Abstract

With global increases in anthropogenic pressures on wildlife populations comes a responsibility to manage them effectively. The assessment of marine ecosystem health is challenging and often relies on monitoring indicator species, such as cetaceans. Most cetaceans are however highly mobile and spend the majority of their time hidden from direct view, resulting in uncertainty on even the most basic population metrics. Here, we discuss the value of long-term and internationally combined stranding records as a valuable source of information on the demographic and mortality trends of the harbour porpoise (Phocoena phocoena) in the North Sea. We analysed stranding records (n = 16,181) from 1990 to 2017 and demonstrate a strong heterogeneous seasonal pattern of strandings throughout the North Sea, indicative of season-specific distribution or habitat use, and season-specific mortality. The annual incidence of strandings has increased since 1990, with a notable steeper rise particularly in the southern North Sea since 2005. A high density of neonatal strandings occurred specifically in the eastern North Sea, indicative of areas important for calving, and large numbers of juvenile males stranded in the southern parts, indicative of a population sink or reflecting higher male dispersion. These findings highlight the power of stranding records to detect potentially vulnerable population groups in time and space. This knowledge is vital for managers and can guide, for example, conservation measures such as the establishment of time-area -specific limits to potentially harmful human activities, aiming to reduce the number and intensity of human -wildlife conflicts.

Abstract

On July 4th 2013 a dead subadult female wolf-like canid was found by the roadside between Luttelgeest and Marknesse in the Noordoostpolder in the central part of the Netherlands. As the last observations of wild wolves in the Netherlands date back to 1869 the discovery of this animal generated a lot of media attention. European wolf populations have been expanding since the 1950s and the first packs recently established themselves in Germany in geographic proximity of the Dutch border, so natural re-appearance of the species in the Netherlands seemed likely. We investigated the taxonomy of the animal, its geographical origin, and its most recent history. Macroscopic and biochemical analyses of the dead animal convincingly showed that it was a purebred wolf, related to populations from eastern Europe. Bullet impacts and shattered fragments found in the chest and flank, and a discrepancy between the timing of the post mortem and rigor mortis intervals indicated that this wolf was shot prior to illegal transport to the Netherlands. The wolf fed on beaver in either the Carpathian mountains or the Eifel which is too far for the animal to have walked from by itself within the 24 hours needed to digest its last meal. These geographical areas are the only regions where haplotypes and 87Sr/86Sr isotopes retrieved from both the dead wolf and the beaver remains in its stomach co-occur. We therefore conclude that the first Dutch wolf found in the Netherlands in 150 years did not enter the Netherlands by itself but sadly proved to be the victim of wildlife crime. Ruim 125 jaar later, op 4 juli 2013, werd een dood subadult vrouwtje gevonden op de weg tussen Luttelgeest en Marknesse in de Noordoostpolder. DNA-analyse wees uit dat het om een raszuivere wolf ging van Oost-Europese afkomst. Het laatst genoten maal bestond uit een bever, die zeker alleen in de Eiffel of Karpaten afkomstig was (isotopen onderzoek). Beide gebieden liggen voor een wolf te ver van de Noordoostpolder vandaan om binnen een dag te belopen. Kogelfragmenten en lijkschouwing wijzen in de richting van afschot, gevolgd door illigaal transport naar Nederland.

Abstract

Tissue microarray (TMA) technology allows analysis of multiple tumour samples simultaneously on a single slide. The aim of the present study was to develop and assess a TMA containing 32 primary canine insulinomas and 13 insulinoma metastases. The results of histopathological and immunohistochemical analyses of triplicate core biopsies were compared with those of individual tissue sections using weighted k statistics. Inter-observer agreement of TMA immunohistochemistry scores were assessed for chromogranin A (CgA), insulin, growth hormone (GH), growth hormone receptor (GHR) and Ki67 index, as well as the prognostic utility of clinicopathological, histopathological and immunohistochemical criteria. There was substantial agreement of scores for histopathological parameters (κ = 0.64-0.70) and a substantial to near-perfect agreement for homogenous immunohistochemical parameters (κ = 0.69-1.00). Except for GH, which demonstrated heterogeneous staining, there was good to excellent inter-observer agreement for all other immunohistochemical staining scores (intra-class correlation coefficients: 0.70-1.00). On univariate analysis, the presence of nuclear atypia was significantly predictive of disease-free intervals (DFls) for canine insulinoma, while tumour size, TNM stage, necrosis and Ki67 index were significant in terms of prognosis, with respect to both DFI and survival time. On multivariate analysis, tumour size and Ki67 index retained predictive power for survival time, as did tumour size for DFI.
