10. Microarray Expression Profile of Circular RNAs in Peripheral Blood Mononuclear Cells from Active Tuberculosis Patients

Abstract

Background/Aims: Dysregulated expression of circular RNAs (circRNAs) was demonstrated to be implicated in many diseases. Here, we aimed to determine circRNA profile in peripheral blood mononuclear cells (PBMCs) from active tuberculosis (TB) patients to identify novel biomarkers for TB. Methods: Expression profile of circRNAs in PBMCs from 3 active pulmonary TB patients and 3 healthy controls were analyzed by microarray assay. Six circRNAs were selected for validation using real time-quantitative PCR (qRT-PCR) in 40 TB patients and 40 control subjects. Receiver operating characteristic (ROC) curve was constructed to evaluate their values in TB diagnosis. Hsa_circRNA_001937 was chosen for further evaluation in an independent cohort consisting of 115 TB, 40 pneumonia, 40 COPD, 40 lung cancer patients and 90 control subjects. An eight-month follow up was performed in 20 newly diagnosed TB patients to investigate the expression change of hsa_circRNA_001937 after chemotherapy. Results: We revealed and confirmed that a number of circRNAs were dysregulated in TB patients. Of the six studied physio circRNAs, the levels of hsa_circRNA_001937, hsa_circRNA_009024 and hsa_ circRNA_005086 were significantly elevated and hsa_circRNA_102101, hsa_circRNA_104964 and hsa_circRNA_104296 were significantly reduced in PBMCs from TB patients as compared to healthy controls. 13. Molecular composition of low-temperature oxidation products of the heavy oil.

Abstract

Low-temperature oxidation (LTO) is the main reaction that affects fuel formation in the in-situ combustion process, which has important significance for the subsequent combustion propulsion and the successful extraction of crude oil. In this study, heavy oil was subjected to LTO reactions at different temperatures. Three types of reaction products with varying oxidation depths were characterized in terms of the number of oxygen atoms and the polarity of the molecule to reveal the low-temperature oxidation process of the heavy oil. Ketone compounds and acid polyoxides in the oil phase and deep oxidation products with a higher number of oxygen atoms in the coke were identified with increasing oxidation depth. The experimental results showed that the oxidation reaction of the heavy oil changed from kinetic-controlled to diffusion-controlled in the open oxidation system of the heavy oil as the oxidation depth increased. The oxidation reaction of the oil phase reached a maximum and stable value in oxygen content. The molecular compositions of the ketone compound and acid polyoxide did not change significantly with further increase in reaction temperature. The molecular compositions of the deep oxidation products with a higher number of oxygen atoms in the coke phase changed significantly. The coke precursor molecules with a lower oxygen content and condensation degree participated in the coke formation, and the oxidation reaction pathway and the complexity of the oxidation product component also increased. Abstract

Chemotherapy is the main clinical treatment method of gastric cancer. Multidrug resistance (MDR) has become a common phenomenon with the development of tumors, which alleviates the effect of chemotherapy and makes it difficult to break the bottleneck of survival rate of advanced gastric cancer. Therefore, the exploration of MDR reversal agents for gastric cancer is the focus and also the difficulty of current treatment. Currently, the researches on the mechanisms of drug resistance in gastric cancer have been continuously deepened, which reveal different pathways and targets of MDR, laying a solid foundation for studying reversal agents. As a kind of natural medicine, traditional Chinese medicine (TCM) owns the characteristics of low toxicity, high safety and effectiveness. It can inhibit the occurrence, growth and metastasis of tumors, and reverse MDR via multiple pathways and mechanisms, the pathological function of which has become a research hotspot in recent years. TCM reversers are mainly divided into Chinese medicine monomers, Chinese patent medicines, and Chinese herbal compounds. With certain quantity and advantage, TCM reversers for MDR play an important role in the clinical treatment and show great potential in gastric cancer.
