Abstract

Background The immunoglobulin J chain (Jchain) is highly expressed in the majority of multiple myeloma (MM), and Jchain-derived peptides presented in HLA molecules may be suitable antigens for T-cell therapy of MM. Methods Using immunopeptidomics, we identified Jchain-derived epitopes presented by MM cells, and pHLA tetramer technology was used to isolate Jchain-specific T-cell clones. Results We identified T cells specific for Jchain peptides presented in HLA-A1, -A24, -A3, and -A11 that recognized and lysed JCHAIN-positive MM cells. TCRs of the most promising T-cell clones were sequenced, cloned into retroviral vectors, and transferred to CD8 T cells. Jchain TCR T cells recognized target cells when JCHAIN and the appropriate HLA restriction alleles were expressed, while JCHAIN or HLA-negative cells, including healthy subsets, were not recognized. Patient-derived JCHAIN-positive MM samples were also lysed by Jchain TCR T cells. In a preclinical in vivo model for established MM, Jchain-A1, -A24, -A3, and -A11 TCR T cells strongly eradicated MM cells, which resulted in 100-fold lower tumor burden in Jchain TCR versus control-treated mice. Conclusions We identified TCRs targeting Jchain-derived peptides presented in four common HLA alleles. All four TCRs demonstrated potent preclinical anti-myeloma activity, encouraging further preclinical testing and ultimately clinical development.

Abstract

The major cause of death after allogeneic Hematopoietic Stem Cell Transplantation (HSCT) for acute myeloid leukemia (AML) is disease relapse. We investigated the expression of Inhibitory Receptors (IR; PD-1/CTLA-4/TIM-3/LAG-3/2B4/KLRG1/GITR) on T cells infiltrating the bone marrow (BM) of 32 AML patients relapsing (median 251 days) or maintaining complete remission (CR; median 1 year) after HSCT. A higher proportion of early-differentiated Memory Stem (T SCM ) and Central Memory BM-T cells express multiple IR in relapsing patients than in CR patients. Exhausted BM-T cells at relapse display a restricted TCR repertoire, impaired effector functions and leukemia-reactive specificities. In 57 patients, early detection of severely exhausted (PD-1 + Eomes + T-bet − ) BM-T SCM predicts relapse. Accordingly, leukemia-specific T cells in patients prone to relapse display exhaustion markers, absent in patients maintaining long-term CR. These results highlight a wide, though reversible, immunological dysfunction in the BM of AML patients relapsing after HSCT and suggest new therapeutic opportunities for the disease.

The data showed that HAMLET accurately detected all fusion genes and overexpression of EVI1 irrespective of 3q26 aberrations. In addition, small variants in 13 genes that are often mutated in AML were called with 99.2% sensitivity and 100% specificity, and tandem duplications in FLT3 and KMT2A were detected by a novel algorithm based on soft-clipped reads with 100% sensitivity and 97.1% specificity. In conclusion, HAMLET has the potential to provide accurate comprehensive diagnostic information relevant for AML classification, risk assessment and targeted therapy on a single technology platform.

Abstract

Conventional dendritic cell (DC) vaccine strategies, in which DCs are loaded with antigens ex vivo, suffer biological issues such as impaired DC migration capacity and laborious GMP production procedures. In a promising alternative, antigens are targeted to DC-associated endocytic receptors in vivo with antibody-antigen conjugates co-administered with toll-like receptor (TLR) agonists as adjuvants. To combine the potential advantages of in vivo targeting of DCs with those of conjugated TLR agonists, we generated a multifunctional antibody construct integrating the DC-specific delivery of viral- or tumor-associated antigens and DC activation by TLR ligation in one molecule. We validated its functionality in vitro and determined if TLR ligation might improve the efficacy of such a molecule. In proof-of-principle studies, an alpha CD40 antibody containing a CMV pp65-derived peptide as an antigen domain (alpha CD40(CMV)) was genetically fused to the TLR5-binding D0/D1 domain of bacterial flagellin (alpha CD40.Flg(CMV)). The analysis of surface maturation markers on immature DCs revealed that fusion of flagellin to alpha CD40(CMV) highly increased DC maturation (3.4-fold elevation of CD80 expression compared to alpha CD40(CMV) alone) by specifically interacting with TLR5. Immature DCs loaded with alpha CD40.Flg(CMV) induced significantly higher CMVNLV-specific T cell activation and proliferation compared to alpha CD40(CMV) in co-culture experiments with allogeneic and autologous T cells (1.8-fold increase in % IFN-gamma/TNF-alpha(+) CD8(+) T cells and 3.9-fold increase in % CMVNLV-specific dextramer(+) CD8(+) T cells). More importantly, we confirmed the beneficial effects of flagellin-dependent DC stimulation using a tumor-specific neoantigen as the antigen domain. Specifically, the acute myeloid leukemia (AML)-specific mutated NPM1 (mNPM1)-derived neoantigen CLAVEEVSL was delivered to DCs in the form of alpha CD40(mNPM1) and alpha CD40.Flg(mNPM1) antibody constructs, making this study the first to investigate mNPM1 in a DC vaccination context. Again, alpha CD40.Flg(mNPM1)-loaded DCs more potently activated allogeneic mNPM1(CLA)-specific T cells compared to alpha CD40(mNPM1). These in vitro results confirmed the functionality of our multifunctional antibody construct and demonstrated that TLR5 ligation improved the efficacy of the molecule. Future mouse studies are required to examine the T cell-activating potential of alpha CD40.Flg(mNPM1) after targeting of dendritic cells in vivo using AML xenograft models.

Abstract

Purpose: The goal of this study was to assess differences in low back stabilization and underlying mechanisms between patients with low back pain (LBP) and healthy controls. It has been hypothesized that inadequate trunk stabilization could contribute to LBP through high tissue strains and/or impingement. Evidence to support this is inconsistent, and not all methods that have been used to study trunk stabilization are equally suitable. We have recently developed a method to assess intrinsic and reflexive contributions to trunk stabilization, which aims to circumvent the limitations of previous studies. Methods: Forty-nine participants suffering from chronic LBP and a control group of fifty healthy subjects participated in this study. Trunk stabilization was measured using force-controlled perturbations directly applied to the trunk. The actuator displacement and contact force between the actuator and subject were measured as well as electromyography (EMG) of the M. Longissimus. Underlying mechanisms were characterized using system identification. Results: LBP patients showed lower admittance, i.e., less displacement per unit of force applied, mainly due to higher position, velocity and acceleration feedback gains. Among patients, lower trunk admittance and higher reflex gains were associated with more negative pain-related cognitions. Conclusion: Trunk stabilization differs between LBP patients and controls, with the same perturbations causing less trunk movement in patients, due to stronger reflexes. We interpret these changes as reflecting protective behavior. Abstract

gamma delta T cells are key players in cancer immune surveillance because of their ability to recognize malignant transformed cells, which makes them promising therapeutic tools in the treatment of cancer. However, the biological mechanisms of how gamma delta T-cell receptors (TCRs) interact with their ligands are poorly understood. Within this context, we describe the novel allo-HLA-restricted and CD8 alpha-dependent V gamma 5V delta 1TCR. In contrast to the previous assumption of the general allo-HLA reactivity of a minor fraction of gamma delta TCRs, we show that classic anti-HLA-directed, gamma delta TCR-mediated reactivity can selectively act on hematological and solid tumor cells, while not harming healthy tissues in vitro and in vivo. We identified the molecular interface with proximity to the peptide-binding groove of HLA-A*24:02 as the essential determinant for recognition and describe the critical role of CD8 as a coreceptor. We conclude that alloreactive gamma delta T-cell repertoires provide therapeutic opportunities, either within the context of haplotransplantation or as individual gamma delta TCRs for genetic engineering of tumor-reactive T cells.

Abstract

Er is een grens aan de hoeveelheid duurzaam te winnen (oever)grondwater in Flevoland. Dit vraagt van de provincie een andere focus voor de lange termijn. Alternatieve bronnen voor drinkwater en waterbesparingsmogelijkheden zijn voldoende voorhanden. Tussen de verschillende oplossingen is bijvoorbeeld onderscheid te maken in de hoeveelheid die winbaar is, beoogde gebruikstoepassingen, ruimtevraag, termijn van inzetbaarheid, robuustheid, maatschappelijke en juridische haalbaarheid. Om te bereiken dat waterbesparing en alternatieve bronnen gemeengoed worden, moeten er daadwerkelijk stappen gezet worden in wet- en regelgeving en in de uitvoering. Het gaat om een verandering in mentaliteit en ook eerste kleine stappen zijn daarin betekenisvol.

Abstract

De Beleidsnota Drinkwater (Rijksoverheid, 2014) agendeert het belang van een betrouwbare openbare drinkwatervoorziening voor de langere termijn. In 2017/2018 is daarom door de Provincie Flevoland en drinkwaterbedrijf Vitens een toekomstverkenning uitgevoerd naar de drinkwatervoorziening op de lange termijn. Vanwege de onzekerheden in de drinkwatervraag is hierbij gebruik gemaakt van het concept ‘adaptieve planning’ (Van der Brugge & Vermooten, 2018). Deze aanpak stelt de Provincie Flevoland en Vitens in staat om steeds opnieuw te bekijken of het aanbod van drinkwater voldoende blijft onder verschillende drinkwatervraagscenario’s en welke maatregelen eventueel genomen kunnen worden. In dit artikel wordt beschreven hoe de Provincie Flevoland en Vitens de methode van adaptieve planning toepassen in de drinkwatersector. Hiertoe zal eerst kort worden ingegaan op de drinkwatersituatie in Flevoland. Vervolgens zal worden beschreven hoe de methode van adaptieve planning is toegepast en hoe de Provincie Flevoland en Vitens het in de praktijk uitvoeren.

Abstract

Flevoland beschikt over een unieke grondwatervoorraad van uitzonderlijke kwaliteit. De provincie heeft voor de ontwikkeling van nieuw drinkwaterbeleid nader onderzoek laten verrichten naar de grondwatervoorraad als bron voor de drinkwatervoorziening. Uit het onderzoek blijkt dat de boringsvrije zone in Zuidelijk Flevoland naar verwachting nog enige ruimte biedt voor extra grondwateronttrekking om de groeiende drinkwatervraag op te vangen. De grenzen van een duurzame benutting van het grondwater komen echter in zicht.
