13. Thermal stability of sputtered Mo/X and W/X (X = BN:O, B4C:O, Si, and C) multilayer soft-x-ray mirrors

Abstract

We prepared eight samples of Mo/X and W/X (X = BN:O, [B.sub.4]C:O, Si, and C) multilayers by magnetron sputtering. Analyses of x-ray photoelectron spectroscopy for the boron nitride and the [B.sub.4]C layers showed the concentration of O to be nonnegligible. We have evaluated the thermal stability by measuring soft-x-ray specular reflectances before and after thermal annealing occurs at temperatures as high as 700 [degrees] C. The results suggest that the thermal stability depends largely on the inclusion of low-density materials and not on the type of metal. Of the four low-density materials studied, BN:O is thermally the most stable, and the Mo/BN:O multilayer, the most stable among the eight samples, shows stability as high as 700 [degrees] C.

Abstract

A nonlinear, least-squares curve-fitting method is described that simultaneously determines the optical constants and the thickness of a very thin (less than or approximately 100-Angstroms) film from reflectance versus angle of incidence (R - Theta) data measured in the soft-x-ray region. The method is applied to R - Theta data obtained for very thin, sputtered films of carbon (65 Angstroms thick) and gold (94 Angstroms thick) at photon energies of 60-900 eV. The results show that the present method is capable of accurately determining the thickness of very thin films even for transparent materials, and that the obtained optical constants are in good agreement with values reported for films with a thickness of 1000 Angstroms.

17. Asymptomatic Pancreatic Metastasis from Renal Cell Carcinoma Diagnosed 21 Years after Nephrectomy

Abstract

This report presents our experience with a case of pancreatic metastasis of renal cell carcinoma (RCC) at a long-term follow-up after nephrectomy. A 73-year-old man underwent nephrectomy for right RCC 21 years ago; computed tomography (CT) scanning on routine follow-up revealed a solid mass in the tail of the pancreas, and magnetic resonance imaging (MRI) showed some tumors in the head and tail of the pancreas. The patient was asymptomatic and allergic to contrast medium. Therefore we could not perform contrast CT/MRI for further examination to diagnose pancreatic tumors. We undertook endoscopic ultrasonography (EUS) and detected a hypervascular and low echoic mass; tumor tissues were obtained by EUS-guided fine-needle aspiration (EUS-FNA). Pathological diagnosis revealed pancreatic metastasis of clear cell RCC; this was similar to the pathological findings of tumor tissues initially obtained by nephrectomy. EUS-FNA was extremely useful for the definitive diagnosis of a rare type of pancreatic tumor.

Abstract

Carbohydrate administration rapidly regulates hepatic mRNA-S14 content. Both sucrose and fructose but not glucose increase the transcription of hepatic mRNA-S14 in vivo. In primary hepatocyle cultures, mRNA-S14 transcription responds to either frustose or glucose. To test the hypothesis that the difference in hexose response is due to differences in cellular metabolism, we studied the regulation of this gene with a transient transfection assay system in Chinese hamster ovary (CHO) cells, hamster pancreatic Β-cells (HIT), and primary hepatocytes. In HIT cells, glucose stimulation of the expression vector pS14CAT (5 kilobases [kb]) containing 4.9 kb of 5' -flanking DNA was threefold greater than frustose. Glucose also gave a fourfold greater response at 27.5 mM than at 2.2 mM. In CHO cells, pS14CAT (5kb) showed a twofold greater response to frustose than to glucose. The differential response to the hexoses in the two cell lines is a result of cell-specific metabolism. Without glucose in the media, both CHO and HIT cells used pyruvate for energy. However, glucose addition to CHO cells enhances glycolysis and hexose shunt pathway activity while inhibiting pyruvate oxidation and S14 gene transcription. In contrast, addition of glucose to HIT cells leads to enhanced tricarboxylic acid cycle activity to oxidize pyruvate and an associated stimulation S14 transcription. We confirmed these conclusions in primary hepatocyte cultures. Addition of 27.5 mM glucose led to a twofold increase in endogenous mRNA-S14 accumulation, a twofold increase in transfected pS14LUC (5kb) activity, and a parallel twofold increase in pyruvate oxidation. Thus, the different transcriptional responses of the S14 gene to fructose and glucose are attributable to cell-specific metabolism. Our data show that the metabolic regulation of mRNA-S14 gene transcription correlates with the rate of cellular pyruvate oxidation. Abstract

The effect of cyclosporin A (CyA) on 2-deoxyglucose (2DG) uptake by soleus muscles of ICR mice was studied in vivo and in vitro. The basal and insulin-stimulated uptakes of 2DG by the muscles as well as the plasma insulin level were significantly decreased by the in vivo treatment of mice with 20 mg/kg/day of CyA for 6 weeks (P less than 0.01 and P less than 0.05), whereas the insulin binding was increased inversely in the muscles from 20 mg/kg/day CyA-treated mice. The insulin-stimulated uptake of 2DG by the muscles was significantly decreased by the in vitro treatment of the muscles with 1, 10 and 100 micrograms/mL of CyA (P less than 0.05 and P less than 0.01, respectively), while the basal uptake of 2DG was not changed by the in vitro treatment of the muscles with CyA. The insulin binding to the muscles was not altered by the in vitro treatment of the muscles with CyA. These findings suggest that CyA affects not only the insulin secretion from the pancreatic islets but also the postbinding mechanisms of insulin action on the glucose transport by the muscles, which may account for a part of the diabetogenic effect of CyA.

Abstract

The effect of polymyxin B on 3-O-methylglucose transport was studied in isolated rat adipocytes and erythrocytes. Polymyxin B (300 micrograms/mL) inhibited basal transport and insulin-stimulated transport of 3-O-methylglucose in adipocytes by 26.1 and 40.1%, respectively. Polymyxin B at concentrations of 300 and 3000 micrograms/mL inhibited transport of 3-O-methylglucose in erythrocytes by 20.0 and 40.8%, respectively. Polymyxin E at a concentration of 3000 micrograms/mL also inhibited, by 40.6%, the transport of 3-O-methylglucose in erythrocytes but 300 micrograms/mL of polymyxin E did not inhibit it significantly. These results indicate that polymyxin B inhibits glucose transport per se, as well as the insulin-dependent stimulation of glucose transport.
