2. A provable two-stage algorithm for penalized hazards regression

Abstract

From an optimizer's perspective, achieving the global optimum for a general nonconvex problem is often provably NP-hard using the classical worst-case analysis. In the case of Cox's proportional hazards model, by taking its statistical model structures into account, we identify local strong convexity near the global optimum, motivated by which we propose to use two convex programs to optimize the folded-concave penalized Cox's proportional hazards regression. Theoretically, we investigate the statistical and computational tradeoffs of the proposed algorithm and establish the strong oracle property of the resulting estimators. 3. Generalized High-Dimensional Trace Regression via Nuclear Norm Regularization

Abstract

We study the generalized trace regression with a near low-rank regression coefficient matrix, which extends notion of sparsity for regression coefficient vectors. Specifically, given a matrix covariate $X$, the probability density function $f(Y\vert X)=c(Y)\exp{(\phi^{-1}\left[-Y\eta^* + b(\eta^*)\right])}$, where $\eta^*=tr({\Theta^*}^TX)$. This model accommodates various types of responses and embraces many important problem setups such as reduced-rank regression, matrix regression that accommodates a panel of regressors, matrix completion, among others. We estimate $\Theta^*$ through minimizing empirical negative log-likelihood plus nuclear norm penalty. We first establish a general theory and then for each specific problem, we derive explicitly the statistical rate of the proposed estimator. They all match the minimax rates in the linear trace regression up to logarithmic factors. Numerical studies confirm the rates we established and demonstrate the advantage of generalized trace regression over linear trace regression when the response is dichotomous. Abstract

Diabetic cardiomyopathy (DCM) is a condition characterized by myocardial dysfunction that occurs in individuals with diabetes, in the absence of coronary artery disease, valve disease, and other conventional cardiovascular risk factors such as hypertension and dyslipidemia. It is considered a significant and consequential complication of diabetes in the field of cardiovascular medicine. The primary pathological manifestations include myocardial hypertrophy, myocardial fibrosis, and impaired ventricular function, which can lead to widespread myocardial necrosis. Ultimately, this can progress to the development of heart failure, arrhythmias, and cardiogenic shock, with severe cases even resulting in sudden cardiac death. Despite several decades of both fundamental and clinical research conducted globally, there are currently no specific targeted therapies available for DCM in clinical practice, and the incidence and mortality rates of heart failure remain persistently high. Thus, this article provides an overview of the current treatment modalities and novel techniques pertaining to DCM, aiming to offer valuable insights and support to researchers dedicated to investigating this complex condition. Resveratrol (RSV), a natural polyphenol, has been proven to have renoprotective effects. In this study, we used a lipopolysaccharide (LPS)-induced rat glomerular mesangial cells (RMCs) model, to elucidate the renoprotective effect of RSV on sphingosine kinase 1 (SphK1)/sphingosine 1-phosphate receptor 2 (S1P2)/NF-κB activation and the expression of downstream inflammatory mediators, such as intercellular adhesion molecule-1 (ICAM-1), inducible nitric oxide synthase (iNOS) and fibronectin (FN) protein expression in RMCs.Methods: Cell proliferation was tested by 3-(4, 5-dimethylthiazol-2-yl)-2, 5- diphenyltetrazolium bromide (MTT). The protein levels of FN, ICAM-1, iNOS, SphK1, S1P2 and NF-κB p65 in RMCs were detected by Western blot. The DNA-binding activity of NF-κB was detected by electrophoretic mobility shift assay (EMSA). SphK1 activity and S1P content were measured by using sphingosine kinase activity assay kit and ELISA assay, respectively.Results: We first found that LPS could stimulate SphK1/S1P axis activation, whereas this occurrence was significantly blocked by RSV pretreatment. RSV obviously repressed LPS-induced upregulated expression of fibronectin (FN), intercellular adhesion molecule-1 (ICAM-1) and inducible nitric oxide synthase (iNOS) in RMCs. Moreover, RSV markedly reduced SphK1 activity and its protein expression, and attenuated S1P content in LPS-induced RMCs. Furthermore, RSV could block LPS-induced upregulation of NF-κB p65 and DNA-binding activity of NF-κB. And this phenomenon was notably attenuated by SphK1 inhibitor and S1P2 inhibitor.Conclusion: RSV inhibited LPS-induced RMCs’ proliferation and inflammation and FN expression by SphK1/S1P2/NF-κB pathway, suggesting that RSV may be independent of its hypoglycemic effect on preventing or delaying the development of mesangial cell fibrosis.Keywords: diabetic nephropathy, resveratrol, lipopolysaccharide, sphingosine kinase 1, sphingosine 1-phosphate, NF-κB

28. Investigating the impact of empagliflozin on the retina of diabetic mice

Abstract

Background Diabetic retinopathy (DR) frequently results in compromised visual function, with hyperglycemia-induced disruption of the blood-retinal barrier (BRB) through various pathways as a critical mechanism. Existing DR treatments fail to address early and potentially reversible microvascular alterations. This study examined the effects of empagliflozin (EMPA), a selective Sodium-glucose transporter 2 (SGLT2) inhibitor, on the retina of db/db mice. The objective of this study is to investigate the potential role of EMPA in the prevention and delay of DR.Methods db/dbmice were randomly assigned to either the EMPA treatment group (db/db?+?Emp) or the model group (db/db), while C57 mice served as the normal control group (C57). Mice in the db/db?+?Emp group received EMPA for eight weeks. Body weight, fasting blood glucose (FBG), and blood VEGF were subsequently measured in all mice, along with the detection of specific inflammatory factors and BRB proteins in the retina. Retinal SGLT2 protein expression was compared using immunohistochemical analysis, and BRB structural changes were observed via electron microscopy.Results EMPA reduced FBG, blood VEGF, and retinal inflammatory factors TNF-a, IL-6, and VEGF levels in the eye tissues of db/dbmice. EMPA also increased Claudin-1, Occludin-1, and ZO-1 levels while decreasing ICAM-1 and Fibronectin, thereby preserving BRB function in db/dbmice. Immunohistochemistry revealed that EMPA reduced SGLT2 expression in the retina of diabetic mice, and electron microscopy demonstrated that EMPA diminished tight junction damage between retinal vascular endothelial cells and prevented retinal vascular basement membrane thickening in diabetic mice.Conclusion EMPA mitigates inflammation and preserves BRB structure and function, suggesting that it may prevent DR or serve as an effective early treatment for DR.

41. Evaluation of the Water Conservation Function of Different Forest Types in Northeastern China.

Abstract

Water conservation is an important function of forest ecosystems, but it is still unclear which forest types function best in this regard. We investigated the water conservation function indicators including the water-holding rate of branches and leaves (BLwr), water-holding capacity of litter (Lwc), water absorption rate of litter (Lwr), soil infiltration rate (Ir), soil and water content (SWC), soil water storage (SWS), and soil organic matter (SOM) accumulation of five forest types (Larix gmelinii forests, Pinus koraiensis forests, Robinia pseudoacacia forests, Pinus tabulaeformis forests, and mixed forests) and evaluated them using the gray correlation method (GCM). The results indicate that the BLwr of five stands in the study area varied from 18.3% to 33.5%. The SWC and SWS of the R. pseudoacacia stand were 13.76% and 178.9 mm, respectively, which was significantly higher than that of the other stands (p < 0.05). The SOM was similar for the R. pseudoacacia (0.23%), mixed forest (0.22%), and L. gmelinii (0.22%) sites. The BLwr, Lwc, Lwr, SWC, and SWS values of broad-leaved tree species were higher than those of the mixed species, followed by those for coniferous tree species. Soil infiltration rate followed the order L. gmelinii > P. koraiensis > mixed forest > P. tabulaeformis > R. pseudoacacia. Based on our results, the R. pseudoacacia stand had the highest water conservation ability, while the lowest performance was found for the P. tabuliformis site. This suggests that, in order to enhance the water conservation function of forests in northeastern China, the focus should be on the establishment of R. pseudoacacia forests. [ABSTRACT FROM AUTHOR]
