Abstract

Platelets are very abundant in the blood, where they play a role in hemostasis, inflammation, and immunity. When activated, platelets undergo a conformational change that allows the release of numerous effector molecules as well as the production of extracellular vesicles, which are circulating submicron vesicles (10 to 1,000 nm in diameter) released into the extracellular space. Extracellular vesicles are formed by the budding of platelet and they carry some of its contents, including nucleic acids, surface proteins, and organelles. While platelets cannot cross tissue barriers, platelet-derived extracellular vesicles can enter the lymph, bone marrow, and synovial fluid. This allows the transfer of diverse contents carried by these platelet-derived vesicles to cell recipients and organs inaccessible to platelets where they can perform many functions. This review highlights the importance of these platelet-derived extracellular vesicles under different physiological and pathophysiological conditions. [ABSTRACT FROM AUTHOR]

Abstract

Platelets were isolated from healthy donors (n = 10, green dots), COVID-19 patients treated with aspirin (n = 10, red dots), or not treated with aspirin (n = 10, blue squares). Given that aspirin was found without effect on platelet functions in certain proinflammatory conditions, [15][16][27] and since it is unclear what mediates platelet activation in COVID-19, the question of whether aspirin would have any effect on platelet activation in COVID-19 patients remained to be addressed. Thus, an aspirin regimen can reduce platelet aggregation induced by low concentrations of thrombin and collagen and the production of TxB SB 2 sb , but the effects of aspirin on platelet aggregation are lost with higher concentrations of agonists. Abstract

The sequencing of the human genome has revealed that the bulk of the genome is transcribed into noncoding RNAs, including microRNAs. These microRNAs are deregulated in cancer where they regulate a wide array of functions associated with tumorigenesis. Glioblastoma (GBM) is the most common and aggressive primary intracranial tumor of the adult brain. miRNAs are short noncoding RNAs that broadly regulate gene expression. They exert their actions via transcriptional, post-transcriptional, and epigenetic mechanisms that are poorly understood. Numerous miRNAs are deregulated in GBM, where their expression levels can serve as biomarkers. They can act as either oncogenes or tumor suppressors in GBM by targeting the expression of numerous tumor-suppressors or oncogenes. miRNAs regulate all GBM malignancy aspects including cell proliferation, survival, motility, migration, invasion, angiogenesis, cancer stem cell biology, microenvironment modulation, immune escape, and therapy resistance. MiRNAs can also be secreted via exosomes or microvesicles in body .uids, where they can be used as diagnosis and/or prognosis biomarkers. Considering their deep involvement in GBM malignancy, numerous studies are currently undertaken to exploit miRNAs as therapeutic agents or targets. 49. Targeting IL-1β in patients with advanced Helicobacter pyloriinfection: a potential therapy for gastric cancer

Abstract

Helicobacter pylori(H. pylori) infection is a causal factor of gastric cancer. Among the cytokines secreted during this infection, IL-1β is highly associated with promotion and progression of gastric cancer. On the therapeutic front, eradication of H. pyloriwas thought to be efficient to restore gastric homeostasis. However, successful H. pylorieradication in patients with advanced stages (intestinal metaplasia) failed to diminish inflammation that is due to heightened Th17 response and elevated IL-1β levels. In fact, association between these two components was established, suggesting that IL-1β is a critical target in these cases. In this review, we will discuss the functional relevance of IL-1β in advanced H. pyloriinfection and how its targeting may bring clinical benefit.
