Toxoplasmosis is caused by Toxoplasma gondii, an apicomplexan parasite that is able to infect any nucleated cell in any warm-blooded animal. Toxoplasma gondii infects around 2 billion people and, whilst only a small percentage of infected people will suffer serious disease, the prevalence of the parasite makes it one of the most damaging zoonotic diseases in the world. Toxoplasmosis is a disease with multiple manifestations: it can cause a fatal encephalitis in immunosuppressed people; if first contracted during pregnancy, it can cause miscarriage or congenital defects in the neonate; and it can cause serious ocular disease, even in immunocompetent people. The disease has a complex epidemiology, being transmitted by ingestion of oocysts that are shed in the faeces of definitive feline hosts and contaminate water, soil and crops, or by consumption of intracellular cysts in undercooked meat from intermediate hosts. In this review we examine current and future approaches to control toxoplasmosis, which encompass a variety of measures that target different components of the life cycle of T. gondii. These include: education programs about the parasite and avoidance of contact with infectious stages; biosecurity and sanitation to ensure food and water safety; chemo- and immunotherapeutics to control active infections and disease; prophylactic options to prevent acquisition of infection by livestock and cyst formation in meat; and vaccines to prevent shedding of oocysts by definitive feline hosts.

Intracellular parasites, such as the apicomplexanToxoplasma gondii, are adept at scavenging nutrients from their host. However, there is little understanding of how parasites sense and respond to the changing nutrient environments they encounter during an infection.TgApiAT1, a member of the apicomplexan ApiAT family of amino acid transporters, is the major uptake route for the essential amino acid L-arginine (Arg) inT. gondii. Here, we show that the abundance ofTgApiAT1, and hence the rate of uptake of Arg, is regulated by the availability of Arg in the parasite’s external environment, increasing in response to decreased [Arg]. Using a luciferase-based ‘biosensor’ strain ofT. gondii, we demonstrate that parasites vary the expression ofTgApiAT1 in different organs within their host, indicating that parasites are able to modulateTgApiAT1-dependent uptake of Arg as they encounter different nutrient environmentsin vivo. Finally, we show that Arg-dependent regulation ofTgApiAT1 expression is post-transcriptional, mediated by an upstream open reading frame (uORF) in theTgApiAT1 transcript, and we provide evidence that the peptide encoded by this uORF is critical for mediating regulation. Together, our data reveal the mechanism by which an apicomplexan parasite responds to changes in the availability of a key nutrient.

Esta tese consiste em estudos sobre a análise de emoções em crianças com desenvolvimento típico e crianças com TEA (idade entre 7 e 12 anos), por meio do imageamento térmico infravermelho (ITIV), uma técnica segura e não obtrusiva (isenta de contato), usada para registrar variações de temperatura em regiões de interesse (RIs) da face, tais como testa, nariz, bochechas, queixo e regiões periorbital e perinasal. Um robô social chamado N-MARIA (Novo-Robô Autônomo Móvel para Interação com Autistas) foi usado como estímulo emocional e mediador de tarefas sociais e pedagógicas. O primeiro estudo avaliou a variação térmica facial para cinco emoções (alegria, tristeza, medo, nojo e surpresa), desencadeadas por estímulos audiovisuais afetivos, em crianças com desenvolvimento típico. O segundo estudo avaliou a variação térmica facial para três emoções (alegria, surpresa e medo), desencadeadas pelo robô social N-MARIA, em crianças com desenvolvimento típico. No terceiro estudo, duas sessões foram realizadas com crianças com TEA, nas quais tarefas sociais e pedagógicas foram avaliadas tendo o robô N-MARIA como ferramenta e mediador da interação com as crianças. Uma análise emocional por variação térmica da face foi possível na segunda sessão, na qual o robô foi o estímulo para desencadear alegria, surpresa ou medo. Além disso, profissionais (professores, terapeuta ocupacional e psicóloga) avaliaram a usabilidade do robô social. Em geral, os resultados mostraram que o ITIV foi uma técnica eficiente para avaliar as emoções por meio de variações térmicas. No primeiro estudo, predominantes decréscimos térmicos foram observados na maioria das RIs, com as maiores variações de emissividade induzidas pelo nojo, felicidade e surpresa, e uma precisão maior que 85% para a classificação das cinco emoções. No segundo estudo, as maiores probabilidades de emoções detectadas pelo sistema de classificação foram para surpresa e alegria, e um aumento significativo de temperatura foi predominante no queixo e nariz. O terceiro estudo realizado com crianças com TEA encontrou aumentos térmicos significativos em todas as RIs e uma classificação com a maior probabilidade para surpresa. N-MARIA foi um estímulo promissor capaz de desencadear emoções positivas em crianças. A interação criança-com-TEA-e-robô foi positiva, com habilidades sociais e tarefas pedagógicas desempenhadas com sucesso pelas crianças. Além disso, a usabilidade do robô avaliada por profissionais alcançou pontuação satisfatória, indicando a N-MARIA como uma potencial ferramenta para terapias.

A recombinant BCG strain expressing the genetically detoxified S1 subunit of pertussis toxin 9K/129G (rBCG-S1PT), previously constructed by our research group, demonstrated the ability to develop high protection in mouse models of pertussis challenge which correlated with the induction of a Th1 immune response pattern. The Th1 immune response induced by rBCG-S1PT treatment was also confirmed in the murine orthotopic bladder cancer model, in which the intravesical instillation of rBCG-S1PT resulted in an improved antitumor effect. Based on these observations, we hypothesize that the reengineering of the S1PT expression in BCG could increase the efficiency of the protective Th1 immune response in order to develop a new alternative of immunotherapy in bladder cancer treatment. Objectives. To construct rBCG strains expressing S1PT from extrachromosomal (rBCG-S1PT) and integrative vectors (rBCG-Sli), or their combination, generating the bivalent strain (rBCG-S1+S1i), and to evaluate the respective immunogenicity of rBCG strains in mice. Methods. Mycobacterial plasmids were constructed by cloning the s1pt gene under integrative and extrachromosomal vectors and used to transform BCG, individually or in combination. Antigen expression and localization were confirmed by Western blot. Mice were immunized with wild-type BCG or the rBCG strains, and cytokines quantification and flow cytometry analysis were performed in splenocytes culture stimulated with mycobacterial-specific proteins. Findings. S1PT expression was confirmed in all rBCG strains. The extrachromosomal vector directs S1PT to the cell wall-associated fraction, while the integrative vector directs its expression mainly to the intracellular fraction. Higher levels of IFN-γ were observed in the splenocytes culture from the group immunized with rBCG-S1i in comparison to BCG or rBCG-S1PT. rBCG-S1+S1i showed higher levels of CD4 + IFN-γ + and double-positive CD4 + IFN-γ + TNF-α + T cells. Conclusions. rBCG-S1+S1i was able to express the two forms of S1PT and elicited higher induction of polyfunctional CD4 + T cells, indicating enhanced immunogenicity and suggesting its use as immunotherapy for bladder cancer.

© 2018 Zhang et al. Staphylococcus aureus is a major cause of morbidity and mortality worldwide. S. aureus colonizes 20 to 80% of humans at any one time and causes a variety of illnesses. Strains that are resistant to common antibiotics further complicate management. S. aureus vaccine development has been unsuccessful so far, largely due to the incomplete understanding of the mechanisms of protection against this pathogen. Here, we studied the role of different aspects of adaptive immunity induced by an S. aureus vaccine in protection against S. aureus bacteremia, dermonecrosis, skin abscess, and gastrointestinal (GI) colonization. We show that, depending on the challenge model, the contributions of vaccine-induced S. aureus-specific antibody and Th1 and Th17 responses to protection are different: antibodies play a major role in reducing mortality during S. aureus bacteremia, whereas Th1 or Th17 responses are essential for prevention of S. aureus skin abscesses and the clearance of bacteria from the GI tract. Both antibody-and T-cell-mediated mechanisms contribute to prevention of S. aureus dermonecrosis. Engagement of all three immune pathways results in the most robust protection under each pathological condition. Therefore, our results suggest that eliciting multipronged humoral and cellular responses to S. aureus antigens may be critical to achieve effective and comprehensive immune defense against this pathogen. IMPORTANCE S. aureus is a leading cause of healthcare-and community-associated bacterial infections. S. aureus causes various illnesses, including bacteremia, meningitis, endocarditis, pneumonia, osteomyelitis, sepsis, and skin and soft tissue infections. S. aureus colonizes between 20 and 80% of humans; carriers are at increased risk for infection and transmission to others. The spread of multidrug-resistant strains limits antibiotic treatment options. Vaccine development against S. aureus has been unsuccessful to date, likely due to an inadequate understanding about the mechanisms of immune defense against this pathogen. The significance of our work is in illustrating the necessity of generating multipronged B-cell, Th1-, and Th17-mediated responses to S. aureus antigens in conferring enhanced and broad protection against S. aureus invasive infection, skin and soft tissue infection, and mucosal colonization. Our work thus, provides important insights for future vaccine development against this pathogen.

Percorrendo a trajet?ria investigativa de pesquisadores voltados ? literatura de fronteira entre o sul do Brasil, Uruguai e Argentina, evidencio como, vinculados ao paradigma da transcultura??o, eles atuaram na cria??o de uma imagem homog?nea da literatura de fronteira da regi?o. Considerando que o imagin?rio da fic??o local na contemporaneidade ? mais diversificado do que tem sido tratado pela cr?tica predominante, proponho-me a desarticular essa imagem. Para tanto, retomo o pensamento cr?tico na Am?rica Latina pela perspectiva de leitura que vem sendo concebida como Pensamento de Fronteira e realizo uma interpreta??o da obra ficcional de Aldyr Garcia Schlee. Nesse percurso de leituras entrecruzadas, entendo e exponho como o escritor, investindo na cria??o de uma performance autoral e na inscri??o de m?ltiplas mem?rias sociais em sua obra ficcional, cria um espa?o simb?lico que n?o somente evidencia a inviabilidade de se restringir a literatura da regi?o ? literatura gauchesca, mas tamb?m sinaliza a inter-rela??o desta com outras regi?es do mundo. In Imaginaries of the South: critical border thinking and the reverse of modernity in Aldyr Garcia Schlee's fiction, I reflected upon the development of critical thinking in Latin America and its implication in the creation of imaginaries. After analyzing the investigations carried out by researchers who focus on border literature in the south of Brazil, Uruguay and Argentina, I showed how they managed to develop a homogeneous image of the border literature in the region while being connected to the paradigm of transculturation. Taking into account that the imaginary of the local fiction in the region is currently more diversified that critics have shown, I aimed at changing this image. Therefore, I analyzed critical thinking in Latin America from the perspective of readings which have been understood as Border Thinking and interpret Aldyr Garcia Schlee's fictional work. In this way, with intercrossing readings, I showed how the writer invests in the development of an authorial performance and in the use of multiple social memories in his fiction, besides creating a symbolic space which not only provides evidence of the inviability towards restricting regional literature to the gaucha literature but also highlights its interrelation and other regions in the world. O objetivo deste trabalho foi avaliar o efeito do caldo de cana desidratado (CCD) no desempenho, na qualidade de carcaça e no índice econômico de codornas (Coturnix coturnix) europeias, no período de 22 a 42 dias de idade. Foram utilizadas 192 codornas de corte com 22 dias de idade, distribuídas em delineamentointeiramente casualizado, com quatro tratamentos e seis repetições de oito aves. Foram adicionados 0, 1,5, 3,0 e 4,5% de CCD na ração. Avaliou-se a viabilidade econômica da inclusão dos níveis de CCD nas rações por meio da margem bruta relativa das dietas. No período de 29 a 35 dias de idade, houve efeito linear crescente para o consumo de ração com o aumento de CCD na ração. No entanto, houve efeito quadrático dos níveis de CCD sobre o peso da carcaça, da coxa+sobrecoxa e da gordura abdominal, e os níveis ótimos estimados foram de 1,69, 2,50 e 2,34%, respectivamente. Houve efeito linear decrescente dos níveis de CCD sobre o peso de peito com e sem pele. A análise econômica indicou maior lucro ao produtor com uso da ração convencional. Entretanto, entre os níveis de inclusão de CCD, o de 1,5% apresentou os melhores resultados, com uma diferença de 0,47% na margem de lucro em relação à ração convencional. A adição de 1,69 e 2,50% de caldo de cana desidratado na ração é recomendada para atingir maior peso de carcaça e coxa+sobrecoxa em codornas de corte, respectivamente.
