Abstract

The structural diversity of glycans on cells-the glycome-is vast and complex to decipher. Glycan arrays display oligosaccharides and are used to report glycan hapten binding epitopes. Glycan arrays are limited resources and present saccharides without the context of other glycans and glycoconjugates. We used maps of glycosylation pathways to generate a library of isogenic HEK293 cells with combinatorially engineered glycosylation capacities designed to display and dissect the genetic, biosynthetic, and structural basis for glycan binding in a natural context. The cell-based glycan array is self-renewable and reports glycosyltransferase genes required (or blocking) for interactions through logical sequential biosynthetic steps, which is predictive of structural glycan features involved and provides instructions for synthesis, recombinant production, and genetic dissection strategies. Broad utility of the cell-based glycan array is demonstrated, and we uncover higher order binding of microbial adhesins to clustered patches of O-glycans organized by their presentation on proteins.

Abstract

The influence of the liquid, its impurities and additives on the ac degradation of impregnated polypropylene (PP) films has been investigated. Aging of polypropylene was performed at 80 [degrees] C, 135 MV/m in model capacitors filled with benzyltoluene saturated with [O.sub.2] in order to increase the aging rate. The thermal oxidation of the liquid has been studied by gas chromatography, the oxidation of PP by FTIR microspectroscopy, the degradation rate of PP films has been evaluated by the variation of their mean breakdown voltage with aging time. All of them are strongly enhanced by the electric field. The oxidation of the liquid which takes place during these experiments influences only slightly the PP degradation rate. It is concluded that these severe conditions of aging do not change the chemical process. They only enhance drastically its propagation rate. Since aging of PP films is due to the scission of macromolecular chains initiated by radicals formed at the electrodes, the influence of the liquid, its components or additives is explained by their ability to react with radicals which initiate the scission of the macromolecules.

Abstract

Measurements of partial discharge (PD) in models of impregnated polypropylene capacitors with and without specific defects have been conducted. Two different regimes of discharges (in the liquid and in gas bubbles) are illustrated. The effect of temperature on the decrease of the insulation property is underlined. Four specific defects have been artificially introduced in the test cell (poor impregnation, bad contact, fold in the polypropylene (PP) film, cavity). The relevance of parameters such as skewness and kurtosis to characterize PD distributions is discussed. New parameters based upon the mean position of the PD in the voltage phase are proposed for a better recognition of the defects inducing PD.

47. Potent Metabolic Sialylation Inhibitors Based on C-5-Modified Fluorinated Sialic Acids

Abstract

Sialic acid sugars on mammalian cells regulate numerous biological processes, while aberrant expression of sialic acid is associated with diseases such as cancer and pathogenic infection. Inhibition of the sialic acid biosynthesis may therefore hold considerable therapeutic potential. To effectively decrease the sialic acid expression, we synthesized C-5-modified 3-fluoro sialic acid sialyltransferase inhibitors. We found that C-5 carbamates significantly enhanced and prolonged the inhibitory activity in multiple mouse and human cell lines. As an underlying mechanism, we have identified that carbamate-modified 3-fluoro sialic acid inhibitors are more efficiently metabolized to their active cytidine monophosphate analogues, reaching higher effective inhibitor concentrations inside cells.

48. In vivo photoacoustics and high frequency ultrasound imaging of mechanical high intensity focused ultrasound (HIFU) ablation

Abstract

The thermal effect of high intensity focused ultrasound (HIFU) has been clinically exploited over a decade, while the mechanical HIFU is still largely confined to laboratory investigations. This is in part due to the lack of adequate imaging techniques to better understand the in-vivo pathological and immunological effects caused by the mechanical treatment. In this work, we explore the use of high frequency ultrasound (US) and photoacoustics (PA) as a potential tool to evaluate the effect of mechanical ablation in-vivo, e.g. boiling histotripsy. Two mice bearing a neuroblastoma tumor in the right leg were ablated using an MRI-HIFU system conceived for small animals and monitored using MRI thermometry. High frequency US and PA imaging were performed before and after the HIFU treatment. Afterwards, the tumor was resected for further assessment and evaluation of the ablated region using histopathology. High frequency US imaging revealed the presence of liquefied regions in the treated area together with fragmentized tissue which appeared with different reflecting proprieties compared to the surrounding tissue. Photoacoustic imaging on the other hand revealed the presence of deoxygenated blood within the tumor after the ablation due to the destruction of blood vessel network while color Doppler imaging confirmed the blood vessel network destruction within the tumor. The treated area and the presence of red blood cells detected by photoacoustics were further confirmed by the histopathology. This feasibility study demonstrates the potential of high frequency US and PA approach for assessing in-vivo the effect of mechanical HIFU tumor ablation.

Abstract

Dendritic cells (DC) are the most potent antigen-presenting cells and form a promising new treatment modality. Fully activated DC loaded with antigen are very useful in stimulating immune responses, in particular those to combat cancer. Immature DC can either cause immunological tolerance or induce regulatory T-cells, opening up future application in transplantation, autoimmunity, and perhaps chronic inflammation. For these clinical applications, generation of DC under Good Manufacturing Practice conditions and without the use of animal products is a main prerequisite. To date, there are many different DC culture protocols, which subsequently lead to different types of DC. Protocols differ not only in DC preparation techniques, but also many other variables, like maturation status, dose and timing interval, route of administration, and antigen loading. DC vaccination, although very promising, is far from standardized. In this chapter we discuss the usefulness of standardized clinical and immunological criteria and the need for careful study design to further optimize the use of dendritic cells and to unequivocally prove their efficacy.
