Abstract Background Acute mountain sickness (AMS) is considered the most common altitude sickness. It can be detrimental to the health of tourists who rapidly ascend high mountains, and can also impair the performance of individuals who move to the plateau for work or education. Acupuncture has been shown to improve AMS as a simple, safe, and effective nonpharmacological method, in case electro-thumbtack needle (ETN) is a more convenient form of acupuncture for both doctor and patient. There are no studies validating the effectiveness of electro-thumbtack needle in improving symptoms in participants with AMS. In this study, we will conduct a randomized controlled trial to evaluate the clinical efficacy and safety of electro-thumbtack needle in participants with AMS. Our hypothesis is that electro-thumbtack needle is safe and effective in treating participants with AMS. Methods This study is a single-center, randomized, single-blinded, and placebo-controlled study involving at least 114 participants who were diagnosed with AMS. The participants randomly assigned in a 1:1 ratio to the electro-thumbtack needle group and the sham acupuncture group. The treatment protocol involved stimulation of seven predefined acupuncture points, including Zhong Wan (RN12), bilateral Nei Guan (PC6), bilateral He Gu (LI4), and bilateral Tai Yang (EX-HN5), for approximately one minute each, with continuous application over 48 h. The primary outcome was improvement in 2018 Lake Louise score (LLS) after 48 h of treatment. Secondary outcome indicators included the incidence of participants with moderate-to-severe AMS (AMS > 5)and AMS, the LLS, visual analogue scale of headache, clinical functioning scores, the Groningen Sleep Quality Survey, the Stanford Somnolence Scale, blood pressure, oxygen saturation, and heart rate, in addition to treatment-related adverse events were also captured. Discussion This trial aims to ascertain the therapeutic benefits of ETN in mitigating AMS symptoms, thereby contributing to the evidence base for traditional medical practices, particularly acupuncture, in high-altitude medicine. Abstract The biodegradable polymer poly-β-hydroxybutyrate (PHB) is a promising carbon source for biological mitigation of nitrogen pollution, a significant problem in aquaculture that physical and chemical methods have not provided a comprehensive solution. Here we investigated the impact of PHB on the zero-water-change largemouth bass culture by 30- and 40-day experiments. PHB loaded into the filter circulation pump at 4 g L− 1, optimum value determined by the first experiment, significantly reduced the levels of nitrate by 99.65%, nitrite by 95.96%, and total nitrogen by 85.22% compared to the control without PHB. PHB also significantly increased denitrifying bacteria (e.g., Proteobacteria and Fusobacteria) and expression of denitrification genes (e.g., nirK and nirS) in the microbial community, improving growth and health parameters of largemouth bass. While the impact may vary in other culture systems, PHB thus demonstrated its remarkable utility in aquaculture, highlighting ecological assessment and application to larger aquaculture operations as future considerations.

The pregnane X receptor (PXR)/nuclear factor-kB (NF-kB) signaling pathway plays a critical role in regulating toxin-induced inflammation and apoptosis in mammals. Whether dietary curcumin (CUR) can prevent copper (Cu)-induced liver injury via this signaling pathway remains to be established in aquatic animals. Juvenile largemouth bass (Micropterus salmoides) were exposed to dietary Cu and CUR treatments for 8 weeks. The results showed that chronic Cu exposure induced oxidative stress, causing liver function damage and liver injury. Cu exposure stimulated inflammation by regulating nf-kb and pro-inflammatory genes such as tnfα and il-1β and promoted apoptotic signals in the liver by modulating bcl2 and casp3 mRNA levels. In addition, Pearson correlation analysis verified that inflammatory and apoptotic responses were important indicators of Cu-induced liver injury. CUR attenuated stress responses by enhancing the antioxidant system. Importantly, CUR significantly stimulated PXR mRNA and protein levels in the Cu + CUR group and suppressed NF-κB activation to inhibit the inflammatory and apoptotic signaling cascade. These results suggest that CUR may be an effective activator of PXR in teleost fishes, exerting cytoprotective effects on Cu-induced liver injury via a PXR-mediated NF-κB repression mechanism. In conclusion, this study is the first to demonstrate that CUR may act as a potent PXR ligand that exerts hepatoprotective effects against Cu-induced liver injury. The findings shed light on the specific regulatory role of the PXR/NF-κB signaling pathway in liver pathogenesis and its potential as a therapeutic target in teleost fishes.

Enteritis often limits intensive aquaculture, where bile acids (BAs), a group of steroid molecules, are used to prevent intestinal diseases. However, the functions of certain BAs are poorly understood. In this study, grass carp with enteritis (EGC) and healthy grass carp (HGC) were sampled, and the BAs' composition was analyzed. Results showed distinct differences in the BA profiles: EGC exhibited significantly elevated levels of taurodeoxycholic acid (TDCA), lithocholic acid (LCA), glycoursodeoxycholic acid (GUDCA), and glycohyocholic acid (GHCA) (P < 0.05), and decreased levels of TCDCA, taurocholic acid (TCA), and cholic acid (CA) in the intestine compared with their levels in HGC. Subsequently, five diets containing four typical BAs (CA, chenodeoxycholic acid [CDCA], deoxycholic acid [DCA], and LCA) at the same concentration (0.1 %) were formulated to feed the juvenile grass carps (6.29 ± 0.05 g) for 8 weeks. DCA alone increased body weight, reduced the feed conversion ratio (FCR), and ameliorated histological intestinal inflammatory hallmarks. Conversely, individual supplementation with CDCA, LCA, or CA decreased body weight and increased FCR and exacerbated the histological hallmarks of enteritis. Furthermore, DCA markedly reduced the mRNA expression levels of endoplasmic reticulum stress (ERS)-related genes in the intestine, including glucose-regulated protein 78 (GPR78), activating transcription factor 6 (ATF6), inositol-requiring enzyme 1, and protein kinase RNA-like ER kinase, as well as genes encoding proinflammatory cytokines, such as interleukin (IL)-1 beta, IL-8, and tumor necrosis factor α (TNF-α). Supplementation with CDCA, LCA, or CA promoted the expression of ERS- and proinflammatory-cytokine-related genes. Additionally, Actinobacteriota abundance negatively correlated with intestinal mRNA expression levels of TNF-α, ATF6, IL-1β, GPR78, and IL-8, and Candidatus-Arthromitus abundance positively correlated with these inflammatory markers (P < 0.05). In conclusion, our results demonstrated that BAs play differential roles in modulating the enteritis process and that DCA may have a specific anti-inflammatory role in grass carp.

Abstract Broad bean (Vicia faba L.) has received attention from the food and animal feed industries as a sustainable, inexpensive, and versatile plant‐based protein source. However, long‐term consumption of broad bean, especially unprocessed ones, can lead to hepatic lipid accumulation. To investigate how broad bean affected hepatic lipid metabolism and its underlying mechanisms, we conducted a multi‐omics analysis of crisp grass carp (Ctenopharyngodon idellus C. et V) model with hepatic steatosis after feeding with single broad bean for 120 days. The glycolysis inhibition, increased de novo lipogenesis, abnormalities in lipid metabolism, and activation of oxidative stress were the key processes involved in broad bean‐induced hepatic lipid accumulation. Disturbances in intestinal flora caused by damage to the intestinal barrier and increased lipopolysaccharide caused by broad bean promoted hepatic lipid accumulation. Reduction of antinutritional factors and balanced diet offer potential strategies to mitigate the liver damage caused by broad bean consumption. Overall, our findings provided a theoretical basis for the wide application of broad bean as animal feeds.

Abstract Electroencephalography (EEG) microstate analysis is a neuroimaging analytical method that has received considerable attention in recent years and is widely used for analysing EEG signals. EEG is easily influenced by internal and external factors, which can affect the repeatability and stability of EEG microstate analysis. However, there have been few reports and publicly available datasets on the repeatability of EEG microstate analysis. In the current study, a 39-year-old healthy male underwent a total of 60 simultaneous electroencephalography and electrocardiogram measurements over a period of three months. After the EEG recording was completed, magnetic resonance imaging (MRI) was also conducted. To date, this EEG dataset has the highest number of repeated measurements for one individual. The dataset can be used to assess the stability and repeatability of EEG microstates and other analytical methods, to decode resting EEG states among subjects with open eyes, and to explore the stability and repeatability of cortical spatiotemporal dynamics through source analysis with individual MRI.

A meniscus injury is a prevalent condition affecting the knee joint. The construction of a subjective prediction model for meniscus injury represents a potentially invaluable diagnostic tool for physicians. Nevertheless, given the variability of pathological manifestations among individual patients, machine learning-based models may produce errors when attempting to predict specific medical records. In order to mitigate this issue, the present study suggests the incorporation of metric learning within the machine learning (ML) modelling process, with the aim of reducing the intra-class spacing of comparable samples and thereby enhancing the classification accuracy of individual medical records. This work has not yet been attempted in the field of knee joint prediction. The findings demonstrate that the adoption of metric learning produces better optimal outcomes. Compared to machine learning baseline models, F1 was increased by 2%.

Habitual dietary changes have the potential to induce alterations in the host’s gut microbiota. Mandarin fish (Siniperca chuatsi), an aquatic vertebrate species with distinct feeding habits, were fed with natural feeds (NF) and artificial feeds (AF) to simulate the effects of natural and processed food consumption on host gut microbiota assemblages. The results showed that the alpha diversity index was reduced in the AF diet treatment, as lower abundance and diversity of the gut microbiota were observed, which could be attributed to the colonized microorganisms of the diet itself and the incorporation of plant-derived proteins or carbohydrates. The β-diversity analysis indicated that the two dietary treatments were associated with distinct bacterial communities. The AF diet had a significantly higher abundance of Bacteroidota and a lower abundance of Actinomycetota, Acidobacteriota, and Chloroflexota compared to the NF group. In addition, Bacteroidota was the biomarker in the gut of mandarin fish from the AF treatment, while Acidobacteriota was distinguished in the NF treatments. Additionally, the increased abundance of Bacteroidota in the AF diet group contributed to the improved fermentation and nutrient assimilation, as supported by the metabolic functional prediction and transcriptome verification. Overall, the present work used the mandarin fish as a vertebrate model to uncover the effects of habitual dietary changes on the evolution of the host microbiota, which may provide potential insights for the substitution of natural foods by processed foods in mammals.

Interstitial fluid, owing to its similarity to blood components and higher sensitivity and specificity, finds widespread application in disease diagnosis and tumor marker detection. However, collecting interstitial fluid, particularly from the deep subcutaneous connective tissue, remains challenging. • This study aimed to compare three different collection methods - push-pull perfusion, multi-filament nylon thread implantation, and tissue centrifugation - for collecting interstitial fluid from the subcutaneous connective tissue layer of mini-pigs. High-performance ion chromatography was employed to analyze the conventional cation components in the samples and compare ion composition analysis between the different methods. • Results indicated that while the distribution of conventional cations in the interstitial fluid collected by the three methods was generally consistent, there were slight variations in the detection rates and concentrations of different ions. Hence, suitable collection methods should be selected based on the ions or collection sites of interest.

Biochar and organic fertilizer can significantly increase soil organic carbon (SOC) and promote agricultural production, but it is still unclear how they affect forest SOC after. Here, low-quality plantation soil was subjected to four distinct fertilization treatments: (CK, without fertilization; BC, tea seed shell biochar alone; OF, tea meal organic fertilizer alone; BCF, tea seed shell biochar plus tea meal organic fertilizer). Cunninghamia lanceolata (Lamb.) Hook and Cyclobalanopsis glauca (Thunb.) Oersted seedlings were then planted in pots at the ratios of 2:0, 1:1, and 0:2 (SS, SQ, QQ) and grown for one year. The results showed that the BCF treatment had the best effect on promoting seedling growth and increasing SOC content. BCF changed soil pH and available nutrient content, resulting in the downregulation of certain oligotrophic groups (Acidobacteria and Basidiomycetes) and the upregulation of eutrophic groups (Ascomycota and Proteobacteria). Key bacterial groups, which were identified by Line Discriminant Analysis Effect Size analysis, were closely associated with microbial biomass carbon (MBC) and SOC. Pearson correlation analysis showed that bacterial community composition exhibited a positive correlation with SOC, MBC, available phosphorus, seedling biomass, and plant height, whereas fungal community composition was predominantly positively correlated with seedling underground biomass. It suggested that environmental differences arising from fertilization and planting patterns selectively promote microbial communities that contribute to organic carbon formation. In summary, the combination of biochar and organic fertilizers would enhance the improvement and adaptation of soil microbial communities, playing a crucial role in increasing forest soil organic carbon and promoting tree growth.

Diets with high carbohydrate (HC) was reported to have influence on appetite and intermediary metabolism in fish. To illustrate whether betaine could improve appetite and glucose-lipid metabolism in aquatic animals, mandarin fish (Siniperca chuatsi) were fed with the HC diets with or without betaine for 8 weeks. The results suggested that betaine enhanced feed intake by regulating the hypothalamic appetite genes. The HC diet-induced downregulation of AMPK and appetite genes was also positively correlated with the decreased autophagy genes, suggesting a possible mechanism that AMPK/mTOR signaling might regulate appetite through autophagy. The HC diet remarkably elevated transcriptional levels of genes related to lipogenesis, while betaine alleviated the HC-induced hepatic lipid deposition. Additionally, betaine supplementation tended to store the energy storage as hepatic glycogen. Our findings proposed the possible mechanism for appetite regulation through autophagy via AMPK/mTOR, and demonstrated the feasibility of betaine as an aquafeed additive to regulate appetite and intermediary metabolism in fish.

In China, aquaculture is the primary source of aquatic products due to the decrease in wild fishery resources. In 2018, the total output of aquatic products in China expanded to 47.6 million tons, accounting for 58% of global aquaculture production. Intensive culture methods generally use significant quantities of feed however, approximately 75% of nitrogen in the feed is retained in aquaculture water, mainly as soluble nitrogen, such as ammonia nitrogen (NH4+-N) and nitrate (NO3–-N), owing to low feed-utilization rates during cultivation. At the same time, fishes generate a substantial amount of excreta, which will cause the increase of nitrogen compounds in water and negatively affects the quality of aquatic products. Serious problems could occur if nitrogen compounds are discharged into the environment, including the eutrophication of rivers, the deterioration of drinking water sources, and hazards to human health. Furthermore, nitrates can form potentially carcinogenic compounds, such as nitrosamines and nitrosamides, and nitrate consumption can cause methemoglobinemia in infants. The Second National Census of Pollution Sources survey showed that the total nitrogen emission from aquaculture was 99 100 tons in 2017. To protect the environment and human health, it is important to remove nitrogen from aquaculture tailwater before discharging it to the surrounding waters. Biological denitrification is considered the most promising approach since nitrate can be reduced to harmless nitrogen gas by bacteria. A sufficient carbon source is necessary during the heterotrophic denitrification process. To solve the problems mentioned above, external liquid carbon sources such as methanol, acetic acid, and glucose are added to the tailwater, but they are costly, require high-energy, and have high operating requirement. In contrast, agricultural wastes as a carbon source have exhibited significant economic advantages and high efficiency. Many aquaculture tailwater treatment systems often face variations in hydraulic retention times (HRT) and influent nitrate concentration (INC), which are caused by acute changes in tailwater characteristics and production, and HRT and INC often exert a profound effect on the treatment performance of biological treatment systems. Extensive research has confirmed that adding agricultural waste (such as corncob, woodchip and rice straw) to municipal sewage and industrial wastewater can effectively improve denitrification efficiency. However, the effect of using agricultural waste as denitrifying carbon source to treat aquaculture tail water remains unclear. Banana stalk (BS), a typical agricultural waste product, is used as a denitrifying carbon source for the first time in this study. The study investigated the effects of HRT and INC on the denitrification performance of BS, and provided a theoretical basis for the application of agricultural waste in aquaculture tailwater treatment. In this study, using BS as a carbon source and a towel as biological carrier, the performance of solid-phase denitrification under dynamic flow conditions was studied by using a 1-D column experiment. In the HRT optimization experiment, INC was maintained at 50 mg/L and operated under four HRTs (16 h, 20 h, 24 h and 28 h) for 14 days. The effluent NO3–-N, nitrite (NO2–-N), NH4+-N, Total nitrogen (TN), Total phosphorus (TP), and chemical oxygen demand (COD) removal efficiency were measured every 2 days. The optimal HRT of BS-DR (banana stalk-denitrification reactor) was optimized by one-way ANOVA analysis. Then, based on the optimization of HRT, the reactor was operated for 14 days under different INC (75 mg/L, 100 mg/L, and 125 mg/L). The sampling time interval and measurement indexes were the same as those of the HRT optimization experiment. The Illumina MiSeq high-throughput sequencing method was used to sequence and analyze the two hyper-variable regions (V3-V4) of the 16S rRNA gene of bacteria in the initial and final stages of the BS-DR. The results indicated that HRT and INC are the key factors affecting the denitrification performance of BS-DR. There was no significant difference in nitrate removal efficiency when the HRT was 20 h (96.71±1.36)%, 24 h (94.57±4.73)%, and 28 h (99.41±0.64)%, but they were significantly higher than that when the HRT was 16 h (87.53±7.95)%. Therefore, the optimal HRT for BS-DR was 20 h, and no nitrite accumulation. The second set of experiments was conducted using the optimal HRT obtained from the first set of experiments. The effluent nitrate concentration (ENC) and nitrate removal rate (NRR) of BS-DR increased significantly with increase in INC (P < 0.05), and the effluent COD decreased with increase in INC, and the proper INC for BS-DR was ≤50 mg/L. It is worth noting that BS-DR could completely remove NH4+-N in both experiments. In addition, HRT significantly affects the removal efficiency of TP, but INC has little effect. According to pyrosequencing analysis, the microbial community structure of BS-DR changed after long-term operation, with the relative abundances of Proteobacteria, Bacteroidetes, Campilobacterota, and Firmicutes increasing to 31.20%, 6.67%, 3.08%, and 4.26%, respectively, ensuring the efficient operation of the reactor. On the contrary, the relative abundances of Halobacterota, Desulfobacterota, Sva0485, Chloroflexi, and Verrucomicrobiota decreased to 10.39%, 5.13%, 2.82%, 2.00%, and 1.17 %, respectively, in the reactor. In addition, at the genus level, most of the dominant bacteria at the end of reactor operation play a role in denitrification and degradation of agricultural waste, which is significantly different from that at the beginning of the reactor operation.

Background : The biophysical properties of the meridian system, an important concept of traditional Chinese medicine, include low impedance, resounding voice, and high acoustic conductance, all of which are helpful for elucidating the essence of meridians. Objectives: To visualize the human pericardium meridian (PC) based on the resounding voice property of meridians. Methods : Visualization of the PC was performed by injection of fluorescein sodium at the PC6 acupoint (Neiguan) on the PC. Before injection, percussion active points (PAPs) were identified by the virtue of their resounding voice properties. After injection, the trajectories of fluorescein migration throughout the body surface were recorded and analyzed. The distribution of fluorescein in the tissue was further studied using cross-sections of hind limbs of mini-pigs, in which fluorescein was injected into low impedance points. Results : The identified PAP lines were colocalized with PC. Following intradermal fluorescein injection, 1-3 fluorescent lines, which were unrelated to the arm veins, were observed in 7 of 10 participants; 85.4% of fluorescent signals were coincident with PAPs and their intensity had a negative correlation with the body mass index (r = -0.56, p = 0.045). Cross-sections showed a Y-shaped fluorescence pattern where the two migration lines on the surface were the two vertices of the “Y.” Conclusion : The trajectories of fluorescein in the body are suggestive of the anatomical structure of meridians. The PC is related to the deep horizontal interstitial channels that connect to the body surface through vertical interstitial spaces. These biophysical properties and techniques for meridian visualization are valuable for revealing the anatomical structure of meridians.

Rapid global industrialization has resulted in irreversible environmental degradation, with one pressing issue being the escalating problem of land contamination induced by heavy metals (HMs). This phenomenon not only inflicts lasting harm on plants and microorganisms within the affected regions but also imposes significant losses on various industrial chains, particularly in the realm of agriculture. A noteworthy illustration is the soil contamination in the Taojia River Basin, Hunan Province, China, attributed to mining activities. Therefore, this study is dedicated to examining the passivation of active-state lead (Pb) and cadmium (Cd) in the cultivated fields of the Taojia River Basin. To achieve this, two passivating agents, namely bentonite (referred to as passivator-P) and an acidifying soil agent known as “Shi Di Jia” (referred to as passivator-S), were employed, along with a control group (CK). The passivation of Pb primarily relies on cation exchange adsorbed on the passivator's surface, while the passivation of Cd is intricately linked to total nitrogen content (TN). Indoor incubation tests were conducted to assess Pb and Cd in the soil. Additionally, an analysis of various soil health indicators and their relationships with microbial dynamics and soil enzymes was undertaken. The findings revealed that passivator-P exhibited greater efficacy in the passivation of Pb at concentrations of 3% and 5%, while higher concentrations of passivator-S demonstrated increased effectiveness in passivating Cd. Concerning bacterial diversity, the soil pH post-passivation showed a positive correlation with the Observed_species index, whereas Pb and Cd exhibited negative associations with the Chao1 index. Regarding fungal diversity, soil TN post-passivation displayed a positive correlation with the Simpson index. It is concluded that adding passivator-P at a 3% concentration proved to be the optimal passivating agent for Pb-contaminated soils in the Taojia River Basin. Although passivator-S demonstrated lesser efficacy in passivating Pb, it proved to be suitable for Cd passivation. These findings offer a crucial reference for passivation passivation remediation in the Taojia River area and other sites with similar characteristics, shedding light on the impacts on soil organisms. The implications underscore the necessity for site-specific remediation strategies and a holistic approach to addressing HMs contamination.

Nitrogen (N) as an essential macronutrient affects the soil nutrient cycle, microbial community abundance, and metabolic function. However, the specific responses of microorganisms and metabolic functions in rhizosphere soil of Phellodendron chinense Schneid seedlings to N addition remain unclear. In this study, four treatments (CK, N5, N10 and N15) were conducted, and the soil physicochemical properties, enzyme activities, microbial community abundances and diversities, metabolism, and gene expressions were investigated in rhizosphere soil of P. chinense Schneid. The results showed that N addition significantly decreased rhizosphere soil pH, among which the effect of N10 treatment was better. N10 treatment significantly increased the contents of available phosphorus (AP), available potassium (AK), ammonium nitrogen (NH4+-N), nitrate nitrogen (NO3−-N) and sucrase (SU) activity, as well as fungal diversity and the relative expression abundances of amoA and phoD genes in rhizosphere soil, but observably decreased the total phosphorus (TP) content, urease (UR) activity and bacterial diversity, among which the pH, soil organic matter (SOM), AP, NH4+-N and NO3−-N were the main environmental factors for affecting rhizosphere soil microbial community structure based on RDA and correlation analyses. Meanwhile, N10 treatment notably enhanced the absolute abundances of the uracil, guanine, indole, prostaglandin F2α and γ-glutamylalanine, while reduced the contents of D-phenylalanine and phenylacetylglycine in rhizosphere soil of P. chinense Schneid seedlings. Furthermore, the soil available nutrients represented a significant correlation with soil metabolites and dominant microorganisms, suggesting that N10 addition effectively regulated microbial community abundance and metabolic functions by enhancing nutrient cycle in the rhizosphere soil of P. chinense Schneid seedlings.

The effects of dietary grape seed proanthocyanidin extract (GSPE) supplementation (0, 100, 200, and 400 mg/kg) on the stress response, antioxidant status, myofiber growth, and meat quality of Nile tilapia were investigated in present study. Dietary GSPE lowered serum stress-related parameters (cortisol, glucose, Na+, and lactate) and improved the total antioxidant capability with enhanced superoxide dismutase and catalase activities and reduced levels of reactive oxygen species. Meanwhile, myoglobin content, pH, color, tenderness, the water-holding capacity were improved. Dietary GSPE also increased the contents of delicious amino acids, flavor amino acids, and total amino acids, C18:3n-3 (ALA), C20:5n-3 (EPA), n-3 poly-unsaturated fatty acids, decreasing off-flavor compounds such as aldehydes and alcohols. Moreover, GSPE supplementation improved myofiber growth and muscle protein deposition likely via the activation of the IGFs/PI3K/Akt/TOR/S6K1/4E-BP1 pathway, contributing to the enhanced growth performance of Nile tilapia (Oreochromis niloticus). Together, GSPE is a promising natural food additive that enhances various important meat quality parameters of fish, and the optimal dietary GSPE requirement for maximum growth and muscle protein deposition was approximately 280 mg/kg.

Abstract FK506 binding proteins (FKBPs) are a highly-conserved group of proteins known to bind to FK506, an immunosuppressive drug. They play different physiological roles, including transcription regulation, protein folding, signal transduction and immunosuppression. A number of FKBP genes have been identified in eukaryotes; however, very little information about these genes has been reported in Locusta migratoria. Here, we identified and characterized 10 FKBP genes from L. migratoria. Phylogenetic analysis and comparison of domain architectures indicated that the LmFKBP family can be divided into two subfamilies and five subclasses. Developmental and tissue expression pattern analysis revealed that all LmFKBPs transcripts, including LmFKBP46, LmFKBP12, LmFKBP47, LmFKBP79, LmFKBP16, LmFKBP24, LmFKBP44b, LmFKBP53, were periodically expressed during different developmental stages and mainly expressed in the fat body, hemolymph, testis, and ovary. In brief, our work depicts a outline but panoramic picture of LmFKBP family in L. migratoria, and provides a solid foundation to further investigate the molecular functions of LmFKBPs.

The normalized difference vegetation index (NDVI) can depict the status of vegetation growth and coverage in grasslands, whereas coarse spatial resolution, cloud cover, and vegetation phenology limit its applicability in fine-scale research, especially in areas covering various vegetation or in fragmented landscapes. In this study, a methodology was developed for obtaining the 30 m annual maximum NDVI to overcome these shortcomings. First, the Landsat NDVI was simulated by fusing Landsat and MODIS NDVI by using the enhanced spatial and temporal adaptive reflectance fusion model (ESTARFM), and then a single-peaked symmetric logistic model was employed to fit the Landsat NDVI data and derive the maximum NDVI in a year. The annual maximum NDVI was then used as a season-independent substitute to monitor grassland variation from 2001 to 2022 in a typical area covering the major vegetation types in the Qinghai Lake Basin. The major conclusions are as follows: (1) Our method for reconstructing the NDVI time series yielded higher accuracy than the existing dataset. The root mean square error (RMSE) for 91.8% of the pixels was less than 0.1. (2) The annual maximum NDVI from 2001 to 2022 exhibited spatial distribution characteristics, with higher values in the northern and southern regions and lower values in the central area. In addition, the earlier vegetation growth maximum dates were related to the vegetation type and accompanied by higher NDVI maxima in the study area. (3) The overall interannual variation showed a slight increasing trend from 2001 to 2022, and the degraded area was characterized as patches and was dominated by Alpine kobresia spp., Forb Meadow, whose change resulted from a combination of permafrost degradation, overgrazing, and rodent infestation and should be given more attention in the Qinghai Lake Basin.

Microcystin-LR (MC-LR) is a cyanobacterial metabolite produced during cyanobacterial blooms and is toxic to aquatic animals, and the liver is the main targeted organ of MC-LR. To comprehensively understand the toxicity mechanism of chronic exposure to environmental levels of MC-LR on the liver of fish, juvenile Nile tilapia were exposed to 0 μg/L (control), 1 μg/L (M1), 3 μg/L (M3), 10 μg/L (M10), and 30 μg/L (M30) MC-LR for 60 days. Then, the liver hepatotoxicity induced by MC-LR exposure was systematically evaluated via histological and biochemical determinations, and the underlying mechanisms were explored through combining analysis of biochemical parameters, multi-omics (transcriptome and metabolome), and gene expression. The results exhibited that chronic MC-LR exposure caused slight liver minor structural damage and lipid accumulation in the M10 group, while resulting in serious histological damage and lipid accumulation in the M30 group, indicating obvious hepatotoxicity, which was confirmed by increased toxicity indexes (i.e., AST, ALT, and AKP). Transcriptomic and metabolomic analysis revealed that chronic MC-LR exposure induced extensive changes in gene expression and metabolites in six typical pathways, including oxidative stress, apoptosis, autophagy, amino acid metabolism, primary bile acid biosynthesis, and lipid metabolism. Taken together, chronic MC-LR exposure induced oxidative stress, apoptosis, and autophagy, inhibited primary bile acid biosynthesis, and caused fatty deposition in the liver of Nile tilapia.

Grasshoppers are important elements for supporting ecosystem services which make it important to understand their spatial distribution characteristics to reduce outbreaks, implement effective control while minimizing non-target impacts. The spatial software package Geographical Detector (GD) was used to rank the interaction and effects of 19 environmental factors on the distribution of grasshopper species were quantified in four types of steppes in Northwestern, China. Results showed that the distribution of the insect was mainly influenced by elevation (q = 0.44). Among the factor interactions, elevation ∩ average annual precipitation (q = 0.795) was the strongest, followed by elevation ∩ soil available nitrogen (q = 0.722). Temperate meadow steppe and temperate steppe were identified as the areas at greatest risk for grasshopper’s occurrence. This research shows that the Geographical Detector could be provided a useful tool to identify the driving forces for grasshopper distribution. Our research also offers a new method to more broadly understand the interactions between different drivers to explain the distribution of other insects.

Abstract Skin blood flux (SkBF) changes caused by drinking cold water are generally associated with vagal tone and osmotic factors in the digestive system. However, there is still a lack of relevant research on whether there are left and right differences in these SkBF change. In the current study, a total of 60 subjects were recruited. Skin blood perfusion of the bilateral lower extremities was recorded simultaneously before and after drinking saline of different temperatures saline by using Laser Doppler flowmetry (LDF). The electrogastrogram (EGG) was also monitored, and the dominant frequency of the EGG and heart rate variability were analyzed. The results indicated that after drinking saline, the laterality index of SkBF at the lower extremities was different and the laterality index changes of SkBF were mainly reflected in the frequency interval V (0.4–1.6 Hz). There was a weak negative correlation between the laterality index of endothelial NO-dependent component and change rate of root mean square of successive differences (RMSSD) after drinking 4 °C saline. However, after drinking 30 °C saline, there was a weak positive correlation between neurogenic component and RMSSD The distribution and regulation of bilateral blood flow are not symmetrical but exhibit a certain laterality.

Solid-phase denitrification shows promise for removing nitrate (NO3−-N) from water. Biological denitrification uses external carbon sources to remove nitrogen from wastewater, among which agriculture waste is considered the most promising source due to its economic and efficiency advantages. Hydraulic retention time (HRT) and influent nitrate concentration (INC) are the main factors influencing biological denitrification. This study explored the effects of HRT and INC on solid-phase denitrification using wheat husk (WH) as a carbon source. A solid-phase denitrification system with WH carbon source was constructed to explore denitrification performance with differing HRT and INC. The optimal HRT and INC of the wheat husk-denitrification reactor (WH-DR) were 32 h and 50 mg/L, respectively. Under these conditions, NO3−-N and total nitrogen removal rates were 97.37 ± 2.68% and 94.08 ± 4.01%, respectively. High-throughput sequencing revealed that the dominant phyla in the WH-DR operation were Proteobacteria, Bacteroidetes, and Campilobacterota. Among the dominant genera, Diaphorobacter (0.85%), Ideonella (0.38%), Thiobacillus (4.22%), and Sulfurifustis (0.60%) have denitrification functions; Spirochaeta (0.47%) is mainly involved in the degradation of WH; and Acidovorax (0.37%) and Azospira (0.86%) can both denitrify and degrade WH. This study determined the optimal HRT and INC for WH-DR and provides a reference for the development and application of WH as a novel, slow-release carbon source in treating aquaculture wastewater.

Faba bean water extract (FBW) and vitamin K3 (VK3) have been demonstrated to improve the muscle textural quality of fish. To better apply these two feed additives in commercial aquaculture setting, four experimental diets (control, commercial feed group; 15% FBW, 15% faba bean water extract group; 2.5% VK3, 2.5% vitamin K3 group; combined group, 15% faba bean water extract + 2.5% vitamin K3 group) were formulated to explore their combined effects of FBW and VK3 on the growth, health status, and muscle textural quality of grass carp. The growth performance, textural quality, intestinal characteristics, and oxidative and immune responses were analyzed on days 40, 80 and 120. The results showed that supplementation with higher doses of FBW and VK3 have no influence on growth-related parameters and immune parameters of grass carp. Notably, compared with the control, fish in the combined group had the highest textural qualities (hardness, chewiness and adhesiveness), followed by those in 15% FBW and 2.5% VK3 groups (P < 0.05). Also, FBW and VK3, to some extent, may lower antioxidative ability of grass carp, as illustrated by lower levels of GSH and CAT in 15% FBW, 2.5% VK3, and combined groups on day 120 (P < 0.05). In addition, enhanced lipase activity was observed in the 15% FBW group. Taken together, the combined supplementation of FBW and VK3 was demonstrated to be a more advanced option than their individual supplementation in a commercial setting owing to the resulting combined effects on both the textural quality and health status of grass carp.

The complete replacement of fresh baits (FB) by formulated artificial feeds (AF) after domestication in mandarin fish (Siniperca chuatsi) has matured in recent years, while the underlying molecular events of long-term AF diet feeding after diet shifts remained unclear. Therefore, mandarin fish, which had been successfully domesticated, were fed with the FB or AF for 180 days to explore the molecular mechanisms underlying the differences due to shifted food habit. Compared to the FB group, slight intestinal histological damage was found in the AF group of mandarin fish. An integrated analysis of miRNA-mRNA sequencing and their interaction networks was conducted between the FB and AF groups. The result showed that a total of 209 differentially expressed mRNAs (DEmRNAs) and 2 differentially expressed miRNAs (DEmiRNAs) with significance were identified. The DEmRNAs were enriched to 39 pathways including tight junction, steroid biosynthesis and protein processing in ER. Oni-Let-7a and aca-Let-7i were the two significantly identified DEmiRNAs, whose target prediction was enriched with GO terms and KEGG pathways including TGFβ signaling pathway, steroid biosynthesis, and retinol metabolism. In general, our research was the first exploration to characterize the interacted miRNA-mRNA sequencing of mandarin fish fed the FB and AF diets, which could reveal the mechanisms of long-term adaptation to new foods in organisms who subjected to a food conversion in their early life. Microbial communities in aquaculture ponds play a critical role in pond productivity and the success of fish production. Nevertheless, our current understanding of the functional diversity of bacterial microbiomes in grass carp pond water and sediments is fragmentary. This study utilized high-throughput sequencing technology and community level physiological profiling (CLPP) methods to investigate microbial communities and metabolic activity of bacterial found in water samples and sediments from eight typical grass carp (Ctenopharyngodon idella) culturing pond in southern China. The correlations between putative function using PICRUSt2 and actual metabolic profiles via Biolog EcoPlates was assessed. Carbohydrates (32.51 %), amino acids (23.13 %), and polymers (16.61 %) were the most dominant carbon sources utilized by the microbial community in the sediment; while carbohydrates (34.72 %), amino acids (25.49 %), and polymers (16.15 %) were also utilized most often in the water samples. In the sediment samples, Proteobacteria (24.67 %), Chloroflexi (15.30 %), and Firmicutes (13.74 %) were the most dominant bacteria phyla. Actinobacteriota (32.62 %), Cyanobacteria (25.06 %), and Proteobacteria (17.51 %) were the most dominant bacteria phyla in the water samples. Moreover, our results suggested that the nitrogen transformations bacterial genera (Bacillus, Bradyrhizobium, Brocadia, Halomonas, and Vogesella) were positively correlated with carbon source utilization of the microbial community in the Biolog EcoPlates in the sediment. Dechloromonas and Pseudomonas were significantly negatively correlated in the water samples. In addition, there was a significant correlation between predicted function and the actual metabolic characteristics of microbial communities in sediment and water samples. The results facilitate detailed perspective of the relationship between the microbial community structure and the function in grass carp ponds and also provide a reference for the selection of suitable carbon sources in aquaculture wastewater treatment technology.

Myocardial infarction (MI) triggers adverse remodeling mechanisms, thus leading to heart failure. Since the application of biomaterial-based scaffolds emerged as a viable approach for providing mechanical support and promoting cell growth, injectable hydrogels have garnered substantial attention in MI treatment because of their minimally invasive administration through injection and diminished risk of infection. To fully understand the interplay between injectable hydrogels and infarcted myocardium repair, this review provides an overview of recent advances in injectable hydrogel-mediated MI therapy, including: I) material designs for repairing the infarcted myocardium, considering the pathophysiological mechanism of MI and design principles for biomaterials in MI treatment; II) the development of injectable functional hydrogels for MI treatment, including conductive, self-healing, drug-loaded, and stimulus-responsive hydrogels; and III) research progress in using injectable hydrogels to restore cardiac function in infarcted myocardium by promoting neovascularization, enhancing cardiomyocyte proliferation, decreasing myocardial fibrosis, and inhibiting excessive inflammation. Overall, this review presents the current state of injectable hydrogel research in MI treatment, offering valuable information to facilitate interdisciplinary knowledge transfer and enable the development of prognostic markers for suitable injectable materials.

​The zero-water exchange aquaculture has been identified as a promising method of farming to decrease the environment pressure of aquaculture and to increase profits. However, the ecological functions (e.g., nitrogen removal) and microbial biodiversity of the zero-water exchange pond aquaculture system are relatively understudied. In the present study, the zero-water exchange pond aquaculture system were constructed to investigated the microbial communities, sediment potential nitrification and denitrification production rates. And five functional genes (AOA amoA, AOB amoA, nirS, nosZ, and hzsB) were used to quantify the abundance of nitrifying and denitrifying microorganisms. The results showed that the sediment of the system had excellent potential nitrification-denitrification performance, with potential nitrification and denitrification rate were 149.77-1024.44 ng N g−1 h−1 and 48.32-145.01 ng N g−1 h−1, respectively. The absolute copy numbers of nitrogen functional genes and total bacterial 16S rRNA were 1.59×105-1.39×109 and 1.55×1010-2.55×1010copies g−1, respectively, with the dominant phyla, i.e., Proteobacteria, Actinobacteriota, Chloroflexi, Cyanobacteria, and Firmicutes. The relative abundances of the genera related to nitrification and denitrification, varied from 0.01% to 0.79% and from 0.01% to 15.54%, respectively. The potential nitrification rate was positively related to the sediment TOC concentration; and the potential denitrification rate had a positive correlation with sediment nitrate concentration. The genera Bacillus positively correlated with sediment NO3‐-N concentration, whereas Flavobacterium and Shewanella positively correlated with sediment NH4+-N concentration, which could be the functional bacteria for nitrogen removal. These findings may shed light on quantitative molecular mechanisms for nitrogen removal in zero-water exchange ponds, providing a sustainable solution to nitrogen pollution problem in the freshwater aquaculture ecosystems.

To investigate the impact of betaine on high-carbohydrate-diet-induced oxidative stress and endoplasmic reticulum (ER) stress, mandarin fish (Siniperca chuatsi) (23.73 ± 0.05 g) were fed with control (NC), betaine (BET), high carbohydrate (HC), and high carbohydrate + betaine (HC + BET) diets for 8 weeks. The results showed that betaine significantly promoted the growth of mandarin fish irrespective of the dietary carbohydrate levels. The HC diet induced oxidative stress, as evidenced by significantly elevated MDA levels. The HC diet significantly stimulated the mRNA levels of genes involved in ER stress (ire1, perk, atf6, xbp1, eif2α, atf4, chop), autophagy (ulk1, becn1, lc3b), and apoptosis (bax). However, betaine mitigated HC-diet-induced oxidative stress by modulating antioxidant enzymes and alleviated ER stress by regulating the mRNA of genes in the PERK-eIF2a-ATF4 pathway. Additionally, betaine significantly reduced the mRNA levels of becn1 and bax, along with the apoptosis rate, indicating a mitigating effect on autophagy and apoptosis. Overall, dietary betaine improved growth, attenuated HC-diet-induced oxidative stress and ER stress, and ultimately alleviated apoptosis in mandarin fish. These findings provide evidence for the use of betaine in aquafeeds to counter disruptive effects due to diets containing high carbohydrate levels.

The simulation and prediction of coastline changes are of great significance for the development and scientific management of coastal zones. Coastline changes are difficult to capture completely but appear significantly periodic over a long time series. In this paper, the transformer model is used to learn the changing trend of the coastline so as to deduce the position of the coastline in the coming year. First, we use the distance regularization level set evolution (DRLSE) model for instantaneous waterline extraction (IWE) from preprocessed Landsat time-series images from 2010–2020 in Weitou Bay, China. Then, tidal correction (TC) is performed on the extracted instantaneous waterline dataset to obtain coastlines projected to a single reference tidal datum. Finally, the coastline datasets from 2010–2019 are used for model training, and the coastline in 2020 is used for accuracy assessment. Three precision evaluation methods, including receiver operating characteristic curve matching, the mean offset, and the root mean square error, were used to verify the predicted coastline data. The receiver operating characteristic curve was specifically designed and improved to evaluate the accuracy of the obtained coastline. Compared with the support vector regression (SVR) and long–short-term memory (LSTM) methods, the results showed that the coastline predicted by the transformer model was the closest to the accurate extracted coastline. The accuracies of the correct values corresponding to SVR, LSTM, and transformer models were 88.27%, 94.08%, and 98.80%, respectively, which indicated the accuracy of the coastline extraction results. Additionally, the mean offset and root mean square error were 0.32 pixels and 0.57 pixels, respectively. In addition, the experimental results showed that tidal correction is important for coastline prediction. Moreover, through field investigations of coastlines, the predicted results obtained for natural coastlines were more accurate, while the predicted results were relatively poor for some artificial coastlines that were intensely influenced by human activities. This study shows that the transformer model can provide natural coastline changes for coastal management.

The ecological process of natural regeneration in forests is achieved by altering the relationship between community diversity and abiotic factors, thereby influencing the structure and functioning of ecosystems. Phoebe bournei is a unique and endangered tree species in China, and due to the rarity of P. bournei (Phoebe bournei) populations, there is limited research on the interaction between plant community diversity and soil environment during its natural regeneration process. Jiangxi Mingyue Mountain Forest Farm is one of the few locations where the natural regeneration of P. bournei communities occurs. From 2018 to 2021, the DBH (diameter at breast height) method was employed to define the stages of P. bournei regeneration. Community tree diversity, productivity, soil nutrients, and microbial diversity were investigated. The results showed the following: (1) P. bournei exhibits a competitive advantage compared to other tree species during natural regeneration, and it becomes the main contributor to community biomass and productivity in the later stages of regeneration. (2) The regeneration process of P. bournei has significant effects on community tree diversity and soil environment. Community diversity and biomass show a trend of initial increase followed by a decrease. Soil moisture content, available phosphorus, and bacterial diversity significantly increase. P. bournei’s competitive advantage is likely derived from its regulation of soil nutrients and microorganisms. (3) Correlation analysis reveals a significant positive correlation between soil microbial diversity and community productivity. Therefore, it is necessary to consider the composition and diversity of underground soil microorganisms in studies aimed at improving the quality of P. bournei forests. In summary, the natural regeneration of P. bournei involves the gradual removal of the original dominant evergreen species in the community by increasing its own competitive advantage and productivity, while continuously regulating community diversity, soil nutrients, and microbial diversity to facilitate the growth and development of P. bournei, ultimately establishing it as the dominant species in the forest community.

IntroductionHuman motivation for happiness involves two main orientations of hedonia and eudaimonia. Numerous studies have shown that hedonic motivation has a much smaller effect on happiness outcomes than eudaimonic motivation, but little is known about the reasons for this. According to the Self-Determination Theory and the Levels of Valence Model, this may be related to the different goal conflicts and mixed emotions elicited by the two motivations. To demonstrate this, the study examined the mediating effect of the above two variables between happiness motivation and life satisfaction. Furthermore, it explained why hedonists are less happy than eudaimonists by comparing the two happiness motivations in terms of their respective path effects.MethodsThe study randomly selected 788 college students from 13 different provinces of China to examine the relationships between hedonic motivation, eudaimonic motivation, goal conflict, mixed emotions, and life satisfaction.ResultsThe result showed that (1) the direct effect of hedonic motivation on life satisfaction was marginally significant, and the effect size was much smaller than that of eudaimonic motivation. (2) The direct and indirect effects of hedonic motivation were the opposite, with a large suppressing effect. In contrast, all paths of eudaimonic motivation positively affected life satisfaction. (3) Hedonic motivation negatively influenced life satisfaction through mixed emotions and the chain mediating effect of goal conflict and mixed emotions, whereas eudaimonic motivation positively influenced life satisfaction through these two mediating paths. (4) The effects on all paths of hedonic motivation were significantly smaller than those of eudaimonic motivation, except for the path mediated by goal conflict.DiscussionThis study explains why hedonists are less happy than eudaimonists from the perspective of goal pursuit, emphasizes the critical role of differences in goal pursuit state and experience between happiness motivation and life satisfaction, and provides new ideas for the study of the influence mechanism of happiness motivation. At the same time, the deficiencies of hedonic motivation and the advantages of eudaimonic motivation presented by the study provide directions for cultivating happiness motivation for adolescents in the practice field.

Aquaculture wastewater (AW) poses a threat to natural aquatic environments. Microecological agents are widely used to regulate and purify AW, with Bacillus being the most common. To evaluate the AW purification effect of adding Bacillus subtilis and Bacillus licheniformis to an AW treatment device, we constructed an experimental device including a small grass carp culture pond and three groups of cuboid reactors. The effects of adding the two strains to the AW treatment reactor on the AW purification effect and the microbiota compositions in the AW and packing surface biofilm were analyzed via high-throughput sequencing of the 16S rRNA gene. Our results showed that adding Bacillus bacteria to reactors improved the total nitrogen (TN) removal efficiency and reduced the chemical oxygen demand (COD). Adding both the B. subtillis and B. licheniformis preparations significantly increased the abundance of Firmicutes in the water microbiota of the reactor at the middle and end stages of the experiment. The addition of Bacillus changed the microbiota composition in the water and packing surface biofilm and significantly increased the abundance of Bacillus at the middle and later stages of the experiment. Therefore, the addition of Bacillus improved the TN removal efficiency in the AW grass carp treatment reactors and significantly reduced the COD in the AW by increasing the abundance of Bacillus and changing the microbiota composition in the system. We provide an effective way for improving the purification capacity of biofilm reactor.

Alpine wetlands are one of the most important ecosystems in the Three Rivers Source Area, China, which plays an important role in regulating the regional hydrological cycle and carbon cycle. Accordingly, Wetland area and its distribution are of great significance for wetland management and scientific research. In our study, a new wetland classification model which based on geomorphological types and combine object-oriented and decision tree classification model (ODTC), and used a new wetland classification system to accurately extract the wetland distributed in the Headwater Area of the Yellow River (HAYR) of the Qinghai-Tibet Plateau (QTP), China. The object-oriented method was first used to segment the image into several areas according to similarity in Pixels and Textures, and then the wetland was extracted through a decision tree constructed based on geomorphological types. The wetland extracted by the model was compared with that by other seven commonly methods, such as support vector machine (SVM) and random forest (RF), and it proved the accuracy was improved by 10%–20%. The overall classification accuracy rate was 98.9%. According to our results, the HAYR’s wetland area is 3142.3 km2, accounting for 16.1% of the study area. Marsh wetlands and flood wetlands accounted for 37.7% and 16.7% respectively. A three-dimensional map of the area showed that alpine wetlands in the research region are distributed around lakes, piedmont groundwater overflow belts, and inter-mountain catchment basin. This phenomenon demonstrates that hydrogeological circumstances influence alpine wetlands’ genesis and evolution. This work provides a new approach to investigating alpine wetlands.

Intensive aquaculture usually produces large volumes of nutrient-rich wastewater, which is essential to treat to avoid eutrophication. This study aimed to evaluate the performance of five continuously aerated biofilm reactors treating simulated high-strength aquaculture wastewater under different dissolved oxygen (DO) levels, and the effects of DO increments on simultaneous nitrification-denitrification (SND). Continuous aeration was beneficial to complete nitrification. Total inorganic nitrogen (TIN), principally ammonium, was mainly removed by SND. The SND rate response to different DO levels was fitted well by the power function of y = 54.81 + 371.58/(1 − 0.16*x)^(−1/0.24) (R2 = 0.897, P = 0.000). When the TIN was removed completely, the optimal SND rate was defined and corresponded to a value of 121.8%. Accordingly, the optimal DO concentration was calculated as 2.10 mg/L, close to the actual level of 1.83 mg/L, at which the highest proportional removals of total nitrogen (58.0%) and TIN (57.3%) were obtained. Phosphorus was also removed by denitrifying polyphosphate-accumulating organisms. HIGHLIGHTS Dissolved ammonium was mainly removed by simultaneous nitrification-denitrification (SND) process.; The optimal SND rate for the current wastewater was 121.8%.; The optimal DO corresponding to the best SND rate was 2.10 mg/L.; Phosphorus was mainly removed by denitrifying phosphate-accumulating organisms (PAOs).;

Abstract Precopulatory courtship plays an essential role in the insemination process and influences postcopulatory behavior between males and females. Male precopulatory oral stimulation of female genitals is rare for invertebrates. Here, we describe an intriguing oral sexual courtship in a cryptic desert beetle Platyope mongolica Faldermann. The males repeatedly contact the female's genitals using their mouths to gain consent to mate. Furthermore, the rate at which males contact the female's genitals relates to the copulation success in a series of observations. However, interference in oral sexual contacts decreased the proportion of successful copulation. Further no‐choice tests found homosexual behavior between males with antenna removed. We report the precopulatory oral sexual behavior and its important role for copulation success in P. mongolica for the first time. These findings highlight the significance of oral sexual courtship in sexual selection.

Crisp grass carp (CGC), the most representative improved varieties of grass carp (GC), features higher muscle hardness after feeding faba bean (Vicia faba L.) for 90–120 days. DNA methylation, a most widely studied epigenetic modification, plays an essential role in muscle development. Previous studies have identified numerous differentially expressed genes (DEGs) between CGC and GC. However, it remains unknown if the expression levels of these DEGs are influenced by DNA methylation. In the present study, we performed a comprehensive analysis of DNA methylation profiles between CGC and GC, and identified important candidate genes related to muscle development coupled with the transcriptome sequencing data. A total of 9,318 differentially methylated genes (DMGs) corresponding to 155,760 differentially methylated regions (DMRs) were identified between the two groups under the CG context in promoter regions. Combined with the transcriptome sequencing data, 14 key genes related to muscle development were identified, eight of which (gsk3b, wnt8a, wnt11, axin2, stat1, stat2, jak2, hsp90) were involved in muscle fiber hyperplasia, six of which (tgf-β1, col1a1, col1a2, col1a3, col4a1, col18a1) were associated with collagen synthesis in crisp grass carp. The difference of methylation levels in the key genes might lead to the expression difference, further resulting in the increase of muscle hardness in crisp grass carp. Overall, this study can help further understand how faba bean modulates muscle development by the epigenetic modifications, providing novel insights into the texture quality improvement in other aquaculture fish species by nutritional programming.

Due to climate change, drought has caused serious impacts on the eco-environment, hydrology and agriculture, and drought events in the Qinghai-Tibet Plateau (QTP) have become more severer and frequent; therefore, understanding the characteristics and variations of drought is crucial to reduce its eco-environmental and socio-economic impacts. This study used the Standardized Precipitation Evapotranspiration Index (SPEI) to identify drought and assessed its interannual changes from 1980 to 2020. Then, the Hurst exponent and intensity analysis were used to identify future drought trend and the characteristics of drought intensity. Moreover, Empirical Orthogonal Function (EOF) analysis was performed to examine the main spatial gathering characteristics. The results indicate that: 1) the QTP was becoming wetter in general, and drying places were mainly distributed in the southeast and northeast of the QTP, as well as the Qaidam Basin accounting for 27% areas of QTP. 2) The trend of wet and dry in the future in most regions would be the same as the present, only 10% of the regions would have the reverse trend. 3) The rate of wet/drought changes was faster in the 1980s and 2000s. 4) The EOF mode1 revealed a gathering distribution structure with negative values in the southeast and east of the QTP and positive values in the center and west. The west was more sensitive to the variation of dry and wet, and most areas will continue to be wet in the future. EOF mode2 and mode3 mainly indicated opposite gathering patterns of southwest-northeast and south-north. The results provide favorable evidence for policymakers to better understand and prevent drought.

Photocatalysis, as a novel technique, has been widely used for antibiotic pollution remediation in wastewater. In the processes of degradation and removal of antibiotics, the impact of photocatalysts on microenvironment is very important but remains poorly understood. In the present study, the effect of typical photocatalyst g-C3N4 (Graphitic carbon nitride) on microbial community was investigated in two sediment types (riverbed sediment and pig-farm sediment) polluted by tetracycline (TC) in central southern China. The riverbed sediment and pig farm sediment samples were respectively exposed to g-C3N4 (25, 75, 125 mg⋅kg–1) and TC (60, 120, 180 mg⋅L–1) treatments alone or combination for 30 days, respectively. The bacterial community and antibiotic resistance genes (ARGs) of the treated sediments were analyzed by Illumina sequencing and metagenomic sequencing. Studies had shown that: TC, g-C3N4, and TC/g-C3N4 have significant effects on the changes of microbial communities and components in riverbed sediment, but they do not exist in pig farm sediment. The most alterations of microbial taxa were Acidobacteriota, Actinobacteriota, and Desulfobacterota in riverbed sediment, and Elusimicrobiota in the pig farm sediment under various treatments. Through network analysis, it was found that the distribution of microorganisms in the pig farm sediment is more complex and more stable. The addition of g-C3N4 reduced the absolute abundance of ARGs in the two examined sediments, but not significantly changed their relative abundance of ARGs. The g-C3N4 application was beneficial to the removal of TC residues and to the prevention of the generation and transmission of ARGs in sediments. Our results suggested that g-C3N4 was a suitable photocatalyst with excellent application prospect for the removal of TC residues and the control of ARGs in environment.

Although many studies have focused on the roles of soil microbes in phosphorus (P) cycling, little is known about the distribution of microbial P cycling genes across soil depths. In this study, metagenomic sequencing was adopted to examine the differences in the abundance of genes and microbial taxa associated with soil P cycling between organic and mineral soil in subtropical forests. The total relative abundance of inorganic P solubilizing genes was the highest, that of P starvation response regulating genes was second, and organic P mineralizing genes was the lowest. The soil organic carbon concentration, N:P ratio, and available P concentration were higher in the organic soil than the mineral soil, resulting in abundances of organic P mineralizing genes (appA and 3-phytase), and inorganic P cycling genes (ppa), whereas those of the inorganic P cycling genes (gcd and pqqC) and the P starvation response regulating gene (phoR) were higher in mineral soil. The four bacteria phyla that related to P cycling, Proteobacteria, Actinobacteria, Bacteroidetes, and Candidatus_Eremiobacteraeota were higher in organic soil; conversely, the three bacteria phyla (Acidobacteria, Verrucomicrobia, and Chloroflexi) and archaea taxa were more abundant in mineral soil. Therefore, we concluded that the distribution of genes and microbial taxa involved in soil P cycling differed among soil depths, providing a depth-resolved scale insight into the underlying mechanisms of P cycling by soil microorganisms in subtropical forests.

Polyculture is an effective way to achieve efficient utilization of nutrient resources in high-density intensive aquaculture systems. In order to study the optimal culture mode of Oreochromis niloticu, Aristichthys nobilis, and Cherax quadricarinatus, the budget of nitrogen and phosphorus in various polyculture systems (CH, CHC1, CHC2, CHC3) was studied with land-based enclosures. The results showed that all the three polyculture systems had higher total yields of cultured animals than the control group (two polyculture systems) (p < 0.05). The co-cultured organisms absorbed artificial feed or organic matter (such as plankton and sediment) from the polyculture system to different degrees. Feed was the main input of nitrogen (98.22–98.33%) and phosphorus (99.43–99.56%) in all systems. Considering the N and P outputs, 46.64–64.58% and 81.60–84.79%, respectively, accumulated in the sediment, and 34.43–52.55% and 14.89–17.30% of the N and P outputs, respectively, were harvested by aquaculture organisms. The pollution production coefficients of TN and TP in the O. niloticus polyculture ponds were 5.35–6.26 g/m2 and 1.17–1.61 g/m2, respectively. The TN production coefficients of O. niloticus and the ternary polyculture groups (CHC1, CHC2, and CHC3) were lower than that of the control group (CH). The TP production coefficients showed the opposite pattern. The N and P utilization efficiencies in the group with the optimal ratio of O. niloticus, A. nobilis, and C. quadricarinatus (4, 0.15, and 3 ind/m2, respectively) were 2.56–12.82% and 6.62–11.03% higher, respectively, compared with those of the other groups. The N utilization efficiency was effectively improved in this group with the optimum stocking density for the polyculture systems, resulting in improved ecological efficiency and economic benefits.

Excessive hepatic lipid accumulation is a common phenomenon in cultured fish; however, its underlying mechanisms are poorly understood. Lipid droplet (LD)-related proteins play vital roles in LD accumulation. Herein, using a zebrafish liver cell line (ZFL), we show that LD accumulation is accompanied by differential expression of seven LD-annotated genes, among which the expression of dehydrogenase/reductase (SDR family) member 3 a/b (dhrs3a/b) increased synchronously. RNAi-mediated knockdown of dhrs3a delayed LD accumulation and downregulated the mRNA expression of peroxisome proliferator-activated receptor gamma (pparg) in cells incubated with fatty acids. Notably, Dhrs3 catalyzed retinene to retinol, the content of which increased in LD-enriched cells. The addition of exogenous retinyl acetate maintained LD accumulation only in cells incubated in a lipid-rich medium. Correspondingly, exogenous retinyl acetate significantly increased pparg mRNA expression levels and altered the lipidome of the cells by increasing the phosphatidylcholine and triacylglycerol contents and decreasing the cardiolipin, phosphatidylinositol, and phosphatidylserine contents. Administration of LW6, an hypoxia-inducible factor 1α (HIF1α) inhibitor, reduced the size and number of LDs in ZFL cells and attenuated hif1αa, hif1αb, dhrs3a, and pparg mRNA expression levels. We propose that the Hif-1α/Dhrs3a pathway participates in LD accumulation in hepatocytes, which induces retinol formation and the Ppar-γ pathway.

Studying the variation in vegetation types within the source region of the Yellow River (SRYR) is of great significance for understanding the response of vegetation to climate change and human activities on the Qinghai-Tibet Plateau (QTP) permafrost. In order to understand the characteristics of the variation in vegetation associations in the SRYR under the influence of climate and human activities, two hyperspectral remote sensing images from HJ-1A in 2013 and OHS-3C in 2020 were used to extract the vegetation types located in the area south of Ngoring Lake, covering 437.11 km2 in Maduo County, from the perspective of vegetation associations. Here, the hybrid spectral CNN (HybridSN) model, which is dependent on both spatial and spectral information, was used for vegetation association classifications. On this basis, the variations in vegetation associations from 2013 to 2020 were studied using the transition matrix, and the variation in noxious weeds across different altitude and slope gradients was analyzed. As an example, Thermopsis lanceolata’s spatial distribution pattern and diffusion mechanism were analyzed. The results showed that (1) in addition to noxious weeds, herbage such as Poa poophagorum, Stipa purpurea, Kobresia humilis, and Carex moorcroftii increased, indicating that the overall ecological environment tended to improve, which may be attributed mainly to the development of a warm and humid climate. (2) Most of the noxious weeds were located at low altitudes with an area increase in the 4250–4400 m altitude range and a decrease in the 4400–4500 m altitude range. More attention should be given to the fact that the noxious weeds area increased from 2.88 km2 to 9.02 km2 between 2013 and 2020, which was much faster than that of herbage and may threaten local livestock development. (3) The Thermopsis lanceolate association characterized by an aggregated distribution tended to spread along roads, herdsmen sites, and degraded swamps, which were mainly affected by human activities and swamp degradation.

The pregnane X receptor (PXR) is a master xenobiotic-sensing receptor in response to toxic byproducts, as well as a key regulator in intermediary lipid metabolism. Therefore, the present study was conducted to investigate the potential role of PXR in mediating the lipid dysregulation and xenobiotic responses under Cu-induced stress in largemouth bass (Micropterus salmoides). Four groups of largemouth bass (52.66 ± 0.03 g) were treated with control, Cu waterborne (9.44 μmol/L), Cu+RIF (Rifampicin, 100 mg/kg, PXR activator), and Cu+KET (Ketoconazole, 20 mg/kg, PXR inhibitor) for 48 h. Results showed that Cu exposure significantly elevated the plasma stress indicators and triggered antioxidant systems to counteract Cu-induced oxidative stress. Acute Cu exposure caused liver steatosis, as indicated by the significantly higher levels of plasma triglycerides (TG), lipid droplets, and mRNA levels of lipogenesis genes in the liver. Liver injuries were detected, as shown by hepatocyte vacuolization and severe apoptotic signals after Cu exposure. Importantly, Cu exposure significantly stimulated mRNA levels of PXR, suggesting the response of this regulator in the xenobiotic response. The pharmacological intervention of PXR by the agonist and antagonist significantly altered hepatic mRNA levels of PXR, implying that RIF and KET were effective agents of PXR in largemouth bass. Administration of RIF significantly exacerbated liver steatosis, and such alterations were dependent on the regulations on pparγ and cd36 rather than srebp1 signaling, which suggested that PXR-PPARγ might be another pathway for Cu-induced lipid deposition in fish. Whereas, KET administration showed reverse effects on lipid metabolism as indicated by the lower hepatic TG levels, suppressed mRNA levels of pparγ and cd36. Activation of PXR stimulated autophagy and inhibited apoptosis, leading to lower hepatic vacuolization; while inhibition of PXR showed higher apoptotic signals, inhibition of autophagic genes and stimulation of apoptotic genes. Taken together, PXR played a cytoprotective role in Cu-induced hepatotoxicity through regulations on autophagy and apoptosis. Overall, our data has demonstrated for the first time on the dual roles of PXR as a co-regulator in mediating xenobiotic responses and lipid metabolism in fish, which implying the potential of PXR as a therapy target for xenobiotics-induced lipid dysregulation and hepatotoxicity.

Numerous studies shown that particulate matter in the ambient environment has a significant impact on the health of the respiratory system. To understand the interrelationships between urban built environment, transportation operations and health, this study proposes an innovative approach that uses real-world GPS datasets to calculate energy consumption and emissions from transportation. The experiment used the traffic operation state in the Fourth Ring Road of Beijing as the research object and tested the impact of using the Regional speed optimization (RSO) strategy based on Macroscopic Fundamental Diagram (MFD) on energy consumption and emissions during peak hours. The impact of traffic emission on the health of roadside pedestrians is also considered. Changes in PM2.5 concentrations around four different built-up areas were calculated and compared. The computational experiments indicate that the PM2.5 pollutants exhausted by the traffic on the Ring Road during peak hours can reach up to 250 μg/m3, while the traffic emission on general roads near residential areas is only 50 μg/m3. Adopting Regional speed optimization can reduce the energy consumption of the road network by up to 18.8%. For roadside runners, the PM2.5 inhalation caused by night running in commercial and recreational areas is about 1.3-2.6 times that of night running in residential areas. Compared with morning or night running, the risk of respiratory disease caused by PM2.5 inhalation was about 10.3% higher than commuter running behavior. The research results provide a useful reference for energy conservation and emission reduction control strategies for different road types in cities and help existing cities to establish a traveler health evaluation system caused by traffic operation.

Abstract Background This study was performed to understand the prevalence of and possible risk factors for cholecystolithiasis in Uyghur, Kazakh, Han, and other ethnic groups in the Xinjiang Uyghur autonomous region of China. Methods Subjects were enrolled using typical case sampling and multistage stratified random sampling. We collected epidemiological data regarding cholecystolithiasis using a standard questionnaire of risk factors for gallbladder disease in Xinjiang. The subjects completed the questionnaire and underwent an abdominal ultrasound examination of the liver and gallbladder. Results This study included 5454 Xinjiang residents aged ≥ 18 years. Soil microorganisms are greatly affected by their microenvironment. To reveal the influence of different land use patterns on the composition and diversity of soil bacterial and fungal communities, this study analyzed microbial (bacteria and fungi) community composition and diversity under different land use patterns (vegetable land, wasteland, woodland, cultivated land) based on 16S rRNA, 18S rRNA, and high-throughput sequencing method in the Taojia River Basin. Spearman analysis and redundancy analysis (RDA) were used to explore the correlation between soil physicochemical properties and soil fungal and bacterial community composition, and a partial least squares path model (PLS-PM) was constructed to express the causal relationship between soil physicochemical properties and soil bacterial and fungal community diversity. The results showed that the soil bacterial species richness was highest in vegetable land and the lowest in the wasteland. Proteobacteria is the dominant phylum (20.69%–32.70%), and Actinobacteria is the dominant class (7.99%–16.95%). The species richness of fungi in woodland was the highest, while was the lowest in cultivated land. The dominant phylum of fungi in vegetable land, woodland, and cultivated land is Mucoromycota, 29.39%, 41.36%, and 22.67%, respectively. Ascomycota (42.16%) is the dominant phylum in wasteland. Sordariomyetes of Ascomycota is the dominant class in wasteland and cultivated land. Mortierellomycetes and Glomeromycetes of Mucoromycota are the dominant class in vegetable land and woodland. The results of the Spearman analysis revealed that the dominant groups in the bacterial and fungal communities had significant correlations with soil pH, clay, and sand (p < 0.01). The RDA results showed that soil clay, pH, and moisture were the key environmental factors affecting the diversity of soil microbial communities. Fungal diversity is more affected by different land use patterns than bacteria. These results provided a theoretical basis for the changes in soil microbial community composition and diversity in river basins.

An ecological public welfare forest is an important basis for the construction of national ecological security. This study took public welfare forests at the provincial level or above in Hunan Province as the research object. Based on the in situ monitoring data and remote sensing data, we constructed a random forest (RF) model for inversing the biomass of public welfare forests with different types. Then, based on the inversion results, we investigated the biomass spatial pattern. Combined with topographical and socio-economic factors, we constructed a geographically weighted regression (GWR) model to analyze the biomass driving factors of different vegetation types in public forests. The results showed the following: (1) The biomass of public welfare forests in Hunan Province presented a strip distribution pattern that gradually increases from the central to the southwest and northeast. The total biomass of public welfare forests in Hunan Province was 338.13 million tons, with an average biomass of 68.31 t·hm−2. In the different types of public welfare forests, the mean biomass of the types were as follows: shrub (4.65 t·hm−2) < broadleaf forest (59.27 t·hm−2) < conifer–broadleaf mixed forest (62.44 t·hm−2) < bamboo forest (71.33 t·hm−2) < coniferous forest (100.33 t·hm−2). (2) Topographic and socio-economic factors have a significant impact on the spatial pattern of biomass in public welfare forests. Slope had the greatest effect on coniferous forest, conifer–broadleaf mixed forest, and shrub forest, while POP had the greatest effect on broadleaf forest and bamboo forest. This study investigates the spatial patterns and driving factors of biomass in public welfare forests at the provincial level, filling the gap in forest biomass monitoring in public welfare forests in Hunan Province. It provides a new method to improve the accuracy of forest biomass estimation and data support for the sustainable management of public welfare forests.

Nitrogen pollution in aquaculture wastewater can pose a significant health and environmental risk if not removed before wastewater is discharged. Biological denitrification uses external carbon sources to remove nitrogen from wastewater; however, these carbon sources are often expensive and require significant energy. In this study, we investigated how six types of agricultural waste can be used as solid carbon sources in biological denitrification. Banana stalk (BS), loofah sponge (LS), sorghum stalk (SS), sweet potato stalk (SPS), watermelon skins (WS) and wheat husk (WH) were studied to determine their capacity to release carbon and improve denitrification efficiency. The results of batch experiments showed that all six agricultural wastes had excellent carbon release capacities, with cumulative chemical oxygen demands of 37.74–535.68 mg/g. During the 168-h reaction, the carbon release process followed the second-order kinetic equation and Ritger-Peppas equation, while carbon release occurred via diffusion. The kinetic equation fitting, scanning electron microscopy, and Fourier transform infrared spectroscopy results showed that LS had the lowest cm and the maximum t1/2 values and only suffered a moderate degree of hydrolysis. It also had the lowest pollutant release rate and cumulative chemical oxygen demand, as well as the most efficient removal of total phosphorous (TP) and total nitrogen (TN). Therefore, we concluded that LS has the lowest potential risk of excess carbon release and capacity for long-lasting and stable carbon release. The WS leachate had the highest TN contents, while the SPS leachate had the highest TP content. In the 181-h denitrification reaction, all six agricultural wastes completely removed nitrate and nitrite; however, SS had the highest denitrification rate, followed by LS, WH, BS, SPS, and WS (2.16, 1.35, 1.35, 1.34, 1.34, and 1.01 mg/(L·h), respectively). The denitrification process followed a zero-order and first-order kinetic equation. These results provide theoretical guidance for effectively selecting agricultural waste as a solid carbon source and improving the denitrification efficiency of aquaculture wastewater treatment.

To address this problem, this work studied the effects of berberine (BBR), a Chinese herbal extract with the serum glucose-lowering activity in mammals, on regulating the serum glucose and intestinal microbiota in largemouth bass. Texture is one of the key quality attributes used in the fresh and processed fish industry to assess product quality and consumer acceptability. To improve the textural quality of tilapia, we formulated the expanded pellet diet (EPD) and pellet diet (PD), both containing faba bean (Vicia faba, FB) water extract, a previously reported potential aquafeed additive to increase flesh texture. The common diet was used as a control. After Nile tilapia (Oreochromis niloticus) were fed three kinds of experimental diet for 120 days, muscle textural quality, growth parameters, oxidative response and immune parameters were analyzed. The results showed that there was no significant difference in the growth parameters between the three groups (P > 0.05).
