Peritoneal adhesions commonly occur following abdominal or pelvic surgery and can cause serious complications. Currently, physical barriers are the primary approach used in clinical practice to prevent adhesion, although their effectiveness is frequently inadequate. In this study, we developed an injectable peptide-loaded hydrogel with multiple functions, including self-fusion, tissue-adhesiveness, anti-inflammation, anti-cell adhesion and anti-angiogenesis. To assess the effectiveness of these hydrogels, which are stabilized by dynamic imine bonds and acetal connections, in preventing postoperative abdominal adhesions, we utilized both a rat abdominal adhesion model and a rat model simulating repeated-injury adhesions. In comparison to the commercially available HA hydrogel, as-prepared hydrogels exhibited significant reductions in inflammation, fibrosis, and angiogenesis, leading to an obvious decrease in peritoneal adhesions. Moreover, this peptide-loaded hydrogel demonstrated an ideal degradation time, maintaining an in vivo viability for about 10 days. We believe this peptide-loaded hydrogel presents a promising solution for the challenging clinical issue of postoperative abdominal adhesions.

Abstract Background Colorectal cancer (CRC) is the third frequently diagnosed cancer with high incidence and mortality rate worldwide. Our previous report has demonstrated that circCOL1A1 (hsa_circ_0044556) functions as an oncogene in CRC, and Gene Ontology (GO) analysis has also revealed the strong association between circCOL1A1 and angiogenesis. However, the mechanism of circCOL1A1 or exosomal circCOL1A1 in CRC angiogenesis remains elusive. Methods Purified exosomes from CRC cells were characterized by nanoparticle tracking analyzing, electron microscopy and western blot. qRT-PCR, immunohistochemistry or western blot were employed to test the expression of circCOL1A1, EIF4A3, Smad pathway and angiogenic markers. Cell proliferation of HUVECs was monitored by CCK-8 assay. The migratory and angiogenic capabilities of HUVECs were detected by wound healing and tube formation assay, respectively. Bioinformatics analysis, RNA immunoprecipitation (RIP), RNA pull-down and FISH assays were used to detect the interactions among circCOL1A1, EIF4A3 and Smad2/3 mRNA. The in vitro findings were verified in xenograft model. Results CRC cell-derived exosomal circCOL1A1 promoted angiogenesis of HUVECs via recruiting EIF4A3. EIF4A3 was elevated in CRC tissues, and it stimulated angiogenesis of HUVECs through directly binding and stabilizing Smad2/3 mRNA. Moreover, exosomal circCOL1A1 promoted angiogenesis via inducing Smad2/3 signaling pathway in vitro, and it also accelerated tumor growth and angiogenesis in vivo. Conclusion CRC cell-derived exosomal circCOL1A1 promoted angiogenesis via recruiting EIF4A3 and activating Smad2/3 signaling.

Remote sensing technology can be used to monitor changes in crop planting areas to guide agricultural production management and help achieve regional carbon neutrality. Agricultural UAV remote sensing technology is efficient, accurate, and flexible, which can quickly collect and transmit high-resolution data in real time to help precision agriculture management. It is widely used in crop monitoring, yield prediction, and irrigation management. However, the application of remote sensing technology faces challenges such as a high imbalance of land cover types, scarcity of labeled samples, and complex and changeable coverage types of long-term remote sensing images, which have brought great limitations to the monitoring of cultivated land cover changes. In order to solve the abovementioned problems, this paper proposed a multi-scale fusion network (MSFNet) model based on multi-scale input and feature fusion based on cultivated land time series images, and further combined MSFNet and Model Diagnostic Meta Learning (MAML) methods, using particle swarm optimization (PSO) to optimize the parameters of the neural network. The proposed method is applied to remote sensing of crops and tomatoes. The experimental results showed that the average accuracy, F1-score, and average IoU of the MSFNet model optimized by PSO + MAML (PSML) were 94.902%, 91.901%, and 90.557%, respectively. Compared with other schemes such as U-Net, PSPNet, and DeepLabv3+, this method has a better effect in solving the problem of complex ground objects and the scarcity of remote sensing image samples and provides technical support for the application of subsequent agricultural UAV remote sensing technology. The study found that the change in different crop planting areas was closely related to different climatic conditions and regional policies, which helps to guide the management of cultivated land use and provides technical support for the realization of regional carbon neutrality.

Summary: CircRNAs are implicated in colorectal cancer (CRC) development and progression. Protein O-fucosyltransferase 1 (POFUT1) plays an oncogenic role via activating Notch1 signaling in CRC. However, the roles of circPOFUT1, which is originated from POFUT1, have not been investigated. Our study showed circPOFUT1 was highly expressed in CRC tissues and cells. CircPOFUT1 enhanced the proliferation, migration and invasion of CRC cells, and promoted tumor growth and liver metastasis in vivo. It also reinforced stemness and chemoresistance of CRC cells. Mechanistically, circPOFUT1 regulated the function of E2F7 via sponging miR-653-5p, thereby transcriptionally inducing WDR66 expression and further promoting metastasis in CRC. On the other hand, circPOFUT1 promoted stemness and chemoresistance of CRC cells via stabilizing BMI1 in an IGF2BP1-dependent manner. In conclusion, circPOFUT1 fosters CRC metastasis and chemoresistance via decoying miR-653-5p/E2F7/WDR66 axis and stabilizing BMI1.

Abstract Patients with metabolic syndrome (MetS) undergoing surgery are at high risk of developing peritoneal adhesions and other severe postoperative complications. However, the single shielding function and absence of physiological activity render conventional methods less useful in preventing adhesions in patients with MetS. To address this challenge, a convenient method is introduced for developing a novel tissue‐adhesive hydrogel called oxidized dextran–metformin (ODE–ME) via Schiff base linkages. This injectable ODE–ME hydrogel exhibits excellent tissue‐adhesive properties and various physiological functions, particularly enhanced antibacterial effects. Furthermore, in vivo experiments demonstrate that the hydrogel can effectively alleviate hyperglycemia, reduce excessive inflammation, and improve fibrinolytic activity in MetS mice, thereby preventing adhesions and promoting incisional healing. The hydrogel concurrently isolates injured tissues and lowers the blood glucose levels immediately after surgery in mice. Therefore, the ODE–ME hydrogel functions as a multifunctional barrier material and has potential for preventing postoperative peritoneal adhesions in patients with MetS in clinical settings.

Abstract On 6 February 2023, a local tsunami was recorded in the southeastern Mediterranean Sea following the Mw 7.7 Turkey–Syria inland strike‐slip earthquake. Due to the lack of underwater observation, the tsunami generation mechanism remains mysterious. To understand the source mechanisms, we analyzed the tsunami waveforms of four nearby tide gauges and located possible sources using a backward tsunami ray tracing approach. We then conducted forward numerical modelings for a range of possible source parameters. We show that there were probably two tsunami sources, inside and outside Iskenderun Bay, which may be related to thick coastal sediments. A source inside the Bay with a characteristic length of 7 km produced dominant periods of 10–30 min with negative initial motion, possibly generated by a landslide. Another source of 6 km length outside the Bay produced dominant periods of 2–10 min with positive initial motions, possibly related with liquefaction.

Abstract Background Colorectal cancer (CRC) is one of the three major cancers in the world and is the cancer with the most liver metastasis. The present study aimed to investigate the role of metallothionein 2A (MT2A) in the modulation of CRC cell proliferation and liver metastasis, as well as its molecular mechanisms. Methods The expression profile of metallothionein 2A (MT2A) in colorectal cancer retrieved from TCGA, GEO and Oncomine database. The biological effect of MT2A overexpression was investigated mainly involving cell proliferation and migration in CRC cells as well as growth and metastasis in CRC animal models. To explore the specific mechanism of MT2A metastasis in CRC, transcriptome sequencing was used to compare the overall expression difference between the control group and the MT2A overexpression group. Results Metallothionein 2A (MT2A) was downregulated in the tumor tissues of patients with CRC compared to adjacent normal tissues and was related to the tumor M stage of patients. MT2A overexpression inhibited CRC cell proliferation and migration in cells, as well as growth and metastasis in CRC animal models. While knockdown of MT2A had the opposite effect in cells. Western blotting confirmed that MT2A overexpression promoted the phosphorylation of MST1, LAST2 and YAP1, thereby inhibiting the Hippo signaling pathway. Additionally, specific inhibitors of MST1/2 inhibited MT2A overexpression-mediated phosphorylation and relieved the inhibition of the Hippo signaling pathway, thus promoting cell proliferation. Immunohistochemistry in subcutaneous grafts and liver metastases further confirmed this result. Conclusions Our results suggested that MT2A is involved in CRC growth and liver metastasis. Therefore, MT2A and MST1 may be potential therapeutic targets for patients with CRC, especially those with liver metastases.

Abstract Background Previous studies have shown that the N6-methyladenosine (m6A) modification enhances the binding ability of mRNAs/long noncoding RNAs (lncRNAs) to microRNAs (miRNAs), but the impact of this modification on the competitive endogenous RNA (ceRNA) function of circular RNAs (circRNAs) is unclear. Methods We used a human circRNA microarray to detect the expression profiles of circRNAs in 3 pairs of cancer and paracancerous tissues from patients with colorectal cancer (CRC) and 3 pairs of peripheral blood specimens from patients with CRC and healthy individuals. The circRNAs highly expressed in both peripheral blood and tumour tissues of patients with CRC, including circALG1, were screened. A quantitative reverse-transcription polymerase chain reaction (qRT-PCR) analysis of an expanded sample size was performed to detect the expression level of circALG1 in peripheral blood and tumour tissues of patients with CRC and determine its correlation with clinicopathological features, and circRNA loop-forming validation and stability assays were then conducted. Transwell assays and a nude mouse cancer metastasis model were used to study the function of circALG1 in CRC and the role of altered m6A modification levels on the regulation of circALG1 function. qRT-PCR, western blot (WB), Transwell, RNA-binding protein immunoprecipitation (RIP), RNA antisense purification (RAP), and dual-luciferase reporter gene assays were performed to analyse the ceRNA mechanism of circALG1 and the effect of the m6A modification of circALG1 on the ceRNA function of this circRNA. Results CircALG1 was highly expressed in both the peripheral blood and tumour tissues of patients with CRC and was closely associated with CRC metastasis. CircALG1 overexpression promoted the migration and invasion of CRC cells, and circALG1 silencing and reduction of the circALG1 m6A modification level inhibited CRC cell migration and invasion. In vivo experiments further confirmed the prometastatic role of circALG1 in CRC. Further mechanistic studies showed that circALG1 upregulated the expression of placental growth factor (PGF) by binding to miR-342-5p and that m6A modification enhanced the binding of circALG1 to miR-342-5p and promoted its ceRNA function. Conclusion M6A modification enhances the binding ability of circALG1 to miR-342-5p to promote the ceRNA function of circALG1, and circALG1 could be a potential therapeutic target in and a prognostic marker for CRC.

Abstract The RNA binding protein PTBP3 was recently reported to play a critical role in multiple cancers, and the molecular mechanisms involved RNA splicing, 3′ end processing and translation. However, the role of PTBP3 in colorectal cancer (CRC) remains poorly explored. Herein, PTBP3 was upregulated in CRC and associated with a poor prognosis. PTBP3 knockdown in colorectal cancer cell lines restricted CRC proliferative capacities in vitro and in vivo. Mechanistically, PTBP3 regulated the expression of the E3 ubiquitin ligase UBE4A by binding the 3′ UTR of its mRNA, preventing its degradation. UBE4A participated in P53 degradation, and PTBP3 knockdown in colorectal cancer cell lines showed increased P53 expression. UBE4A overexpression rescued PTBP3 knockdown-induced inhibition of CRC cell proliferation and P53 expression. Our results demonstrated that PTBP3 plays an essential role in CRC cell proliferation by stabilizing UBE4A to regulate P53 expression and may serve as a new prognostic biomarker and effective therapeutic target for CRC.

In horizontal drilling, high frictional forces adversely influence operation efficiency. The influence of the rate of change in the overall hole trajectory angle (dogleg severity) is often ignored in the prediction of frictional force in drilling program design, and there is a shortage of quantitative analysis. Aiming at this issue, a mathematical model for computing the radial force caused by axial tension, string bending, and gravity of the string was established. It is found that the axial tension and the gravity of the string are the main forces affecting the radial force of the string, and the bending radial force of the medium-long-radius horizontal well is no more than 14% of the gravity of the string. The influence of dogleg severity on the friction in the vertical, building, and horizontal sections was analysed, and the equations for computing the friction were also established. Based on case analysis, it is suggested that dogleg severity should be kept within a lower limit in the vertical section of horizontal wells with a greater TVD (true vertical depth) because the greater the TVD, the higher the add-on factor related to string tension. For example, in the upper well section with a vertical depth of 3000 m, the add-on factor related to string tension is 0.5, and dogleg severity at 1000 m should be controlled within 0.5°/30 m. Lower building rate should be adopted in the early building section, and appropriately higher building rate can be adopted later. Sliding drilling should be practiced as less as possible provided that target hitting is guaranteed, and combined drilling should be used more. In horizontal wells, dogleg severity at certain point of the horizontal section should be controlled inversely proportional to the drilling drag caused by the subsequent points. The closer to target A (the starting point of the horizontal section), the higher the control requirement for dogleg severity, and it can be appropriately reduced in subsequent well section. If the horizontal interval length Lh is 1000 m and the friction coefficient μ is 0.3, then dogleg severity ϑ should be controlled within 1°/30 m at point A and within 2°/30 m at the middle.

Fibrotic diseases pose significant clinical challenges due to their broadness and complexity. Thus, a better understanding of fibrogenesis and the development of more effective treatments is imperative. Recent evidence suggests a significant antifibrotic potential of an endogenous glycoprotein, endostatin. While endostatin has been widely studied for its role as an anticancer adjuvant by inhibiting tumor angiogenesis, its possible implication in fibrosis remains largely unclear. Here, we review the role of endostatin in various cellular processes and highlight its antifibrotic activity. We hypothesize that endostatin conveys a homeostatic function in the process of fibrosis by regulating (a) TGF-β1 and its downstream signaling; (b) RhoA/ROCK pathway; (c) NF-κB signaling pathway; (d) expression of EGR-1; (e) PDGF/PDGFR pathway; (f) autophagy-related pathways; (g) pathways associated with cell proliferation and apoptosis. Finally, we propose a schematic model of the antifibrotic roles and mechanisms of endostatin; also, we outline future research directions of endostatin and aim to present a potential therapeutic approach for fibrosis.

BackgroundFerroptosis is a newly defined form of programmed cell death that plays an important role in many cancers. However, ferroptosis-related lncRNAs (FRLs) involved in the regulation of colon cancer are not thoroughly understood. This study aimed to identify a prognostic FRL signature in colon cancer and explore its potential molecular function.MethodsRNA-seq data and relevant clinical information were obtained from The Cancer Genome Atlas (TCGA) database, and a list of ferroptosis-related genes was extracted from the FerrDb website. Analysis of differentially expressed FRLs was performed using the ‘limma’ package in R software. By implementing coexpression analysis and univariate Cox analysis, we then identified prognostic FRLs. Using Cox regression analysis with the least absolute shrinkage and selection operator (LASSO) algorithm, we constructed a prognostic model based on 4 FRLs. We evaluated the prognostic power of this model using Kaplan–Meier (K-M) survival curve analysis and receiver operating characteristic (ROC) curve analysis. Moreover, the relationships between the signature and immune landscape, somatic mutation and drug sensitivity were explored. Finally, in vitro experiments were conducted to validate the functions of AP003555.1 and AC000584.1.ResultsA 4-FRL signature was constructed. Two risk groups were classified based on the risk score calculated by this signature. The signature-based risk score exhibited a more powerful capacity for survival prediction than traditional clinicopathological features in colon patients. Additionally, we observed a significant difference in immune cells, such as CD4+ and CD8+ T cells and macrophages, between the two groups. Moreover, the high-risk group exhibited lower IC50 values for certain chemotherapy drugs, such as cisplatin, docetaxel, bleomycin or axitinib. Finally, the in vitro experiments showed that ferroptosis processes were suppressed after AP003555.1 and AC000584.1 knockdown.ConclusionThe proposed 4-FRL signature is a promising biomarker to predict clinical outcomes and therapeutic responses in colon cancer patients.

A YDC type hydraulic hammer is a new valve-type double action hydraulic hammer suitable for oil and gas well drilling. It is hard to find out the optimal matching relationship among various factors based on experience and experiments, for the matching relationships of inner pressure is complex and the impact performance is influenced by many factors. In this paper, the operating principle of a YDC type hydraulic hammer was investigated, the force applied to the main moving components (valve core and hammer) was analyzed and a dynamic model of valve core and hammer in each operating stage was established. Then, a hydraulic hammer performance optimization design software was developed on the Matlab software platform, and the performance parameters calculated by the software were compared with the laboratory test results. The following research results were obtained. Firstly, single impact energy, impact frequency and impact power increase with the increase of pump displacement or the decrease of flow bean diameter, and they increase firstly and then decrease with the increase of area difference between the upper and lower chambers. Secondly, with the increase of hammer weight, single impact energy and impact power increase, but the impact frequency decreases slowly. Thirdly, with the increase of hammer travel, single impact energy presents an increasing trend, impact frequency presents a decreasing trend and impact power basically remains unchanged. Fourthly, with the increase of valve core weight, single impact energy presents an increasing trend, while both impact frequency and impact power decrease. Fifthly, the parameter combination corresponding to the optimal single impact energy and impact power is A5B1C5D4E3F2, and the effect of displacement on single impact energy and impact power is the greatest. It is concluded that under the existing displacement and pressure of drilling pumps, the impact performance of the hydraulic hammer can be increased effectively by improving the structure of the hydraulic hammer and thus increasing its work displacement. Research on DNA methylation offers great potential for the identification of biomarkers that can be applied for accurately assessing an individual's risk for cancer. In this article, we try to find the ideal epigenetic genes involved in colorectal cancer (CRC) based on a CRC database and our CRC cohort. The top 20 genes with an extremely high frequency of hypermethylation in CRC were identified in the latest database. Remarkably, 3 HOXA genes were included in this list and ranked at the top. The percentage of methylation in the HOXA5, HOXA2, and HOXA6 genes in CRC were up to 67.62, 58.36, and 31.32%, respectively, and ranked first in CRC among all human tumor tissues. Paired colorectal tumor samples and adjacent non-tumor colorectal tissue samples and four CRC cell lines were selected for MethylTarget™ assays. The results demonstrated that CRC tissues and cells had a stronger methylation status around the 3 HOXA gene promoter regions compared with adjacent non-tumor colonic tissue samples. The Receiver operator characteristic curve (ROC) curves for HOXA genes show excellent diagnostic ability in distinguishing tissue from healthy individuals and CRC patients, especially for Stage I patients (AUC = 0.9979 in HOXA2, 0.9309 in HOXA5, and 0.8025 in HOXA6). An association analysis between the methylation pattern of HOXA genes and clinical indicators was performed and found that HOXA2 methylation was significantly associated with age, N, stage, M, lymphovascular invasion, perineural invasion, lymph node number. HOXA5 methylation was associated with age, T, M, stage, and tumor status, and HOXA6 methylation was associated with age and KRAS mutation. Notably, we found that the highest methylation of HOXA5 and HOXA2 occurs in the early stages of colorectal cancer tissues such as stage I, N0, MO, and non-invasive tissues. The methylation levels declined as tumors progressed. However, methylation level at any stage of the tumor was still significantly higher than in normal tissues (p < 0.0001). The mRNA of the 3 HOXA genes was downregulated in early tumor stages due to hypermethylation of CpG islands adjacent to the promoters of the genes. In addition, hypermethylation of HOXA5 and HOXA6 mainly occurred in patients < 60 years old and with MSI-L, MSS, CIMP.L and non-CIMP tumors. Together, this suggests that epigenetic silencing of 3 adjacent HOXA genes may be an important event in the progression of colorectal cancer.

The Derbugan metallogenic belt is located in the eastern Mongolia-Xing'an orogenic belt, between the Ertix-Zuolun-Heihe suture zone and Mongolia-Okhotsk suture zone. Affected by Yanshanian movement, magmatic intrusion is heavy in the area, accompanied by intense gold mineralization in Derbugan area. Through the study of Shabaosi, Xiajibaogou and Xiaoyinuogaigou gold deposits, the paper analyzes the metallogenic characteristics and tectonic background of the gold deposits. The deposits can be classified into four types, including epi-mesothermal altered rock type hosted by sandstone, epithermal type hosted by volcanic rock, magmatic hydrothermal altered rock type related to deep intrusive complex, and tectonic-hydrothermal altered rock type occurred in ductile shear zone. The mineralization time of different types of gold deposits is concentrated mainly in Late Jurassic-Early Cretaceous of Yanshanian. According to the H-O-Pb isotope results, the ore-forming hydrothermal fluid is mainly the mixtures of magmatic water and atmospheric precipitation, with the metallogenic material from mantle-derived magma, mixed with minor crustal material. Different types of gold deposits have certain internal genetic relations and belong to the product of magmatic evolution at various stages in the same magmatic source region. The metallogenesis of the endogenetic deposits of nonferrous and precious metals in the region is characteristically distributed at the intersections of a series of parallel NE-trending structural belts and NW-trending secondary structures.
