Increasingly, safety and liability critical applications require GNSS-like positioning metrics in environments where GNSS cannot work. Indoor navigation for the vision impaired and other mobility restricted individuals, emergency responders and asset tracking in buildings demand levels of positioning accuracy and integrity that cannot be satisfied by current indoor positioning technologies and techniques. This paper presents the challenges facing positioning technologies for indoor positioning and presents innovative algorithms and approaches that aim to enhance performance in these difficult environments. The overall aim is to achieve GNSS-like performance in terms of autonomous, global, infrastructure free, portable and cost efficient. Preliminary results from a real-world experimental campaign conducted as part of the joint FIG Working Group 5.5 and IAG Sub-commission 4.1 on multi-sensor systems, demonstrate performance improvements based on differential Wi-Fi (DWi-Fi) and cooperative positioning techniques. The techniques, experimental schema and initial results will be fully documented in this paper.

Com o objetivo de atualizar o estudo da situação da hemoterapia no Estado de São Paulo, Brasil foi realizado inquérito em 1990, comparando-o com outro realizado em 1988. Foram pesquisados 79 municípios, sendo que (77,4%) realizavam transfusões de sangue, envolvendo 104 serviços, dos quais 13,5% se utilizavam de sangue proveniente de Hemocentros. Em 1988, o inquérito envolveu 57 municípios, 40 com atividade hemoterápica e 71 serviços, sendo que em 8,5% foi constatada a participação de Hemocentros. O número de doadores, transfusões e prevalência de sorologia positiva para a doença de Chagas foi de, respectivamente: 105.170, 79.544 e 1,24%, enquanto que em 1988 estes números foram: 51.614, 49.211 e 1,52%. Avaliando a triagem sorológica evidenciaram-se 94,2%; 94,2%; 94,2%; 94,3% e 20,2% dos serviços que realizavam, respectivamente, testes sorológicos para doença de Chagas, sífilis, hepatite, HIV e malária, enquanto que em 1988, 69,0%; 70,4%; 67,6%; 63,4% e 16,9% dos serviços empregavam tais testes, respectivamente. Em relação à triagem sorológica específica para doença de Chagas, observou-se que: a reação de fixação do complemento era realizada por 4,9% dos serviços; látex por 4,0%, imunofluorescência indireta 80%; hemaglutinação direta 24%; hemaglutinação indireta 69% e ELISA 59,0%. No inquérito precedente apenas 14,3% dos serviços realizavam teste ELISA, e a fixação do complemento era utilizada em 28,6% dos serviços. Os resultados mostram que os serviços estão utilizando, a cada dia, técnicas mais sensíveis na rotina sorológica. Foi observado ainda que em 1988, 77,6% de todos os serviços estudados realizavam apenas uma técnica para o diagnóstico da doença de Chagas, enquanto que em 1990, 92,9% empregavam duas ou mais diferentes técnicas. Conclui-se que a qualidade da hemoterapia praticada no Estado de São Paulo foi significativamente melhorada nos dois últimos anos.

Pyrrolidone carboxyl peptidase (EC 3.4.11.8) is an exopeptidase commonly called PYRase, which hydrolytically removes the pGlu-proteins. pGlu also known as pyrrolidone carboxylic acid may occur naturally by an enzymatic procedure or may occur as an artifact in proteins or peptides. The enzymatic synthesis of pGlu suggests that this residue may have important biological and physiological functions. Several studies are consistent with this supposition. PYRase has been found in a variety of bacteria, and in plant, animal, and human tissues. For over two decades, biochemical and enzymatic properties of PYRase have been investigated. At least two classes of PYRase have been characterized. The first one includes the bacterial and animal type I PYRases and the second one the animal type II and serum PYRases. Enzymes from these two classes present differences in their molecular weight and in their enzymatic properties. Recently, the genes of PYRases from four bacteria have been cloned and characterized, allowing the study of the primary structure of these enzymes, and their over-expression in heterelogous organisms. Comparison of the primary structure of these enzymes revealed striking homologies. Type I PYRases and bacterial PYRases are generally soluble enzymes, whereas type II PYRases are membrane-bound enzymes. PYRase II appears to play as important a physiological role as other neuropeptide degrading enzymes. However, the role of type I and bacterial PYRases remains unclear. The primary application of PYRase has been its utilization for some protein or peptide sequencing. Development of chromogenic substrates for this enzyme has allowed its use in bacterial diagnosis.

Bacillus subtilis pyrrolidone carboxyl peptidase (Pcp) overexpressed in Escherichia coli was purified to homogeneity in less than 12 h using ammonium sulphate precipitation and hydrophobic interaction chromatography. The enzyme, which removes amino-terminal L-pyroglutamic acid from peptides, appears to be a tetramer of 25,200 molecular mass subunits. The protein cross-reacted with polyclonal antibodies raised against Pcp from Streptococcus pyogenes. The overexpressed enzyme exhibits an absolute substrate specificity towards N-terminal pyroglutamyl residues with a Michaelis constant of 1.04 mM for L-pyroglutamyl-beta-naphthylamide. The enzyme could be used for the removal of pyroglutamyl residues that block amino termini of proteins and peptides before performing Edman sequential degradation.

Pyrrolidone carboxyl peptidase (EC 3.4.11.8) (Pcp), an enzyme which selectively removes pyrrolidone carboxylic acid (PCA) from some PCA-peptides and -proteins, was demonstrated in bacteria and in plant, animal and human tissues. In this paper we describe the purification to homogeneity of the enzyme of Streptococcus pyogenes, over-expressed in Escherichia coli. This was achieved, for the first time in one step, by hydrophobic interaction chromatography. Analysis under non-denaturing conditions revealed a molecular mass of 85 kDa and in the presence of sodium dodecyl sulfate gave a molecular mass of 23.5 kDa. Investigations on enzymatic properties showed that the Pcp over-expressed in E. coli disclosed properties similar to those found for the enzyme extracted from S. pyogenes or for some other Pcps studied previously. Thus the over-expressed enzyme should serve as a suitable source for N-terminal unblocking prior to some PCA protein sequencing.

Pyrrolidone carboxyl peptidase (EC 3.4.11.8) (Pcp) is an enzyme that catalyzes the removal of the N-terminal pyroglutamyl group from some peptides or proteins. Its value in protein chemistry and bacterial diagnosis makes this enzyme an interesting subject of study. The present paper reports for the first time the cloning and characterization of a pyrrolidone carboxyl peptidase gene (pcp). This gene is present in a single copy in the genome of Bacillus subtilis as indicated by Southern blot hybridization analysis. The pcp transcripts were analyzed in Escherichia coli by Northern blot hybridization and S1 nuclease mapping. The deduced amino acid sequence predicts a protein of 215 amino acids with a calculated molecular weight of 23,777 Da. The pcp gene has been over-expressed in E. coli, allowing the identification and partial characterization of Pcp protein.

The authors report a case of false aneurysm of the posterior subaortic region of the left ventricle after a technically difficult surgical decalcification in a case of aortic stenosis. At reoperation, the surgeon used the femoral artery and both vena cavae for canulation. A direct approach of this "vascular" tumour, which was exterior to the great vessels, confirmed the diagnosis and enabled the localisation of the internal orifice inside the left ventricle. This was closed simply by suturing. No previous cases of this kind were found in the literature. This report underlines the necessity of taking the greatest care when decalcifying this region in order to avoid this type of serious complication.

The effect of one tablet of placebo daily for 8 days on blood pressure was studied in 90 hypertensive patients, using the blood pressure profile method, with a 12-hour recording period (from 8 a.m. to 8 p.m) and readings every 15 minutes (Dinamam 845). Statistical analysis of the results showed no significant difference in diastolic, systolic and mean blood pressure in 50% of the cases. A significant decrease of about 10% in blood pressure without change in variability was observed in more than 20% (complete placebo effect) and in about 30% (total placebo effect) of the cases. A significant increase of about 8% on blood pressure was recorded in less than 20% of the cases. Almost 25% of the patients had a significant decrease in heart rate. There was no correlation between heart rate and blood pressure under placebo. These criteria, which are both qualitative and quantitative, can be taken as reference when the blood pressure profile method is used to evaluate the effectiveness of antihypertensive drugs.
