Abstract

BACKGROUND Plant bugs (Lygus spp.) and thrips (Thrips spp.) are two of the most economically important insect pest groups impacting cotton production in the USA today, but are not controlled by current transgenic cotton varieties. Thus, seed or foliar‐applied chemical insecticides are typically required to protect cotton from these pest groups. Currently, these pests are resistant to several insecticides, resulting in fewer options for economically viable management. Previous publications documented the efficacy of transgenic cotton event MON 88702 against plant bugs and thrips in limited laboratory and field studies. Here, we report results from multi‐location and multi‐year field studies demonstrating efficacy provided by MON 88702 against various levels of these pests. RESULTS MON 88702 provided a significant reduction in numbers of Lygus nymphs and subsequent yield advantage. MON 88702 also had fewer thrips and minimal injury. The level of control demonstrated by this transgenic trait was significantly better compared with its non‐transgenic near‐isoline, DP393, receiving insecticides at current commercial rates. CONCLUSION The level of efficacy demonstrated here suggests that MON 88702, when incorporated into existing IPM programs, could become a valuable additional tool for management of Lygus and thrips in cotton agroecosystems experiencing challenges of resistance to existing chemical control strategies. © 2018 The Authors. Abstract

Cloning and sequencing of the choM structural gene in plant protoplasts and bacteria reveal that the gene encodes a cholesterol oxidase and has a primary translation product with a 547-amino-acid protein. The first 43 amino acids represent a secretory signal peptide whose expression in Escherichia coli yields a protein with insecticidal and enzymatic characteristics. The gene expression in tobacco protoplasts results in an enzymatically active cholesterol oxidase in the presence or absence of the signal peptide sequence.

Abstract

We have expressed truncated forms of the insect control protein genes of Bacillus thuringiensis var. kurstaki HD-1(cryIA(b)) and HD-73 (cryIA(c)) in cotton plants at levels that provided effective control of agronomically important lepidopteran insect pests. Total protection from insect damage of leaf tissue from these plants was observed in laboratory assays when tested with two lepidopteran insects, an insect relatively sensitive to the B.t.k. insect control protein, Trichoplusia ni (cabbage looper) and an insect that is 100 fold less sensitive, Spodoptera exigua (beet armyworm). Whole plants, assayed under conditions of high insect pressure with Heliothis zea (cotton bollworm) showed effective square and boll protection. Immunological analysis of the cotton plants indicated that the insect control protein represented 0.05% to 0.1% of the total soluble protein. We view these results as a major step towards the agricultural use of genetically modified plants with insect resistance in this valuable, high acreage crop.

Abstract

Transgenic tomato plants expressing a lepidopteran-specific insect control protein from Bacillus thuringiensis var. kurstaki HD-1 (B.t.k.) were field tested in 1987 and 1988 to evaluate their efficacy under normal field conditions. When infested with eggs of the tobacco hornworm (Manduca sexta), the transgenic plants showed only very limited feeding damage on the leaves near the inoculated sites soon after the larvae hatched, and sustained no further damage. In contrast, nontransgenic control plants suffered heavy feeding damage and were almost completely defoliated within two weeks. Significant control of tomato fruitworm (Heliothis zea) and tomato pinworm (Keiferia lycopersicella) was also observed. These results demonstrate that transgenic tomato plants containing a gene encoding a B.t.k. protein can provide control of lepidopteran insect pests under field conditions.
