1. Ka/C dual frequency ranging system for ocean altimetry satellite and analysis of ionospheric error

Abstract

In view of the future development trend of ocean altimetry satellites, the design idea of Ka/C dual frequency combination system is proposed, that is, the mode of using an altimeter antenna to contain Ka and C frequencies (35.7 and 5.3 GHz, respectively) for combined ranging. First, the random error model of altimeter measuring the distance from the altimeter phase center to the sea surface is given, and the analysis shows that the ionospheric correction error is one of the important factors. Second, the calculation of typical ionospheric parameters shows that the influence of ionospheric higher order terms on altimeter ranging is below the mm level, and its influence can be ignored. The rigorous expression for the first-order and second-order term correction of ionosphere error are given, respectively, by using dual frequency and three frequencies. The computational experiment of Jason-2 and SARAL satellite show that the maximum magnitude of ionospheric error correction in Ku band can reach 11 cm, while the maximum magnitude of ionospheric error correction in Ka band can reach 2 cm. For high-precision applications, ionospheric corrections must be made for single Ka frequency. After Ka/C dual frequency combination is adopted, error correction can be directly conducted without global ionospheric map (GIM) model, and its accuracy will be further improved than GIM model. Under 1 Hz sampling conditions, the ionospheric correction accuracy of Ka/C combination can be better than 2.5 mm, basically eliminating the influence of the ionosphere, and the total ranging accuracy can reach 3.5 cm considering the propagation error.

Abstract

Rhizosphere microorganisms are essential parts in maintaining soil ecological functions. Reforestation using genetically modified trees might have great potential to enhance tree production in biotic and abiotic stress, however, their long-term impact on rhizosphere microorganisms is scant. In this study, we studied soil enzyme activities and composition of rhizosphere microorganisms in 2-year-old transgenic PaGLK overexpression (OE), repressed expression (RE) and wild-type (WT) poplar (P. alba × P.berlinensis). The root exudates of PaGLK transgenic poplar (P.alba × P. berlinensis) were analyzed by liquid chromatography-mass spectrometry (LC-MS). The results showed that there were significant difference for soil sucrase, urease, catalase, neutral protease and cellulase between the transgenic and WT lines at different growth periods. Alpha diversity analysis showed that bacterial community abundance and diversity for RE lines were significantly lower than WT (p < 0.05), while RE lines for fungi were significantly higher than WT lines. At the genus level, Burkholderia was the dominant group of rhizosphere bacterial community, and the relative abundance for RE was significantly higher than WT. Tomentella was the dominant group for fungi community. Serendipita for RE was significantly higher than WT and OE. Main metabolite contents of (S)-ACPA, geniposidic acid, agnuside, hydroquinone and pyranocoumarins were significantly different among transgenic lines. These results suggest that transgenic activities have effects on root exudates, rhizosphere soil enzyme activities and soil microbial community composition, but long term effects need to be further investigated.

Abstract

Atmospheric rivers (ARs) reaching high-latitudes in summer contribute to the majority of climatological poleward water vapor transport into the Arctic. This transport has exhibited long term changes over the past decades, which cannot be entirely explained by anthropogenic forcing according to ensemble model responses. Here, through observational analyses and model experiments in which winds are adjusted to match observations, we demonstrate that low-frequency, large-scale circulation changes in the Arctic play a decisive role in regulating AR activity and thus inducing the recent upsurge of this activity in the region. It is estimated that the trend in summertime AR activity may contribute to 36% of the increasing trend of atmospheric summer moisture over the entire Arctic since 1979 and account for over half of the humidity trends in certain areas experiencing significant recent warming, such as western Greenland, northern Europe, and eastern Siberia. This indicates that AR activity, mostly driven by strong synoptic weather systems often regarded as stochastic, may serve as a vital mechanism in regulating long term moisture variability in the Arctic.

Abstract

The thighbone is the largest bone supporting the lower body. If the thighbone fracture is not treated in time, it will lead to lifelong inability to walk. Correct diagnosis of thighbone disease is very important in orthopedic medicine. Deep learning is promoting the development of fracture detection technology. However, the existing computer aided diagnosis (CAD) methods baesd on deep learning rely on a large number of manually labeled data, and labeling these data costs a lot of time and energy. Therefore, we develop a object detection method with limited labeled image quantity and apply it to the thighbone fracture localization. In this work, we build a semi-supervised object detection(SSOD) framework based on single-stage detector, which including three modules: adaptive difficult sample oriented (ADSO) module, Fusion Box and deformable expand encoder (Dex encoder). ADSO module takes the classification score as the label reliability evaluation criterion by weighting, Fusion Box is designed to merge similar pseudo boxes into a reliable box for box regression and Dex encoder is proposed to enhance the adaptability of image augmentation. The experiment is conducted on the thighbone fracture dataset, which includes 3484 training thigh fracture images and 358 testing thigh fracture images. The experimental results show that the proposed method achieves the state-of-the-art AP in thighbone fracture detection at different labeled data rates, i.e. 1%, 5% and 10%. Abstract

PurposeFoveoschisis involves the pathologic splitting of retinal layers at the fovea, which may occur congenitally in X-linked retinoschisis (XLRS) or as an acquired complication of myopia. XLRS is attributed to functional loss of the retinal adhesion protein retinoschisin 1 (RS1), but the pathophysiology of myopic foveoschisis is unclear due to the lack of animal models. Here, we characterized a novel nonhuman primate model of myopic foveoschisis through clinical examination and multimodal imaging followed by morphologic, cellular, and transcriptional profiling of retinal tissues and genetic analysis.MethodsWe identified a rhesus macaque with behavioral and anatomic features of myopic foveoschisis, and monitored disease progression over 14 months by fundus photography, fluorescein angiography, and optical coherence tomography (OCT). After necropsy, we evaluated anatomic and cellular changes by immunohistochemistry and transcriptomic changes using single-nuclei RNA-sequencing (snRNA-seq). Finally, we performed Sanger and whole exome sequencing with focus on the RS1 gene.ResultsAffected eyes demonstrated posterior hyaloid traction and progressive splitting of the outer plexiform layer on OCT. Immunohistochemistry showed increased GFAP expression in Müller glia and loss of ramified Iba-1+ microglia, suggesting macro- and microglial activation with minimal photoreceptor alterations. SnRNA-seq revealed gene expression changes predominantly in cones and retinal ganglion cells involving chromatin modification, suggestive of cellular stress at the fovea. No defects in the RS1 gene or its expression were detected.ConclusionsThis nonhuman primate model of foveoschisis reveals insights into how acquired myopic traction leads to phenotypically similar morphologic and cellular changes as congenital XLRS without alterations in RS1.

Abstract

PurposeTo investigate baseline mesopic microperimetry (MP) and spectral domain optical coherence tomography (OCT) in the Rate of Progression in USH2A-related Retinal Degeneration (RUSH2A) study.DesignNatural history study METHODS: Setting: 16 clinical sites in Europe and North AmericaStudy Population: Participants with Usher syndrome type 2 (USH2) (N = 80) or autosomal recessive nonsyndromic RP (ARRP) (N = 47) associated with biallelic disease-causing sequence variants in USH2AObservation Procedures: General linear models were used to assess characteristics including disease duration, MP mean sensitivity and OCT intact ellipsoid zone (EZ) area. The associations between mean sensitivity and EZ area with other measures, including best corrected visual acuity (BCVA) and central subfield thickness (CST) within the central 1 mm, were assessed using Spearman correlation coefficients.Main outcome measuresMean sensitivity on MP; EZ area and CST on OCT.ResultsAll participants (N = 127) had OCT, while MP was obtained at selected sites (N = 93). Participants with Usher syndrome type 2 (USH2, N = 80) and nonsyndromic autosomal recessive Retinitis Pigmentosa (ARRP, N = 47) had the following similar measurements: EZ area (median (interquartile range [IQR]): 1.4 (0.4, 3.1) mm2 vs 2.3 (0.7, 5.7) mm2) and CST (median (IQR): 247 (223, 280) µm vs 261 (246, 288), and mean sensitivity (median (IQR): 3.5 (2.1, 8.4) dB vs 5.1 (2.9, 9.0) dB). Longer disease duration was associated with smaller EZ area (P < 0.001) and lower mean sensitivity (P = 0.01). Better BCVA, larger EZ area, and larger CST were correlated with greater mean sensitivity (r > 0.3 and P < 0.01). Better BCVA and larger CST were associated with larger EZ area (r > 0.6 and P < 0.001).ConclusionsLonger disease duration correlated with more severe retinal structure and function abnormalities, and there were associations between MP and OCT metrics. Monitoring changes in retinal structure-function relationships during disease progression will provide important insights into disease mechanism in USH2A-related retinal degeneration.

Abstract

Observation-based climate model evaluation and future projections help policymakers in developing action plans for efficient management of water resources and mitigation of the impacts of hazardous extremes. Apart from this socioeconomic importance, the scientific value cannot be overstated, especially in light of the upcoming Fifth U.S. National Climate Assessment (NCA) report. In this study, we evaluate the realism of hydroclimate variability in the historical simulations of a suite of coupled general circulation models (CGCMs) participating in the fifth and sixth phases of the Coupled Model Intercomparison Project (CMIP5 and CMIP6). Our results demonstrate systematic biases in the simulated seasonal precipitation-most prominently, wet bias over the mountainous western United States in winter, and dry bias over the U.S. central plains in summer. A distinctive feature of this work is our focus on the examination of the atmospheric water budget, in particular the relative importance of remote and local contributions-convergence of moisture fluxes and local land surface processes (evapotranspiration) respectively-in helping produce precipitation. This diagnosis reveals that the leading contribution of the remote influence in winter is overestimated by the CMIP6 multimodel mean (MMM), whereas the local influence, which is more influential in summer, is underestimated. Our results aid in understanding the drivers of seasonal precipitation over the United States, where precipitation will likely increase by the end of the century but with significant model disagreement for the summer and fall. In support of ongoing NCA efforts, our study aims to contribute a comprehensive, regional-level analysis of the moisture budget and emphasizes the importance of realistically simulating its major components in CGCMs.

Abstract

PurposeUveal coloboma is a congenital eye malformation caused by failure of the optic fissure to close in early human development. Despite significant progress in identifying genes whose regulation is important for executing this closure, mutations are detected in a minority of cases using known gene panels, implying additional genetic complexity. We have previously shown knockdown of znf503 (the ortholog of mouse Zfp503) in zebrafish causes coloboma. Here we characterize Zfp503 knockout (KO) mice and evaluate transcriptomic profiling of mutant versus wild-type (WT) retinal pigment epithelium (RPE)/choroid.MethodsZfp503 KO mice were generated by gene targeting using homologous recombination. Embryos were characterized grossly and histologically. Patterns and level of developmentally relevant proteins/genes were examined with immunostaining/in situ hybridization. The transcriptomic profile of E11.5 KO RPE/choroid was compared to that of WT.ResultsZfp503 is dynamically expressed in developing mouse eyes, and loss of its expression results in uveal coloboma. KO embryos exhibit altered mRNA levels and expression patterns of several key transcription factors involved in eye development, including Otx2, Mitf, Pax6, Pax2, Vax1, and Vax2, resulting in a failure to maintain the presumptive RPE, as evidenced by reduced melanin pigmentation and its differentiation into a neural retina-like lineage. Comparison of RNA sequencing data from WT and KO E11.5 embryos demonstrated reduced expression of melanin-related genes and significant overlap with genes known to be dynamically regulated at the optic fissure.ConclusionsThese results demonstrate a critical role of Zfp503 in maintaining RPE fate and optic fissure closure.

Abstract

We assessed genotype-phenotype correlations among the visual, auditory, and olfactory phenotypes of 127 participants with Usher syndrome (USH2) (n =80) or nonsyndromic autosomal recessive retinitis pigmentosa (ARRP) (n = 47) due to USH2A variants, using clinical data and molecular diagnostics from the Rate of Progression in USH2A Related Retinal Degeneration (RUSH2A) study. USH2A truncating alleles were associated with USH2 and had a dose-dependent effect on hearing loss severity with no effect on visual loss severity within the USH2 subgroup. A group of missense alleles in an interfibronectin domain appeared to be hypomorphic in ARRP. These alleles were associated with later age of onset, larger visual field area, better sensitivity thresholds, and better electroretinographic responses. No effect of genotype on the severity of olfactory deficits was observed. This study unveils a unique, tissue-specific USH2A allelic hierarchy with important prognostic implications for patient counseling and treatment trial endpoints. These findings may inform clinical care or research approaches in others with allelic disorders or pleiotropic phenotypes.

Abstract

The Energy Exascale Earth System Model (E3SM) project is an ongoing, state-of-the-science Earth system modeling, simulation, and prediction project developed by the US Department of Energy (DOE). With an emphasis on supporting the DOE's energy mission, understanding and quantifying how well the model simulates water cycle processes is of particular importance. Here, we evaluate E3SM version 1.0 (v1.0) for its ability to represent atmospheric rivers (ARs), which play significant roles in water vapor transport and precipitation. The characteristics and precipitation associated with global ARs in E3SM at standard resolution (1×1) are compared to the Modern-Era Retrospective analysis for Research and Applications, version 2 (MERRA2). Global patterns of AR frequencies in E3SM show high degrees of correlation (≥0.97) with MERRA2 and low mean absolute errors (MAEs;

Abstract

For disorders with X-linked inheritance, variants may be transmitted through multiple generations of carrier females before an affected male is ascertained. Pathogenic RS1 variants exclusively cause X-linked retinoschisis (XLRS). While RS1 is constrained to variation, recurrent variants are frequently observed in unrelated probands. Here, we investigate recurrent pathogenic variants to determine the relative burden of mutational hotspot and founder allele events to this phenomenon. A cohort RS1 variant analysis and standardized classification, including variant enrichment in the XLRS cohort and in RS1 functional domains, were performed on 332 unrelated XLRS probands. A total of 108 unique RS1 variants were identified. A subset of 19 recurrently observed RS1 variants were evaluated in 190 probands by a haplotype analysis, using microsatellite and single nucleotide polymorphisms. Fourteen variants had at least two probands with common variant-specific haplotypes over ~1.95 centimorgans (cM) flanking RS1. Overall, 99/190 of reportedly unrelated probands had 25 distinct shared haplotypes. Examination of this XLRS cohort for common RS1 haplotypes indicates that the founder effect plays a significant role in this disorder, including variants in mutational hotspots. This improves the accuracy of clinical variant classification and may be generalizable to other X-linked disorders.

48. Averages and nonvanishing of central values of triple product $L$-functions

Abstract

Let $f,g,h$ be three normalized cusp newforms of weight $2k$ on $\Gamma_0(N)$ which are eigenforms of Hecke operators. We use Ichino's period formula combined with a relative trace formula to show exact averages of $L(3k-1,f\times g\times h)$. We also present some applications of the average formulas to the nonvanishing problem, giving a lower bound on the number of nonvanishing central $L$-values when one of the forms is fixed.
