- Abstract
The authors discovered a new population of Hieracium lucidumGuss., a species hitherto considered strictly endemic to Monte Gallo. Useful elements for the ecological characterisation of the new population are provided. The stretch of carbonate coastline between the Egadi Islands and the western portion of the Province of Palermo displays a very high wealth of valuable floristic elements and represents an unicum in terms of both phytogeography and ecology. Considering the extreme fragmentation of the distribution pattern of many endemic and exclusive species that characterise this district, as in the case of H. lucidum, the authors suggest upgrading the strategies to protect the local botanical heritage, going beyond the species approach and thinking on a wider territorial scale.

Colonization is crucial to habitat restoration projects that rely on the spontaneous regeneration of the original vegetation. However, as a previously declining plant species spreads again, the likelihood of founder effects increases through recurrent population founding and associated serial bottlenecks. We related Amplified Fragment Length Polymorphism markers genetic variation and fitness to colonization history for all extant populations of the outcrossing terrestrial orchid Dactylorhiza incarnata in an isolated coastal dune complex. Around 1970, D. incarnata suffered a severe bottleneck yet ultimately persisted and gradually spread throughout the spatially segregated dune slacks, aided by the restoration of an open vegetation. Genetic assignment demonstrated dispersal to vacant sites from few nearby extant populations and very limited inflow from outside the spatially isolated reserve. Results further indicated that recurrent founding from few local sources resulted in the loss of genetic diversity and promoted genetic divergence (F(ST) = 0.35) among populations, but did not influence population fitness. The few source populations initially available and the limited inflow of genes from outside the study reserve, as a consequence of habitat degradation and spatial isolation, may have magnified the genetic effects of recurrent population founding.

Olea europaea L. subsp. laperrinei (Oleaceae) is an endemic taxon of the mountainous regions of central Sahara, consisting of currently fragmented and small relict populations. The tree can propagate vegetatively or by seed, but no recent natural regeneration was observed in the Algerian massive populations, some of which are considered threatened with extinction. Sterile triploid individuals were also identified in some populations showing increasing vigour. As a result of its long persistence and despite its rarity, the Laperrine's olive is an iconic component of Saharan mountain ecosystems. The aim of this study is to develop an efficient micropropagation protocol for both diploid and triploid Laperrine's olive to safeguard and preserve this genetic resource. Best shoot propagation was obtained on a modified Murashige and Skoog medium supplemented with 9.2 μM zeatin. Best rooting rate of regenerated shoots was achieved on the same culture medium supplemented with 4.8 μM indole-3-butyric acid. In absence of morphological changes of in vitro regenerated plants acclimatized to the greenhouse, genetic conformity was assessed by simple sequence repeat screening. Our results suggest that in vitro propagation could be a useful tool for conservation of both diploid and triploid threatened Laperrine's olive. [ABSTRACT FROM AUTHOR]
