18. Magnetic nano composites for gallic acid delivery

Abstract

A new magnetic layered double hydroxide (MLDH) composed of Fe, Mg, and Al was designed to improve gallic acid delivery to breast tumors. The metals formed the octahedrons within the layers, while the gallate ions were dissolved in the aqueous interlayer space. The drug loading was 30 %, determined with UV/Vis spectrophotometry. The material exhibited superparamagnetic behavior with a saturated magnetization of 5.5 emu/gr. This magnetic property enables the compound to be directed towards the therapeutic target using an external magnetic field, while the superparamagnetic attribute prevents the material from remaining magnetized in the body after the external magnetic field is removed. The diameter of the composites, measured from TEM images, was 110 nm: small enough for systemic circulation. For comparison, a conventional layered double hydroxide composed of Mg and Al was loaded with gallic acid and impregnated with Fe3O4magnetic nanoparticles (LDH-NP). The drug loading was slightly lower (23 %), and the material demonstrated a combination of superparamagnetic and ferromagnetic behavior. However, the saturated magnetization was higher, 15 emu/gr for the superparamagnetic-like component and 13 emu/gr for the ferromagnetic-like component. From TEM images, the diameter of the composites was 57 nm. Drug release was studied with Franz diffusion cells. Both systems showed fast release at the lysosomal pH (4.8), but the MLDH avoids the release at the blood pH (7.4) more than the LDH-NP. Lastly, the systems were tested against breast cancer cells. After 48 h, the new composite demonstrated a 77 % reduction in cell viability, while the conventional composite impregnated with NPs showed a 66 % reduction. The designed Fe/Mg/Al magnetic nano composite loaded with gallic acid is easier to manufacture and holds promise for breast cancer treatments.

Abstract

El propóleos es una mezcla de composición química compleja que posee propiedades biológicas de gran interés para fines terapéuticos. Existen distintos tipos de propóleos que difieren en su composición química de acuerdo a su origen botánico. En la actualidad se carece de un estándar de calidad de propóleos. Por esta razón, en este trabajo fueron determinados los caracteres organolépticos, propiedades físico- químicas y el contenido total de compuestos fenólicos y flavonoides de 15 muestras de propóleos recolectadas de diferentes regiones de Chaco, Corrientes, Santiago del Estero y Mendoza, para así brindar una estandarización que permita producir medicamentos de propóleos de seguridad y eficacia demostrable. Se encontró gran variación en las propiedades de los diferentes propóleos estudiados, incluso de aquellos provenientes de una misma región. Los propóleos difirieron principalmente en su contenido de compuestos fenólicos y flavonoides, siendo significativamente superiores en los propóleos originarios de Mendoza, donde la vegetación más abundante pertenece a Populus sp., Propolis is a mixture of complex chemical composition that possesses biological properties of great interest for therapeutic purposes. Different types of propolis exist, which differ in its chemical composition according to its botanical origin. At present, there is not a quality standard for propolis. For this reason, in this work the organoleptic characteristics, the physicochemical properties, the total content of phenolic compounds and the flavonoid composition of 15 samples of propolis collected from different regions of Chaco, Corrientes, Santiago del Estero, and Mendoza were determined, for providing an standardization that allows the production of effective and safe propolis products. It was found great variation in the properties of the different studied propolis, even of those from the same region. Abstract

Abstract: The complex formation of chlorhexidine digluconate (CHX-G2) with hydroxypropyl-β-cyclodextrin (HPβCD) was studied using NMR spectroscopy. The results revealed that this surfactant agent shows an monomer/aggregate equilibrium, which is dependent on the concentration of this drug. This equilibrium can be modified by the presence of HPβCD, which reduces the aggregation of the CHX-G2 molecules. An inclusion process of the CHX-G2 aromatic residue within the cyclodextrin cavity was confirmed by 2D ROESY spectroscopy. 1H NMR titration studies of CHX-G2 with HPβCD in D2O confirmed the formation of higher order complexes between CHX-G2 and HPβCD. Moreover, the addition of HPβCD into CHX-G2 solutions forms insoluble aggregates. Such insoluble aggregates may result in the stacking of CHX-G2 molecules on the surface of the CHX-G2:HPβCD complexes. [Copyright &y& Elsevier]
