Abstract

The mechanisms governing adipose tissue macrophage (ATM) metabolic adaptation during diet-induced obesity (DIO) are poorly understood. In obese adipose tissue, ATMs are exposed to lipid fluxes, which can influence the activation of specific inflammatory and metabolic programs and contribute to the development of obesity-associated insulin resistance and other metabolic disorders. In the present study, we demonstrate that the membrane ATP-binding cassette g1 (Abcg1) transporter controls the ATM functional response to fatty acids (FAs) carried by triglyceride-rich lipoproteins, which are abundant in high-energy diets. Mice genetically lacking Abcg1 in the myeloid lineage presented an ameliorated inflammatory status in adipose tissue and reduced insulin resistance. Abcg1-deficient ATMs exhibited a less inflammatory phenotype accompanied by a low bioenergetic profile and modified FA metabolism. A closer look at the ATM lipidome revealed a shift in the handling of FA pools, including a redirection of saturated FAs from membrane phospholipids to lipid droplets, leading to a reduction in membrane rigidity and neutralization of proinflammatory FAs. ATMs from human individuals with obesity presented the same reciprocal relationship between ABCG1 expression and this inflammatory and metabolic status. Abolition of this protective, anti-inflammatory phenotype in Abcg1-deficient ATMs was achieved through restoration of lipoprotein lipase (Lpl) activity, thus delineating the importance of the Abcg1/Lpl axis in controlling ATM metabolic inflammation. Overall, our study identifies the rewiring of FA pools by Abcg1 as a major pathway orchestrating ATM plasticity and insulin resistance in DIO. 18. Breathlessness and 'exacerbation' questions predictive for incident COPD (MARKO study): data after two years of follow-up

Abstract

Aims: To determine the predictability of the MARKO questionnaire and/or its domains, individually or in combination with other markers and characteristics (age, gender, smoking history, lung function, 6-min walk test (6 MWT), exhaled breath temperature (EBT), and hsCRP for the incident chronic obstructive pulmonary disease (COPD) in subjects at risk over 2 years follow-up period).Participants and Methods: Patients, smokers/ex-smokers with >20 pack-years, aged 40-65 years of both sexes were recruited and followed for 2 years. After recruitment and signing the informed consent at the GP, a detailed diagnostic workout was done by the pulmonologist; they completed three self-assessment questionnaires-MARKO, SGRQ and CAT, detailed history and physical, laboratory (CBC, hsCRP), lung function tests with bronchodilator and EBT. At the 2 year follow-up visit they performed: the same three self-assessment questionnaires, history and physical, lung function tests and EBT.Results: A sample of 320 subjects (41.9% male), mean (SD) age 51.9 (7.4) years with 36.4 (17.4) pack-years of smoking was reassessed after 2.1 years. Exploratory factor analysis of MARKO questionnaire isolated three distinct domains (breathlessness and fatigue, "exacerbations", cough and expectorations). We have determined a rate for incident COPD that was 4.911/100 person-years (95% CI [3.436-6.816]). We found out that questions about breathlessness and "exacerbations", and male sex were predictive of incident COPD after two years follow-up (AUC 0.79, 95% CI [0.74-0.84], p < 0.001). When only active smokers were analyzed a change in EBT after a cigarette (Delta EBT) was added to a previous model (AUC 0.83, 95% CI [0.78-0.88], p < 0.001).Conclusion: Our preliminary data shows that the MARKO questionnaire combined with EBT (change after a cigarette smoke) could potentially serve as early markers of future COPD in smokers., Funding Agencies|GlaxoSmithKline [CRT114338]

Abstract

Aberrant plasma protein glycosylation is associated with a wide range of diseases, including diabetes, cardiovascular, and immunological disorders. To investigate plasma protein glycosylation alterations due to weight loss and successive weight-maintenance diets, 1850 glycomes from participants of the Diogenes study were analyzed using Ultra-High-Performance Liquid Chromatography (UHPLC). The Diogenes study is a large dietary intervention study in which participants were subjected to a low-calorie diet (LCD) followed by one of five different weight-maintenance diets in a period of 6 months. The most notable alterations of the plasma glycome were 8 weeks after the subjects engaged in the LCD; a significant increase in low-branched glycan structures, accompanied by a decrease in high-branched glycan structures. After the LCD period, there was also a significant rise in N-glycan structures with antennary fucose. Interestingly, we did not observe significant changes between different diets, and almost all effects we observed immediately after the LCD period were annulled during the weight-maintenance diets period.

Abstract

Immunoglobulin G (IgG) is the most abundant antibody in the blood and plays a critical role in host immune defence against infectious pathogens. IgG glycosylation is known to modulate IgG effector functions and is involved in the development and progression of disease. Because the reactivity of salivary IgG mirrors that of plasma IgG, saliva is an attractive alternative to blood for studies in which antibody analysis is intended to provide an indication of a subject's immune status. Therefore, in this study, we described a rapid and effective method for comparative N-glycome analysis of IgG from human plasma and saliva samples. Plasma and saliva were collected from 9 healthy volunteers within a 2- hour time window. IgGs were affinity-purified from the two biofluids with protein G-agarose beads and then denatured and enzymatically deglycosylated. The released N-glycans were analysed by ultra-high performance liquid chromatography with hydrophilic interaction analysis (HILIC-UPLC). IgG from saliva exhibited reduced galactosylation and sialylation compared with IgG from plasma, whereas core fucosylation remained unchanged. The slightly proinflammatory profile of salivary IgG may be due to higher exposure to pathogens compared with plasma IgG. This study provides an additional tool for the analysis of salivary IgG glycosylation and highlights its potential as a surrogate biomarker for the study of plasma IgG.

Over half of all proteins are altered by covalently bound glycans that are crucial for maintaining a normal physiological role of glycoproteins. Alterations in sialylation and fucosylation of circulating glycoproteins have recently been shown to be affected by the diet, however, this is the first study that considered plasma proteins’ susceptibility to different dietary regimes for weight control after the initial weight loss. To investigate plasma protein glycosylation alterations due to weight loss and successive weight-maintenance diets, 1850 glycomes from participants of the Diogenes study were analyzed using Ultra-High-Performance Liquid Chromatography (UHPLC). The most notable alteration of the plasma glycome was 8 weeks after the subjects engaged in the LCD ; a significant increase of low-branched glycan structures, accompanied by a decrease of high- branched glycan structures. After the LCD period, there was also a significant rise in fucosylated N-glycan structures, both core and antennary. Moreover, we have observed a significant decrease in trigalactosylated and trisialylated glycans and a concomitant increase in tetragalactosylated and tetrasialylated glycan structures. Interestingly, we did not observe significant changes between different diets, and almost all effects we have observed immediately after the LCD period were annulled during the weight maintenance diets.

Korištenje uređaja koji je povezan na internet korisnike automatski izlaže mogućem napadu i gubitku osobnih podataka koji su pohranjeni na tom uređaju. Potrebno je implementirati metode zaštite protiv mogućih napada. U radu je opisan postupak implementacije vatrozida u običnu korisničku mrežu te podešavanje njegovih mogućnosti. Vatrozid je najbolje sigurnosno rješenje budući da ima mogućnosti prijavljivanja uljeza i/ili zlonamjernog softvera u sustavu kao i mogućnost njihovog zaustavljanja. Implementacijom vatrozida je admnisnitrator dobio nadzor nad cjelokupnom mrežom. Usage of devices that have the ability to connect to the internet automatically puts users in danger by exposing them to cyber-attacks. It is necessary to implement security methods against possible cyber-attacks. This paper explains the firewall implementation procedure as well as modification of its services. Firewall is the best security solution because of its ability to locate and stop attackers and their malware. By implementing the firewall, administrator gains control of the entire network.

Abstract

Obesity-induced inflammation usually starts with the activation of the innate immune system and consequently activates the adaptive immune system which leads to the production of a more inflammatory repertoire of cytokines and pathogenic immunoglobulin G (IgG) antibodies. Each IgG molecule has an N-glycosylation site on each of its heavy chains that influences its structural stability, conformation and half-life, as well as effector functions. For example, a higher proportion of sialic acid and galactose residues is associated with anti-inflammatory activity. IgG glycome has already been well studied and its alterations correlate with different diseases and conditions, including high body mass index (BMI) and central adiposity. Still, this is the first study that studied its susceptibility to different dietary regimes for weight control after the initial weight loss. To explore IgG glycosylation alterations due to weight loss and subsequent weight-maintenance diets, we analyzed 1850 IgG glycomes from subjects of the Diogenes study, one of the largest dietary intervention studies in which subjects underwent a low-calorie diet (LCD) followed by one of five weight-maintenance diets (low protein/low glycaemic index, low protein/high glycaemic index, high protein/low glycaemic index, high protein/high glycaemic index and control) in a period of 6 months when the subjects were vulnerable to regain the weight. The most prominent alteration of the glycome was 8 weeks after the subjects underwent LCD ; a statistically significant decrease of agalactosylated and accompanying increase of sialylated N-glycans in IgG glycome presents shifting from pro- to the anti-inflammatory activity of IgG. These observations were expected knowing that apart from being an energy storage depot, adipose tissue is also an endocrine organ that produces adipokines such as interleukin (IL-6) and tumor necrosis factor-α (TNF-α), cytokines already well-linked with inflammation. Since adipose tissue production is affected by the degree of adiposity, the attenuation of systematic inflammation is an obvious outcome after a reduction diet. Unfortunately, we did not observe significant changes between different diets, and an increased abundance of glycans with bisecting GlcNAc and decreased abundance of sialylated ones, which were present regardless of diet type, suggest a pro- inflammatory direction of IgG glycome alteration in the period of weight-maintaining or regaining.

Povećanjem brzina kretanja zrakoplova manifestiraju se pojave koje itekako imaju utjecaj na letne karakteristike zrakoplova. Stlačivost zraka se više ne zanemaruje prilikom izračuna. Pojave poput udarnih valova, odvajanje strujnica, zračnog zida, vibracija itd. se počinju javljati u područjima okozvučnog strujanja. Kako bi zrakoplovi postizali što bolje performanse na tim brzinama i brzinama većima od brzine zvuka, kontinuirano se radi na poboljšanju aeroprofila i ostali aerodinamičkih površina. Poznavanje tih pojava je također vrlo bitan čimbenik u osiguravanju tih performansi.

Abstract

Invertases are glycosidases applied for synthesis of alkyl glycosides that are important and effective surfactants. Stability of invertases in the environment with increased content of organic solvent is crucial for increase of productivity of glycosidases. Their stabilityis significantly influenced by N-glycosylation. However, yeast N-glycosylation pathways may synthesize plethora of N-glycan structures. A total natural crude mixture of invertase glycoforms (EINV) extracted from Saccharomyces cerevisiae was subfractionatedby anion-exchange chromatography on industrial monolithic supports to obtain different glycoforms (EINV1–EINV3). Separated glycoforms exhibited different stabilities in wateralcohol solutions that are in direct correlation with the amount of phosphate bound to N-glycans. Observed differences in stability of different invertase glycoforms were used to improve productivity of methyl β-d-fructofuranoside (MF) synthesis. The efficiency and yield of MF synthesis were improved more than 50% when the most stabile glycoform bearing the lowest amount of phosphorylated N-glycans is selected and utilized. These data underline the importance of analysis of glycan structures attached to glycoproteins, demonstrate different impact of N-glycans on the surface charge and enzyme stability inregard to particular reaction environment, and provide a platform for improvement of yield of industrial enzymatic synthesis by chromatographic selection of glycoforms on monolithic supports.

Abstract

Immunoglobulin G (IgG) is the most abundant serum antibody which structural characteristics and effector functions are modulated through the attachment of various sugar moieties called glycans. Composition of the IgG N-glycome changes with age of an individual and in different diseases. Variability of IgG glycosylation within a population is well studied and is known to be affected by both genetic and environmental factors. However, global inter-population differences in IgG glycosylation have never been properly addressed. Here we present population-specific N-glycosylation patterns of IgG, analyzed in 5 different populations totaling 10,482 IgG glycomes, and of IgG’s fragment crystallizable region (Fc), analyzed in 2,579 samples from 27 populations sampled across the world. Country of residence associated with many N-glycan features and the strongest association was with monogalactosylation where it explained 38% of variability. IgG monogalactosylation strongly correlated with the development level of a country, defined by United Nations health and socioeconomic development indicators, and with the expected lifespan. Subjects from developing countries had low levels of IgG galactosylation, characteristic for inflammation and ageing. Our results suggest that citizens of developing countries may be exposed to environmental factors that can cause low-grade chronic inflammation and the apparent increase in biological age.
