Abstract

Background: Cleft lip and palate is an anomaly that affects both women and men. It is considered to be among the most frequent congenital abnormalities and is related to modifications in chromosomal DNA and multiple genetic alterations. This anomaly can also be associated with various environmental factors, such as tobacco and alcohol consumption, medication use, and exposure to different environmental and industrial toxic substances. The objective of this study was to document the frequency of risk factors related to cleft lip and palate through a systematic review of Mexican studies. Methods: In this systematic review, a bibliographic search was conducted following PRISMA guidelines in the databases Scielo, ScienceDirect, PubMed, and EBSCO. Keywords related to cleft lip and palate, epidemiology, and risk factors were used. In all, 3 independent reviewers (J.A.S.L., S.L.V., and N.M.F.) selected and evaluated a total of 17 articles included in this analysis, achieving a coefficient of κ = 0.84. Results: The analysis revealed that the highest frequency of conducted studies was in the State of Mexico. The most common risk factors identified were environmental, pharmacological, consumption habits, and gynecological factors. Conclusions: Identifying the main risk factors for cleft lip and palate in the Mexican population will enable the implementation of preventive measures aimed at reducing exposure to these factors. Abstract

Background: The development and establishment of oral squamous cell carcinoma are confined to carcinogenesis, which involves oxidative stress via oxygen-free radical production as a hydroxyl radical (HO•), considered the most important cause of oxidative damage to basic biomolecules since it targets DNA strands. 8-Hydroxy-2'-deoxyguanosine (8-OHdG) is considered a free radical with a promutagenic capacity due to its ability to pair with adenosine instead of cytosine during replication. Material and Methods: We collected 30 paraffin-embedded tissue samples of OSCC from patients treated between 2013 and 2018. We recorded risk habits, disease stage, disease free survival and death with at least 3 years of followup. 8-Hydroxyguanosine was evaluated by immunohistochemistry and subsequently classified as weak-moderate or strong positive expression. Additionally, we noted whether it was expressed in the cytoplasm and/or nucleus. Results: Most of the cases expressed 8-OHdG with a strong intensity (80%). All neoplastic cells were preferentially stained in only the cytoplasm (70.0%), but nuclear positivity was found in 30%, independent of the intensity. Based on the location in the cytoplasm and/or nucleus, tumors >4 cm showed a high frequency (95.5%) of 8-OHdG expression in only the cytoplasm, with a significant difference (p value ≤ 0.001). Additionally, overall survival was affected when immunoexpression was present in the cytoplasm and nucleus because all deaths were in this group were statistically significant (p value = 0.001). Conclusions: All tumors showed DNA oxidative damage, and 8-OHdG was preferentially expressed in the cytoplasm. Abstract

Los principales factores de riesgo con los que se asocia el carcinoma oral de células escamosas son el hábito de tabaco y alcohol. La inflamación también es un factor de riesgo importante en el desarrollo del cáncer, ésta posiblemente inducida por el acúmulo de bacterias que se refleja en la placa bacteriana (biofilm microbiano) y posterior cálculo dental debido a una higiene oral deficiente Introduction: The primary risk factors to which Oral Squamous Cell Carcinoma is associated are related with tobacco and alcohol habits. Inflammation is an important risk factor in the development of cancer, induced, perhaps because of the cumulus of bacteria that is reflected in dental plaque and posterior calculus to a poor oral hygiene. Objective: The purpose of this study was to associate poor oral hygiene as a risk factor for Oral Squamous Cell Carcinoma. Material y methods: Case-control study in which the simplified index of oral hygiene to develop dental plaque and dental calculus was used as a measurement instrument. Tobacco and alcohol habits were registered. The risk was estimated obtaining odds ratio and the statistical significance was taken through χ2. Results: Poor oral hygiene with high levels of dental plaque were identified as a risk to the presence of Oral Squamous Cell Carcinoma, the known risk factors (tobacco and alcohol) were also identified in our population, however the risk was lower in contrast to poor oral hygiene. Conclusion: Poor oral hygiene is a significant risk factor that contributes to the presence of oral cancer and can be bigger in comparison with tobacco and alcohol use.

Abstract

The article presents a study which evaluated the presence of taste dysfunction and the influence that predisposing factors have on subjects with gustatory impairment, with particular regard to those positive for SARS-CoV-2. Topics include evaluation of gustatory function, taste dysfunction according to SARS-CoV-2 status and differences between predisposing factors, and finding on the frequency of taste disturbances according to the results of the analysis.

Abstract

Background: Ep-CAM, a transmembrane glycoprotein expressed in most epithelium in normal conditions, has diverse roles in these tissues, including in cell adhesion, proliferation, differentiation, cell cycle regu-lation, migration and intracellular signaling. It is also over-expressed in most malignant neoplasia, partic-ipating in the initiation, progression, and metastatic dissemination of the tumor. The expression and roles of this protein in oral neoplasia, particularly in odontogenic tumors, remain unestablished. The objective of this study consisted in analyzing the expression of this protein in ameloblastoma and tooth germ. Material and Methods: Ep-CAM (MOC-31) expression was evaluated by immunohistochemistry in tooth germs (TG) (n = 16) ameloblastomas (AM) (n = 60) and 2 ameloblastic carcinomas. Sections were visualized in their totality with an optical microscope, and positivity observed in cell membrane and cytoplasm was graded according to the following semi-quantitative scale: Neg, "essentially unstained", for negative sections or staining <5% of cells; + for staining of 5-50% of cells; ++ for staining >50% of cells. Results: Most tooth germs expressed MOC-31 (81.3%), strong staining was observed both in the inner epithelium of the enamel organ and in the adjacent stellate reticulum. 16.7% of the AM cases showed MOC-31 expression, the immunoexpression expression was diffuse at the cytoplasmic and membrane level. The only two cases of ameloblastic carcinoma included were strong positive to MOC-31. No correlation was observed between protein expression and gender, age, clinical variants, or histological subtypes. Conclusions: Overexpression was found in TG and ameloblastic carcinoma compared to AM; further studies with different experimental strategies are suggested to clarify the biological significance of this finding. Abstract

Comunicación breve Actinic prurigo (AP) is an idiopathic photodermatosis; the initial manifestations usually occur during the first decades of life but can appear at any age. Cases are usually diagnosed late once the lesions have exacerbated; due to the extensive involvement of the vermilion border and the etiology, it has been confused with and related to a potentially malignant process. Syndecan-1 and E-cadherin were positive in the epidermis, with moderate-to-intense staining in 100% of samples. Ki67 and MCM3 were expressed in the lower third of the epidermis and showed greater immunolabeling in samples that contained lymphoid follicles (Ki 67: epidermis [17.7% ± 6.79%] and dermis [7.73% ± 6.69%]; MCM3: epidermis [22.92% ± 10.12%] and dermis [6.13% ± 6.27%]). In conclusión AP is a disease in which there is no evidence that the lesions are potentially cancerous. AP cheilitis should not be confused with actinic cheilitis because they are separate entities.

Abstract

La diabetes mellitus es considerada un problema de salud pública en todo el mundo. Este padecimiento afecta órganos y sistemas, incluyendo la cavidad oral; sin embargo, hay pocos informes en la literatura de los efectos de la diabetes mellitus en la cavidad bucal en la población del estado de Durango. Objetivo: El objetivo de este estudio fue determinar la asociación de diversas enfermedades orales, el índice de masa corporal, así como el nivel de glucosa en sangre en pacientes con diabetes mellitus. Material y métodos: Se realizó un estudio de casos y controles para identifi car la prevalencia de las enfermedades orales en individuos con y sin diabetes mellitus. Los sujetos con diabetes mellitus tipo 1 o 2 se defi nieron como casos y aquellos con otras enfermedades fueron defi nidos como controles. El índice de masa corporal y la concentración de glucosa en sangre fueron evaluados en todos los sujetos. El análisis estadístico incluyó las pruebas chi cuadrada y promedio y desviación estándar de los datos. Resultados: Se evaluaron 312 personas, 38 pacientes con diabetes mellitus tipo 1, 79 con diabetes mellitus tipo 2 y 195 sin diabetes mellitus. Diferencias signifi cativas fueron encontradas en individuos con DM2, que mostraron una mayor frecuencia de periodontitis, así como diferencias signifi cativas entre este tipo de diabetes y niveles elevados de glucosa en sangre. Además, la presencia de obesidad e hipertensión arterial en los sujetos con diabetes tipo 2 se mostró como un riesgo para el desarrollo de periodontitis. Conclusiones: La diabetes mellitus tipo 2 puede ser un factor de riesgo para la enfermedad periodontal, y el estado de hiperglucemia aumentó el riesgo de padecer periodontitis cuando la obesidad y la hipertensión arterial estaban presentes en personas con diabetes mellitus tipo 2.

Abstract

Los carcinomas de cabeza y cuello son relativamente frecuentes, presentan diversos tipos histológicos y características clínicas, mismas que se asocian con una variedad de riesgos de mortalidad. Objetivo: El propósito de este trabajo fue examinar en un periodo de cinco años carcinomas de cabeza y cuello tratados en el Centro Estatal de Cancerología. Material y métodos: Este estudio se realizó de manera retrospectiva, se revisaron factores de riesgo y tratamientos, se hicieron análisis univariados y multivariados para evaluar factores pronóstico y supervivencia. Resultados: Se obtuvieron un total de 32 expedientes que cumplían con las características del estudio, el periodo de seguimiento fue de 2 a 32 meses. La variante histológica predominante fue carcinoma epidermoide, el tratamiento primario más común fue cirugía. Abstract

GPN‐loop GTPases 1 and 3 are required for RNA polymerase II (RNAPII) nuclear import. Gpn1 and Gpn3 display some sequence similarity, physically associate, and their protein expression levels are mutually dependent in human cells. We performed here Fluorescence Resonance Energy Transfer (FRET), molecular modeling, and cell biology experiments to understand, and eventually disrupt, human Gpn1–Gpn3 interaction in live HEK293‐AD cells. Transiently expressed EYFP‐Gpn1 and Gpn3‐CFP generated a strong FRET signal, indicative of a very close proximity, in the cytoplasm of HEK293‐AD cells. Molecular modeling of the human Gpn1–Gpn3 heterodimer based on the crystallographic structure of Npa3, the Saccharomyces cerevisiae Gpn1 ortholog, revealed that human Gpn1 and Gpn3 associate through a large interaction surface formed by internal α‐helix 7, insertion 2, and the GPN‐loop from each protein. In site‐directed mutagenesis experiments of interface residues, we identified the W132D and M227D EYFP‐Gpn1 mutants as defective to produce a FRET signal when coexpressed with Gpn3‐CFP. Simultaneous but not individual expression of Gpn1 and Gpn3, with either or both proteins fused to EYFP, retained RNAPII in the cytoplasm and markedly inhibited global transcription in HEK293‐AD cells. Interestingly, the W132D and M227D Gpn1 mutants that showed an impaired ability to interact with Gpn3 by FRET were also unable to delocalize RNAPII in this assay, indicating that an intact Gpn1–Gpn3 interaction is required to display the dominant‐negative effect on endogenous Gpn1/Gpn3 function we described here.
