The diffuse and invisible pollution caused by microplastic (MP) represents a current and global concern. While the fragmentation of plastic debris into smaller particles occurs in rivers, few data about MP effects are available on freshwater streams compared to the marine environment. Also, still too few studies combine in situ approach with the assessment of the effects of MP on organisms. Plastic-Loire project, funded by the urban community of Angers Loire Métropole, demonstrates its innovation through the complementary field/laboratory approach that aims at characterizing in situ contamination of MP in the Loire river and at evaluating MP effects on a freshwater bivalve. Firstly, MP primary characteristics (concentration, size, type, shape and colour) were determined both in sediment and in biotic matrices (polychaetes and bivalves) sampled in the Loire, longest river in France. In parallel, the ecotoxicity of MP was assessed on bivalves C. fluminea in laboratory. Organisms were exposed during 21 days to field derived MP, at 4 concentrations (0.008, 10, 100 and 500 µg.L-1). The exposure experiment was realized from an ecological realistic point of view: exposure of organisms to MP with a shape, a composition and a range of concentration similar to those found in the environment. While the condition index and the filtration capacity were not impacted by a MP exposure, the measured biochemical biomarkers involved in immune responses (CAT, GST, AchE and MDA) presented significant differences in digestive mass and gills of the organisms according to the MP concentration. Active biomonitoring involves exposure of calibrate and well-known organisms to environmental pollution and is increasingly used in ecological studies. Despite of the advantages of caging (standardization of some biotic and abiotic parameters), caged organisms are subject to some constraints which can influence the organisms’ responses to pollution and therefore can be considered as confounding factors. In the way to make biomarkers efficient tools in active biomonitoring, it is necessary to precisely characterize the possible effects of constraints linked to caging on organism physiology. For this purpose, several studies were conducted on threeBspined sticklebacks (Gasterosteus aculeatus) to specify the impacts of i) fish-density inside cages, ii) short-transport after caging, iii) movement limitation and iv) food access restriction induced by caging. The optimization of caging conditions has been validated by assessment of several biomarkers including innate immune responses, antioxidant systems, metabolic detoxification, synaptic transmission and energy metabolism. Results have shown that a high density of fish (> 2.31 kg/m3) and a short transport after caging have induced an oxidative stress manifested by an overproduction of reactive oxygen species and a stimulation of the antioxidant system. The confinement stress had small impact on sticklebacks’ biomarkers. However, this stress seemed to accentuate the negative effects of food restriction on these biomarkers, particularly during their breeding period. Out of breeding time, most of investigate biomarkers were not impacted by food access restriction. These works have allowed to specify the conditions of application and interpretation of biomarkers during active monitoring using stickleback to ensure a good diagnosis of the quality of water bodies with this species.

Les stratégies de biosurveillance active ont maintes fois été utilisées dans les milieux aquatiques avec divers organismes tels que les gammares, les bivalves ou encore les poissons. Elles présentent la particularité de pouvoir exposer à une pollution environnementale des organismes calibrés et dont les antécédents sont bien connus, sans nécessairement avoir à les retrouver sur les sites étudiés. Cependant, malgré les avantages de cette approche (contrôle des paramètres biotiques et abiotiques), il peut être pertinent de vérifier la plasticité des organismes face à de l'encagement. Afin de limiter les erreurs d'interprétation, les effets des contraintes liées à l'encagement doivent être caractérisés. Plusieurs études basées sur l'épinoche à trois épines (Gasterosteus aculeatus) ont ainsi été réalisées afin d'évaluer l'impact de certains de ces facteurs sur une batterie de biomarqueurs représentative de plusieurs fonctions biologiques (système immunitaire inné, système antioxydant et de détoxification, paramètres énergétiques, transmission synaptique). Les premiers travaux ont porté sur les effets de la densité de poissons par cage et le transport des organismes avant et après l'encagement [2]. Les résultats ont montré qu'une densité élevée (> 2,31 kg/m3 ) et le transport induisaient un stress oxydatif caractérisé par une surproduction d'espèces réactives de l'oxygène ainsi qu'une stimulation du système antioxydant. À la suite de cette étude, d'autres facteurs tels que le confinement et la restriction d'accès à la nourriture ont été investigués. Les résultats ont montré que le confinement impactait peu les biomarqueurs étudiés. En revanche, cette contrainte a semblé accentuer les effets néfastes de la restriction alimentaire, surtout pendant la période de reproduction (printemps-été). En dehors de cette période, la plupart des biomarqueurs n'ont pas été impactés par la limitation d'accès à la nourriture. L'ensemble de ces travaux a permis de préciser les conditions d'encagement des épinoches en vue de leur utilisation en biosurveillance. La démarche a ensuite été déployée in situ en aval d'une station d'épuration (Sacy-le-Grand, Oise) afin d'évaluer, en particulier, l'effet de la mise en place d'un système de lagunage. Les épinoches encagées en entrée de lagunage ont présenté une inhibition du système immunitaire inné ainsi qu'une augmentation de certains paramètres liés au stress oxydant. Cet impact de l'effluent de STEP était atténué en sortie de lagunage, mettant en évidence l'effet bénéfique de cette installation.
