The Small Hive Beetle (SHB), Aethina tumida, is an invasive pest of honey bee colonies that causes significant damage to the beekeeping sector. SHB was detected in southern Italy (EU) in 2014 and despite adopted eradication measures, is still present there. After three years of observations of SHB in Calabria (2014-2016), we provide here some practical tips for improving control measures. A new time-saving colony examination method, including the use of an internal divider reduced the time needed for hive inspections by 31.86 % on average. Prioritizating the inspection of pollen and honey combs rather than brood combs is advised. Sentinel apiaries with no more than five colonies without supers are suggested for each beekeeping location in order to attract and to monitor the early appearance of SHB. The use of these methods will enable early detection and prompt control measures application before this destructive pest can spread in the region.

- Abstract
Background With numerous endemic subspecies representing four of its five evolutionary lineages, Europe holds a large fraction of Apis mellifera genetic diversity. This diversity and the natural distribution range have been altered by anthropogenic factors. The conservation of this natural heritage relies on the availability of accurate tools for subspecies diagnosis. Based on pool-sequence data from 2145 worker bees representing 22 populations sampled across Europe, we employed two highly discriminative approaches (PCA and F-ST) to select the most informative SNPs for ancestry inference. Results Using a supervised machine learning (ML) approach and a set of 3896 genotyped individuals, we could show that the 4094 selected single nucleotide polymorphisms (SNPs) provide an accurate prediction of ancestry inference in European honey bees. The best ML model was Linear Support Vector Classifier (Linear SVC) which correctly assigned most individuals to one of the 14 subspecies or different genetic origins with a mean accuracy of 96.2% +/- 0.8 SD. A total of 3.8% of test individuals were misclassified, most probably due to limited differentiation between the subspecies caused by close geographical proximity, or human interference of genetic integrity of reference subspecies, or a combination thereof. Conclusions The diagnostic tool presented here will contribute to a sustainable conservation and support breeding activities in order to preserve the genetic heritage of European honey bees.

The socio-economic impacts of COVID-19 on society have yet to be truly revealed; there is no doubt that the pandemic has severely affected the daily lives of most of humanity. It is to be expected that the research activities of scientists could be impacted to varying degrees, but no data exist on how COVID-19 has affected research specifically. Here, we show that the still ongoing COVID-19 pandemic has already diversely and negatively affected bee research at a global level. An online survey disseminated through the global COLOSS honey bee research association showed that every participant (n = 230 from 56 countries) reported an impact on one or more of their activities. Activities that require travelling or the physical presence of people (meetings and conferences, teaching and extension) were affected the most, but also laboratory and field activities, daily operations, supervision and other activities were affected to varying degrees. Since the basic activities are very similar for many research fields, it appears as if our findings for bee research can be extrapolated to other fields. In the light of our data, we recommend that stakeholders such as governments and funding bodies who support research should facilitate the wide implementation of web-based information technology required for efficient online communication for research and education, as well as adequately loosened restriction measures with respect to field and laboratory work. Finally, increased flexibility in administration and extension of research grants and fellowships seem to be needed. It is apparent that adequate responses by all stakeholders are required to limit the impact of COVID-19 and future pandemics on bee science and other research fields.

The European Commission has asked the EFSA to evaluate the risk for animal health related to the presence of hydroxymethylfurfural (HMF) in honey bee feed. HMF is a degradation product of particular sugars and can be present in bee feed. HMF is of low acute toxicity in bees but causes increased mortality upon chronic exposure. A benchmark dose lower limit 10% (BMDL10) of 1.16 μg HMF per bee per day has been calculated from mortalities observed in a 20‐day study and established as a Reference Point covering also mortality in larvae, drones and queens for which no or insufficient toxicity data were available. Winter bees have a much longer lifespan than summer bees and HMF shows clear time reinforced toxicity (TRT) characteristics. Therefore, additional Reference Point intervals of 0.21–3.1, 0.091–1.1 and 0.019–0.35 µg HMF/bee per day were calculated based on extrapolation to exposure durations of 50, 90 and 180 days, respectively. A total of 219 analytical data of HMF concentrations in bee feed from EU Member States and 88 from Industry were available. Exposure estimates of worker bees and larvae ranged between 0.1 and 0.48, and between 0.1 and 0.51 μg HMF/per day, respectively. They were well below the BMDL10 of 1.16 μg HMF/bee per day, and thus, no concern was identified. However, when accounting for TRT, the probability that exposures were below established reference point intervals was assessed to be extremely unlikely to almost certain depending on exposure duration. A concern for bee health was identified when bees are exposed to HMF contaminated bee feed for several months. [ABSTRACT FROM AUTHOR]

- Abstract
This short article presents loss rates of honey bee colonies over winter 2017/18 from 36 countries, including 33 in Europe, from data collected using the standardized COLOSS questionnaire. The 25,363 beekeepers supplying data passing consistency checks in total wintered 544,879 colonies, and reported 26,379 (4.8%, 95% CI 4.7-5.0%) colonies with unsolvable queen problems, 54,525 (10.0%, 95% CI 9.8-10.2%) dead colonies after winter and another 8,220 colonies (1.5%, 95% CI 1.4-1.6%) lost through natural disaster. This gave an overall loss rate of 16.4% (95% CI 16.1-16.6%) of honey bee colonies during winter 2017/18, but this varied greatly from 2.0 to 32.8% between countries. The included map shows relative risks of winter loss at regional level. The analysis using the total data-set confirmed findings from earlier surveys that smaller beekeeping operations with at most 50 colonies suffer significantly higher losses than larger operations (p < .001). Beekeepers migrating their colonies had significantly lower losses than those not migrating (p < .001), a different finding from previous research. Evaluation of six different forage sources as potential risk factors for colony loss indicated that intensive foraging on any of five of these plant sources (Orchards, Oilseed Rape, Maize, Heather and Autumn Forage Crops) was associated with significantly higher winter losses. This finding requires further study and explanation. A table is included giving detailed results of loss rates and the impact of the tested forage sources for each country and overall.

- Abstract
In this short note we present comparable loss rates of honey bee colonies during winter 2016/2017 from 27 European countries plus Algeria, Israel and Mexico, obtained with the COLOSS questionnaire. The 14,813 beekeepers providing valid loss data collectively wintered 425,762 colonies, and reported 21,887 (5.1%, 95% confidence interval 5.0-5.3%) colonies with unsolvable queen problems and 60,227 (14.1%, 95% CI 13.8-14.4%) dead colonies after winter. Additionally we asked for colonies lost due to natural disaster, which made up another 6,903 colonies (1.6%, 95% CI 1.5-1.7%). This results in an overall loss rate of 20.9% (95% CI 20.6-21.3%) of honey bee colonies during winter 2016/2017, with marked differences among countries. The overall analysis showed that small operations suffered higher losses than larger ones (p < 0.001). Overall migratory beekeeping had no significant effect on the risk of winter loss, though there was an effect in several countries. A table is presented giving detailed results from 30 countries. A map is also included, showing relative risk of colony winter loss at regional level.

- Abstract
We analyzed the morphological changes on the hepato-nephrocitic system (HNS) of Africanized honey bees exposed to thiamethoxam (TMX) and to Nosema ceranae (NOS) in isolation and co-exposure. We also analyzed the relative amount of hemocytes in TMX and/or Nosema-exposed bees. Newly emerged bees were exposed individually per os to TMX, at doses of 0.0856 ng/bee or 0.00856 ng/bee, or to TMX doses and 60,000 spores of N. ceranae per bee. Eight days after the single exposure, the results showed that TMX was extremely deleterious to the HNS at 0.00856 ng/bee, dramatically inducing morphological changes in HNS cells (trophocytes and oenocytes) and increasing hemocyte number. However, the treatment to TMX at 0.00856 ng/bee with additional NOS inoculation provoked the highest critical damage observed on HNS, collapsing the trophocytes, reducing the surface area of oenocytes, and inducing cell death in pericardial cells. Therefore, a sublethal dose of TMX, applied alone or simultaneously with NOS inoculation, triggered the disruption of the HNS and induced progressive damage on worker bees. The results demonstrated an interaction between biotic and abiotic agents in Africanized bees at individual level, whose impact at the colonial level remains to be assessed under field conditions. - Abstract
In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. - Abstract
A practical method to control the destructive varroa mite, Varroa destructorAnderson and Trueman in honey bee colonies consisted of monitoring with powdered sugar and treatment with oxalic acid (OA). We tested mite populations within 28 honey bee colonies on adult bees by a using a powdered sugar shake method. Subsequently, the same colonies were treated with 2.9% oxalic acid and 31.9% sucrose-in-water solution (w/w) four times, with a final varroa treatment using Apistan strips. On average 66.10 ± 35.23% and 94.64 ± 9.56% mites were detected using this simple sugar shake method in August and October, 2015. Furthermore, positive correlations were found between relative number of infested honey bees detected by powdered sugar shake and washing bees with alcohol plus soapy water (r = 0.90 and r = 0.62). Temperature and humidity seems to be crucial in determining the effectiveness of the powdered sugar shake method. For control of the mites, four consecutive OA treatments performed during the winter months in colonies containing capped brood had an average of 228 ± 226.93, 167 ± 313.79, 92 ± 160.50, and 27 ± 42.19 varroa mortality, respectively. Combination of four successive OA applications by trickling onto adult bees between combs in situ, during warm temperate winter conditions ensured a cumulative efficacy of 98%. Based on this level of mortality, it can be concluded that using the powdered sugar shake method to establish adult bee infestation levels in colonies along with an OA treatment may provide adequate varroa control in certain situations.

SUMMARYThe prothoracic and propodeal spiracles of three honey bee strains used commercially in Greece (from N Greece, N Italy and Slovenia) were examined with a light microscope in order to detect possible morphological differences between the strains. This is an attempt to explain the apparent resistance against Acarapis woodishown by the Greek Macedonian (N Greece) strain but not by the others. The propodeal spiracles of honey bees from N Greece and N Italy were examined, in addition, using a scanning electron microscope. Certain dimensions, like the length and the area of aperture in the prothoracic spiracle, were found to be significantly larger in honey bees from N Italy and Slovenia compared with the honey bees from N Greece. The length of aperture was found to be 648 μm in the honey bees from N Greece, while it was 13% larger in the honey bees from N Italy and 10% larger in the honey bees from Slovenia; the area of the aperture was found to be 103000 μm2in the honey bees from N Greece, while it was 26% larger in the honey bees from N Italy and 27% larger in the honey bees from Slovenia. The dimensions of the propodeal spiracle were found to be similar between the strains examined. The fact that the prothoracic spiracle of honey bees from N Greece was significantly smaller compared to other honey bee groups (from N Italy and Slovenia), is discussed as a possible mechanism for resistance developed by this strain against Acarapis woodi.

38. Acaricidal effect of oxalic acid in honeybee (apis mellifera</it>) colonies

- Abstract
Three oxalic acid (OA) solutions were applied to 24 honeybee colonies to test acaricidal effects on Varroa destructor. Daily natural mite drop per colony averaged 0.52. Higher mite mortality (18.33) was found after three August OA treatments. The mean efficacy's of the three water solutions of OA/sucrose (w/w), 3.4% /47.6% , 3.7% /26.1% , and 2.9% /31.9% applied in the presence of brood, was 52.28% , 40.66% and 39.16% respectively. A significantly higher efficacy was recorded when 3.4% /47.6% was applied in comparison to 2.9% /31.9% solution. There was no difference in efficacy between OA solutions administered during a broodless period on October 28. The average efficacy in all colonies was 99.44% . The results suggest that OA has limited acaricidal effect in colonies with brood, but it is highly effective in a broodless period.

peer-reviewed, Background: With numerous endemic subspecies representing four of its five evolutionary lineages, Europe holds a large fraction of Apis mellifera genetic diversity. This diversity and the natural distribution range have been altered by anthropogenic factors. The conservation of this natural heritage relies on the availability of accurate tools for subspecies diagnosis. Based on pool-sequence data from 2145 worker bees representing 22 populations sampled across Europe, we employed two highly discriminative approaches (PCA and FST) to select the most informative SNPs for ancestry inference. Results: Using a supervised machine learning (ML) approach and a set of 3896 genotyped individuals, we could show that the 4094 selected single nucleotide polymorphisms (SNPs) provide an accurate prediction of ancestry inference in European honey bees. The best ML model was Linear Support Vector Classifier (Linear SVC) which correctly assigned most individuals to one of the 14 subspecies or different genetic origins with a mean accuracy of 96.2% ± 0.8 SD. A total of 3.8% of test individuals were misclassified, most probably due to limited differentiation between the subspecies caused by close geographical proximity, or human interference of genetic integrity of reference subspecies, or a combination there of. Conclusions: The diagnostic tool presented here will contribute to a sustainable conservation and support breeding activities in order to preserve the genetic heritage of European honey bees.

peer-reviewed, This short article presents loss rates of honey bee colonies over winter 2017/18 from 36 countries, including 33 in Europe, from data collected using the standardized COLOSS questionnaire. The 25,363 beekeepers supplying data passing consistency checks in total wintered 544,879 colonies, and reported 26,379 (4.8%, 95% CI 4.7–5.0%) colonies with unsolvable queen problems, 54,525 (10.0%, 95% CI 9.8–10.2%) dead colonies after winter and another 8,220 colonies (1.5%, 95% CI 1.4–1.6%) lost through natural disaster. This gave an overall loss rate of 16.4% (95% CI 16.1–16.6%) of honey bee colonies during winter 2017/18, but this varied greatly from 2.0 to 32.8% between countries. The included map shows relative risks of winter loss at regional level. The analysis using the total data-set confirmed findings from earlier surveys that smaller beekeeping operations with at most 50 colonies suffer significantly higher losses than larger operations (p<.001). Beekeepers migrating their colonies had significantly lower losses than those not migrating (p<.001), a different finding from previous research. Evaluation of six different forage sources as potential risk factors for colony loss indicated that intensive foraging on any of five of these plant sources (Orchards, Oilseed Rape, Maize, Heather and Autumn Forage Crops) was associated with significantly higher winter losses. This finding requires further study and explanation. A table is included giving detailed results of loss rates and the impact of the tested forage sources for each country and overall.
41. A CASE STUDY OF ACUTE TOXICITY IN HONEYBEE COLONIES INDUCED BY ACARICIDE COUMAPHOS.

Abnormal bee behavior was observed 4 hours after coumaphos CheckMite+ strips had been inserted between two brood frames in each colony. Bees started to leave the hives, flew extensively around the hives, clustered on the front hive wall and dropped down in the grass. Workers also gathered there in smaller clusters and were dying in the surrounding of the treated hives with extended wings, and curved, shortened and tremored abdomens. Bees were also clustering on the back hive doors and walls in the innerside of the hives. Brood combs in the lower hive compartment were scared, and dead workers were found on the hive bottom board. Treated colonies were reduced for approximately 1/3 of adult bees. Workers were sampled from the bottom board, brood and honey compartment sand grass in front of the hives. Weper formed GC analyses, and quantify coumaphos by gas chromatography- electron capture detection (GC-ECD) with the limit quantification (LOQ) of 30 ppb. Coumaphos was found in workers sampled in brood compartment, honey compartment and in front of the hives at the levels of 1771, 606 and 514 ppb (µg/kg) respectively, which did not exceed the oral dose LD50. Nevertheless, it would be possible that some bees were exposed to much higher pesticide doses, as miticides do not distribute evenly within the bee colony. Bees can receive 3.2 µg of coumaphos in colonies treated with Checkmite+ (Haarmann et al., 2002) and in the previous study we showed that 2 µg of coumaphos already affects food transfer (trophallaxis), foraging activity, homing and learning ability of workers which could interfere with colony fitness. The interactions between pesticides used for crop protection and mite stresses are likely contributing factors. This supports a hypothesis that no factor alone is theoretically responsible for the dramatic worker mortality of Checkmite+ treated honey bee colonies. Pesticide treatments can induce toxic effects in honeybee colonies and reduce bee population. Application of acaricides in honeybee colonies should thus be applied with a great consideration. One of the potent tools in investigating the impact of environmental stress on honeybees is the in-vitro rearing of larvae. Young larvae are removed from the colony and can be treated with various doses of pesticides, other substances from the environment, pathogens or can be exposed to physical stressors. The application of this method was previously discussed in a workshop in Austria (2010) and also mediated in a short-term-scientific-mission (STSM). In November 2011, a follow-up workshop will be held in France. Another topic of interest of work group (WG) 3 is honeybee colony vitality, which was addressed at workshop in Wageningen (Netherlands) in June/July 2011. Here the establishment and validation of methods to assess honeybee vitality and health at individual and colony level was discussed. One aim is the development of a "toolkit" to determine the colony vitality as result of parasite infestation, feed / environment, diseases and (chronic) exposure to pesticides. The action of the herbicide 2,4-dichlorophenoxyacetic acid (2,4-D) on the isolated heart of the frog (Rana ridibunda) and two insects, the honeybee (Apis mellifera macedonica) and the beetle (Tenebrio molitor), was investigated using basic electrophysiological methods. The results of this study showed that a concentration of 1 μM 2,4-D was required to reduce the force and the frequency of the isolated heart of the honeybee to about 70% of the initial contraction in less than 20 min. To cause the same effects on the atria of the frog, 45 μM 2,4-D was required and on the isolated heart of the beetle, over 1000 μM of 2,4-D. The presence of an extensive system of gap junctions found in the honeybee is most probably the cause of the unusual sensitivity of its heart to 2,4-D, compared with the heart of the beetle, where no gap junctions were identified. - Abstract
Honey bee colonies were exposed to pesticides used in agriculture or within bee hives by beekeepers: coumaphos; diazinon; amitraz or fluvalinate. Samples of bee workers, larvae and royal jelly were analysed using Gas Chromatography-Electron Capture Detection (GC-ECD). Amitraz was quantified using High Performance Liquid Chromatography (HPLC), and Gas Chromatography-Tandem Mass Spectrometry (GC/MS/MS) was used for quantification of diazinon. Sixth day after treatment, coumaphos was found in the royal jelly (250 ng/g) secreted by nurse workers and fluvalinate was found in both bee heads (105 ng/g, 8 days after treatment) and in larvae (110 ng/g, 4 days after treatment). Amitraz residues in all sampled material were below the level of detection of 10 ng/g. Diazinon was not detected in any of the analysed samples. The large quantities of fluvalinate found in bee heads and larvae, the coumaphos residues in royal jelly, and additional potential sub-lethal effects on individual honey bees or brood are discussed.

- Abstract
Honey bee (Apis mellifera carnica) colonies were placed in two apple orchards treated with the insecticides diazinon and thiacloprid and the fungicide difenoconazole in accordance with a Protection Treatment Plan in the spring of 2007. Pollen and bee bread were collected from combs inside the hives. The residue of diazinon in pollen loads 10 days after orchard treatment was 0.09 mg/kg, and the same amount of residue was found in bee bread 16 days after treatment. In pollen loads 6 days after application 0.03 mg/kg of thiacloprid residues and 0.01 mg/kg of difenoconazole were found on the first day after application. Possible sub-lethal effects on individual honey bees and brood are discussed.

- Abstract
This experiment assessed the efficacy of rotenone and oxalic acid (OA) in an aqueous sugar solution in controlling the honeybee mite Varroa destructor. Colonies were populated with mite-infested brood combs and worker bees. Three rotenone or OA treatments administered during the period with capped brood on 31 July, 14 and 18 August resulted in an average efficacy of 24.10%. In untreated colonies mite mortality averaged 5.40%. No significant differences (P>0.05) were found between the rotenone and OA treatments. Three OA treatments administered on 9, 12 and 18 September resulted in a 77.93% mite mortality. An increase in mite drop (P<0.05) was observed at 2 and 4 days after each treatment. OA applications in broodless colonies resulted in significantly (P<0.001) higher mite mortality rates (98.65% average) than the three treatments of rotenone or OA in colonies with capped brood. The dynamics of mite mortality after each rotenone or OA treatment are discussed in this study.

Twenty-four honeybee (Apis mellifera) colonies were used to monitor the efficacy of a solution of 2.9% oxalic acid (OA) and 31.9% sugar against the mite Varroa destructor. Mite mortality was established prior to and after OA treatments, which were conducted in August and September. The treatments resulted in 37% mite mortality as opposed to 1.11% in the controls. OA treatment conducted in September on previously untreated colonies resulted in 25% mite mortality. OA treatments in October and November resulted in approximately 97% mite mortality. These results suggest that OA is effective during the broodless period and less effective when applied to colonies with capped broods. The possible use of OA against the Varroa mite in honeybee colonies as an alternative to routine chemical treatments is discussed., (Copyright 2002 Published by Elsevier Science Ltd.)
