Search

Observing the world on a global scale can help us understand better the role of water and water resource management utilities in a climate change context that engage us all. The usage of machine learning algorithms on open data measurements and statistical indicators can help us understand the behavioral changes in seasons and better prepare. These are complemented by powerful text mining algorithms that mine worldwide news, social media, published research and patented innovation towards best practices from success stories. In this paper, we propose a data-driven global observatory that puts together the different perspectives of media, science, statistics and sensing over heterogeneous data sources and text mining algorithms. We also discuss the implementation of this global observatory in the context of epidemic intelligence, monitoring the impact of climate change, and the value of this global solution in local contexts and priorities.

V diplomski nalogi sem se ukvarjala z ustvarjanjem serije risb in kolažev in ilustracije po besedilni predlogi. Za izhodišče pri iskanju besedilne predloge sem izbrala poezijo in kratko prozo mlajše generacije slovenskih avtorjev. Raziskala sem besedila in hkrati svoj likovni jezik, ki sem ga razvijala glede na izbrane besedilne predloge. V teoretskem delu diplomske naloge pa sem se ukvarjala z zgodovinskim pregledom ilustracije v povezavi s poezijo in ilustriranih izdaj poezije za odrasle ter z definicijo, zgodovino in vizualnimi karakteristikami vizualne in konkretne poezije. Raziskovala sem tudi format in medij umetniškega zina, njegovo definicijo, pozicijo v likovni umetnosti ter okolju, v katerem nastaja. V praktičnemu delu naloge sem formirala vizualne podobe in vizualno predstavila izbrane pesmi, zbrane v obliki zina. Ukvarjala sem se s povezavo ilustracije in poezije ter njuno likovno močjo in ugotovila, da lahko stojita sami zase, vendar se združeni še bolj dopolnjujeta. In my bachelor’s thesis, I was involved in creating a series of drawings, collages, and illustrations based on a text template. I chose poetry and short prose from the younger generation of Slovenian authors as a starting point in my search for a textual proposal. I researched the texts and, at the same time, my artistic language which I developed according to the selected textual proposals. In the theoretical part of the bachelor’s thesis, I dealt with a historical review of illustration in connection with poetry and illustrated editions of poetry for adults, as well as with the definition, history, and visual characteristics of visual and concrete poetry. I also researched the format and medium of the art zine, its definition, position in fine art, and the environment in which it is created. In the practical part of the assignment, I formed visual images and visually presented selected poems collected in the form of a zine. I dealt with the connection between illustration and poetry and their artistic power and realized that they can stand on their own. Combined, however, they complement each other even more.

SummaryPhosphatidylinositol(4,5)bisphosphate (PI(4,5)P2) is an important signaling phospholipid that is required for regulated exocytosis and some forms of endocytosis. The two processes share a topologically similar pore structure that connects the vesicle lumen with the outside. Widening of the fusion pore during exocytosis leads to cargo release, while its closure initiates kiss&run or cavicapture endocytosis. We show here, using live cell TIRF microscopy of insulin granule exocytosis, that transient accumulation of PI(4,5)P2 at the release site recruits components of the endocytic fission machinery, and stalls the late fusion pore expansion that is required for peptide release. The absence of clathrin differentiates this mechanism from clathrin-mediated endocytosis. The PI(4,5)P2 transients result from local phosphatidylinositol-phosphate-5-kinase-1c (PIP5K1c) activity, and knockdown of PIP5K1c, or optogenetic ablation of PI(4,5)P2 promotes fusion pore expansion. Thus, local phospholipid signaling controls fusion pore expansion peptide release through an unconventional endocytic mechanism.

Celovita presoja vplivov na okolje (CPVO) je postopek, s katerim je mogoče ugotoviti in oceniti vplive načrtovanih ukrepov načrta na okolje ter vključenost zahtev drugih inštitucij v načrt. Skladno z Zakonom o varstvu okolja (ZVO-2) je treba CPVO izvesti tudi za načrte s področja gozdarstva. V okviru obnove območnih gozdnogospodarskih načrtov (GGN GGO 2021–2030) ter lovsko upravljavskih načrtov (LUN LUO 2021–2030) je bil za omenjene načrte prvič izveden postopek CPVO, in sicer en postopek za vseh štirinajst GGN GGO 2021–2030 ter en postopek za petnajst LUN LUO 2021–2030. Postopek CPVO se je začel z vsebinjenjem, v okviru katerega je bila na podlagi opredelitve pomembnih vplivov plana skupaj s pristojnim ministrstvom in mnenjedajalci sprejeta odločitev o vsebinah, ki se bodo presojale v nadaljevanju postopka CPVO. Za vsak sklop načrtov je bilo pripravljeno okoljsko poročilo z dodatkom, v katerem so se opredelili, opisali in ovrednotili pomembni vplivi izvedbe obeh sklopov načrtov na okolje, ohranjanje narave, varstvo človekovega zdravja in kulturne dediščine. Vpliv izvedbe GGN GGO 2021–2030 na okoljska cilja Trajnostna raba in varstvo naravnih virov ter Blaženje in prilagajanje na podnebne spremembe je bil zaradi številnih pozitivnih učinkov načrtov ocenjen kot pozitiven (ocena A). Vpliv izvedbe GGN GGO 2021–2030 na okoljska cilja Ohranjena kulturna dediščina in Ohranjena narava je bil zaradi nekaterih pomanjkljivosti v načrtih ocenjen kot C – nebistven vpliv ob upoštevanju omilitvenih ukrepov. Ocenjeno je bilo, da bodo z izvedbo vsakega LUN LUO 2021–2030 nastali nebistveni negativni vplivi (ocena B) na okoljski cilj Trajnostna raba in varstvo naravnih virov in da bo vpliv izvedbe vseh petnajstih LUN LUO 2021–2030 na okoljski cilj Ohranjena narava nebistven zaradi izvedbe omilitvenih ukrepov (ocena C). V obeh okoljskih poročilih so bili navedeni omilitveni ukrepi, ki so bili vgrajeni v osnutke načrtov. CPVO je dolgotrajen in zapleten postopek, ki znatno podaljšuje čas sprejemanja območnih načrtov. Vendar pa zaradi poglobljene presoje mnenjedajalcev in presojevalcev daje načrtom bistveno večjo težo. Hkrati se s CPVO izboljšuje vpetost gozdarskega in lovskega načrtovanja v okoljsko politiko Slovenije, kar daje obema sektorjema večjo veljavo. Strategic environmental assessment (SEA) is a procedure aimed at identifying and assessing the impacts of a plan in order to ensure that the principles of sustainable development, integrity and prevention are implemented. In accordance with the Law on protection of the environment (ZVO-2), SEA must also be carried out for plans in the field of forestry. In the frame of revision of regional forest management (GGN GGO 2021–2030) and game management plans (LUN LUO 2021–2030), SEA was carried out, namely one for all 14 GGN GGO 2021–2030 and one for 15 LUN LUO 2021–2030. SEA procedure started with the scoping, in the context of which decisions on the issues that were to be assessed in the latter stages of the SEA were determined together with competent ministry and the opinion makers. For each set of plans, an environmental report including appropriate assessment was drafted, in which the important impacts of the implementation of both sets of plans on the environment, nature conservation, protection of human health and cultural heritage were defined, described and assessed. The impact of the implementation of GGN GGO 2021–2030 on the environmental objectives Sustainable use and protection of natural resources and Mitigation and adaptation to climate changes was assessed as positive (grade A) due to many positive impacts of the plans. The impact of the implementation of GGN GGO 2021–2030 on the environmental objectives Preserved cultural heritage and Preserved nature was assessed with the grade C (non-material impact due to the implementation of mitigation measures) due to some shortcomings in the plans. It was assessed that the implementation of each LUN LUO 2021–2030 will lead to nonmaterial impacts (grade B) on the environmental objective Sustainable use and protection of natural resources and that the implementation of 15 LUN LUO 2021–2030 on the environmental objective Preserved nature will be non-material due to the implementation of mitigation measures (grade C). The defined mitigation measures were integrated into the draft plans. SEA is a long-term and complex procedure, which significantly prolongs the period of adaptation of plans. However, it gives more weight to the plans due to the in-depth assessment by opinion makers and assessors. In addition, SEA improves the inclusion of forestry and game management planning in the environmental policy of Slovenia, which gives both sectors greater validity.

V letih 2020, 2021 in 2022 je potekala obnova območnih gozdnogospodarskih in lovskoupravljavskih načrtov (v nadaljevanju območni načrti), ki bodo usmerjali razvoj slovenskih gozdov do leta 2030. Zaradi vse zahtevnejšega upravljanja gozdnih ekosistemov in usklajevanja številnih interesov je sodelovanje deležnikov v postopku priprave in sprejemanja območnih načrtov ključnega pomena. Pri tokratni obnovi območnih načrtov je bil velik poudarek ravno na vključevanju lastnikov gozdov in drugih deležnikov v proces izdelave in sprejemanja načrtov. Formalno določeno participacijo smo nadgradili z aktivnejšim vključevanjem zainteresirane javnosti prek številnih predstavitev, delavnic, anket in sestankov. V letih 2020 in 2021 je bilo tako v sklopu priprave območnih načrtov, poleg zbiranja pobud in javne razprave ter javne predstavitve načrtov, dodatno organiziranih 73 delavnic na regionalni in nacionalni ravni, na katerih je sodelovalo več kot dva tisoč udeležencev. Zaradi preprečevalnih ukrepov zaradi epidemije bolezni covid-19 je večina participativnih aktivnosti v letih 2020 in 2021 potekala v spletni obliki, kar je pospešilo digitalizacijo gozdarstva. Nastala so številna orodja za digitalno sodelovanje, in sicer od novih programskih rešitev na spletni strani do široke implementacije in uporabe orodij za digitalno sodelovanje. Rezultati participativnih aktivnosti so bili vgrajeni v osnutke območnih načrtov. Nadgradnja participativnega pristopa in vključevanje zainteresirane javnosti že v fazi priprave osnutkov območnih načrtov sta prispevala k večji usklajenosti gozdnogospodarskega in lovskoupravljavskega dela območnih načrtov, večji kakovosti in izvedljivosti načrtov ter usklajevanju ekonomskih, ekoloških in socialnih vidikov upravljanja z gozdnimi ekosistemi. In the years 2020, 2021 and 2022, the regional forest management and game management plans (hereafter referred to as regional plans) are being revised they will guide the development of Slovenian forests until 2030. Due to increasing challenges in managing forest ecosystems and the need for harmonization of various interests, participation of stakeholders in the process of preparation of regional plans is of outmost importance. In the current revision of regional plans, a great deal of emphasis was given to the inclusion of forest owners and other stakeholders in the process of preparation of the plans. Formally defined participation was upgraded with more active involvement of the interested public through various presentations, workshops, surveys and meetings. In 2020 and 2021, in addition to the collection of initiatives and public debate and public presentation of plans, 73 workshops on regional and national level with 2000 participants were additionally organized as part of preparation of regional plans. Due to preventive measures due to covid-19, most of the participatory activities in 2020 and 2021 were carried out online, which accelerated the digitalization of forestry. Various tools for digital participation have been developed, from new software solutions on the website to the wide implementation and the use of tools for digital participation. The results of participatory activities were integrated in the drafts of regional plans. The upgrade of participatory approach and the inclusion of the interested public in an early phase of plan preparation contributed to greater coordination of forest management and game management part of the regional plans, to the greater quality and feasibility of the plans and to the harmonization of economic, ecological and social aspects of managing forest ecosystems.

With the awareness of a changing climate impacting our sustainability, and in line with the European Green Deal initiative or the Sustainable Development Goal 6 addressing water, the industry, society and local governments are requiring reliable and comprehensive technology that can provide them an overview to water events to anticipate problems and the tools to analyse best practices appropriate to solve them. This paper presents the NAIADES Water Observatory (NOW), a digital solution offering a series of analysis and visualisations of water-related topics, helping users to extract important insights in relation to the water sector. Taking advantage of heterogeneous data sources, from the media and social media landscape, to published research and global/local indicators. Through collaboration with local water resource management institutions, the NWO was configured to local priorities and ingests local datasets to better fit the needs of decision-makers

In some lysosomal storage diseases (LSD) cholesterol accumulates in vesicles. Whether increased vesicle cholesterol affects vesicle fusion with the plasmalemma, where the fusion pore, a channel between the vesicle lumen and the extracellular space, is formed, is unknown. Super-resolution microscopy revealed that after stimulation of exocytosis, pituitary lactotroph vesicles discharge cholesterol which transfers to the plasmalemma. Cholesterol depletion in lactotrophs and astrocytes, both exhibiting Ca$^{2+}$-dependent exocytosis regulated by distinct Ca$^{2+}$ sources, evokes vesicle secretion. Although this treatment enhanced cytosolic levels of Ca$^{2+}$ in lactotrophs but decreased it in astrocytes, this indicates that cholesterol may well directly define the fusion pore. In an attempt to explain this mechanism, a new model of cholesterol-dependent fusion pore regulation is proposed. High-resolution membrane capacitance measurements, used to monitor fusion pore conductance, a parameter related to fusion pore diameter, confirm that at resting conditions reducing cholesterol increases, while enrichment with cholesterol decreases the conductance of the fusion pore. In resting fibroblasts, lacking the Npc1 protein, a cellular model of LSD in which cholesterol accumulates in vesicles, the fusion pore conductance is smaller than in controls, showing that vesicle cholesterol controls fusion pore and is relevant for pathophysiology of LSD.

Uvajanje informacijskih tehnologij v arheološko delo je vplivalo v 21. stoletju na to, da sta Londonska listina in Seviljska načela 3D modeliranje in digitalizacijo priporočili kot mednarodni standard arheološke dokumentacije. 3D modeli so trajni nosilci številnih informacij o predmetu, ki jih lahko arheologi preiskujejo, vrednotijo in primerjajo, ne da bi pri tem predmete poškodovali ali uničili. Večina 3D modelov je v arheologiji izdelana s fotogrametrijo, snemalniki na strukturirano svetlobo ali z laserskimi snemalniki. Delo hipotetično predpostavlja, da površinski 3D modeli ne zagotavljajo potrebnih informacij za celostno obravnavo in vrednotenje arheoloških ostalin. Popolnejšo informacijo lahko zagotovi le prostorninski 3D model. Tega je mogoče neinvazivno rekonstruirati iz mikrotomografskih slik. V magistrskem delu sta opisana primera zasnove dveh iterativnih algoritmov za rekonstrukcijo prostorninskih 3D modelov iz dvodimenzionalnih mikrotomografskih slik. Algoritma sta izdelana s programskim paketom za numerično analizo MatLab. Prilagojena sta konkretnim arheološkim ciljem raziskovanja (deformacijska in volumetrična analiza 4D analiza prepoznavanje in mikrolokacija poškodb, deformacij, razpok in lomov). Oblak točk in triangulacijska mreža prostorninskega 3D modela vsebujeta veliko število informacij, ki kvalitativno dopolnjujejo informacije površinskega 3D modela in optimirajo arheološki dokumentalistični zapis. Testiranje algoritmov na izbranih arheoloških predmetih je potrdilo njuno uporabnost za celostno vrednotenje arheoloških objektov. Algoritma pa sta lahko tudi koristno orodje v postopkih izbora ustreznejših metod in tehnik konserviranja in restavriranja ostalin kulturne dediščine. The introduction of information technology into archaeological work has in the 21st century influenced The London Charter and The Seville Principles in such a way that they recommended 3D modeling and digitization as an international standard for archaeological documentation. 3D models are permanent carriers of a wealth of information about an object that archaeologists can investigate, evaluate, and compare without damaging or destroying the objects. Most 3D models in archeology are made with photogrammetry, structured light recorders or with laser recorders. This work hypothetically assumes that surface 3D models do not provide the necessary information for a comprehensive treatment and evaluation of archaeological remains. Only a volumetric 3D model can provide more complete information. This can be non-invasively reconstructed from microtomographic images. The master’s thesis describes the design of two iterative algorithms for the reconstruction of volumetric 3D models from two-dimensional microtomographic images. The algorithms are developed using the MatLab numerical analysis software package. They are adapted to the specific archaeological objectives of the research (deformation and volumetric analysis 4D analysis identification and microlocation of damage, deformations, cracks and fractures). The point cloud and the triangulation network of the volumetric 3D model contains a large amount of information that qualitatively complements the information of the surface 3D model and optimizes the archaeological documentary record. Testing of the algorithms on selected archaeological objects confirmed their usefulness for the comprehensive evaluation of archaeological objects. Algorithms can also be a useful tool in the process of selecting more appropriate methods and techniques for the conser-vation and restoration of cultural heritage remains.
