Most paleoclimate studies of Mainland Southeast Asia hydroclimate focus on the summer monsoon, with few studies investigating rainfall in other seasons. Here, we present a multiproxy stalagmite record (45,000 to 4,000 years) from central Vietnam, a region that receives most of its annual rainfall in autumn (September-November). We find evidence of a prolonged dry period spanning the last glacial maximum that is punctuated by an abrupt shift to wetter conditions during the deglaciation at ~14 ka. Paired with climate model simulations, we show that sea-level change drives autumn monsoon rainfall variability on glacial-orbital timescales. Consistent with the dry signal in the stalagmite record, climate model simulations reveal that lower glacial sea level exposes land in the Gulf of Tonkin and along the South China Shelf, reducing convection and moisture delivery to central Vietnam. When sea level rises and these landmasses flood at ~14 ka, moisture delivery to central Vietnam increases, causing an abrupt shift from dry to wet conditions. On millennial timescales, we find signatures of well-known Heinrich Stadials (HS) (dry conditions) and Dansgaard-Oeschger Events (wet conditions). Model simulations show that during the dry HS, changes in sea surface temperature related to meltwater forcing cause the formation of an anomalous anticyclone in the Western Pacific, which advects dry air across central Vietnam, decreasing autumn rainfall. Notably, sea level modulates the magnitude of millennial-scale dry and wet phases by muting dry events and enhancing wet events during periods of low sea level, highlighting the importance of this mechanism to autumn monsoon variability.

Trophic position is a fundamental characteristic of animals, yet it is unknown in many extinct species. In this study, we ground-truth the 15N/14N ratio of enameloid-bound organic matter (δ15NEB) as a trophic level proxy by comparison to dentin collagen δ15N and apply this method to the fossil record to reconstruct the trophic level of the megatooth sharks (genus Otodus). These sharks evolved in the Cenozoic, culminating in Otodus megalodon, a shark with a maximum body size of more than 15 m, which went extinct 3.5 million years ago. Very high δ15NEB values (22.9 ± 4.4‰) of O. megalodon from the Miocene and Pliocene show that it occupied a higher trophic level than is known for any marine species, extinct or extant. δ15NEB also indicates a dietary shift in sharks of the megatooth lineage as they evolved toward the gigantic O. megalodon, with the highest trophic level apparently reached earlier than peak size.

The interpretation of palaeoclimate archives based on oxygen isotopes depends critically on a detailed understanding of processes controlling the isotopic composition of precipitation. In the summer monsoonal realm, like Southeast Asia, seasonally and interannually depleted oxygen isotope ratios in precipitation have been linked to the summer monsoon strength. However, in some regions, such as central Vietnam, the majority of precipitation falls outside the summer monsoon period. We investigate processes controlling stable isotopes in precipitation from central Vietnam by combining moisture uptake calculations with monthly stable isotope data observed over five years. We find that the isotopic seasonal cycle in this region is driven by a shift in moisture source from the Indian Ocean to the South China Sea. This shift is reflected in oxygen isotope ratios with low values (- 8 to - 10‰) during summer and high values during spring/winter (0 to - 3‰), while 70% of the annual rainfall occurs during autumn. Interannual changes in precipitation isotopes in central Vietnam are governed by the timing of the seasonal onset and withdrawal of the Intertropical Convergence Zone, which controls the amount of vapour contributed from each source.

Between 5 and 4 thousand years ago, crippling megadroughts led to the disruption of ancient civilizations across parts of Africa and Asia, yet the extent of these climate extremes in mainland Southeast Asia (MSEA) has never been defined. This is despite archeological evidence showing a shift in human settlement patterns across the region during this period. We report evidence from stalagmite climate records indicating a major decrease of monsoon rainfall in MSEA during the mid- to late Holocene, coincident with African monsoon failure during the end of the Green Sahara. Through a set of modeling experiments, we show that reduced vegetation and increased dust loads during the Green Sahara termination shifted the Walker circulation eastward and cooled the Indian Ocean, causing a reduction in monsoon rainfall in MSEA. Our results indicate that vegetation-dust climate feedbacks from Sahara drying may have been the catalyst for societal shifts in MSEA via ocean-atmospheric teleconnections.

Speleothem oxygen isotope records have revolutionized our understanding of the paleo East Asian monsoon, yet there is fundamental disagreement on what they represent in terms of the hydroclimate changes. We report a multiproxy speleothem record of monsoon evolution during the last deglaciation from the middle Yangtze region, which indicates a wetter central eastern China during North Atlantic cooling episodes, despite the oxygen isotopic record suggesting a weaker monsoon. We show that this apparent contradiction can be resolved if the changes are interpreted as a lengthening of the Meiyu rains and shortened post-Meiyu stage, in accordance with a recent hypothesis. Model simulations support this interpretation and further reveal the role of the westerlies in communicating the North Atlantic influence to the East Asian climate.

The late Neogene megatooth shark, Otodus megalodon (Lamniformes: Otodontidae), is mostly known for its gigantic teeth and vertebrae. Re-examination of the rock matrix surrounding a previously described associated tooth set of O. megalodon from the upper Miocene of Japan resulted in the observation of numerous fragments of tessellated calcified cartilage and placoid scales. The morphology of each tessera and the arrangement of overall tessellated calcified cartilage are practically identical to those of extant chondrichthyans. Many placoid scales possess pronounced, rather broadly-spaced keels. A quantitative relationship between interkeel distances of keeled scales and reported cruising speeds across extant pelagic lamniforms and carcharhiniforms suggests that O. megalodon with a representative interkeel distance of ca. 100 µm was not a fast swimmer. We propose that O. megalodon was generally a slow cruising shark with occasional burst swimming for prey capture, where much of its metabolic heat through regional endothermy was possibly used to facilitate the digestion of large pieces of ingested meat as well as absorbing and processing nutrients. If so, the relative importance of the functional roles of regional endothermy possibly shifted from maintaining high cruising speeds to visceral food processing through the evolution towards gigantism in otodontids. Interdecadal modes of tropical Pacific ocean-atmosphere circulation have a strong influence on global temperature, yet the extent to which these phenomena influence global climate on multicentury timescales is still poorly known. Here we present a 2,000-year, multiproxy reconstruction of western Pacific hydroclimate from two speleothem records for southeastern Indonesia. The composite record shows pronounced shifts in monsoon rainfall that are antiphased with precipitation records for East Asia and the central-eastern equatorial Pacific. These meridional and zonal patterns are best explained by a poleward expansion of the Australasian Intertropical Convergence Zone and weakening of the Pacific Walker circulation (PWC) between ∼1000 and 1500 CE Conversely, an equatorward contraction of the Intertropical Convergence Zone and strengthened PWC occurred between ∼1500 and 1900 CE. Our findings, together with climate model simulations, highlight the likelihood that century-scale variations in tropical Pacific climate modes can significantly modulate radiatively forced shifts in global temperature.

The 8.2 ka event is the most significant global climate anomaly of the Holocene epoch, but a lack of records from Mainland Southeast Asia (MSEA) currently limits our understanding of the spatial and temporal extent of the climate response. A newly developed speleothem record from Tham Doun Mai Cave, Northern Laos provides the first high‐resolution record of this event in MSEA. Our multiproxy record (δ18O, δ13C, Mg/Ca, Sr/Ca, and petrographic data), anchored in time by 9 U‐Th ages, reveals a significant reduction in local rainfall amount and weakening of the monsoon at the event onset at ∼8.29 ± 0.03 ka BP. This response lasts for a minimum of ∼170 years, similar to event length estimates from other speleothem δ18O monsoon records. Interestingly, however, our δ13C and Mg/Ca data, proxies for local hydrology, show that abrupt changes to local rainfall amounts began decades earlier (∼70 years) than registered in the δ18O. Moreover, the δ13C and Mg/Ca also show that reductions in rainfall continued for at least ∼200 years longer than the weakening of the monsoon inferred from the δ18O. Our interpretations suggest that drier conditions brought on by the 8.2 ka event in MSEA were felt beyond the temporal boundaries defined by δ18O‐inferred monsoon intensity, and an initial wet period (or precursor event) may have preceded the local drying. Most existing Asian Monsoon proxy records of the 8.2 ka event may lack the resolution and/or multiproxy information necessary to establish local and regional hydrological sensitivity to abrupt climate change. Plain Language Summary: The most significant global climate event in the last ∼11,000 years occurred ∼8,200 years ago (the "8.2 ka event"). Many globally distributed records of past climate (paleoclimate) document significant changes during the event, but there are few from Mainland Southeast Asia available to reconstruct the climate impacts in this region. We present a new speleothem (cave sample) record from Tham Doun Mai Cave, Northern Laos that uses multiple geochemical and physical signals (proxies) to document local and regional climate effects of the 8.2 ka event at a high resolution. Using oxygen isotopes, we find that the 8.2 ka event weakened regional monsoon intensity for at least ∼170 years, which resembles findings from other studies. However, other proxies suggest local rainfall changed abruptly decades before the signal of monsoon weakening. Additionally, local drying may have lasted much longer than current estimates of 8.2 ka event responses. Most proxy records of the 8.2 ka event in monsoon regions lack similar multiproxy information and/or do not have high enough resolution to fully capture the climatic response. Additional records like ours from monsoon regions may assist in establishing how sensitive local and regional rainfall is to abrupt climate change. The late Neogene megatooth shark, Otodus megalodon, is known mainly from its gigantic teeth and possibly reached 18–20 m in total length (TL). We re-examine the previously proposed body size trends and nursery areas of O. megalodon by confining the previously used samples to upper anterior teeth offering more reliable TL estimates, and by taking paleolatitudes and sea-surface temperatures into consideration. We demonstrate that individuals of O. megalodon are on average larger in cooler water than those in warmer water – a pattern attributable to Bergmann's rule showing a latitudinal body size gradient at least for the eastern Pacific late Miocene and the western Atlantic early Pliocene assemblages. While it is still possible that neonatal O. megalodon could have utilised nursery areas, the previously identified paleo-nursery areas based on body size may reflect temperature-dependent trends rather than the inferred reproductive strategy. Thus, the gigantism of O. megalodon in cooler waters was possibly further enhanced by its cooler environment. If so, the corollary of this study is that not all populations of O. megalodon likely grew to gigantic sizes equally, where the common notion that the species reached 18–20 m TL should be applied primarily to populations in cooler environments. Fluorescent labeling is widely used in biological and chemical analysis, and the drive for increased throughput is stretching multiplexing capabilities to the limit. The limiting factor in multiplexed analyses is the ability to subsequently deconvolute the signals. Consequently, alternative approaches for interpreting complex data sets are required to allow individual components to be identified. Here we have investigated the application of a novel approach to multiplexed analysis that does not rely on multivariate curve resolution to achieve signal deconvolution. The approach calculates a sample-specific confidence interval for a multivariate (partial least-squares regression (PLSR)) prediction, thereby enabling the estimation of the presence or absence of each fluorophore based on the total spectral signal. This approach could potentially be applied to any multiplexed measurement system and has the advantage over the current algorithm-based methods that the requirement for resolution of spectral peaks is not central to the method. Here, PLSR was used to obtain the concentrations for up to eight dye-labeled oligonucleotides at levels of (0.6-5.3) x [10.sup.-6] M. The sample-specific prediction intervals show good discrimination for the presence/absence of seven of the eight labeled oligonucleotides with efficiencies ranging from ~91 to 100%.

Extinct lamniform sharks (Elasmobranchii: Lamniformes) are well represented in the late Mesozoic‒Cenozoic fossil record, yet their biology is poorly understood because they are mostly represented only by their teeth. Here, we present measurements taken from specimens of all 13 species of extant macrophagous lamniforms to generate functions that would allow estimations of body, jaw, and dentition lengths of extinct macrophagous lamniforms from their teeth. These quantitative functions enable us to examine the body size distribution of all known macrophagous lamniform genera over geologic time. Our study reveals that small body size is plesiomorphic for Lamniformes. There are four genera that included at least one member that reached >6 m during both the Mesozoic and Cenozoic, most of which are endothermic. The largest form of the genus Otodus, O. megalodon ('megatooth shark') that reached at least 14 m, is truly an outlier considering that all other known macrophagous lamniforms have a general size limit of 7 m. Endothermy has previously been proposed to be the evolutionary driver for gigantism in Lamniformes. However, we contend that ovoviviparous reproduction involving intrauterine cannibalism, a possible synapomorphy of Lamniformes, to be another plausible driver for the evolution of endothermy achieved by certain lamniform taxa.
