We tested the effects of prolonged voluntary wheel running on the muscle function of mdx mice treated with one of two different microdystrophin constructs. At 7 weeks of age mdx mice were injected with a single dose of AAV9-CK8-microdystrophin with (gene therapy 1, GT1) or without (gene therapy 2, GT2) the nNOS-binding domain and were assigned to one of four gene therapy treated groups: mdxRGT1 (run, GT1), mdxGT1 (no run, GT1), or mdxRGT2 (run,GT2), mdxGT2 (no run, GT2). There were two mdx untreated groups injected with excipient: mdxR (run, no gene therapy) and mdx (no run, no gene therapy). A third no treatment group, Wildtype (WT) received no injection and did not run. mdxRGT1, mdxRGT2 and mdxR performed voluntary wheel running for 52 weeks; WT and remaining mdx groups were cage active. Robust expression of microdystrophin occurred in diaphragm, quadriceps, and heart muscles of all treated mice. Dystrophic muscle pathology was high in diaphragms of non-treated mdx and mdxR mice and improved in all treated groups. Endurance capacity was rescued by both voluntary wheel running and gene therapy alone, but their combination was most beneficial. All treated groups increased in vivo plantarflexor torque over both mdx and mdxR mice. mdx and mdxR mice displayed ∼3-fold lower diaphragm force and power compared to WT values. Treated groups demonstrated partial improvements in diaphragm force and power, with mdxRGT2 mice experiencing the greatest improvement at ∼60% of WT values. Evaluation of oxidative red quadriceps fibers revealed the greatest improvements in mitochondrial respiration in mdxRGT1 mice, reaching WT levels. Interestingly, mdxGT2 mice displayed diaphragm mitochondrial respiration values similar to WT but mdxRGT2 animals showed relative decreases compared to the no run group. Collectively, these data demonstrate that either microdystrophin construct combined with voluntary wheel running increased in vivo maximal muscle strength, power, and endurance. However

Nonclassical light sources are highly sought-after as they are an integral part of quantum communication and quantum computation devices. Typical sources use bulk nonlinear crystals that rely on stringent phase-matching conditions, limiting the operating wavelength and bandwidth. In this work, we demonstrate the generation of photon pairs from a freestanding lithium niobate microcube at the telecommunication wavelength of 1.56 µm through the spontaneous parametric down-conversion process. The maximum photon pair generation rate obtained from a single microcube with the size of 3.6 µm is 490 Hz, resulting in an efficiency of 20.6 GHz/Wm, which is three orders of magnitude larger than the efficiency of biphoton generation in bulk nonlinear crystals. The microcubes are synthesized through a solvothermal method, offering the possibility for scalable devices via bottom-up assembly on any substrates. Our work constitutes an important step forward in the realization of compact nonclassical light sources with a wide bandwidth for various quantum applications.

Espectrógrafo Multi-objecto InfraRojo (EMIR), currently entering into its fabrication and AIV phase, will be one of the first common user instruments for the GTC, the 10 m telescope under construction by GRANTECAN at the Roque de los Muchachos Observatory (Canary Islands, Spain). EMIR is being built by a Consortium of Spanish and French institutes led by the Instituto de Astrofísica de Canarias (IAC). EMIR is designed to realize one of the central goals of 10 m class telescopes, allowing observers to obtain spectra for large numbers of faint sources in an time-efficient manner. EMIR is primarily designed to be operated as a MOS in the K band, but offers a wide range of observing modes, including imaging and spectroscopy, both long slit and multiobject, in the wavelength range 0.9-2.5 µm. It is equipped with two innovative subsystems: a robotic reconfigurable multislits mask and dispersive elements formed by the combination of high quality diffraction grating and conventional prisms, both at the heart of the instrument. The present status of development, expected performances, schedule, and plans for scientific exploitation are described and discussed. Semiconductor saturable absorber mirror (SESAM) devices have become a key component of ultrafast passive mode-locked laser sources. Here we describe in more detail how the key SESAM parameters such as saturation fluence, modulation depth, and nonsaturable losses are measured with a high accuracy. These parameters need to be known and controlled to obtain stable pulse generation for a given laser. A high-precision, wide dynamic range setup is required to measure this nonlinear reflectivity of saturable absorbers. The challenge to measure a low modulation depth and key measures necessary to obtain an accurate calibration are described in detail. The model function for the nonlinear reflectivity is based on a simple two-level travelling wave system. We include spatial beam profiles, nonsaturable losses and higher-order absorption, such as two-photon absorption and other induced absorption. One of the most application-relevant milestones that remain to be achieved in the field of passively mode-locked surface-emitting semiconductor lasers is the integration of the semiconductor absorber into the gain structure, enabling the realization of ultra-compact high-repetition-rate laser devices suitable for wafer-scale integration. We have recently succeeded in fabricating the key element in this concept, a quantum-dot-based saturable absorber with a very low saturation fluence, which for the first time allows stable mode locking of surface-emitting semiconductor lasers with the same mode areas on gain and absorber. The reflectivity of a semiconductor saturable absorber mirror (SESAM) is generally expected to increase with increasing pulse energy. However, for higher pulse energies the reflectivity can decrease again; we call this a ‘roll-over' of the nonlinear reflectivity curve caused by inverse saturable absorption. We show for several SESAMs that the measured roll-over is consistent with two-photon absorption only for short (femtosecond) pulses, while a stronger (yet unidentified) kind of nonlinear absorption is dominant for longer (picosecond) pulses. These inverse saturable absorption effects have important technological consequences, e.g. for the Q-switching dynamics of passively mode-locked lasers. A simple equation using only measurable SESAM parameters and including inverse saturable absorption is derived for the Q-switched mode-locking threshold. We present various data and discuss the sometimes detrimental effects of this roll-over for femtosecond high repetition rate lasers, as well as the potentially very useful consequences for passively mode-locked multi-GHz lasers. We also discuss strategies to enhance or reduce this induced absorption by using different SESAM designs or semiconductor materials
