury for its lower content of cis-SG and trans-SG than its root counterparts.Conclusion: The potential risks for PMC-induced liver injury should be fully aware of.

ObjectivesThe purpose of this study was to investigate the baseline independent risk factors for predicting 6-month mortality of patients with anti-melanoma differentiation-associated gene 5 (anti-MDA5)-positive dermatomyositis (DM) and develop a matrix prediction model formed by these risk factors.MethodsThe hospitalized patients with DM who completed at least 6-month follow-up were recruited as a derivation cohort. The primary exposure was defined as positive anti-MDA5 at the baseline. The primary outcome was all-cause 6-month mortality after enrollment. A matrix prediction model was developed in the derivation cohort, and another published cohort was used for external validation.ResultsIn derivation cohort, 82 patients with DM were enrolled (mean age of onset 50 ± 11 years and 63% women), with 40 (49%) showing positive anti-MDA5. Gottron sign/papules (OR: 5.135, 95%CI: 1.489–17.708), arthritis (OR: 5.184, 95%CI: 1.455–18.467), interstitial lung disease (OR: 7.034, 95%CI: 1.157–42.785), and higher level of C4 (OR: 1.010, 95%CI: 1.002–1.017) were the independent associators with positive anti-MDA5 in patients with DM. Patients with anti-MDA5-positive DM had significant higher 6-month all-cause mortality than those with anti-MDA5-negative (30 vs. 0%). Among the patients with anti-MDA5-positive DM, compared to the survivors, non-survivors had significantly advanced age of onset (59 ± 6 years vs. 46 ± 9 years), higher rates of fever (75 vs. 18%), positive carcinoma embryonic antigen (CEA, 75 vs. 14%), higher level of ferritin (median 2,858 ug/L vs. 619 ug/L, all p < 0.05). A stepwise multivariate Cox regression showed that ferritin ≥1,250 μg/L (HR: 10.4, 95%CI: 1.8–59.9), fever (HR: 11.2, 95%CI: 2.5–49.9), and positive CEA (HR: 5.2, 95%CI: 1.0–25.7) were the independent risk factors of 6-month mortality. A matrix prediction model was built to stratify patients with anti-MDA5-positive DM into different subgroups with various probabilities of 6-month mortality risk. In an external validation cohort, the observed 6-month all-cause mortality was 78% in high-risk group, 43% in moderate-risk group, and 25% in low-risk group, which shows good accuracy of the model.ConclusionBaseline characteristics such as fever, ferritin ≥1,250 μg/L, and positive CEA are the independent risk factors for 6-month all-cause mortality in patients with anti-MDA5-positive DM. A novel matrix prediction model composed of these three clinical indicators is first proposed to provide a chance for the exploration of individual treatment strategies in anti-MDA5-positive DM subgroups with various probabilities of mortality risk.

Fully bioresorbable scaffolds have been designed to overcome the limitations of traditional drug-eluting stents (DESs), which permanently cage the native vessel wall and pose possible complications. The ultrathin-strut designed sirolimus-eluting iron bioresorbable coronary scaffold system (IBS) shows comparable mechanical properties to traditional DESs and exhibits an adaptive degradation profile during target vessel healing, which makes it a promising candidate in all-comers patient population. For implanted medical devices, magnetic resonance (MR) imaging properties, including MR safety and compatibility, should be evaluated before its clinical use, especially for devices with intrinsic ferromagnetism. In this study, MR safety and compatibility of the IBS scaffold were evaluated based on a series of well-designed in-vitro, ex-vivo and in-vivo experiments, considering possible risks, including scaffold movement, over-heating, image artifact, and possible vessel injury, under typical MR condition. Traditional ASTM standards for MR safety and compatibility evaluation of intravascular devices were referred, but not only limited to that. The unique time-relevant MR properties of bioresorbable scaffolds were also discussed. Possible forces imposed on the scaffold during MR scanning and MR image artifacts gradually decreased along with scaffold degradation/absorption. Rigorous experiments designed based on a scientifically based rationale revealed that the IBS scaffold is MR conditional, though not MR compatible before complete absorption. The methodology used in the present study can give insight into the MR evaluation of magnetic scaffolds (bioresorbable) or stents (permanent).

This paper presents a terrain-related method to simultaneously correct the global error trends and local linear/nonlinear terrain-related errors of the Shuttle Radar Topography Mission (SRTM) digital elevation model (DEM), which have not been focused in most of the previous methods. To meet this goal, an adaptive strategy for modelling the SRTM DEM errors is first proposed, especially over mountainous areas, based on the Bayesian information criterion. Then, the M-estimator, instead of the ordinary least squares solver, is utilized to estimate the model parameters of the constructed model to improve the estimation robustness. The proposed method was tested over the Zhangjiajie areas of China, where the ground surface terrain varies from plains to steep mountains. The results show that the errors of the SRTM DEM over the region of interest decrease from 10.1 m to approximately 8.1 m after correction with the proposed method, indicating an improvement of approximately 20%. In addition, compared with two existing and common methods that can respectively correct the global error trends and local linear terrain-related errors of the SRTM DEM, the accuracy of the corrected SRTM DEM improves by approximately 29% and 27.9% over mountainous areas with slopes larger than 20° when the proposed method is used. Moreover, the proposed method will be beneficial to the correction of other airborne or spaceborne DEM products, especially over mountainous areas.

AbstractLarge-scale geothermal production based on a multiwell layout has been promoted by the development of clean energy. However, the theories about the effects of this geothermal production pattern on the temperature field of geothermal reservoirs are yet to be improved, which hinders the design optimization of the deployment schemes of geothermal wells. This study proposed a 3D geothermal multiwell simulation method covering the heat transfer in wellbores by introducing the geothermal well theory of 1D line elements, thus improving the calculation efficiency without reducing the calculation accuracy. By taking the geothermal development using multiple wells in the Xianxian geothermal field, China, as an example, this study firstly analyzed the evolution of the temperature field in geothermal reservoirs during the geothermal exploitation based on a multiwell layout. Then, it revealed the influencing mechanisms of geothermal multiwell effect, reinjection depth, and well spacing on the thermal breakthrough of exploitation wells. Finally, it proposed an optimized design method for the geothermal development based on a multiwell layout. All these will provide scientific support for the large-scale geothermal production based on exploitation-reinjection combination using multiple wells.
