A significant amount of natural gas resources has been discovered in the Ordovician reservoirs under a thick layer of gypsum-salt strata as a result of ongoing research into deep natural gas in China’s Ordos Basin, however, the genetic type and source of the Ordovician sub-salt natural gas remain contentious. These limits understanding how the geochemical properties of the natural gas in Ordos Basin have evolved and how to best guide future exploration and development. Based on the components, carbon and hydrogen isotopic ratios of natural gas from the latest exploration wells, combined with the geological background, this study systematically analyzed the genetic types and sources of sub-salt natural gas of the central and eastern Ordos Basin. The results show that the Ordovician natural gas is mainly composed of hydrocarbon gases with low content of heavy hydrocarbons and dryness (CCH4/CCH4+) above 0.95, belonging to typical dry gas. Non-hydrocarbon gases mainly include N2 and CO2, and some natural gas samples contained H2S. The Ordovician sub-salt natural gas is primarily a self-generating and self-preserving oil-cracking gas with relatively high maturity (Ro ranging from 1.5% to 2.1%) mixed with coal-derived gas generated from upper-Paleozoic coal with relatively low maturity (Ro ranging from 1.1% to 1.5%). Due to the dual-source hydrocarbon supply, the Ordovician sub-salt reservoirs provide favorable gas accumulation conditions and development prospects. This study offers a reasonable explanation for the anomalous geochemical characteristics of the Ordovician sub-salt reservoirs in the Ordos Basin, and also may serve as a guide for future exploration of natural gas in the carbonate gypsum-salt sedimentary system in other basins.

Abstract Small extracellular vesicles (sEVs)-derived circular RNAs (circRNAs) could regulate gene expression in recipient cells, and dysregulation of sEVs-derived circRNAs has been implicated in several diseases. However, the expression and function of sEVs-derived circRNAs in coronary heart atherosclerotic disease (CAD) remain unknown. In this study, we investigated global changes in the expression patterns of circRNAs in sEVs from CAD-related monocytes and identified circNPHP4 as a significantly upregulated circRNA. Knockdown of circNPHP4 inhibited heterogeneous adhesion between monocytes and coronary artery endothelial cells and reduced ICAM-1 and VCAM-1 expression. Investigations of the underlying mechanisms revealed that circNPHP4 contains a functional miR-1231-binding site. Mutation of the circNPHP4-binding sites in miR-1231 abolished the interaction, as indicated by a luciferase reporter assay. Furthermore, circNPHP4 affected the expression of miR-1231 and its target gene EGFR. Overexpression of miR-1231 blocked the inhibitory effect of circNPHP4 on heterogeneous adhesion. Moreover, downregulation of miR-1231 restored heterogeneous adhesion upon inhibition by circNPHP4 silencing. Additionally, circNPHP4 overexpression was correlated with aggressive clinicopathological characteristics in CAD patients. A multivariate logistic regression model and bootstrapping validation showed that circNPHP4 overexpression had a good risk prediction capability for CAD. The decision curve analysis revealed that using the CAD nomogram that included circNPHP4 overexpression to predict the risk of CAD was beneficial. Our results suggest that sEVs-derived circNPHP4 can serve as a potential target for CAD treatments or as a potential diagnostic marker for CAD patients.

BackgroundColorectal cancer (CRC) is regarded as one of the most common malignancies in the world. MiR-1-3p was reported to be a tumor suppressor in CRC. However, the mechanisms have not been fully elucidated.MethodsTo identify CRC-associated miRNA, microarray data set GSE30454 was downloaded from the Gene Expression Omnibus database (GEO), and miR-1-3p was screened out as a candidate. The expression of miR-1-3p was detected using quantitative real-time polymerase chain reaction (qRT-PCR) in CRC cell lines and tissues. CCK-8 assay and transwell invasion assay were performed to determine CRC cell line proliferation and invasion, respectively. The levels of YWHAZ and EMT-associated proteins were detected using western blotting.ResultsBioinformatic analysis showed that miR-1-3p was downregulated in CRC tissues, which is verified by our experimental validation. The overexpression of miR-1-3p significantly suppressed CRC cell proliferation and invasion. Further studies showed that YWHAZ was a direct target of miR-1-3p and mediated epithelial–mesenchymal transition (EMT) modulated by miR-1-3p.ConclusionOur results demonstrated that miR-1-3p suppresses colorectal cancer cell proliferation and metastasis through regulating YWHAZ-mediated EMT, which may support a novel therapeutic strategy for CRC patients.

Reuse of plastic IC packages disassembled from printed circuit boards (PCBs) has significant environmental benefits and economic value. The interface delamination caused by moisture diffusion is the main failure mode of IC packages during the disassembling process, which greatly reduces the reusability and reliability of disassembled IC packages. Exploring moisture diffusion mechanism is a prerequisite to optimize prebaking processes before disassembling that is an effective way to avoid the interface delamination. To this end, a computational model with variable boundary conditions is developed based on the different combination state of water in IC packages. The distribution characteristics and mechanism of moisture diffusion behavior are analyzed including the humidity distribution field and the relation among baking temperature, water loss rate, and baking time during baking process, and then the results are validated by FEA simulation based on the improved definition of relative moisture concentration. Baking under variable temperature is proposed and compared with the baking process and baking efficiency under constant temperature to find out the optimized baking parameters. Finally, a set of curves which indicate the relation between baking energy consumption and temperature are determined under actual industrial baking experiments, which could be used as references to develop industrial standards for PCB disassembling process.
