Abstract

A physicochemical characterization of the process-related impurities associated with the synthesis of pemetrexed disodium was performed. The possibility of pemetrexed impurities forming has been mentioned in literature, but no study on their structure has been published yet. This paper describes the development of the synthesis methods for these compounds and discusses their structure elucidation on the basis of two-dimensional NMR experiments and MS data. The identification of these impurities should be useful for the quality control during the production of the pemetrexed disodium salt. [ABSTRACT FROM AUTHOR]

Abstract

A two-step chemical process for controlled degradation of escin, affording a mixture of olean-12-ene sapogenins, was elaborated and scaled up. The main component of the mixture--protoescigenin--was isolated and purified, in the form of its corresponding monohydrate, without resource to chromatographic methods. This material was further converted into the high purity 3,24;16,22-di-O,O-isopropylidene derivative in a validated large scale laboratory process. [ABSTRACT FROM AUTHOR]

8. Synthesis and Stereochemical Properties of “Extended” Biphenols Bridged by ortho-, meta-, and para-Phenylene Spacers

Abstract

A series of isomeric biphenols based on para1, meta2, and ortho3 terphenyl backbones was synthesized. Suzuki crosscoupling methodology was employed for the construction of the terphenyl backbone of their methylprotected precursors respectively 8, 13, and 16. Using K2CO3as the base, the best reaction conditions involved DMF at 100 °C as solvent. Anhydrous conditions greatly improved the yields of the sterically crowded systems particularly 16. orthoTerphenyls 3and 16exist as mixtures of synantiatropisomers in solution. Compound 16crystallizes in the antiinform. DFT calculations at the B3LYP6311+Gd,p level indicate that for both compounds the antiinform is more stable than the synform by 6.07 and 2.07 kJ mol–1for 16and 3, respectively. These ΔΔG0values are higher by the same factor of two than those obtained in solution at 240 K 3.17 and 0.93 kJ mol–1. The atropisomeric properties of 16and 3are finally discussed in the context of orthoterphenyls in the literature. © WileyVCH Verlag GmbH & Co. KGaA, 69451 Weinheim, Germany, 2009
