Although Astrospartus mediterraneus (Risso, 1826) is an emblematic Mediterranean species, limited information is currently available, as it has not received much attention from the marine scientific community to date. In this context, an unusually high abundance of this basket star was observed in 2018 as part of the by-catch of local artisanal fishers in the Cap de Creus area (NW Mediterranean Sea). Indeed, fishers reported that this species had increased in abundance and expanded its distribution in recent years, ultimately interfering with their fishing activity. As such, this study aimed to elucidate the abundance, distribution, size, and structure of A. mediterraneus populations, as well as to evaluate the potential impact this species has on artisanal fisheries’ performance in the study area. To such aim, this work benefited from the Local Ecological Knowledge (LEK) of the fishers, by-catch analyses of regular fishing events and video transects recorded employing Remote Operated Vehicles (ROV). Results show that A. mediterraneus has a specific distribution in the study area, mainly driven by habitat requirements. High abundances of the basket star were associated with rocky substrates with the presence of gorgonians, located between 50 and 80 m depth and preferentially occurringon sloping areas. Despite the lack of long-time monitoring data, A. mediterraneus' aggregations recorded in the studied area are the densest ones so far reported for the whole Mediterranean Sea with peakvalues of 18 ind. m−2 and a mean density of 0.45 ind. m −2 ± 0.71 ind. m−2. Additionally, the average size of their central disks (2.67 ± 0.97 cm), suggests that this is rather young population, which could be possibly linked with the beginning of a massive outbreak, which would be in accordance with the fishers’ own experiences, as it seems apparent that the species has been on the rise in recent years. The genus Sebastes is a morphologically and ecologically diverse genus of rockfish characterized by high longevity, late-maturity and low natural mortality. On the northwest Atlantic continental shelf, the Acadian redfish (Sebastes fasciatus ) is the most common rockfish species above 300 m depth. This species has been widely exploited resulting in the depletion or collapse of most of its stocks. Management of long-lived species with intricate life-history characteristics is challenging and requires highly integrated biological and oceanographic monitoring, which allow the identification of environmental drivers and demographic and behavioral trends. The present study uses high-temporal resolution imaging and environmental data, acquired with an autonomous lander deployed for 10-months at the Sambro Bank Sponge Conservation Area (Scotian Shelf) to elucidate S. fasciatus temporal dynamics and behavioral trends in response to near-bed environmental conditions. S. fasciatus , mostly displayed passive locomotion and static behaviors, in common with other shelf-dwelling Sebastes species. Structural complexity provided by sponges positively influenced S. fasciatus presence. Fish used sponges to avoid being dragged by bottom currents. Hydrodynamics appear to act as a synchronizing factor conditioning its swimming behavior. S. fasciatus total counts exhibited a seasonal shift in rhythm’s phase likely reflecting changes in lifestyle requirements. This study provides new insights on S. fasciatus dynamics and behavior. Nonintrusive monitoring approaches, such as the one used in this study, will be key to monitor this threatened species populations. Especially, since it is expected that S. fasciatus will experience distribution shifts to higher latitudes due to future climate stressors.

A new species of sea anemone (Actiniaria, Metridioidea) is described and illustrated from cold-water coral environments at 600 m depth at the Blanes Canyon in the north-western Mediterranean Sea. Litophellia enoplosa sp. nov. is characterized by a column divisible into scapus and scapulus, the former with cuticle forming tenaculi, a mesogleal marginal sphincter muscle, more mesenteries proximally than distally arranged octomerously, and a single pair of directive mesenteries; it has strong retractor and parietobasilar muscles, and acontia with basitrichs and p-mastigophores B2a. Litophellia enoplosa sp. nov. is the second species described in the genus and it can be distinguished from the other species by the cuticle in the column, size ranges of the cnidae, and geographic and bathymetric distribution.

Deep-sea ecosystems of the Iberian margin have been widely impacted over the past decades, but the limited knowledge on their biodiversity and functioning limits our ability to contribute to their conservation. So far, in the Gulf of Cadiz, research has mostly been focused on megabenthic assemblages associated to mud volcanoes . However, several other geomorphological structures have remained widely unexplored. Here, by means of a quantitative analysis of 17 video transects conducted between 220 and 980 m depth, we investigated megabenthic assemblages associated to canyons, contouritic channels, contouritic furrows and open slopes. We report the presence of 8 different assemblages, segregated as a result of the different substrates and geomorphologic features. Megabenthic assemblages on hard substrates were characterized by mono or multispecific sponge assemblages. Soft bottoms hosted crinoid beds,pennatulacean meadows and fields of the gorgonian Radicipes gracilis. These results highlight the high diversity of megabenthic assemblages in deep-sea ecosystems of the Gulf of Cadiz and suggest that most of the geomorphological features that remained so far unexplored represent vulnerable marine ecosystems deser

The processes that control diversification and speciation in deep-sea species are poorly known. Here, we analyzed data produced by Restriction-Site Associated DNA Sequencing (RAD-Seq) of octocorals in the genus Paramuricea to elucidate diversification patterns andexamine the role of environmental gradients in their evolution. The genus Paramuricea evolved around 8 MYA, with a high probability of a broad ancestral depth range from mesophotic depths to the deep sea. At around 1-2 MYA, the genus diversified across the continental slope of the deep North Atlantic, supporting the depth-differentiation hypothesis, with no invasions back into shallower depths (< 200 m). Diversification in the deep sea generally occurred from shallower, warmer waters to deeper, colder depths of the lower continental slope. We also found that the vertical structure of water masses was influential in shaping phylogeographic patterns across the North Atlantic Ocean, with clades found in either upper/intermediate or intermediate/deep water masses. Our data suggest that species diverged first because of environmental conditions, including depth, temperature, and/or water mass, and then diversified into different geographical regions multiple times. Our results highlight therole of the environment in driving the evolution and distribution of Paramuricea throughout the deep sea. Furthermore, our study supports prior work showing the utility of genomic approaches over the conventionally-used DNA barcodes in octocoral species delimitation.

In addition, primary endpoints in kidney transplantation studies seek to demonstrate the short-term efficacy and safety of clinical interventions. Although such endpoints might demonstrate short-term improvement in specific aspects of graft function or incidence of rejection, such findings do not automatically translate into meaningful long-term graft survival benefits. Combining many factors into a well-validated model is therefore more likely to predict long-term outcome and better reflect the complexity of late graft failure than using single endpoints. If conditional marketing authorization could be considered for therapies that aim to improve long-term outcomes following kidney transplantation, then the surrogate endpoint for graft failure in clinical trial settings needs clearer definition. This Consensus Report considers the potential benefits and drawbacks of several candidate surrogate endpoints (including estimated glomerular filtration rate, proteinuria, histological lesions, and donor-specific anti-human leukocyte antigen antibodies) and composite scoring systems. The content was created from information prepared by a working group within the European Society for Organ Transplantation (ESOT). The group submitted a Broad Scientific Advice request to the European Medicines Agency (EMA), June 2020: the request focused on clinical trial design and endpoints in kidney transplantation. Following discussion and refinement, the EMA made final recommendations to ESOT in December 2020 regarding the potential to use surrogate endpoints in clinical studies that aim to improving late graft failure.

Kidney dysfunction is reflected in the glomerular filtration rate (GFR), and although a predefined (e.g., 50%) reduction in GFR was recommended as an endpoint by the European Medicines Agency (EMA) in 2016, many other endpoints are also included in clinical trials. End-stage renal disease is strongly associated with a change in estimated (e)GFR, and eGFR trajectories or slopes are increasingly used as endpoints in clinical intervention trials in chronic kidney disease (CKD). Similar approaches could be considered for clinical trials in kidney transplantation, although several factors should be taken into account. The present Consensus Report was developed from documentation produced by the European Society for Organ Transplantation (ESOT) as part of a Broad Scientific Advice request that ESOT submitted to the EMA in 2020. This paper provides a contemporary discussion of primary endpoints used in clinical trials involving CKD, including proteinuria and albuminuria, and evaluates the validity of these concepts as endpoints for clinical trials in kidney transplantation.

Late graft failure remains a serious, burdensome, and life-threatening condition for recipients. This article has been developed from content prepared by members of a working group within the European Society for Organ Transplantation (ESOT) for a Broad Scientific Advice request, submitted by ESOT to the European Medicines Agency (EMA), and reviewed by the EMA in 2020. The article presents the rationale for using surrogate endpoints in clinical trials aiming at improving late graft failure rates, to enable novel kidney transplantation therapies to be considered for CMA and improve access to medicines. The paper also provides background data to illustrate the relationship between primary and surrogate endpoints. Developing surrogate endpoints and a CMA strategy could be particularly beneficial for studies where the use of primary endpoints would yield insufficient statistical power or insufficient indication of long-term benefit following transplantation.

Despite cold-water coral (CWC) reefs being considered biodiversity hotspots, very little is known about the main processes driving their morphological development. Indeed, there is a considerable knowledge gap in quantitative experimental studies that help understand the interaction between reef morphology, near-bed hydrodynamics, coral growth, and (food) particle transport processes. In the present study, we performed a 2-month long flume experiment in which living coral nubbins were placed on a reef patch to determine the effect of a unidirectional flow on the growth and physiological condition of Lophelia pertusa. Measurements revealed how the presence of coral framework increased current speed and turbulence above the frontal part of the reef patch, while conditions immediately behind it were characterised by an almost stagnant flow and reduced turbulence. Owing to the higher current speeds that likely promoted a higher food encounter rate and intake of ions involved in the calcification process, the coral nubbins located on the upstream part of the reef presented a significantly enhanced average growth and a lower expression of stress-related enzymes than the downstream ones. Yet, further experiments would be needed to fully quantify how the variations in water hydrodynamics modify particle encounter and ion intake rates by coral nubbins located in different parts of a reef, and how such discrepancies may ultimately affect coral growth. Nonetheless, the results acquired here denote that a reef influenced by a unidirectional water flow would grow into the current: a pattern of reef development that coincides with that of actual coral reefs located in similar water flow settings. Ultimately, the results of this study suggest that at the local scale coral reef morphology has a direct effect on coral growth thus, indicating that the spatial patterns of living CWC colonies in reef patches are the result of spatial self-organisation.

The Norway lobster, Nephrops norvegicus, supports a key European fishery. Stock assessments for this species are mostly based on trawling and UnderWater TeleVision (UWTV) surveys. However, N. norvegicus are burrowing organisms and these survey methods are unable to sample or observe individuals in their burrows. To account for this, UWTV surveys generally assume that “1 burrow system = 1 animal”, due to the territorial behavior of N. norvegicus. Nevertheless, this assumption still requires in-situ validation. Here, we outline how to improve the accuracy of current stock assessments for N. norvegicus with novel ecological monitoring technologies, including: robotic fixed and mobile camera-platforms, telemetry, environmental DNA (eDNA), and Artificial Intelligence (AI). First, we outline the present status and threat for overexploitation in N. norvegicus stocks. Then, we discuss how the burrowing behavior of N. norvegicus biases current stock assessment methods. We propose that state-of-the-art stationary and mobile robotic platforms endowed with innovative sensors and complemented with AItools could be used to count both animals and burrows systems in-situ, as well as to provide key insights into burrowing behavior. Next, we illustrate how multiparametric monitoring can be incorporated into assessments of physiology and burrowing behavior. Finally, we develop a flowchart for the appropriate treatment of multiparametric biological and environmental data required to improve current stock assessment methods.

Sea pens (Octocorallia, Pennatulacea) are a specialized and morphologically distinct group of octocorals. The majority of them have adapted to survive on soft sediments with the help of an anchoring muscular peduncle. The whip-like sea pen Protoptilum carpenteri is considered a deep-sea North Atlantic species, which recently has been documented also in the Mediterranean Sea, where its actual distribution and abundance are still unknown. Even less is known about its ecology and behaviour, its reactions after disturbance, and its possible escape strategies. Several species of pennatulaceans can withdraw partially or completely into the sediment, following an apparent rhythmic but unsynchronized procedure that is usually preceded by the closure of the autozooids and the expulsion of the water contained within the colony. The present study reports and discusses forthe first time the extremely fast withdrawal behaviour of P. cf. carpenteri after in situ disturbance.

Cold-water coral (CWC) habitats dwell on continental shelves, slopes, seamounts, and ridge systems around the world’s oceans from 50 to 4000 m depth, providing heterogeneous habitats which support a myriad of associated fauna. These highly diverse ecosystems are threatened by human stressors such as fishing activities, gas and oil exploitation, and climate change. Since their life-history traits such as long lifespan and slow growth rates make CWCs very vulnerable to potential threats, it is a foremost challenge to explore the viability of restoration actions to enhance and speed up their recovery. In contrast to terrestrial and shallow-water marine ecosystems, ecological restoration in deep marine environments has received minimal attention. This review, by means of a systematic literature search, aims to identify CWC restoration challenges, assess the most suitable techniques to restore them, and discuss future perspectives. Outcomes from the few restoration actions performed to date on CWCs, which have lasted between 1 to 4 years, provide evidence of the feasibility of coral transplantation and artificial reef deployments. Scientific efforts should focus on testing novel and creative restoration techniques, especially to scale up to the spatial and temporal scales of impacts. There is still a general lack of knowledge about the biological, ecological and habitat characteristics of CWC species exploration of which would aid the development of effective restoration measures. To ensure the long-term viability and success of any restoration action it is essential to include holistic and long-term monitoring programs, and to ideally combine active restoration with natural spontaneous regeneration (i.e., passive restoration) strategies such as the implementation of deep-sea marine protected areas (MPAs). We conclude that a combination of passive and active restoration approaches with involvement of local society would be the best optimal option to achieve and ensure CW

Cold-water coral reefs (CWC) are known to be biodiversity hotspots, however, the sponge assemblages found to dwell within these habitats haven not been studied in depth to date in the Mediterranean Sea. The present article provides the first insight on the associated sponge fauna of the recently discovered CWC communities on the Catalan Margin and, to a lesser extent, the Cabliers Coral Mound Province, while also reviewing the current knowledge of the sponge fauna dwelling in all the Mediterranean CWC provinces. In regards to the studied areas, some rare species are cited forthe first time in the Mediterranean or redescribed, while two of them,Hamacantha (Hamacantha) hortae sp. nov. and Spongosorites cabliersi sp. nov. are new to science. At a basin scale, Mediterranean CWC appear as poriferan biodiversity hotspots, yet current diversity values on each site rather represent a small fraction of its actual fauna. Additionally, the existence of an endemic sponge fauna exclusively dwelling on CWC is refuted. Nonetheless, the sponge fauna thriving in Mediterranean CWC appears to be unique, and different from that of other Atlantic regions. Finally, with the current knowledge, the sponge fauna from the Mediterranean CWC is grouped in three distinguishable clusters (Alboran Sea, Western and Eastern Mediterranean), which appears to be determined by the basins water circulation, specially the Levantine Intermediate Water and the Atlantic Water following a western-eastern pattern from the Strait of Gibraltar to the Adriatic Sea. Overall, sponge living in Mediterranean CWC are still poorly explored in most areas, yet they appear to be good candidates for biogeographical studies.
