Abstract

Percutaneous coronary diagnosis and intervention via distal transradial artery approach has become one of the hot research topics in the field of coronary angiography and intervention. Compared with transradial approach, the distal transradial artery approach has the advantages of higher patient comfort and fewer related complications; however, due to the tortuosity of radial artery and relatively small size of distal radial artery, there is a significant learning curve for distal transradial artery approach puncture. This paper analyzes and summarizes the common causes (mainly including patient factors and operational factors) and treatment strategies of distal transradial artery approach puncture failure based on the experience of more than 2 000 cases, in order to provide a reference for improving the success rate of puncture and promoting the application of distal transradial artery approach.

Abstract

Carbon monoxide (CO) plays a vital role in the biochemical processes of marine microorganisms, climate regulation, and global carbon cycle. In this study, seasonal and spatial variations of CO and oceanographic factors influencing these variations were investigated in the East China Sea (ECS) during two cruises from 16 to 30 May 2020 and from 8 to 30 October 2021. ECS was as a net source of atmospheric CO in both spring and autumn. The average values of seawater CO concentrations ([CO]surf), atmospheric CO mixing ratio ([CO]atm) and sea‐to‐air flux of CO were 2.22 ± 0.47 nmol L−1 and 0.97 ± 0.86 nmol L−1, 182.8 ± 71.8 ppbv and 102.7 ± 38.8 ppbv, 96.14 ± 25.82 nmol m−2 hr−1 and 31.96 ± 35.61 nmol m−2 hr−1 in spring and autumn, respectively. A steady‐state model was used to estimate the biogeochemical cycling rates of [CO]atm over the ECS. The effect of CO sea‐to‐air fluxes on atmospheric ·OH concentration was likely decreased from spring to autumn. Photoirradiation experiments further showed that the actual contributions of ultraviolet radiation and photosynthetically active radiation to CO photoproduction in the surface waters were 72.2% ± 13.8% and 27.8% ± 13.8%, respectively. The sea‐to‐air fluxes of CO in autumn were 5.15 μmol m−2 day−1 lower than in spring, which were due to larger bacterial consumption and lower [CO]surf in autumn in the ECS. Further, this study is expected to improve our understanding of the biogeochemical processes of CO in the eastern marginal seas of China. Plain Language Summary: Over the past decade, there has been a significant increase in global carbon emissions. CO is a potent greenhouse gas, and chromophoric dissolved organic matter (CDOM) photochemical processes control CO emission from the ocean to the atmosphere. Coastal waters dominate global oceanic carbon emissions, but the dynamics, controls and roles of CO remain largely unconstrained in the marginal seas around China. Here, we conducted a variety of biogeochemical analyses to investigate the controls of CO dynamics cycling in the East China Sea during spring and autumn. [CO]surf, [CO]atm, and sea‐to‐air flux of CO showed significant seasonal variability between spring and autumn, with higher values observed in spring. A variety of environmental factors such as CDOM concentration, wind speed and direction, physical mixing, air‐sea exchange, bacterial consumption, and dark production could affect CO distribution. Compared to air‐sea exchange, bacterial consumption represented the main CO‐sink and contributed over 90% of the total CO loss. These results provide a supplement to the global database of CO cycling processes and improve our current understanding of CO seasonal distribution in coastal ecosystems. 44. Characterizing novel molecular mechanisms involved in breast cancer tumourigenesis/progression and tamoxifen resistance

Abstract

Breast cancer is the most common cancer among women. Breast cancer tumorigenesis could be due to loss/mutation of BRCA1, yet the mechanisms underlying BRCA1-associated tumorigenesis are not well understood. Tamxoifen stands in the first-line for treatment of oestrogen receptor (ER) positive breast cancers but drug resistance significantly compromises its clinical efficacy in preventing cancer progression and improving survival. The mechanism of tamoxifen resistance is not fully understood and there is no clinical in-use biomarker that can reliably predict tamoxifen response. My PhD study aims at characterizing novel molecular pathways that are involved in breast cancer tumorigenesis/progression and tamoxifen resistance, focusing on two gaps in breast cancer research: 1) the role of BRCA1-mediated epigenetic regulation of gene expression; 2) identifying biomarker that can be used for prediction of tamoxifen resistance. In chapter 3, regulation of tumour suppressor FOXO3a by BRCA1 through EZH2-mediated epigenetic mechanism is studied. Knockdown or overexpression of BRCA1 suppresses or induces FOXO3a expression at both mRNA and protein levels, respectively. In addition, FOXO3a gene expression is negatively regulated by EZH2 in basal cell lines where BRCA1 is suppressed or mutated but not in BRCA1-wild-type MCF-7 cells, as chemical inhibition or siRNA-mediated depletion of EZH2 enhanced FOXO3a expression in basal cell lines. Moreover, ChIP assays showed that DNMT1, DNMT3a and DNMT3b and H3K27me3 are recruited to FOXO3a promoter in basal cell lines but not in MCF-7, and depletion of BRCA1 in MCF-7 induces the deposition of DNMTs and H3K27me3 to FOXO3a promoter. It is also found BRCA1 suppressed cancer cell proliferation at least partially through FOXO3a. Consistent with the in vitro data, FOXO3a promoter methylation scores are significantly higher in BRCA1-muated tumours comparing with non-BRCA1 mutated tumours. These data collectively suggest that BRCA1 can supress promoter methylation and enhance transcription of FOXO3a indirectly through binding to EZH2 and restraining its methyltransferase activity. The findings could contribute to better understanding of BRCA1-associated tumorigenesis and the regulation of FOXO3a in breast cancer. In chapter 4, the role of BQ323636.1, a splice variant of NCOR2 in tamoxifen resistance is studied and its efficacy in predicting tamoxifen resistance in breast cancer patients is also evaluated. It was found that overexpression of BQ323636.1 conferred resistance to tamoxifen in both breast cancer cell lines and orthotopic mouse model. Mechanistic studies showed that BQ323636.1 overexpression compromised the suppressive role of NCOR2 in regulating oestrogen-response element activity shown by luciferase assay and rescues the transcriptional suppression of tamoxifen on ER-target genes. Co- immunoprecipitation also showed that BQ323636.1 could bind to NCOR2 and inhibit the formation of co-repressor complex, which could explain how BQ323636.1 interferes with functions of NCOR2. Unlike NOCR2 that is localized in the nucleus, BQ323636.1 is found to be expressed in both nucleus and cytoplasm while nuclear BQ323636.1 level is significantly higher in tamoxifen resistant cells. More importantly, in human breast cancer tissue microarray, nuclear BQ323636.1 overexpression is significantly associated with tamoxifen resistance as well as patients' survival. The findings lead to the proposal of using BQ323636.1 as a biomarker for prediction of tamoxifen resistance.

Abstract

Evaluating the effects of wall restoration on ancient buildings has been a difficult task, and it is important that the overall appearance of the restored walls of ancient buildings is similar, harmonious, and uniform. This paper used a hue–saturation–value (HSV) color space and Circular Local Binary Pattern (CLBP) to analyze the comprehensive similarity between a restored wall and the original walls in Qi Li Ancient Town. The results show that the values of the comprehensive similarity calculation of ancient buildings based on the color and texture were consistent with the actual situation. The method is suitable for evaluating the degree of matching between wall repair materials and the appearance of the original wall materials of ancient buildings, and it can also be used to assess the comprehensive similarity between the repair materials and the original building walls before carrying out the wall repair in order to select more suitable materials for wall repair and achieve the best repair effect. And it is flexible and objective compared to human judgement. Through the accurate restoration of ancient buildings, not only can we protect cultural heritage and continue the historical lineage, we can also enhance the aesthetic value of buildings and meet people's needs for historical and cultural tracing.
