Abstract

Fusarium oxysporum causes devastating vascular wilt diseases in numerous crop species, resulting in substantial yield losses. The Arabidopsis thaliana-F. oxysporum f.sp. conglutinans (FOC) model system enables the identification of meaningful genotype–phenotype correlations and was applied in this study to evaluate the effects of overexpressing an NLR gene (AsTIR19) from Arachis stenosperma against pathogen infection. AsTIR19 overexpression (OE) lines exhibited enhanced resistance to FOC without any discernible phenotype penalties. To elucidate the underlying resistance mechanisms mediated by AsTIR19 overexpression, we conducted whole transcriptome sequencing of an AsTIR19-OE line and non-transgenic wild-type (WT) plants inoculated and non-inoculated with FOC using Illumina HiSeq4000. Comparative analysis revealed 778 differentially expressed genes (DEGs) attributed to transgene overexpression, while fungal inoculation induced 434 DEGs in the OE line, with many falling into defense-related Gene Ontology (GO) categories. GO and KEGG enrichment analysis showed that DEGs were enriched in the phenylpropanoid and flavonoid pathways in the OE plants. This comprehensive transcriptomic analysis underscores how AsTIR19 overexpression reprograms transcriptional networks, modulating the expression of stress-responsive genes across diverse metabolic pathways. These findings provide valuable insights into the molecular mechanisms underlying the role of this NLR gene under stress conditions, highlighting its potential to enhance resistance to Fusarium oxysporum.

Abstract Background Digital health technologies have expanded tremendously in the last two decades, creating an emerging research and clinical field. They are regarded as cost‐effective, and their use in healthcare is prioritized by many countries. However, the constant evolution of these technologies has led to an abundance of related literature. Thus, we conducted an umbrella review to identify and characterize digital supportive care interventions for patients with cancer and their relatives. Methods A preregistered umbrella review was conducted (PROSPERO registration number CRD42022333110). Five databases were searched (Embase, PsycINFO, PubMed, CINAHL, and the Cochrane Library). To be considered, studies had to be systematic reviews or meta‐analyses, be performed on pediatric or adult patients with cancer or survivors or their relatives, report results on web‐based or app‐based supportive care interventions, and measure psychological, functional, or behavioral variables or quality of life related to cancer. The methodological quality of the studies was assessed using the AMSTAR‐2 tool. Findings Twenty eligible studies were identified. Most of the included studies reported results from adult patients with cancer. Globally, digital interventions were shown to be effective for physical activity in patients with cancer but had mixed results regarding emotional outcomes and quality of life. Additionally, a lack of methodological quality was noted for most of the included reviews. Discussion Digital supportive care interventions could be an effective tool in cancer care for some outcomes. Recommendations have been formulated for further research in this field using adapted methodologies for the development of digital health interventions.

Abstract

In the last decade, the evolution of oncofertility has sparked a resurgence of interest in in vitro maturation (IVM) due to its suitability in certain oncological scenarios where controlled ovarian hyperstimulation may not be feasible. The retrieval of immature cumulus–oocyte complexes from small antral follicles, regardless of the menstrual cycle phase, presents a swift opportunity to vitrify mature oocytes or embryos post-IVM in urgent situations or when stimulation is not advisable. Harvesting immature cumulus–oocyte complexes and immature oocytes can be achieved transvaginally or directly in the laboratory from extracorporeal ovarian tissue. Although IVM has transitioned from an experimental status due to safety validations, it relies on the intricate process of oocyte maturation. Despite successful live births resulting from IVM in fertility preservation contexts, the comparatively lower developmental competence of in vitro matured oocytes highlights the necessity to enhance IVM culture systems. Recent advancements in IVM systems hold promise in bolstering oocyte competence post-IVM, thereby narrowing the gap between IVM and outcomes from ovarian stimulation. Additionally, for optimizing the chances of conception in cancer survivors, the combination of IVM and ovarian tissue cryopreservation stands as the favored choice when ovarian stimulation is unfeasible.

[...], Trademark scholars love to hate the merchandising right (i.e., the use of trademark law to give trademark owners control over product markets in which the trademark is the good--e.g., a BOSTON RED SOX baseball cap). We think that trademark law should protect consumer interests. If no one thinks that sports teams manufacture their own merchandise, then there's no possibility of source confusion. Rather than benefitting consumers, the merchandising right artificially increases consumer costs by giving trademark holders an unwarranted monopoly over the use of their marks as products. Nobody cares. Whatever law professors may think, people--and importantly, judges--generally believe that trademark holders should control merchandising markets. Hard-wired moral intuitions suggest that the resulting profits are a fair reward for creating popular brands and that others should not "free ride" off of these efforts. These intuitions are resistant to argument. We are therefore likely stuck with the merchandising right. Nonetheless, the merchandising right is inconsistent with fundamental trademark doctrine. Accommodating it creates difficulties that ripple throughout trademark law and reach cases that have nothing to do with merchandising logos. The problem is especially acute in those cases in which it is the trademark owner who is trying to capture an unearned benefit. These cases turn the intuitions behind the merchandising right on their head, but courts do not have a vocabulary for distinguishing them from "traditional" merchandising disputes. The result is a muddle that affects trademark law as a whole. We might ameliorate the problem by taking anti-free riding moral intuitions seriously. If they are to be part of trademark law, then they may be used to limit the merchandising right and reconcile it to trademark law. To be sure, living with the merchandising right is hardly an optimal result, but if the right is here to stay, we may as well contain its scope and craft a trademark doctrine that comports with the moral intuitions of the judges who implement it.

Abstract

Cupuassu (Theobroma grandiflorum) is a fruit tree native to the Amazon region, presenting high social and economic value. Besides, owing to its suitability for agroforestry cultivation, cupuassu is useful for the conservation of the Amazon Forest. Cupuassu plantations are severely affected by Moniliophthora perniciosa. Thus, to gain insights into resistance against M. perniciosa, transcriptomes of susceptible (SG) and resistant (RG) genotypes of cupuassu were analyzed at the early stage of infection using RNA sequencing. A total of 21,441 unigenes were identified, and differentially expressed genes were detected in intra- (440) and inter-genotype (301) analyses. Gene expression was altered at 24 h after inoculation (HAI) in SG. This alteration was prominent at 48 HAI in RG. These datasets allowed the identification of genes potentially involved in defense mechanisms. Phytohormone signature analysis revealed a significant effect of hormones on genotype responses. The present study is the first large-scale transcriptomic analysis of cupuassu.
