5. Asymptomatic Cryptosporidium infections in ewes and lambs are a source of environmental contamination with zoonotic genotypes of Cryptosporidium parvum

Abstract

Protozoan parasites of the Cryptosporidium genus cause severe cryptosporidiosis in newborn lambs. However, asymptomatic infections also occur frequently in lambs and ewes. In sheep, the most commonly detected Cryptosporidium species are C. ubiquitum, C. xiaoi and C. parvum. Due to a lack of relevant information about such infections in France, we investigated the situation on five dairy sheep farms in the Pyrénées-Atlantiques Department in south-western France in December 2017. Individual fecal samples were collected from 79 female lambs (5–17 days old) and their mothers (72 ewes). Oocysts were screened using Heine staining before and after Bailenger concentrations. Cryptosporidium species identification and genotyping were performed using real-time PCR and gp60 gene sequencing. No cases of clinical cryptosporidiosis were observed in the 79 lambs. Microscopically, Cryptosporidium spp. oocysts were observed in only one lamb on one farm (prevalence 1.3%) and one ewe on another farm (prevalence 1.4%). By contrast, Cryptosporidium spp. DNA was detected in 17 ewes (prevalence ranging from 10.5% to 50% depending on the farm) and in 36 lambs (prevalence ranging from 0% to 77.8% depending on the farm). Only zoonotic Cryptosporidium parvum IId and IIa genotypes were identified when genotyping was possible. Cryptosporidium ubiquitum and C. xiaoi were detected on one and three farms, respectively. We conclude that healthy young lambs and their mothers during the peripartum period could be a source of environmental contamination with oocysts.

Abstract

International audience; Breeding sheep for enhanced resistance to gastrointestinal parasites is a promising strategy to limit the use of anthelmintics as parasite resistance to these molecules is now widespread. This paper presents genetic parameters estimated for resistance and resilience traits in the Blond-faced Manech dairy sheep breed. Two datasets were used. First, rams of the selection scheme were experimentally infected twice with L3 larvae of Haemonchus contortus. Fecal egg counts (FEC) and packed cell volume (PCV) loss were measured 30 days after each infection. Secondly, FEC and PCV of naturally infected ewes were recorded in spring, summer and fall of 2015 and 2016 in farms (1 to 6 measures per ewe). Genetic parameters were estimated for each infection conditions but also between natural and artificial infections thanks to the pedigree connections between the two datasets. In experimental conditions, FEC of rams were moderately heritable in second infection (heritability of 0.35) and were strongly correlated to FEC in first infection (genetic correlation of 0.92). For the ewes in natural conditions, FEC heritability was 0.18. Finally, the genetic correlation between FEC of experimentally challenged rams and FEC of the naturally infected ewes was 0.56-0.84. Our results suggest that genetic selection for enhanced resistance to gastro-intestinal nematodes is feasible using FEC in experimental infection measured on rams to improve the parasitism resistance of ewes on farms. It is of interest for the breeders’ organization to measure FEC on rams in order to have standardized measures and allow a better diffusion of the genetic progress in farms.

Abstract

National audience; Face à l’apparition de multirésistances aux anthelminthiques chez les strongles digestifs des petits ruminants, il est urgent de mettre en application une lutte intégrée contre ces parasites. L’objectif est d’utiliser les traitements antiparasitaires actuels de façon raisonnée, aussi souvent que nécessaire mais aussi peu que possible. Une première étape consiste à identifier les molécules anthelminthiques encore efficaces dans une exploitation par un test FECRT simplifié. Puis traitements ciblés ou traitements sélectifs pourront être proposés. D’autres approches (champignons nématophages, plantes à effets anthelminthiques…) sont en cours d’étude pour limiter la contamination des pâturages. Les systèmes de pâturage eux-mêmes, qui ont un effet majeur sur les populations de strongles digestifs, font l’objet d’une attention toute particulière depuis peu (pâturage cellulaire). Enfin le renforcement de la résistance de l’hôte par la vaccination ou par la sélection génétique d’animaux résistants ouvre des perspectives intéressantes.

Abstract

International audience; Gastro-intestinal nematodes are of major concern to sheep health worldwide. Identifying the causative genes responsible for resistance would augment the efficiency of selection. Previously, back-cross (BC) sheep (Black Belly * Romane breeds) were genotyped with a 50K SNP chip and were measured for faecal egg count after two successive experimental challenges by H. contortus. The most significant QTL were identified on chromosomes 5, 12, 13, 21. A customized assay including 1000 SNP was created to increase the density ofmarker coverage in these regions and others major QTL regions affecting parasitism resistance in natural infection conditions. In the present study, 277 Romane lambs were experimentally challenged with H contortusand genotyped with the dedicated 1000 SNP. Six out of the eight tested QTL regions have an effect in the Romane pure Breed population.

Abstract

Gastrointestinal nematodes are one of the main health issues in sheep breeding. To identify loci affecting the resistance to Haemonchus contortus, a genome scan was carried out using 1,275 Romane × Martinik Black Belly backcross lambs. The entire population was challenged with Haemonchus contortus in 2 consecutive experimental infections, and fecal egg counts (FEC) and packed cell volumes were measured. A subgroup of 332 lambs with extreme FEC was necropsied to determine the total worm burden, length of female worms, sex ratio in the worm population, abomasal pH, and serum and mucosal G immunoglobulins (IgG) responses. Pepsinogen concentration was measured in another subset of 229 lambs. For QTL detection, 160 microsatellite markers were used as well as the Illumina OvineSNP50 BeadChip that provided 42,469 SNP markers after quality control. Linkage, association, and joint linkage and association analyses were performed with the QTLMAP software. Linkage disequilibrium (LD) was estimated within each pure breed, and association analyses were carried out either considering or not the breed origin of the haplotypes. Four QTL regions on sheep chromosomes (OAR)5, 12, 13, and 21 were identified as key players among many other QTL with small to moderate effects. A QTL on OAR21 affecting pepsinogen concentration exactly matched the pepsinogen (PGA5) locus. A 10-Mbp region affecting FEC after the 1st and 2nd infections was found on OAR12. The SNP markers outperformed microsatellites in the linkage analysis. Taking advantage of the LD helped to refine the locations of the QTL mapped on OAR5 and 13.

National audience; Les strongyloses gastro-intestinales occasionnent des pertes de production laitière et de la mortalité chez les petits ruminants laitiers. Les ovins laitiers de races Manech Tête Rousse (MTR), Manech Tête Noire (MTN) et Basco-Béarnaise (BB) sont soumis à de fortes pressions parasitaires car les conditions climatiques des PyrénéesAtlantiques sont très favorables à ces parasites. L’utilisation exclusive des Benzimidazoles (seuls anthelminthiques autorisés en lactation chez la brebis) a conduit à l’apparition de résistance dans les populations de strongles d’où la nécessité de proposer des méthodes alternatives aux traitements chimiques. La sélection génétique d’animaux résistants aux parasites est une approche prometteuse, le regroupement de béliers dans des centres d’élevage offrant l’opportunité d’effectuer cette sélection sur les futurs mâles d’insémination artificielle. Cet article fait le point sur la sélection sur phénotypes chez des béliers du Centre Ovin d’Ordiarp. Les jeunes béliers d’Ordiarp n’étant pas élevés au pâturage, des infestations expérimentales avec le nématode hématophage Haemonchus contortus ont été réalisées. Une première année a permis de tester différents protocoles d’épreuve et d’évaluer l’impact des infestations sur leurs paramètres hématologiques. L’intensité d’excrétion d’oeufs par gramme de fèces (OPG) et l’hématocrite ont été mesurés. La seconde année a consisté à éprouver tous les béliers MTR, MTN et BB d’un même millésime avec le protocole retenu. Ce protocole consiste en deux infestations expérimentales uniques, successives, interrompues par un traitement au bout de 30 jours et séparées de quinze jours. L’infestation des béliers lors de la seconde année a permis de générer une grande variabilité individuelle des excrétions d’oeufs dans les trois races en première infestation : 1120 +/- 700 (MTR, N = 103), 960 +/- 920 (MTN, N = 37), 550 +/- 590 (BB, N = 36) comme en seconde infestation : 970 +/- 825 (MTR), 800 +/- 740 (MTN), 990 +/- 1400 (BB). L’impact sur l’hématocrite était minime (pertes de 3 à 6% seulement selon les races). Une sélection sur phénotypes de la résistance aux strongles gastro-intestinaux semble réalisable en centre de béliers. Abstract

Protozoan parasites of the Cryptosporidiumgenus cause severe cryptosporidiosis in newborn lambs. However, asymptomatic infections also occur frequently in lambs and ewes. In sheep, the most commonly detected Cryptosporidiumspecies are C. ubiquitum, C. xiaoiand C. parvum.Due to a lack of relevant information about such infections in France, we investigated the situation on five dairy sheep farms in the Pyrénées-Atlantiques Department in south-western France in December 2017. Individual fecal samples were collected from 79 female lambs (5–17 days old) and their mothers (72 ewes). Oocysts were screened using Heine staining before and after Bailenger concentrations. Cryptosporidiumspecies identification and genotyping were performed using real-time PCR and gp60gene sequencing. No cases of clinical cryptosporidiosis were observed in the 79 lambs. Microscopically, Cryptosporidiumspp. oocysts were observed in only one lamb on one farm (prevalence 1.3%) and one ewe on another farm (prevalence 1.4%). By contrast, Cryptosporidiumspp. DNA was detected in 17 ewes (prevalence ranging from 10.5% to 50% depending on the farm) and in 36 lambs (prevalence ranging from 0% to 77.8% depending on the farm). Only zoonotic Cryptosporidium parvumIId and IIa genotypes were identified when genotyping was possible. Cryptosporidium ubiquitumand C. xiaoiwere detected on one and three farms, respectively. We conclude that healthy young lambs and their mothers during the peripartum period could be a source of environmental contamination with oocysts.

Abstract

Selection of resistant animals and host immunization have been proposed as alternative methods for the control of gastrointestinal nematode parasites. However, a better knowledge of the mechanisms involved in protective immunity against these parasites is required for the development of optimal strategies. In this study, 3 month old INRA 401 lambs (n = 81) were allocated into three groups: uninfected control, challenged either once (primary-infected animals) or twice (previously-infected animals) with 10 000 Haemonchus contortus L3. Uninfected control and challenged animals were sequentially sacrificed at 3, 7, 15 and 28 days post challenge. In both challenged groups, a clear Th2-oriented immune response was observed in the abomasal lymph node and mucosa. IL-4 and IL-13 mRNA over-expression, recruitment of eosinophils, mast cells and globule leukocytes and production of specific systemic IgG and mucosal IgA were observed earlier in previously-infected animals than in primary-infected ones. At 28 days post infection, no differences between intensities of these responses were observed between the challenged groups. Worm establishment rates were similar in previously-infected and primary-infected lambs. However, reductions of worm development, female fecundity and fecal egg output were observed in previously-infected sheep. We conclude that H. contortus infection in young INRA 401 lambs induced an unequivocal Th2 immune response resulting in the regulation of worm egg production without affecting their establishment.

47. Lack of Efficacy of Albendazole against Dicrocoelium dendriticum Infection in a Sheep Farm in France.

Abstract

Dicrocoeliosis is a common parasitic disease in European sheep farming. The prevalence of infection by this parasite can reach almost 70% in areas where the environment is favorable to intermediate hosts. In France, only one drug is currently available for the treatment of dicrocoeliosis: albendazole at a dose of 15 mg/kg in a single administration. However, a control coproscopy following a routine treatment led us to suspect that the efficacy of albendazole against Dicrocoelium dendriticum had diminished. Therefore, we carried out an efficacy test on 15 animals by treating them with albendazole at a dose of 15 mg/kg and performing a coproscopy on D0 and a control coproscopy 14 days later. We obtained a 39% reduction in the excretion of D. dendriticum eggs. This shows a reduction in the expected efficacy of albendazole, which is normally more than 90% in other studies involving this molecule at a dosage of 15 mg/kg. These results are of major concern as albendazole is currently the only drug available in France to treat dicrocoeliosis.

Abstract

Different authors have reported that eosinophils are capable of immobilising infective larvae of different species of nematodes in vitro. However, classifying larvae as mobile or immobile is so subjective that it does not always mean all apparently immobile larvae are dead or those that are mobile are capable of surviving further immune responses if administered to their natural hosts. The objective of this experimental study was therefore to substantiate the role of eosinophils in the killing of Haemonchus contortus infective larvae by comparing the infectivity in sheep of larvae that had been incubated with eosinophil-enriched cell suspensions with control larvae. Since it was not possible to isolate pure eosinophils from sheep blood, we were compelled to evaluate the effects of other blood cells contaminating our eosinophil-enriched suspensions. Although eosinophils and neutrophils were the only cells found adherent to H. contortus infective larvae in vitro, induced eosinophils in the presence of immune serum were primarily responsible for the drastic reduction in larval motility compared to the minor effects of neutrophils and mononuclear cells. Corresponding reductions in faecal egg count and worm numbers were observed when the incubated larvae were transferred intra-abomasally to sheep. Interestingly, the proportion of larvae that failed to establish was much higher following incubation with induced eosinophils compared with other cells or with immune serum alone. Although this study did not address the in vivo role of eosinophils in sheep, the results strongly indicate that sheep blood eosinophils have a larval killing potential in vitro, and a larval mobility test alone may not fully explain the level of damage inflicted on the larvae.
