7. RASGRP1 targeted by H3K27me3 regulates myoblast proliferation and differentiation in mice and pigs

Abstract

Skeletal muscle is not only the largest organ in the body that is responsible for locomotion and exercise but also crucial for maintaining the body’s energy metabolism and endocrine secretion. The trimethylation of histone H3 lysine 27 (H3K27me3) is one of the most important histone modifications that participates in muscle development regulation by repressing the transcription of genes. Previous studies indicate that the RASGRP1 gene is regulated by H3K27me3 in embryonic muscle development in pigs, but its function and regulatory role in myogenesis are still unclear. In this study, we verify the crucial role of H3K27me3 in RASGRP1 regulation. The gain/loss function of RASGRP1 in myogenesis regulation is performed using mouse myoblast C2C12 cells and primarily isolated porcine skeletal muscle satellite cells (PSCs). The results of qPCR, western blot analysis, EdU staining, CCK-8 assay and immunofluorescence staining show that overexpression of RASGRP1 promotes cell proliferation and differentiation in both skeletal muscle cell models, while knockdown of RASGRP1 leads to the opposite results. These findings indicate that RASGRP1 plays an important regulatory role in myogenesis in both mice and pigs.

Abstract

For each non-constant $q$ in the set of $n$-variable Boolean functions, the {\em $q$-transform} of a Boolean function $f$ is related to the Hamming distances from $f$ to the functions obtainable from $q$ by nonsingular linear change of basis. Klapper conjectured that no Boolean function exists with its $q$-transform coefficients equal to $\pm 2^{n/2}$ (such function is called $q$-bent). In our early work, we only gave partial results to confirm this conjecture for small $n$. Here we prove thoroughly that the conjecture is true by investigating the nonexistence of the partial difference sets in Abelian groups with special parameters. We also introduce a new family of functions called almost $q$-bent functions, which are close to $q$-bentness.

Abstract

The iron‐armored chain horse is a military strategy in ancient China to enhance the combat force of the entire army, especially when individual fighters are vulnerable to being picked off one by one. Herein, this strategy is adopted to generate highly efficient metal‐free blue room temperature phosphorescence (RTP) using the world's largest production of dicarboxylate acid. By encapsulating the commercially available chemical disodium terephthalate (DTPA) within the iron armor of α‐cyclodextrin (α‐CD), and subsequently linking the iron‐armored DTPA@α‐CD horses into a crystalline array mediated by hydrogen bonds and Na+ coordination, a highly confined and segregated arrangement of DTPA is achieved, leading to heavy atom‐free deep‐blue phosphorescence with a maximum efficiency of 83.3%. Since the crystalline array of the iron‐armored chain horse DTPA@α‐CD can be facilely obtained through water evaporation, the current phosphorescent material allows for rapid, high‐throughput solution processing of deep‐blue afterglow display, offering an economic and environmentally benign approach for deep‐blue RTP materials and facilitating their application. Abstract

Birth weight is a complex multifactorial trait relevant to health states and disease risks in later life. The placenta is essential for proper fetal growth and facilitates gas, nutrient, and waste exchange between the mother and developing fetus. How changes in placental DNA methylation affect fetal birth weight remains to be fully elucidated. In this study, we used whole-genome bisulfite sequencing and RNA sequencing to reveal a global map of DNA methylation and gene expression changes between the placentas of highest birth weight and lowest birth weight piglets in the same litters. The transcriptome analysis identified 1682 differential expressed genes and revealed key transcriptional properties in distinct placentas. We also identified key transcription factors that may drive the differences in DNA methylome patterns between placentas. The decrease in DNA methylation level in the promoter was associated with the transcriptional activation of genes associated with angiogenesis, extracellular matrix remodeling, and transmembrane transport. Our results revealed the regulatory role of DNA methylation in gene transcription activity leading to the differences in placental morphological structures and birth weights of piglets. These results could provide novel clues to clarify the underlying regulatory mechanisms of placental development and fetal growth. [ABSTRACT FROM AUTHOR]

Abstract

Major depressive disorder is a common and devastating psychiatric disease, and the prevalence and burden are substantially increasing worldwide. Multiple studies of depression patients have implicated glucose metabolic dysfunction in the pathophysiology of depression. However, the molecular mechanisms by which glucose and related metabolic pathways modulate depressive-like behaviors are largely uncharacterized. Uridine diphosphate N-acetylglucosamine (UDP-GlcNAc) is a glucose metabolite with pivotal functions as a donor molecule for O-GlcNAcylation. O-GlcNAc transferase (OGT), a key enzyme in protein O-GlcNAcylation, catalyzes protein posttranslational modification by O-GlcNAc and acts as a stress sensor. Here, we show that Ogt mRNA was increased in depression patients and that astroglial OGT expression was specifically upregulated in the medial prefrontal cortex (mPFC) of susceptible mice after chronic social-defeat stress. The selective deletion of astrocytic OGT resulted in antidepressant-like effects, and moreover, astrocytic OGT in the mPFC bidirectionally regulated vulnerability to social stress. Furthermore, OGT modulated glutamatergic synaptic transmission through O-GlcNAcylation of glutamate transporter-1 (GLT-1) in astrocytes. OGT astrocyte-specific knockout preserved the neuronal morphology atrophy and [Ca.sup.2+] activity deficits caused by chronic stress and resulted in antidepressant effects.
