Bone density (BD) is an important factor in osteoporotic fracture risk in humans. However, BD is a complex trait confounded by environmental influences and polygenic inheritance. Sheep provide a potentially useful model for studying differences in BD, as they provide a means of circumventing complex environmental factors and are a similar weight to humans. The aims of this study were to establish whether there is genetic variation in BD in sheep and then to localise quantitative trait loci (QTLs) associated with this variation. We also aimed to evaluate the relationship between fat and muscle body components and BD in sheep. Results showed that there was significant (P < 0.01) genetic variation among Coopworth sheep sires for BD. This genetic difference was correlated (P < 0.01) with body weight and muscle mass. A number of QTLs exceeding the suggestive threshold were identified (nine in total). Of these, two (chromosomes 1, P < 0.05; chromosome 24, P < 0.01) were significant using genome-wide permutation significance thresholds (2000 iterations). The position of the QTL on chromosome 24 coincided with a number of other body composition QTLs, indicating possible pleiotropic effects or the presence of multiple genes affecting body composition at that site. This study shows that sheep are potentially a useful model for studying the genetics of BD.

A systematic, house-based serological survey for Trypanosoma cruzi seroreactivity was conducted in three contiguous communities in Olopa municipality, Chiquimula Department, Guatemala. Blood samples from a total of 292 individuals in 63 households were examined by enzyme-linked immunosorbent assay. The seropositive rate ranged from 0% to 20.8% for the three communities, with a mean of 15.1%. Log-linear models showed that seroprevalence was significantly related to age (P < 0.005) but not to sex. However, when the age group with the lowest prevalence (1-9 years) was excluded from the analysis, age was not a significant factor (P = 0.55). Data from a stratified sample collected at the same time were combined with those of the systematic sample to analyze the relationship between seropositivity and possible explanatory variables. Log-linear models, based on 586 individuals in 129 households from the two surveys, revealed a significant positive association between seropositivity and thatched roofs (P = 0.01).

Human abdominal angiostrongyliasis is a potentially fatal disease caused by Angiostrongylus costaricensis, a nematode found in the Americas. During the period of December 1994 through August 1995, an outbreak of this disease occurred in Guatemala. We identified 22 cases of abdominal angiostrongyliasis and conducted a matched case-control study to identify risk factors for illness. The median age of the 18 cases enrolled in the study was 37 years (range, 9-68 years), and 11 (61.1%) were male. Consumption of the following six raw food items was associated with angiostrongyliasis: mint (odds ratio [OR], 6.9; 95% confidence interval [CI], 1.5-66.0), shrimp (OR, infinite; 95% CI, 1.4 to infinite), and four kinds of ceviche that reportedly contained raw mint (OR for consumption of mint or ceviche that contained mint, 7.0; 95% CI, 1.0-315). We conclude that raw mint was the likely vehicle of infection for this outbreak. To our knowledge, this is the first reported outbreak of abdominal angiostrongyliasis and the first time that a specific food item has been epidemiologically linked to the disease.

The evolution of the population densities of Bulinus truncatus and Bulinus forskalii has been studied during 18 months in a number of sites of the Sudan-Sahelian zone of Cameroon, in relation to certain environmental factors. Only the altitude of the water in the sites has an influence on the density of the snails. The populations of B. truncatus present a greater proportion of young molluscs during the entire year, with the exception of the last months of the dry season. The population dynamics of B. forskalii varies depending on whether the environment is temporarily stagnant, temporarily running or stable. The proportions of B. truncatus and B. forskalii that emit cercariae of schistosomes are 1.2% and 0.8% respectively. Study of the adult worms obtained after infestation of the laboratory mammals show that B. truncatus emits cercariae of Schistosoma haematobium at the end of the dry season and at the beginning of the rainy season. We have not been able to identify precisely the cercariae emitted by B. forskalii, which raises the necessity of further systematic studies being carried out on the schistosomes of Cameroon.

The population dynamics of Bulinus globosus was followed at seasonal and perennial habitats in three villages in the Extreme North Province, Cameroon, known to have high prevalences of Schistosoma haematobium. The study covered 2 rainy and 1 dry season between June 1987 to November 1988. Of several environmental factors monitored, only rainfall and the resulting changes in water level were strongly correlated with changes in snail density, and even then only when values were at their extremes. In seasonal pools, adult snails emerge from estivation and begin laying eggs shortly after the first rains. A decline in the number of larger snails 3 to 6 weeks later suggests a die off of the founding population. In general, the population density increases modestly during the rainy season and then rapidly declines after torrential rains have ended in September or October. Adults are again dominate just before the habitats dry up. In seasonal streams, population densities fluctuate dramatically in association with rainfall and subsequent flooding, and populations stabilize only after the torrential rains have ceased. Unlike populations in temporary ponds, young snails constituted a large percentage of the population very late in the season. In perennial habitats, seasonal fluctuations in population density were related to manipulation of water levels in a rice irrigation scheme. The lowest densities occurred when the water levels were at their most extreme levels, between November and April. But declines in the population densities often preceded significant changes in the water level, indicating that this factor alone is not responsible for these declines. Of the B. globosus shedding schistosome cercariae, more than 90% were infected with S. haematobium and the remainder with S. bovis. Of 153 infected B. globosus, 87% were found in seasonal habitats. In those sites the first infected snails were found in July but the large numbers were not present until late August and September. Infected snails declined rapidly before the final decline in the general population of B. globosus in November and December. In perennial habitats, infected snails were most common in May and June, just before and early in the rainy season. These findings may prove useful in developing more effective schistosomiasis control measures in sahelian climates.

Terminal-spined schistosome eggs were found in both stool and urine specimens of a high proportion of infected individuals in Kinding Njabi, a village located in a savannah/forest ecotone of Center Province, Cameroon. Infections with terminal-spined schistosomes are not common in that region and first appeared in Kinding Njabi in the mid-1980s. The village population, determined by house-to-house census in 1986, was 345. Of 302 stool samples examined by a thick smear technique, 58 (19%) were positive, and of 320 urine samples examined by sedimentation, 40 (12%) were positive. Twenty-six (37%) of a total of 71 infected persons passed eggs in both stool and urine. Egg size distribution and morphology were similar to previous reports for Schistosoma haematobium/S. intercalatum hybrids. Bulinus forskalii was the only snail found during extensive surveys in and around Kinding Njabi; infected snails were recovered from several temporary habitats. This focus is of particular interest because it represents the first report of transmission of hybrid schistosomes outside of the hybrid zone. Interviews with villagers and local health officials suggest that schistosomiasis was introduced by immigrants from Loum, a town approximately 100 km southwest of Kinding Njabi, where S. haematobium, S. intercalatum, and their hybrid are known to exist. Results from snail host specificity studies using the Kinding Njabi parasite showed that stool-derived miracidia were compatible only with B. forskalii, the exclusive host for S. intercalatum in Cameroon. Urine-derived miracidia were compatible only with B. truncatus, a S. haematobium host in Cameroon.(ABSTRACT TRUNCATED AT 250 WORDS)

The status of schistosomiasis in Cameroon was examined in a nationwide survey of 5th grade schoolchildren. Five hundred twelve schools were surveyed; 19,524 urine and 22,166 stool samples were examined. The 3 northern provinces, which comprised 29% of the population, had 87% of all urinary and 82% of all intestinal cases. These provinces have a low seasonal rainfall. The presence of temporary bodies of water and of molluscan intermediate hosts adapted to this environment permits intense transmission of schistosomiasis haematobium and mansoni. In the rest of the country, the distribution of Schistosoma haematobium and S. mansoni was highly focal. S. intercalatum endemic areas were restricted to the equatorial forest and were small with low prevalences and intensities.

A nationwide survey for snail hosts of human schistosomes was carried out in Cameroon between 1985 and 1988. In total, 668 sites at 432 locations were sampled. In the arid, northern half of the country (tropical climatic zone), where both intestinal and urinary schistosomiasis are hyperendemic, Biomphalaria pfeifferi was the only Schistosoma mansoni host and Bulinus globusus and B. senegalensis the most common S. haematobium hosts. In that region, these snails occurred almost exclusively in temporary bodies of water. Taking into account results from a companion study on the distribution of schistosomiasis in humans, our results clearly show that temporary water bodies in the tropical zone are the principal foci of transmission. These findings disagree with commonly held views about schistosome transmission in Cameroon. B. truncatus, a S. haematobium host, was also present in the tropical zone but was found principally in perennial habitats. Although some perennial habitats were important transmission sites, they represent only a small portion of the overall problem. B. truncatus is the principal S. haematobium host in the wetter southern half of the country where schistosomiasis haematobium is highly focal. Biom. camerunensis was far more common than Biom. pfeifferi in the South but did not occur where S. mansoni prevalence rates were high; thus it appears to be a poor host. B. forskalii, the sole host of S. intercalatum in Cameroon, occurs widely throughout the country; however, the schistosome is restricted to a small region in the South.

Schistosoma malayensis n. sp., a member of the Schistosoma japonicum complex is described from Rattus muelleri in Peninsular Malaysia and 2 strains are characterized. The only morphological differences noted among adults from natural hosts were that S. malayensis are in general smaller than S. mekongi and S. japonicum. But these differences may be the result of host-induced variations and therefore are of little taxonomic value. To minimize the effects of host-induced variations, adult worms recovered from laboratory mice with similar worm burdens at 50-56 days postinfection were compared. These comparisons revealed only minor morphometric differences among these 3 species. Schistosoma malayensis eggs from naturally and experimentally infected hosts are most similar to those of S. mekongi, with eggs of both species being, in general, smaller than those of S. japonicum. The egg index for S. malayensis is usually higher than for S. japonicum and lower than for S. mekongi. Differences were noted in the developmental rates in mice for 2 isolates of S. malayensis, S. mekongi, and S. japonicum (Philippine strain), but relatively large differences observed between isolates of S. malayensis indicate that, in this case, the developmental rate is not a useful taxonomic character. Schistosoma malayensis is erected principally on the basis of differences, reported elsewhere, in the life histories and in the electrophoretic migration patterns of isoenzymes of adult worms as compared to S. mekongi and S. japonicum. These comparisons indicate that S. malayensis is more closely related to S. mekongi than to S. japonicum.

Tapeworms were recovered from a 10-month-old girl in Bangkok, Thailand, following anthelmintic treatment. The worms were identified as members of family Anoplocephalidae, genus Mathevotaenia, which has not previously been reported from humans. The worm most closely resembles M. symmetrica, a cosmopolitan parasite of rodents with insects serving as intermediate hosts.

A stable population at risk of Malaysian schistosomiasis was studied. Census results indicated that approximately one-fourth of the inhabitants used a stream where Schistosoma malayensis-infected snails were present as their principal source of water for bathing, drinking, and household tasks. The general population also contacted this stream when fording it or while fishing. Serological surveys using enzyme-linked immunosorbent assay (ELISA) and the circumoval precipitin (COP) test revealed six (9%) and three (4%) positives, respectively, among 67 persons examined. No schistosome ova were found in a general survey of 56 persons which included five ELISA positive and two COP test positive patients. ELISA and COP test prevalences among those dependent on the foci of transmission for water, 13 and 7% respectively, were only slightly higher than prevalences among the remainder of the population, 8 and 4% respectively. These results indicate that even among a stable population at risk of Malaysian schistosomiasis the prevalence is low. Our findings support the hypothesis that S. malayensis is a zoonotic infection in man and that it is unlikely to become a significant public health problem.

The surface of adult Schistosoma japonicum-like (Malaysian) was studied by scanning electron microscopy. The basic pattern of surface microtopography is similar to other strains of S. japonicum as previously reported. However, among male member there are some unique differences in the types, number and distribution of surface papillae and morphology of ridges. Three kinds of papillae were observed: (1) the large fungiform papillae (3.5-4 micron in diameter, most without cilia) are more numerous than in other strains of S. japonicum, they concentrate on the lateral aspect of the anterior and middle parts close to the edge of the gynecophoral canal, and on the dorso-lateral aspect of the posterior part towards the tail tip; (2) the small hemispherical papillae (1.5-2 micron in diameter, all bearing cilia) are especially numerous in the suckers, the gynecophoral canal and parts of the tegument around the suckers and close to the tail tip; on the rest of the surface they are evenly distributed; (3) the cratered papillae (3-4 micron in diameter, about half having cilia) are more numerous than on other strains, they concentrate on the lateral aspect of the middle part and on the edges of the gynecophoral canal. The surface ridges (about 0.2-0.3 micron in width) are tall, highly branching and perforated; they are most developed in the middle part. Spines were observed only in the suckers and the gynecophoral canals. In contrast to the male, the female has numerous spines on all parts of the surface except the most anterior, where a large number of long cilia were observed. All three kinds of papillae were present; fungiform papillae are more numerous than in females of other strains; they concentrate on the latero-dorsal aspect of middle and posterior parts, and around the excretory pore. Ridges are much less developed than in the male and are prominent only in the middle part.

Using ELISA and COPT diagnostic tests, serological evidence of Malaysian schistosomiasis was discovered among Orang Asli populations from three areas in Peninsular Malaysia. Serum samples collected in 1975 indicated an ELISA-positive prevalence of 25% and a COPT prevalence of 11% from Pos Iskandar, Pahang and an ELISA prevalence of 13% and a COPT of 4% from Bukit Lanjan, Selangor. Resurveys at these site in 1982-1984 showed a continued presence of serological positive individuals but prevalence rates were markedly lower: 7% and 1% for ELISA and 4% and 2% for COPT at Pos Iskandar and Bukit Lanjan respectively. Snail hosts were not found at either site. The source of infection for persons living in these lowland areas remains unknown. In a third area, Kuala Tahan, Pahang, located in the foothills of the central mountain range, foci of transmission have been found near to Orang Asli settlements. The serological prevalence rate among Negrito Orang Asli in that study area was 9% for ELISA and 4% for COPT. Thirty-three of 36 COPT-positive sera produced vacuolated bleb precipates and in 31 these were the only reactions seen. The high percentage of positives producing only these precipates suggests that among Orang Asli schistosomiasis patients such reactions are not an indication of recently acquired infection as has been reported for schistosomiasis patients in the Philippines.

Schistosoma sinensium Pao, 1959, was first isolated from an unidentified snail in Szechuan Province, China, and was described on the basis of adult worms and eggs recovered from experimentally infected mice. We discovered snails, Tricula bollingi Davis, and rodents, Rattus rattus (L.) and Crocidura attenuata Milne-Edwards, infected with S. sinensium near Fang in northwest Thailand. Adult worms were recovered from small veins immediately adjacent to the small intestine. Eggs first appeared in the feces of experimentally infected mice at 25 days postinfection. Cercariae first emerged from experimentally infected T. bollingi at 40 days postinfection. Shortly after emerging, cercariae were found at the water surface, their bodies flat against the meniscus and tails hanging free in the water column. Our morphometric results for adults and eggs from naturally infected hosts agree well with those presented in the original description. The occurrence of a lateral spine on the egg of this Asian schistosome has created some confusion as to the affinities of S. sinensium. A comparison of 8 character states, including snail hosts, cercarial behavior, geographical distribution, and morphology of life cycle stages, indicates that S. sinensium is more closely related to S. japonicum Katsurada than it is to S. mansoni Sambon.
