International audience; For several decades the "prawn-in-the-tube" procedure has been extensively used in the exploration of behavioral plasticity and its neural correlates in cuttlefish. Although the nature of the task has been characterized, the effect of reinforcement and the extent of different cues cuttlefish can use to solve and memorize the task remain unclear. To determine whether cuttlefish learned to inhibit predatory behavior because of pain incurred when the tentacles hit the glass tube, the shrimp prey (typically attacked with a tentacle strike) was replaced by crabs (normally caught by a jumping strategy, using all eight arms together, which is thought less likely to be painful). We showed that the cuttlefish is still capable of learning inhibition of predatory behavior when it adopts another catching strategy, which suggests that pain from the tentacles hitting the tube has little effect on the learning process. The two latest experiments have shown that cuttlefish do not learn to inhibit predatory behavior towards a specific type of prey, but rather learn and memorize visual (light polarization) and tactile information from the glass tube. The "prawn-in-the-tube" procedure is a powerful and user-friendly tool in the investigation of the processing and retention of multisensory information in invertebrates. Our recent findings now open up new areas of investigation into the neural correlates of learning and memory processes in cuttlefish.

WOS:000389670100001; International audience; Vertebrates with laterally placed eyes typically exhibit preferential eye use for ecological activities such as scanning for predators or prey. Processing visual information predominately through the left or right visual field has been associated with specialized function of the left and right brain. Lateralized vertebrates often share a general pattern of lateralized brain function at the population level, whereby the left hemisphere controls routine behaviors and the right hemisphere controls emergency responses. Recent studies have shown evidence of preferential eye use in some invertebrates, but whether the visual fields are predominately associated with specific ecological activities remains untested. We used the European common cuttlefish. Sepia officinalis, to investigate whether the visual field they use is the same, or different, during anti-predatory, and predatory behavior. To test for lateralization of anti-predatory behavior, individual cuttlefish were placed in a new environment with opaque walls, thereby obliging them to choose which eye to orient away from the opaque wall to scan for potential predators (i.e., vigilant scanning). To test for lateralization of predatory behavior, individual cuttlefish were placed in the apex of an isosceles triangular arena and presented with two shrimp in opposite vertexes, thus requiring the cuttlefish to choose between attacking a prey item to the left or to the right of them. Cuttlefish were significantly more likely to favor the left visual field to scan for potential predators and the right visual field for prey attack. Moreover, individual cuttlefish that were leftward directed for vigilant scanning were predominately rightward directed for prey attack. Lateralized individuals also showed faster decision-making when presented with prey simultaneously. Cuttlefish appear to have opposite directions of lateralization for anti-predatory and predatory behavior, suggesting that there is functional specialization of each optic lobe (i.e., brain structures implicated in visual processing). These results are discussed in relation to the role of lateralized brain function and the evolution of population level lateralization.

International audience; Juvenile cuttlefish spontaneously prefer shrimp to crabs. This preference can be changed by early visual learning during embryonic and postembryonic life and has been related to a form of food imprinting. Nevertheless, it is unknown whether generalization occurs in conjunction with this early learning process. We found that inexperienced hatchling cuttlefish preferred 'black' to 'white crab' for their first meal, the black ink envelope of cuttlefish eggs did not prevent embryos discriminating between black and white crabs and embryonic or postembryonic visual familiarization to 'white crabs' induced a subsequent visual preference for 'white crabs' over 'black crabs' in the hatchling's first meal. Finally, juvenile cuttlefish previously pre- or postnatally exposed to 'white crabs' preferred 'black crabs' to shrimp, indicating for the first time that prey generalization occurs as early as the embryonic stages in cuttlefish. Such cognitive abilities could confer important adaptive advantages in processing information about prey likely to be available in the egg-laying environment at hatching and in the course of juvenile dispersal.

International audience; Cuttlefish are sensitive to linear polarization of light, a sensitivity that they use in predation and possibly in intraspecific communication. It has also been shown that cuttlefish are able to solve a maze using visual landmarks. In this study, cuttlefish were trained to solve a Y-maze with the e-vector of a polarized light and landmarks as redundant spatial information. The results showed that cuttlefish can use the e-vector orientation and landmarks in parallel to orient and that they are able to use either type of cue when the other one is missing. When they faced conflicting spatial information in the experimental apparatus, the majority of cuttlefish followed the e-vector rather than landmarks. Differences in response latencies in the different conditions of testing (training with both types of cue, tests with single cue or with conflicting information) were observed and discussed in terms of decision making. The ability to use near field and far field information may enable animals to interpret the partially occluded underwater light field.

International audience; The interaction between depression and stroke is highly complex. Post-stroke depression (PSD) is among the most frequent neuropsychiatric consequences of stroke. Depression also negatively impacts stroke outcome with increased morbidity, mortality and poorer functional recovery. Antidepressants such as the commonly prescribed selective serotonin reuptake inhibitors improve stroke outcome, an effect that may extend far beyond depression, e.g., to motor recovery. The main biological theory of PSD is the amine hypothesis. Conceivably, ischaemic lesions interrupt the projections ascending from midbrain and brainstem, leading to a decreased bioavailability of the biogenic amines--serotonin (5HT), dopamine (DA) and norepinephrine (NE). Acetylcholine would also be involved. So far, preclinical and translational research on PSD is largely lacking. The implementation and characterization of suitable animal models is clearly a major prerequisite for deeper insights into the biological basis of post-stroke mood disturbances. Equally importantly, experimental models may also pave the way for the discovery of novel therapeutic targets. If we cannot prevent stroke, we shall try to limit its long-term consequences. This review therefore presents animal models of PSD and summarizes potential underlying mechanisms including genomic signatures, neurotransmitter and neurotrophin signalling, hippocampal neurogenesis, cellular plasticity in the ischaemic lesion, secondary degenerative changes, activation of the hypothalamo-pituitary-adrenal (HPA) axis and neuroinflammation. As stroke is a disease of the elderly, great clinical benefit may especially accrue from deciphering and targeting basic mechanisms underlying PSD in aged animals.

International audience; INTRODUCTION: Effects of 3-month continuous environmental enrichment (EE) on cognitive abilities and on theta burst-related synaptic plasticity of CA1 hippocampal neuronal networks have been assessed in 6- and 20-month old NMRI female mice. RESULTS: EE decreased anxiety-like behavior and improved learning and memory performances in adult but not in aged mice. Electrophysiological results in CA1 hippocampal slices showed that basal synaptic transmission was not affected by EE in adult mice whereas it was partially improved in aged animals, even though not sufficient to rescue the decrease related to aging. Besides, no effect of EE on N-methyl-d-aspartate receptor activation and theta-burst-induced long-term potentiation was found in adult or aged animals. DISCUSSION: These results indicate that continuous EE is able to improve cognitive abilities in adult NMRI female mice, that does not correlate with changes in theta burst-related synaptic plasticity within neuronal networks. In addition, the lack of effects in aged animals suggests the existence of a critical delay for the beneficial effects of EE on cognitive aging.

International audience; 5-Hydroxytryptamine subtype-4 (5-HT(4)) receptors have stimulated considerable interest amongst scientists and clinicians owing to their importance in neurophysiology and potential as therapeutic targets. A comparative analysis of hierarchical methods applied to data from one thousand 5-HT(4) receptor-ligand binding interactions was carried out. The chemical structures were described as chemical and pharmacophore fingerprints. The definitions of indices, related to the quality of the hierarchies in being able to distinguish between active and inactive compounds, revealed two interesting hierarchies with the Unity (1 active cluster) and pharmacophore fingerprints (4 active clusters). The results of this study also showed the importance of correct choice of metrics as well as the effectiveness of a new alternative of the Ward clustering algorithm named Energy (Minimum E-Distance method). In parallel, the relationship between these classifications and a previously defined 3D 5-HT(4) antagonist pharmacophore was established.

WOS:000379002800010; International audience; A great deal is known about development in Sepia officinalis; however, much of this knowledge comes from animals incubated in laboratory conditions. Since cuttlefish are behaviorally plastic and known as embryos to perceive environmental stimuli from within the egg, we wondered if they are affected by incubation environment and thus whether laboratory-incubated cuttlefish exhibit natural behavior. We investigated the effects of incubation environment on hatchling size, defense and predation behavior in Sepia officinalis by comparing artificially-incubated hatchlings to naturally-incubated ones. Contrary to our expectations, no significant differences were apparent in hatchling size, disruptive body patterning and predation. A significant difference did exist between groups in one type of body patterning: Artificially-incubated hatchlings appeared to be better at producing a uniform body pattern than naturally-incubated individuals, possibly due to their prenatal experience with a homogeneous artificial substrate. This difference suggests some caution when interpreting experiments utilizing laboratory-incubated hatchlings, but overall, the limited effect of artificial incubation conditions demonstrated in this experiment bolsters confidence in previous behavioral results. These results are also promising for cuttlefish culture; eggs that would otherwise be lost as bycatch could instead be cultured artificially.

International audience; BACKGROUND:Binge drinking is popular and highly prevalent in teenagers. However, the long-term cognitive and neurobiological consequences of such practices are not yet fully understood. In this context, we therefore assessed in mice whether a chronic intermittent alcohol (CIA) exposure in adolescence had long-term consequences on object discrimination and memory performances, emotional behaviors, brain activity, and morphology.METHODS:C57BL/6JRj mice were treated with either saline or ethanol (EtOH) (2 g/kg/d, i.p., from postnatal days [PND] 30 to PND 44 every other day). The day following the last administration or later in adulthood (PND 71) mice were tested for different behavioral tests (novel object recognition, spontaneous alternation, light-dark box, elevated plus-maze, actimeter test), to assess object recognition, working memory performances, anxiety-like behavior, and locomotor activity. We also investigated neuronal activation of hippocampus, prefrontal and perirhinal cortices, and anatomical changes using immediate-early gene expression and longitudinal brain magnetic resonance imaging.RESULTS:Our results showed that adolescent mice exposed to CIA present a critical and persistent impairment of short-term object recognition performances. By contrast, spatial working memory was not impaired, nor was anxiety-like behavior. This altered object discrimination was associated with a biphasic change in neuronal activity in the hippocampus but without morphological changes. Indeed, c-Fos expression was specifically increased in the dorsal dentate gyrus (DG) of the hippocampus after the binge exposure, but then became significantly lower in adulthood both in the DG and the CA1 part of the hippocampus compared with adult saline pretreated mice.CONCLUSIONS:These findings provide evidence for adolescent vulnerability to the effects of intermittent binge EtOH exposure on object discrimination and hippocampal activity with long-lasting consequences.

90 pages, This paper is the result of an international initiative and is a first attempt to develop guidelines for the care and welfare of cephalopods (i.e. nautilus, cuttlefish, squid and octopus) following the inclusion of this Class of ∼700 known living invertebrate species in Directive 2010/63/EU. It aims to provide information for investigators, animal care committees, facility managers and animal care staff which will assist in improving both the care given to cephalopods, and the manner in which experimental procedures are carried out. Topics covered include: implications of the Directive for cephalopod research; project application requirements and the authorisation process; the application of the 3Rs principles; the need for harm-benefit assessment and severity classification. Guidelines and species-specific requirements are provided on: i. supply, capture and transport; ii. environmental characteristics and design of facilities (e.g. water quality control, lighting requirements, vibration/noise sensitivity); iii. accommodation and care (including tank design), animal handling, feeding and environmental enrichment; iv. assessment of health and welfare (e.g. monitoring biomarkers, physical and behavioural signs); v. approaches to severity assessment; vi. disease (causes, prevention and treatment); vii. scientific procedures, general anaesthesia and analgesia, methods of humane killing and confirmation of death. Sections covering risk assessment for operators and education and training requirements for carers, researchers and veterinarians are also included. Detailed aspects of care and welfare requirements for the main laboratory species currently used are summarised in Appendices. Knowledge gaps are highlighted to prompt research to enhance the evidence base for future revision of these guidelines., CephRes, AISAL and FELASA support this work. The world is three-dimensional; hence, even surface-bound animals need to learn vertical spatial information. Separate encoding of vertical and horizontal spatial information seems to be the common strategy regardless of the locomotory style of animals. However, a difference seems to exist in the way freely moving species, such as fish, learn and integrate spatial information as opposed to surface-bound species, which prioritize the horizontal dimension and encode it with a higher resolution. Thus, the locomotory style of an animal may shape how spatial information is learned and prioritized. An alternative hypothesis relates the preference for vertical information to the ability to sense hydrostatic pressure, a prominent cue unique to this dimension. Cuttlefish are mostly benthic animals, but they can move freely in a volume. Therefore, they present an optimal model to examine these hypotheses. We tested whether cuttlefish could separately recall the vertical and horizontal components of a learned two-dimensional target, and whether they have a preference for vertical or horizontal information. Sepia officinalis cuttlefish were trained to select one of two visual cues set along a 45 deg diagonal. The animals were then tested with the two visual cues arranged in a horizontal, vertical or opposite 45 deg configuration. We found that cuttlefish use vertical and horizontal spatial cues separately, and that they prefer vertical information to horizontal information. We propose that, as in fish, the availability of hydrostatic pressure, combined with the ecological value of vertical movements, determines the importance of vertical information.

WOS:000353599600013; International audience; A common trait of numerous memory disorders is the impairment of episodic memory. Episodic memory is a delay-dependant memory, especially associating three components, the "what", "where" and "when" of a unique event. To investigate underlying mechanisms of such memory, several tests, mainly based on object exploration behaviour, have been set up in rodents. Recently, a three-trial object recognition task has been proposed to evaluate simultaneously the different components of episodic-like memory in rodents. However, to date, the time course of each memory component in this paradigm is not known. We characterised here the time course of memory decay in adult mice during the three-trial object recognition task, with inter-trial interval (ITI) ranging from 1 h to 4 h. We found that, with 1 hand 2 h, but not 4h ITI, mice spent more time to explore the displaced "old object" relative to the displaced "recent object", reflecting memory for "what and when". Concomitantly, animals exhibited more exploration time for the displaced "old object" relative to the stationary "old object", reflecting memory for "what and where". These results provide strong evidence that mice establish an integrated memory for unique experience consisting of the "what", "where" and "when" that can persist until 2 h ITI. International audience; The effect of environment on the maturation of sand digging behaviour in cuttlefish was studied. Sand digging behaviour of cuttlefish individually reared on sand was daily observed in their rearing tanks (first study). Other cuttlefish were individually reared from hatching to 2 weeks of life in different conditions (Group A, on a sandy substrate and group B, without sand). At days 0, 3, 6, 9, 12 and 15, cuttlefish from Groups A and B were placed in a novel tank, the bottom of which was covered by sand (second study). The first study shows that more and more cuttlefish sand dig in their rearing tank during the first 6 days of life. The second study shows that, confronted with a novel sand bottom, cuttlefish from Group A show shorter latencies of sand digging and they cover more completely than do cuttlefish from Group B. This indicates that the developmental changes in sand digging appear not totally pre-programmed, but at least partially experience-dependent. Presence of sand in rearing tanks may allow cuttlefish to acquire experience of digging to make this behaviour more efficient.

WOS:000348100700001; International audience; Elderly persons often face biological, psychological or social changes over time that may cause discomfort or morbidity. While some cognitive domains remain stable over time, others undergo a decline. Spatial navigation is a complex cognitive function essential for independence, safety and quality of life. While egocentric (body centered) navigation is quite preserved during aging, allocentric (externally centered) navigation based on a cognitive map using distant landmarks declines with age. Recent preclinical studies showed that serotonergic 5-HT7 receptors are localized in brain regions associated with allocentric spatial navigation processing. Behavioral assessments with pharmacological or genetic tools have confirmed the role of 5-HT7 receptors in allocentric navigation. Moreover, few data suggested a selective age-related decrease in the expression of 5-HT7 receptors in pivotal brain structures implicated in allocentric navigation such as the hippocampal CA3 region. We aim to provide a short overview of the potential role of 5-HT7 receptors in spatial navigation, and to argue for their interests as therapeutic targets against age-related cognitive decline.

Iron dyshomeostasis is proving increasingly likely to be involved in the pathology of Alzheimer's disease (AD); yet, its mechanism is not well understood. Here, we investigated the AD-related mechanism(s) of iron-sulfate exposure in vitro and in vivo, using cultured primary cortical neurons and APP/PS1 AD-model mice, respectively. In both systems, we observed iron-induced disruptions of amyloid precursor protein (APP) processing, neuronal signaling, and cognitive behavior. Iron overload increased production of amyloidogenic KPI-APP and amyloid beta. Further, this APP misprocessing was blocked by MK-801 in vitro, suggesting the effect was N-methyl-D-aspartate receptor (NMDAR) dependent. Calcium imaging confirmed that 24 hours iron exposure led to disrupted synaptic signaling by augmenting GluN2B-containing NMDAR expression-GluN2B messenger RNA and protein levels were increased and promoting excessing extrasynaptic NMDAR signaling. The disrupted GluN2B expression was concurrent with diminished expression of the splicing factors, sc35 and hnRNPA1. In APP/PS1 mice, chronic iron treatment led to hastened progression of cognitive impairment with the novel object recognition discrimination index, revealing a deficit at the age of 4 months, concomitant with augmented GluN2B expression. Together, these data suggest iron-induced APP misprocessing and hastened cognitive decline occur through inordinate extrasynaptic NMDAR activation.

International audience; Antidepressants released in the environment have the potential to generate neural disrupting effects in non-target organisms, yet their putative effects on behaviors have never been studied in cephalopod molluscs. This study assessed the impact of the antidepressant fluoxetine (FLX) on the efficiency of cryptic behaviors (body patterns on uniform, checkerboard and sandy substrates), locomotor activity, and brain chemistry in young cuttlefish exposed to environmental concentrations (1 and 100 ng L−1 of FLX) during the perinatal period. Behavioral responses of cuttlefish were monitored at hatching and two weeks later, and brain monoamine contents were quantified at one month of age. FLX significantly altered the camouflage efficiencies on uniform and sandy backgrounds only at the lowest concentration, but not at 100 ng L−1. Hatchlings exposed to 1 ng L−1 of FLX exhibited a duration exposure-dependent decrease in the uniform camouflage. They also showed a significant increase of the frequency of sand digging behaviors which might make them highly visible to predators in nature. When tested again two weeks later, cuttlefish seemed to have recovered and no more behavioral alterations were observed showing a transitory effect of the antidepressant. FLX did not affect the levels of serotonin, norepinephrine and their metabolites; however, it seemed to influence dopaminergic activity between the two FLX-exposed groups. The results show for the time that environmentally realistic concentrations of a single SSRI significantly impair the cryptic performances of newly hatched cuttlefish, and may ultimately reduce their chance for survival.

In recent years, preclinical and clinical studies have generated considerable interest in the development of histamine H3 receptor (H3R) antagonists as novel treatment for degenerative disorders associated with impaired cholinergic function. To identify novel scaffolds for H3R antagonism, a common feature-based pharmacophore model was developed and used to screen the 17,194 compounds of the CERMN (Centre d'Etudes et de Recherche sur le Médicament de Normandie) chemical library. Out of 268 virtual hits which have been gathered in 34 clusters, we were particularly interested in tricyclic derivatives also exhibiting a potent 5HT4R affinity. Benzo[h][1,6]naphthyridine derivatives showed the highest H3R affinity, and compound 17 (H3R Ki = 41.6 nM; 5-HT4R Ki = 208 nM) completely reversed the amnesiant effect of scopolamine at 3 mg/kg in a spatial working memory experiment. For the first time we demonstrated the feasibility to combine H3R and 5-HT4R activities in a single molecule, raising the exciting possibility that dual H3R antagonist/5HT4R agonist have potential for the treatment of neurodegenerative diseases such as Alzheimer's disease.

International audience; When shown prawns in a glass tube, cuttlefish quickly learn to inhibit their predatory behavior. By using a visual learning paradigm, we studied training and retention performances of cuttlefish aged from 8 to 90 days. We found an improvement in the acquisition of learning abilities during the first 2 months of life as well as an increase of 24-hr retention performance between 30 and 90 days of age. Using morphometric measurements of different lobes of the central nervous system, we correlated the emergence of these learning abilities with the postembryonic development of related nervous structures. Our results show that only the growth of the superior frontal and vertical lobes appears to be significantly correlated with the improvement of learning and long-term retention performances. Thus, as found in earlier data collected in Octopus, the vertical lobe complex of the cuttlefish seems to be involved in these learning processes.

International audience; We appreciate the commentary made by Sumpter and Margiotta-Casaluci (in this issue) who are participating in the brainstorming around the effects of antidepressants on non-target aquatic organisms based on two recent articles published in Aquatic Toxicology, our paper Di Poi et al. (2013) and Franzellitti's article (Franzellitti et al., 2013).

A series of substituted 6-nitrophenylpropanamide derivatives (1-20) were synthesized using either the TDAE strategy or classical organic reactions. All these compounds were characterized by fusion point, (1)H NMR, (13)C NMR, elemental analysis or mass spectrometry data. Because of their structural analogy with recently published compounds possessing antinociceptive properties, our derivatives were screened for peripheral analgesic activities on acetic acid-induced writhing in mice. Compound 13 showed the best result at 100 μmol/kg ip (50% inhibition vs 59% for aspirin). This antinociceptive activity was maintained after oral administration (40% inhibition vs 31.6% for aspirin). Both hot-plate and actimetry-based tests were non-significant suggesting the analgesic activity of 13 linked to a peripheral mechanism.

International audience; From January 2013 scientific projects involving cephalopods became regulated by Directive 2010/63/EU, but at present there is little guidance specifically for cephalopods on a number of key requirements of the Directive, including: recognition of pain, suffering and distress and implementation of humane end-points; anaesthesia and analgesia, and humane killing. This paper critically reviews these key areas prior to the development of guidelines and makes recommendations, including identifying topics for further research. In particular: a) Evidence on how cephalopods might experience pain is reviewed; and a draft scheme of behavioural and physiological criteria for recognising and assessing pain, suffering and distress in cephalopods used in scientific procedures is presented and discussed. b) Agents and protocols currently used for general anaesthesia and analgesia are evaluated. Magnesium chloride, ethanol and clove oil are the most frequently used agents, but their efficacy and potential for induction of aversion need to be systematically investigated, according to the species of cephalopod and factors such as body weight, sex and water temperature. Means of sedating animals prior to anaesthesia should be investigated. Criteria for assessing depth of anaesthesia, including depression of ventilation, decrease in chromatophore tone (paling), reduced arm activity, tone and sucker adhesiveness, loss of normal posture and righting reflex, and loss of response to a noxious stimulus, are discussed. c) Analgesia should be provided for cephalopods used in scientific procedures, whenever this would be the case for vertebrates. However, research is needed to evaluate effective agents and administration routes for cephalopods. d) Techniques for local anaesthesia need to be defined and evaluated. e) Currently used methods of killing and criteria for confirmation of death in cephalopods are evaluated. Based on present knowledge, a protocol for humane killing of cephalopods is proposed. However, further evaluation is needed, along with development of humane methods of killing that will not compromise study of the brain. On humane grounds: i. mechanical (as opposed to chemical) methods of killing should not be used on conscious cephalopods (unless specifically authorised by the national competent authority); and ii. hatchlings and larvae should be killed by overdose of anaesthetic and not by immersion in tissue fixative. Key gaps in current knowledge are also highlighted, so as to encourage research that will contribute to the evidence base needed to develop guidelines to the Directive.

International audience; The use of polarization vision to improve visibility through turbid water has been proposed by different researchers, but has rarely been tested behaviorally. Cephalopods perceive the polarization of light but the function of this sensitivity is not fully understood yet. In this two-phase study, we examined the ability of cuttlefish to detect an artificial stimulus through water of different turbidity, with or without polarization information. We tested responses of Sepia prashadi and Sepia pharaonis to computer generated looming stimuli, presented through different levels of water turbidity. Stimuli were presented with a modified LCD monitor delivering polarized pattern that could not be detected without a polarization analyzer, and via a CRT monitor delivering an unpolarized, intensity contrasted image. The results showed that both cuttlefish can use their polarization vision to detect objects, such as a predator. Both species detected the pattern when it included a polarization component, through higher turbidity than when it contained only intensity information. Cuttlefish are nectobenthic organisms which probably often experience low visibility conditions. Therefore the ability to see further into turbid water and to better detect prey and predator would be beneficial for their survival.

International audience; This chapter summarizes the literature on the anatomical and functional organization of the cuttlefish brain, with a focus on the structures involved in learning and memory processes (namely the vertical lobe system and optic lobes). Also, different learning paradigms that are commonly used in Sepia officinalis are described with, when possible, their neural correlates. Recent work on the early development of brain and memory is also reviewed. Some research directions to follow in the field of neurobiology of learning and memory in cuttlefish are suggested to better understand the extraordinary behavioral plasticity of these sophisticated invertebrates.

International audience; Cuttlefish possess the complex ability to identify approaching threats and then to selectively express the appropriate defense. We examined the visual cues used by Sepia officinalis cuttlefish during predator detection and the responses they selected. Using computer-generated stimuli, we set out to quantitate the deimatic responses to artificial looming stimuli of different shapes and contrasts. Defensive behavior gradually intensified as geometrical shapes resembled an image of a fish. Therefore, in addition to an object's size or its sudden increase in size, cuttlefish use form recognition to identify a threat. Cuttlefish demonstrated equal performance in predator detection trough clear water when presented with intensity versus polarization contrasts. However, when the water turbidity increased, the cuttlefish still detected looming fish shapes based on polarization contrast even when intensity information alone did not suffice. These results demonstrate the interplay between intensity and polarization information transmission and processing in the spatial domain. As nectobenthic organisms, cuttlefish probably experience low visibility conditions on a regular basis. The ability to see further into turbid water and to better detect an approaching object would be beneficial for their survival.

International audience; Predation pressure acts on the behaviour and morphology of prey species. In fish, the degree of lateralization varies between high- and low-predation populations. While lateralization appears to be widespread in invertebrates, we do not know whether heredity and early experience interact during development as in vertebrates. Here we show, for the first time, that an exposure to predator odour prior to hatching modulates visual lateralization in newly hatched cuttlefish. Only cuttlefish that have been exposed to predator odour display a left-turning bias when tested with blank seawater in a T-shaped apparatus. Exposure to predator odour all the incubation long could appear as an acute predictor of a high-predation surrounding environment. In addition, cuttlefish of all groups display a left-turning preference when tested with predator odour in the apparatus. This suggests the ability of cuttlefish to innately recognize predator odour. To our knowledge, this is the first clear demonstration that lateralization is vulnerable to ecological challenges encountered during embryonic life, and that environmental stimulation of the embryo through the olfactory system could influence the development of subsequent visual lateralization.

SummaryPolarization sensitivity (PS) is a characteristic of the visual system of cephalopods. It has been well documented in adult cuttlefish which use PS in a large range of tasks such as communication, orientation and predation. Since cuttlefish do not benefit from parental care, their visual system and motion detection ability need to be efficient from hatching to enable them to detect prey or predators. We studied the maturation and functionality of polarization sensitivity in newly hatched cuttlefish. In a first experiment, we examined the response of juvenile cuttlefish from hatching to the age of one month, towards a moving, vertically-oriented grating (contrasting and polarized stripes) using an optomotor response apparatus. Cuttlefish showed differences in maturation of polarization vs. intensity contrast motion detection. In a second experiment, we examined the involvement of polarization information in prey preference and detection in cuttlefish of the same age. Cuttlefish chose preferentially not to attack transparent prey in which the polarization contrast has been removed with a depolarizing filter. Performances of prey detection based on intensity contrast improved with age. Polarization contrast can help cuttlefish detect transparent prey. Our results suggest that polarization is not a simple modulation of intensity information, but rather that it is processed as a distinct channel of visual information. Both intensity and polarization sensitivity are functional, though not fully matured, in newly hatched cuttlefish and seem to help in prey detection.

International audience; Inbred LOU/C/Jall rats are currently described as a model of successful aging. These rats have a longer healthy median lifespan than many other strains, do not develop obesity, diabetes, or tumor and more importantly they do not show cognitive decline with aging. This is the first study to examine gene expression changes in the inbred LOU/C/Jall rat hippocampus and frontal cortex. Microarray data from animals aged 5 and 26 months were compared to that obtained from the classical Wistar rat strain to potentially identify only the genes associated with successful aging. We have thus identified a set of 15 genes in the hippocampus and 70 genes in the frontal cortex that could be grouped into several clearly delineated clusters of highly correlated genes associated with a diversity of biological processes, including regulation of plasticity, inflammatory response, metabolic, catabolic and homeostatic processes, and transcription. Such a multiplicity of gene networks and diversity of biological functions were not observed in the Wistar rat strain. The gene expression profiles identified in aged the LOU/C/Jall rats' hippocampus and frontal cortex should be related to their intact cognitive abilities, such as those assessed through spontaneous alternation.

International audience; Recent ecotoxicological studies highlight the increasing presence of pharmaceuticals discharged in the aquatic environment. Amongst them is the antidepressant fluoxetine (FLX), a selective serotonin reuptake inhibitor, primarily indicated for treatment of depression. The effect of chronic exposure to FLX on memory processing in 1-month-old cuttlefish Sepia officinalis was evaluated. Three groups of new-borns were reared in different conditions: one control group (no FLX) and two groups exposed to environmental concentrations of FLX (1 and 100ng/L) from 15 days pre-hatching to 1 month post-hatching. Acquisition and retention performances were assessed using the 'prawn-in-the-tube' procedure. Perinatal exposure to fluoxetine led to significant changes in memory processing of the animals. The lowest observed effect concentration of this antidepressant on learning and retention was 1ng/L which is under the range of environmental contamination. Cuttlefish exposed at low FLX concentration had impaired acquisition capabilities and animals exposed at high FLX concentration displayed a deficit of memory retention compared to the control group that had nonimpaired initial acquisition and retention performances. The results subsequently suggested that FLX-induced changes in cognitive capacities could potentially lead to inappropriate predatory behaviors in the natural environment. The study provides the basis for future studies on how pharmaceutical contaminants disrupt cognition in ecologically and economically relevant marine invertebrates.

International audience; The common cuttlefish, Sepia officinalis (cephalopod mollusc) has recently become a relevant model for studying the setting-up of brain asymmetry among invertebrates. As the animals age from 3 to 30 days post hatching, they progressively develop a left-turning bias resulting from an eye-use preference. The aim of this study is to investigate whether anatomical (vertical, peduncle, inferior buccal, and optic lobes) or neurochemical (monoamines in optic lobes) brain asymmetries are present in the cuttlefish brain at 3 or at 30 post hatching days; and whether these correlate with side-turning preferences. We here find brain and behavioral asymmetry only at 30 post hatching days. Cuttlefish displayed a significant population bias towards a larger right peduncle lobe, and higher monoamine concentration in the left optic lobe (i.e. serotonin, dopamine and noradrenaline). None of these brain asymmetries were correlated to the studied side-turning bias. However, we found individual variation in the magnitude of the vertical and optic lobes asymmetry. A striking correlation was found with the behavioral results: the larger the right optic lobe and the right part of the vertical lobe, the stronger the bias to turn leftwards. To our knowledge, this is the first study to demonstrate a relationship at the individual level between brain and behavioral asymmetries in invertebrates.

International audience; RATIONALE: Blockade of 5-HT6 receptors (5-HT6R) is known to improve cognitive performances in the rodent. This improvement has been hypothesized to be the result, at least in part, of a modulation of the cholinergic neurotransmission. OBJECTIVE: We assessed the effects of 5-HT6R blockade on selected types of memory relevant to functional deficits of ageing and neurodegenerative diseases, in mice that present a scopolamine-induced cholinergic disruption of memory. METHOD: Following the selection of an adequate dose of scopolamine to induce cognitive deficits, we have studied the effects of the selective 5-HT6R antagonist SB-271046, alone or in combination with scopolamine, on working memory (spontaneous alternation task in the T-maze), recognition memory (place recognition) and aversive learning (passive avoidance). RESULTS: SB-271046 alone failed to affect working memory, recognition memory and aversive learning performances. In contrast, SB-271046 was able to reverse the scopolamine-induced deficits in working memory (only at 30 mg kg⁻¹) and those of acquisition and retrieval of aversive learning (dose-dependent effect); scopolamine-induced deficits in episodic-like memory (acquisition and retrieval) were partially counteracted by 5-HT6R blockade. CONCLUSION: The modulation between 5-HT6R and the cholinergic system appears to be predominant for working memory and aversive learning, but not for other types of memory (i.e. episodic-like memory). Interactions between 5-HT6R and alternative neurotransmission systems (i.e. glutamatergic system) should be further studied. The respective involvement of these interactions in the memory disorders related to ageing and neurodegenerative diseases is of pivotal importance regarding the possible use of 5-HT6R antagonists in the treatment of memory disorders in humans.

International audience; Cuttlefish rapidly change their appearance in order to camouflage on a given background in response to visual parameters, giving us access to their visual perception. Recently, it was shown that isolated edge information is sufficient to elicit a body pattern very similar to that used when a whole object is present. Here, we examined contour completion in cuttlefish by assaying body pattern responses to artificial backgrounds of 'objects' formed from fragmented circles, these same fragments rotated on their axis, and with the fragments scattered over the background, as well as positive (full circles) and negative (homogenous background) controls. The animals displayed similar responses to the full and fragmented circles, but used a different body pattern in response to the rotated and scattered fragments. This suggests that they completed the broken circles and recognized them as whole objects, whereas rotated and scattered fragments were instead interpreted as small, individual objects in their own right. We discuss our findings in the context of achieving accurate camouflage in the benthic shallow-water environment.

International audience; The behavioural lateralization observed in several invertebrate species appears to parallel the phenomenon seen in vertebrates. However, it is not yet clear whether lateralization in invertebrates varies within an individual across development as it does in vertebrates. We examined when during postembryonic development juvenile cuttlefish begin to show side-turning preferences in a T-shaped apparatus. We also determined whether or not the direction of turning was stimulus dependent by providing, or not providing, shelters in the two choice arms of the apparatus. We found that as the animals aged from 3 to 45 days posthatch they progressively developed a left-turning bias but only when shelters were provided in the apparatus. We suggest that left-turning biases in cuttlefish result from an eye use preference. Emergence of visual lateralization at the time of dispersal may afford juveniles greater behavioural efficiency by enabling them to look out for escape routes while hunting. The left visual field may be associated with rapid responses as in many species of vertebrates. Determining the role of the right visual field of cuttlefish when looking for prey may provide evidence for homology between lateralization in invertebrates and vertebrates.

International audience; RATIONALE: Selective serotonin reuptake inhibitors (SSRIs) such as fluoxetine are increasingly used for the treatment of depression in children. Limited data are, however, available on their effects on brain development and their efficacy remains debated. Moreover, previous experimental studies are seriously hampered in their clinical relevance. OBJECTIVES: The aim of the present study was to investigate putative age-related effects of a chronic treatment with fluoxetine (5 mg/kg, either orally or i.p. for 3 weeks, 1 week washout) using conventional methods (behavioral testing and binding assay using [(123)I]β-CIT) and a novel magnetic resonance imaging (MRI) approach. METHODS: Behavior was assessed, as well as serotonin transporter (SERT) availability and function through ex vivo binding assays and in vivo pharmacological MRI (phMRI) with an acute fluoxetine challenge (10 mg/kg oral or 5 mg/kg i.v.) in adolescent and adult rats. RESULTS: Fluoxetine caused an increase in anxiety-like behavior in treated adult, but not adolescent, rats. On the binding assays, we observed increased SERT densities in most cortical brain regions and hypothalamus in adolescent, but not adult, treated rats. Finally, reductions in brain activation were observed with phMRI following treatment, in both adult and adolescent treated animals. CONCLUSION: Collectively, our data indicate that the short-term effects of fluoxetine on the 5-HT system may be age-dependent. These findings could reflect structural and functional rearrangements in the developing brain that do not occur in the matured rat brain. phMRI possibly will be well suited to study this important issue in the pediatric population.

International audience; Whereas severe hypoxia is known to contribute to neuronal death and to lead to neurological disturbances, mild hypoxia can also induce beneficial effects through endogenous adaptive responses. The aim of this study was to investigate the effects of mild hypoxia (8% O(2)) on cognitive and emotional behavior in the adult mouse. To this end, mice were submitted to repeated mild hypoxia exposure or normoxia during 6 weeks and underwent behavioral testing during the last 3 weeks. Hypoxia decreased anxiety-like behavior in the light/dark transition test, enhanced, albeit modestly, non-spatial recognition memory, but did not alter spontaneous locomotor activity, nor spatial learning. On additional mice, whole brain adrenomedullin mRNA expression was found to be increased at D1, D25 and D41 after hypoxia initiation and vascular endothelial growth factor (VEGF) mRNA expression was increased at only on D41. This work shows that repeated mild hypoxic exposure decreases anxiety-related behavior and points out hypoxia inducible factor-1 (HIF-1) target genes, particularly adrenomedullin, as potential mediator candidate.

International audience; Environmental enrichment is known to improve learning and memory in adult rodents. Whereas the morphological changes underlying these beneficial effects are well documented, few studies have addressed the influence of this housing condition on the neuronal networks underlying memory processes. We assessed the effects of environmental enrichment on behavioural performances and brain metabolic activation during a memory task in mice. Adult mice were housed in standard (SC) or enriched (EC) conditions for 3 weeks. Then, recent and remote memory performances were measured in the passive avoidance test. After testing, brain metabolic activation was assessed through cytochrome oxidase (CO) activity. EC improved recent memory, in association with an increased metabolic activation in the frontal and prefrontal cortices and a decreased activation in the baso-lateral amygdala and the hippocampus. EC did not improve remote memory, and globally decreased CO activity. Our findings suggest the involvement of regions of pivotal importance during recent memory, such as the frontal cortex, in the beneficial effects of EC.

International audience; BACKGROUND: We aimed to further understand the relationship between cAMP concentration and mnesic performance. METHODS AND FINDINGS: Rats were injected with milrinone (PDE3 inhibitor, 0.3 mg/kg, i.p.), rolipram (PDE4 inhibitor, 0.3 mg/kg, i.p.) and/or the selective 5-HT4R agonist RS 67333 (1 mg/kg, i.p.) before testing in the object recognition paradigm. Cyclic AMP concentrations were measured in brain structures linked to episodic-like memory (i.e. hippocampus, prefrontal and perirhinal cortices) before or after either the sample or the testing phase. Except in the hippocampus of rolipram treated-rats, all treatment increased cAMP levels in each brain sub-region studied before the sample phase. After the sample phase, cAMP levels were significantly increased in hippocampus (1.8 fold), prefrontal (1.3 fold) and perirhinal (1.3 fold) cortices from controls rat while decreased in prefrontal cortex (∼0.83 to 0.62 fold) from drug-treated rats (except for milrinone+RS 67333 treatment). After the testing phase, cAMP concentrations were still increased in both the hippocampus (2.76 fold) and the perirhinal cortex (2.1 fold) from controls animals. Minor increase were reported in hippocampus and perirhinal cortex from both rolipram (respectively, 1.44 fold and 1.70 fold) and milrinone (respectively 1.46 fold and 1.56 fold)-treated rat. Following the paradigm, cAMP levels were significantly lower in the hippocampus, prefrontal and perirhinal cortices from drug-treated rat when compared to controls animals, however, only drug-treated rats spent longer time exploring the novel object during the testing phase (inter-phase interval of 4 h). CONCLUSIONS: Our results strongly suggest that a "pre-sample" early increase in cAMP levels followed by a specific lowering of cAMP concentrations in each brain sub-region linked to the object recognition paradigm support learning efficacy after a middle-term delay.

SUMMARYIt is now well established that prenatal sensory experience affects development itself and has long-term consequences in terms of postnatal behavior. This study focused on the functionality of the sensory system in cuttlefish in ovo. Embryos of stage 23, 25 and 30 received a tactile, chemical or visual stimulus. An increase of mantle contraction rhythm was taken to indicate a behavioral response to the stimulus. We clearly demonstrated that tactile and chemical systems are functional from stage 23, whereas the visual system is functional only from stage 25. At stage 25 and 30, embryos were also exposed to a repeated light stimulus. Stage 30 embryos were capable of habituation, showing a progressive decrease in contractions across stimulations. This process was not due to fatigue as we observed response recovery after a dishabituation tactile stimulus. This study is the first to show that cuttlefish embryos behaviorally respond to stimuli of different modalities and that the visual system is the last to become functional during embryonic development, as in vertebrate embryos. It also provides new evidence that the memory system develops in ovo in cuttlefish.

International audience; Hypoxic-ischemic (HI) brain injury occurring during the perinatal period is still a major cause of mortality and morbidity. We assessed the impact of maternal hypertension, the most common medical disorder of pregnancy, on the anatomical and functional consequences of HI insult in the immature brain. Rat pups from spontaneously hypertensive (SHR) and normotensive (Wistar Kyoto - WKY) dams were subjected to HI brain damage at post-natal day 7 (P7). Brain lesion and functional deficits were analyzed from 10 min to 35 days after HI, using magnetic resonance imaging (MRI), sensorimotor and cognitive tests. MRI data revealed that SHR pups displayed less brain damage than WKY, attested by an initial smaller lesion followed by a reduced tissue loss at chronic stage (57.1±21.6 and 31.1±27% ipsilateral hemisphere atrophy in WKY and SHR, respectively). Behavioral analyses showed less HI-induced behavioral deficits in motor coordination (rotarod test) and spatial learning (Morris water maze test) in pups from hypertensive dams compared to those from normotensive ones. The data suggest that maternal hypertension causes prenatal stress that may render the immature brain more resistant to subsequent hypoxia-ischemia, related to a preconditioning phenomenon.

International audience; Although environmental enrichment is well known to improve learning and memory in rodents, the underlying neuronal networks' plasticity remains poorly described. Modifications of the brain activation pattern by enriched condition (EC), especially in the frontal cortex and the baso-lateral amygdala, have been reported during an aversive memory task in rodents. The aims of our study were to examine 1) whether EC modulates episodic-like memory in an object recognition task and 2) whether EC modulates the task-induced neuronal networks. To this end, adult male mice were housed either in standard condition (SC) or in EC for three weeks before behavioral experiments (n = 12/group). Memory performances were examined in an object recognition task performed in a Y-maze with a 2-hour or 24-hour delay between presentation and test (inter-session intervals, ISI). To characterize the mechanisms underlying the promnesiant effect of EC, the brain activation profile was assessed after either the presentation or the test sessions using immunohistochemical techniques with c-Fos as a neuronal activation marker. EC did not modulate memory performances after a 2 h-ISI, but extended object recognition memory to a 24 h-ISI. In contrast, SC mice did not discriminate the novel object at this ISI. Compared to SC mice, no activation related to the presentation session was found in selected brain regions of EC mice (in particular, no effect was found in the hippocampus and the perirhinal cortex and a reduced activation was found in the baso-lateral amygdala). On the other hand, an activation of the hippocampus and the infralimbic cortex was observed after the test session for EC, but not SC mice. These results suggest that the persistence of object recognition memory in EC could be related to a reorganization of neuronal networks occurring as early as the memory encoding.

International audience; Facing inefficiency of current treatments to cure Alzheimer disease (AD), a pharmacological approach is now emerging on the assumption that a single compound may be able to hit multiple targets, namely Multi-Target-Directed Ligands (MTDLs). Displaying numerous advantages, several MTDL for AD have been recently described but none associating an inhibition of AChE and an activation of 5-HT(4)R. The aim of this study was to validate the concept of a synergistic action of these two targets on episodic-like memory performances in mice. Among potent molecules, RS67333, a reference 5-HT(4)R agonist and donepezil (DNPZ), a reference acetylcholinesterase inhibitor, have been particularly chosen because of their close chemical structure. Administered separately, RS67333 (0.3 and 1mg/kg) and DNPZ (1mg/kg) improved recognition performances compared to saline treated animals but not with lower doses. Co-administration of subactive doses of RS67333 (0.1mg/kg) and DNPZ (0.3mg/kg) improved memory, moreover, this improvement is prevented if a 5-HT(4)R antagonist (GR125487, 10mg/kg) is also administered. Activation of 5-HT(4)R combined with inhibition of AChE with subactive doses of RS67333 and of DNPZ has synergistic effects on memory performances in mice. These molecules having close chemical structures, the synergistic effect of their combination affords new hope to chemist for the synthesis of MTDL.

International audience; BACKGROUND: To what extent are the properties of neuronal networks constrained by computational considerations? Comparative analysis of the vertical lobe (VL) system, a brain structure involved in learning and memory, in two phylogenetically close cephalopod mollusks, Octopus vulgaris and the cuttlefish Sepia officinalis, provides a surprising answer to this question. RESULTS: We show that in both the octopus and the cuttlefish the VL is characterized by the same simple fan-out fan-in connectivity architecture, composed of the same three neuron types. Yet, the sites of short- and long-term synaptic plasticity and neuromodulation are different. In the octopus, synaptic plasticity occurs at the fan-out glutamatergic synaptic layer, whereas in the cuttlefish plasticity is found at the fan-in cholinergic synaptic layer. CONCLUSIONS: Does this dramatic difference in physiology imply a difference in function? Not necessarily. We show that the physiological properties of the VL neurons, particularly the linear input-output relations of the intermediate layer neurons, allow the two different networks to perform the same computation. The convergence of different networks to the same computational capacity indicates that it is the computation, not the specific properties of the network, that is self-organized or selected for by evolutionary pressure.

International audience; In terms of visual perception, coral reefs are structurally complex habitats. Therefore, visual stimuli that invoke territorial behavior in fish, causing them to respond to potential intruders and competitors, may be fragmented. Amodal completion was recently shown in a fish species. Here, we presented a mirror covered by occluders, with different squared openings arranged in a vertical or a horizontal broken line or a checkerboard formation, within their natural territories in the reef. Single small openings in the occluders did not trigger an attack on the reflected image, while specific configurations of them did so, suggesting that fish recognized an intruder in the fragmented image. The results suggest the use of amodal completion by fish although other possibilities cannot be rejected.

International audience; Erythropoietin (EPO), a well known haematopoietic growth factor, possesses neuroprotective and neurotrophic effects which have been recently reported to improve cognition and to modulate emotional processing. We investigated the effects of EPO and of its non-erythropoietic carbamylated derivative (CEPO) on memory- and emotion-related behaviour in the adult mouse. Locomotor activity, memory performances (place and object recognition tasks), anxiety- (light/dark transition test) and despair-like behaviours (tail suspension test) were assessed over 6 weeks of repeated EPO or CEPO administration (40 μg/kg, twice a week). Given the potential involvement of hippocampal neurogenesis in memory, we also assessed the effects of EPO and CEPO on neurogenesis in the dentate gyrus. Both treatments improved spatial and non-spatial recognition memory and increased the number of NeuN/BrdU double-labeled cells in the dentate gyrus. These effects seem to be, at least partly, independent from an haematopoietic action since administration of CEPO leads to the similar results. Moreover, CEPO decreased, albeit modestly, despair-related behaviour and tended to decrease anxiety-like behaviour. These results suggest that CEPO is as an attractive molecule for the treatment of neuropsychiatric diseases associating memory and/or emotional disorders.

International audience; Environmental factors are involved to various degrees in psychiatric diseases. Unfortunately, early-life manipulations have been rarely investigated in mice. Interestingly, given the advances in genetics, combination of environmental and genetic factors to get construct validity is now possible. Herein, spontaneous activity, anxiety-like behaviour, social behaviour and short term spatial working memory were assessed in mice after maternal separation or social isolation. Of note, social withdrawal was observed in both models suggesting that this aspect needs to be better considered in future studies, particularly in testing new treatments for schizophrenia.

International audience; Newly-hatched cuttlefish can use the same defensive behavior as adults: camouflage and burying. One alternative method involves the use of shelters to avoid potential predators. This behavior, though frequently observed in hatchlings and juveniles, has never been studied in cuttlefish. Here, we shall address the importance of embryonic visual experience on the choice of shelters in hatchlings. For this, we incubated eggs in three different visual environments: (i) with white shelters, (ii) with black shelters, (iii) without shelters. After hatching, the cuttlefish were individually placed in a tank containing both black and white shelters. Both naive cuttlefish and those exposed to black shelters prior to hatching preferred hiding under the black shelters. The cuttlefish exposed to white shelters prior to hatching did not show any preference in shelter color. This study is the first to report the use of shelters in hatchling cuttlefish, their preference regarding black or white shelters, and the influence of embryonic exposure on this preference. Behavioral functions of this kind of embryonic learning are discussed.

International audience; Developmental studies have shown that environmental stimulation received by a developing sensory system can alter the developmental outcome of both that sensory system and other aspects of the nervous system. We investigated the ecologically relevant question of whether prior exposure to prey early in development within one sensory modality could influence later prey choice within a different sensory modality. Cuttlefish are visual predators; they can detect prey odors but attacks on prey cannot be elicited without visual stimulation. Cuttlefish eggs were exposed to the odor of shrimp (preferred prey), crabs (non-preferred prey), mollusks (non-prey), or a seawater control (no prey). Seven days after hatching, prey preferences were tested with a visual choice test between crabs and shrimp. Hatchlings exposed to crabs odors and the seawater control were significantly more likely to attack shrimp. Hatchlings exposed to mollusk odors showed no visual prey preference, while those exposed to shrimp preferentially attacked crabs. These results demonstrate a complex relationship between an early sensory exposure and later prey preference

International audience; BACKGROUND AND PURPOSE: Human and rat 5-HT(7) receptors were studied with a particular emphasis on the molecular interactions involved in ligand binding, searching for an explanation to the interspecies selectivity observed for a set of compounds. We performed affinity studies, molecular modelling and site-directed mutagenesis, with special focus on residue Phe(7.38) of the human 5-HT(7) receptor [Cys(7.38) in rat]. EXPERIMENTAL APPROACH: Competition binding studies were performed for seven 5-HT(7) receptor ligands at three different 5-HT(7) receptors. The functional behaviour was evaluated by measuring 5-carboxytryptamine-stimulated cAMP production. Computational simulations were carried out to explore the structural bases in ligand binding observed for these compounds. KEY RESULTS: Competition experiments showed a remarkable selectivity for the human receptor when compared with the rat receptor. These results indicate that mutating Cys to Phe at position 7.38 profoundly affects the binding affinities at the 5-HT(7) receptor. Computational simulations provide a structural interpretation for this key finding. Pharmacological characterization of compounds mr25020, mr25040 and mr25053 revealed a competitive antagonistic behaviour. Compounds mr22423, mr22433, mr23284 and mr25052 behaved as partial agonists. CONCLUSIONS AND IMPLICATIONS: We propose that the interspecies difference in binding affinities observed for the compounds at human and rat 5-HT(7) receptors is due to the nature of the residue at position 7.38. Our molecular modelling simulations suggest that Phe(7.38) in the human receptor is integrated in the hydrophobic pocket in the central part of the binding site [Phe(6.51)-Phe(6.52)] and allows a tighter binding of the ligands when compared with the rat receptor.

International audience; We found that recombinant human erythropoietin (rhEPO) reduced significantly the development of brain edema in a rat model of diffuse traumatic brain injury (TBI) (impact-acceleration model). In this study, we investigated the molecular and intracellular changes potentially involved in these immediate effects. Brain tissue nitric oxide (NO) synthesis, phosphorylation level of two protein kinases (extracellular-regulated kinase (ERK)-1/-2 and Akt), and brain water content were measured 1 (H1) and 2 h (H2) after insult. Posttraumatic administration of rhEPO (5,000 IU/kg body weight, intravenously, 30 mins after injury) reduced TBI-induced upregulation of ERK phosphorylation, although it increased Akt phosphorylation at H1. These early molecular changes were associated with a reduction in brain NO synthesis at H1 and with an attenuation of brain edema at H2. Intraventricular administration of the ERK-1/-2 inhibitor, U0126, or the Akt inhibitor, LY294002, before injury showed that ERK was required for brain edema formation, and that rhEPO-induced reduction of edema could involve the ERK pathway. These results were obtained in the absence of any evidence of blood-brain barrier damage on contrast-enhanced magnetic resonance images. The findings of our study indicate that the anti edematous effect of rhEPO could be mediated through an early inhibition of ERK phosphorylation after diffuse TBI.
